Influence Of Dietary Component Manipulation And Feed Management Strategies On Growth And Rumen Development Of Weaned Dairy Heifers, Tana Shea Dennis

Open Access Dissertations

Well-developed replacement heifers provide a central foundation for the continued success of the dairy industry. Emphasis on improving pre-weaned calf nutrition has predominated in the industry, but opportunities exist to improve post-weaning heifer nutrition and management. We aimed to evaluate common feed management strategies seen in the industry and their effects on growth, feed efficiency (G:F), and rumen development of calves from birth to 8 mo of age using pen- and individually-fed animal trials. Little information exists regarding post-weaning performance and rumen development of calves fed conventional or high planes of nutrition pre-weaning; therefore, we evaluated two milk replacer feeding …

Evaluation Of Negative Energy And Mineral Balance In Periparturient Holstein-Friesian Cattle, Ameer Ahmed Abdelhameed

Open Access Dissertations

Most cows experience a state of negative energy and mineral balance during the periparturient period. This imbalance is a major problem in the dairy industry and it is more frequently seen in multiparous cattle due to their high milk production. A large amount of fat and protein is mobilized in order to address the energy deficit during the periparturient period. The ability to accurately monitor energetics and electrolytes status using accurate, rapid, and low cost point-of-care instruments will assist in optimizing animal health and productivity.

This dissertation contains three studies that evaluated point-of-care instruments for measuring blood and plasma glucose, …

Investigation Of Dried Distillers Grains With Solubles (Ddgs) Susceptibility To Red Flour Beetle (Tribolium Castaneum (Herbst)) Infestation, Mahsa Fardisi Apr 2015

Investigation Of Dried Distillers Grains With Solubles (Ddgs) Susceptibility To Red Flour Beetle (Tribolium Castaneum (Herbst)) Infestation, Mahsa Fardisi

Open Access Dissertations

Protecting livestock feed from insect infestation is a common challenge that feed facility managers and farmers have to deal with. It becomes an issue of importance because insect infestation degrades feed quality and quantity. Dried Distillers Grains with Solubles (DDGS) is added to livestock feed because of its high protein content. As DDGS usage in animal feed increases, understanding the susceptibility of DDGS, when mixed with livestock feed, to insect infestation is necessary for safe feed storage. This research addressed four questions. The first looked at the susceptibility of DDGS, when stored as a raw ingredient, to Tribolium castaneum (Herbst) …

Propionate Induces Gluconeogenesis In Dairy Cattle Through Direct Activation Of The Bovine Cytosolic Phosphoenolpyruvate Carboxykinase Gene Promoter, Qian Zhang Apr 2015

Propionate Induces Gluconeogenesis In Dairy Cattle Through Direct Activation Of The Bovine Cytosolic Phosphoenolpyruvate Carboxykinase Gene Promoter, Qian Zhang

Open Access Dissertations

Phosphoenolpyruvate carboxykinase (PCK, GTP; EC4.1.1.32) catalyzes the conversion of oxaloacetate (OAA) to phosphoenolpyruvate (PEP), a critical reaction for gluconeogenesis. The two isoforms of PCK, namely cytosolic PCK (PCK1), and the mitochondrial PCK (PCK2) are abundantly expressed in liver and kidney. Increased PCK1 activity and mRNA expression in liver is observed as feed intake recovers during early lactation in dairy cows, a condition linked to increased ruminal propionate production. The central hypothesis of this dissertation is that propionate regulates its own metabolism in liver of dairy cattle through regulation of the PCK1 gene. The objectives of this dissertation were to determine: …

Prediction Of Parturition And Dystocia In Holstein-Friesian Cattle, And Cesarean Section In Dystocic Beef Cattle, Wen Han Mark Hiew

Open Access Dissertations

Dystocia is a major problem in the dairy industry as it causes livestock and economic loss. It is more frequently seen in primiparous cattle compared to their multiparous counterparts due to their smaller stature and the slow maturation of pelvic dimensions. In some instances, human intervention of the parturition process is imperative to avoid pain, injury, and mortality of the neonate and the dam. The ability to accurately predict dystocia and the time of parturition will ensure that timely assistance can be given to animals that are of high risk of dystocia.^ The present thesis contains four studies that explore …

