Detecting Viral Particles In Vitro Using Photoacoustic Flow Cytometry, Anie-Pier Samson
Detecting Viral Particles In Vitro Using Photoacoustic Flow Cytometry, Anie-Pier Samson
Electronic Theses and Dissertations
The Covid-19 pandemic is a powerful example of just how damaging the rapid spread of an unknown virus can be. Viruses have the ability to spread rapidly amongst individuals if not treated and controlled. The first key step towards treatment is the timely and specific diagnosis of the virus causing the infection. An innovative method for the rapid detection of viral particles in solution consists of tagging the viral particles paired with using photoacoustic flow cytometry to irradiate the particles and get a signal. The high affinity of Streptavidin for Biotin can be used to bind Streptavidin-coated microspheres to biotinylated …
Real-Time Monitoring Of Cell Death Progress Using Capacitance Spectroscopy, Suyang Wu
Real-Time Monitoring Of Cell Death Progress Using Capacitance Spectroscopy, Suyang Wu
Electronic Theses and Dissertations
Biologics, including the monoclonal antibody (mAb), has experienced rapid development in the last decade. However, the price of biologics is often prohibitively high because of the low process efficiency. Delaying the inevitable cell death improves the productivity of upstream bioprocessing, whose success relies on monitoring the cell death onset that indicates the timing for preventive actions.
This study proposes to develop a real-time monitoring model that quantifies the dying cell percentage in lab-scale bioreactors using capacitance spectroscopy. The capacitance spectroscopy contains cell death-related information due to various physical properties changes during the cell death process, e.g., cytoplasmic conductivity change. The …

Investigation Of Mass-Manufactured Nanostructured Material For Contamination Detection Via Structural Color Analysis, Oisin Biswas
Electronic Theses and Dissertations
The spread of the SARS-CoV-2 virus has underlined the importance of monitoring surface contamination of infectious elements on commonly used surfaces. Current monitoring methods of surfaces are both lengthy and expensive. The work in this project took advantage of the optical phenomenon of structural color to develop a rapid, low-cost, and contactless method of surface contamination detection. To accomplish this, a mass-produced material imprinted with a nanostructured pattern capable of exhibiting this structural color phenomenon was used. Structural color, or the bands of color that appear as different wavelengths of light reflect off a textured surface, can vary as a …
Gradient Generating Microfluidic Coculture System For Disease Modeling And Neural Development, Phaneendra Chennampally
Gradient Generating Microfluidic Coculture System For Disease Modeling And Neural Development, Phaneendra Chennampally
Electronic Theses and Dissertations
Cellular microenvironment or cell niche plays an important role in developmental biology and disease pathophysiology. Physical or chemical signals in microenvironment drive the cellular activity. These signaling molecules are generated from the surrounding cells/tissues as part of intercellular communication; a fundamental property of a cell. Dynamic profile of these signaling molecules in the microenvironment plays a pivotal role in transfer of molecular information from cell to cell in disease proliferation or fate determination. Recapitulating these signaling cues in an in vitro study is difficult to achieve using standard cell culture techniques. However microfluidic systems are capable of addressing these issues, …
Computational Modeling Of 3d-Printed Lactobacillus-Antibiotic Scaffolds For Bacterial Vaginosis., Veeresh P. Rai
Computational Modeling Of 3d-Printed Lactobacillus-Antibiotic Scaffolds For Bacterial Vaginosis., Veeresh P. Rai
Electronic Theses and Dissertations
The standard treatment for bacterial vaginosis (BV) is currently antibiotics, such as metronidazole, clindamycin, or tinidazole. These antibiotics are highly effective in getting rid of bacteria in the female reproductive tract (FRT); however, there are some bacteria that provide benefits to the FRT which also get expunged. While there are many strains of bacteria that play a beneficial role in the FRT, lactobacilli are among the most important. These bacteria are responsible for maintaining a healthy environment in the FRT via pH regulation by lactic acid metabolism. Antibiotics eliminate all bacteria from an environment, including lactobacilli, and as a result, …
Update Of Multicellular Data Standard To Match Physicell "Compact" Output., Reid A Honeycutt
Update Of Multicellular Data Standard To Match Physicell "Compact" Output., Reid A Honeycutt
Electronic Theses and Dissertations
In the field of multicellular biology, it is currently very difficult for researchers to share digital data, due to a lack of standards for said data. Each laboratory will collect data from a particular project, which will describe only the features relevant to their research, and then present that data using written descriptions, charts, graphs, and images. This makes it difficult for the data produced by these studies be leveraged in other research efforts by scientists studying similar phenomena. An open-source, universal standard that attempts to meet this need is the MultiCellular Data Standard (MultiCellDS). MultiCellDS is described as a …
Development Of A Novel Haptic Feedback System For Gait Training Applications, Mohsen Alizadeh Noghani
Development Of A Novel Haptic Feedback System For Gait Training Applications, Mohsen Alizadeh Noghani
Electronic Theses and Dissertations
Until recently, study and correction of motor or gait functions required costly sensors and measurement setups (e.g., optical motion capture systems) which were only available in laboratories or clinical environments. However, due to (1) the growing availability and affordability of inertial measurement units (IMUs) with high accuracy, and (2) progress in wireless, high bandwidth, and energy-efficient networking technologies such as Bluetooth Low Energy (BLE), it is now possible to measure and provide feedback in real-time for biomechanical parameters outside of those specialized settings. To enable gait training without an expert who can provide verbal feedback, augmented feedback, which is divided …