Cellular And Molecular Regulation Of Skeletal Muscle Regeneration, Chunhui Jiang Oct 2014

Skeletal muscle regeneration provides a powerful model to study the cellular and molecular mechanism governing stem cell function since the whole process is mediated by a population of muscle-resident stem cells called satellite cells. The overall aim of this dissertation is to explore the roles of Notch signaling and a novel gene (Bex1) in satellite cell function and skeletal muscle regeneration. ^ The first part of this thesis is to examine how the Notch signaling pathway regulates the satellite cell self-renewal in a disease model. The MDX mouse is a well-established model for Duchenne muscular dystrophy (DMD) and characterized by …

The Genetic Architecture Of Juvenile Migration In Rainbow Trout (Oncorhynchus Mykiss), Benjamin Charles Hecht Oct 2013

The Genetic Architecture Of Juvenile Migration In Rainbow Trout (Oncorhynchus Mykiss), Benjamin Charles Hecht

Open Access Dissertations

Animal migrations play a critical role in the health and balance of ecological systems and in the evolution and diversification of species, and this is perhaps best displayed amongst salmonid fishes (salmon, trout, and char) who exhibit variation in the propensity to migrate both within and among species. Rainbow and steelhead trout (Oncorhynchus mykiss), members of the salmonid family of fishes, capture this variation throughout their native range. Some populations and ecotypes will remain resident in freshwater habitats throughout their life history; while others have the ability to embark on tremendous marine migrations. Those that migrate undergo a suite of …

Store-Operated Calcium Entry And Its Function During Fertilization In Porcine Eggs, Chunmin Wang Oct 2013

Store-Operated Calcium Entry And Its Function During Fertilization In Porcine Eggs, Chunmin Wang

Open Access Dissertations

In a variety of mammalian cells, the depletion of intracellular Ca2+ stores leads to Ca2+ influx across the plasma membrane though a process known as store-operated Ca2+ entry. It not only plays a critical role in the replenishment of the Ca2+ stores but it is also responsible for the maintenance of long-lasting high intracellular Ca2+ levels and sustaining repetitive Ca2+ oscillations. During fertilization a Ca2+ signal, which can take the form of a single or multiple Ca2+ elevations (depending on the species) is the universal trigger for the egg-to-embryo transition. For sustaining …

Genes With Physiological Roles In Callipyge Muscle Hypertrophy, Hui Yu Oct 2013

Genes With Physiological Roles In Callipyge Muscle Hypertrophy, Hui Yu

Open Access Dissertations

Callipyge sheep is an excellent model to study genes regulation in muscle growth since the up-regulation of DLK1 and/or RTL1 results in extreme postnatal muscle hypertrophy in loin and hindquarters. DLK1 and/or RTL1 are the primary inducers of muscle hypertrophy due to the inheritance model for the callipyge phenotype. The overall aim of this dissertation is to study the physiological pathways responding to the up-regulation of DLK1 and / or RTL1 in the hypertrophied muscles. Microarray analysis of gene expression identified 375 genes that were differentially expressed in callipyge semimembranosus. Twenty-five transcripts were further verified by quantitative PCR in two …

Notchless Interacts With Multiple Signaling Pathways During Mouse Peri-Implantation Development, Chiao-Ling Lo Oct 2013

Notchless Interacts With Multiple Signaling Pathways During Mouse Peri-Implantation Development, Chiao-Ling Lo

Open Access Dissertations

During peri-implantation, the embryo transitions from a suspension environment in the fallopian tubes to an adherent system within the uterus. Successful transition requires maternal and fetal signaling cascades that establish maternal-fetal boundaries. Failure is common, as ~ 25% of all human pregnancies terminate during these steps. A large-scale mutation study in mice produced two mutants (l11Jus1 and l11Jus4) that are excellent models of this transition. l11Jus1 and l11Jus4 contain missense mutations in the Notchless homolog 1 (Drosophila) (Nle1) gene. NLE1 is thought to signal via the canonical NOTCH pathway in vertebrates. Although in invertebrates and lower vertebrates, NOTCH signaling directs …