Segmentation And Statistical Analysis Of Imaged And Simulated 3d Chromosome Territories, Hannah Varney
Electronic Theses and Dissertations
With advances in microscopy in the 21st century, more accurate measurements of the morphology and position of chromosome territories (CTs) during interphase within the cell nucleus can be obtained. These measurements have determined that CTs are non-randomly positioned, and their position and shape can change during diseases like breast cancer, pancreatic cancer, and Huntington’s Disease. The 2.5D Wavelet Transform Modulus Maxima segmentation method uses multi-scale edge detection to accurately segment objects in 3D images to find variables such as surface area, volume, and diameter without assuming a spherical or ellipsoidal shape. Measurements of centroid distance, shell distance and radial distance …
Development Of A Microfluidic Viscoelastic Hemostatic Assay For Real-Timeviscosity Measurements Of Blood, Shay Kent
Electronic Theses and Dissertations
Blood coagulation disorders are malfunctions in the body’s ability to control blood clotting. It can result in either insufficient clotting causing an increased risk of bleeding or excessive clotting obstructing blood flow. The rapid and accurate diagnosis of coagulopathies is an important, unmet need in the clinical setting. Rapidly identifying the source of bleeding, either acquired or inherited, is critical to reduce the risk of major blood loss and deliver personalized hemostatic therapies. Viscoelastic hemostatic assays, or VHAs, deliver an effective solution to the diagnosis of coagulopathies by evaluating global hemostatic function using whole blood rather than plasma. VHAs are …

Refinement And Automation Using Algorithmic Control Of Breathforce, A Respiratory Training System For Patients With Spinal Cord Injuries., Anna Goestenkors
Electronic Theses and Dissertations
Spinal cord injuries (SCI) can lead to impaired respiratory and cardiovascular function and a general decrease in lung compliance. This can complicate breathing as well as impair the ability to sigh, cough, and clear secretions, leading to increased risk of respiratory infections. Respiratory training has been shown to combat these effects. BreathForce is under active development to create a user-centric inspiratory-expiratory device that is an affordable option for at-home training. This study reports on the refinement of valve design and automation incorporated into BreathForce to enhance and enforce clinical practices and processes as part of the respiratory training protocol used …
Bilateral Ground Reaction Force Jumping Asymmetry And Performance, Keith Painter
Bilateral Ground Reaction Force Jumping Asymmetry And Performance, Keith Painter
Electronic Theses and Dissertations
The prevalence of asymmetry in performance research has increased in recent years with mixed results. Much of the performance research has focused on unilateral jumping activities attempting to show relationships to other performance variables. However, bilateral ground reaction forces (bGRF) from jumps are more frequently assessed in athlete monitoring programs and the asymmetry from those jumps could be a simple addition to data already being collected. Research into bGRF asymmetries is lacking and no studies have addressed longitudinal changes. Additionally, research into the relationship of asymmetries to performance have infrequently used athletes. For these reasons, this dissertation will focus on …
Development And Evaluation Of A Body Weight Support Treadmill For Use With Locomotor Training On Pediatric Spinal Cord Injury Patients., Winston T Rauch
Development And Evaluation Of A Body Weight Support Treadmill For Use With Locomotor Training On Pediatric Spinal Cord Injury Patients., Winston T Rauch
Electronic Theses and Dissertations
The consequences of spinal cord injury (SCI) are devastating regardless of the age of a patient. When the injury occurs in children five years old or younger, however, the impact is magnified due to the inevitable development of scoliosis (96%) and hip dysplasia (57%) (Schottler et al., 2012). To reduce occurrence of these complications and improve the quality of life for these patients, specialized activity-based therapies such as locomotor training (LT) are being increasingly used to improve overall trunk control and muscle activity in the lower extremities (Harkema et al., 2012; Howland et al., 2014). The aim of this therapy …

Development Of A Pneumatically Controllable Microdroplet Generator With Electrical Sensing, Gnanesh Nagesh
Electronic Theses and Dissertations
Microfluidic droplet generation is popular in lab-on-a-chip based biochemical analysis because it can provide precise and high throughput fluids in the form of small droplets. This thesis presents a T-junction microdroplet generator with pneumatic actuation for regulating droplet size and a capacitance-based sensor with real-time sensing capability for characterizing droplet composition and size. The multi-layer device developed in this thesis is compatible with rapid manufacturing using a desktop-based laser cutter to fabricate the fluidic and pneumatic layers. A finite element based numerical model was developed to predict the best operating and geometric parameters for droplet generation. It was revealed that …
Manipulation Of Particles And Fluid With Surface Acoustic Waves, Joel Scott Tewksbury
Manipulation Of Particles And Fluid With Surface Acoustic Waves, Joel Scott Tewksbury
Electronic Theses and Dissertations
Small biomolecules can be challenging and expensive to isolate and manipulate, but they may be the key for developing more efficient diagnostic tools for the detection of biomarkers, such as those associated with pancreatic cancer. Pancreatic cancer has a survival rate of less than 10% five years after diagnosis. For this reason, the research and establishment of cheap, effective screening tools are highly desired. One promising method of screening is separating and identifying biomarkers in a patient’s blood that are indicative of pancreatic cancer. The purpose of this project is to develop a system using surface acoustic waves (SAWs), in …

Machine Learning Applied To Colloidal Properties Of Perfluorocarbon Nanoemulsions For Imaging In Ards/Ali, Marco Hosfeld
Electronic Theses and Dissertations
Acute Respiratory distress Syndrome (ARDS) and Acute Lung Injury (ALI) are inflammatory lung pathologies consisting of non-hydrostatic pulmonary edema leading to hypoxia and impaired gas exchange in the lungs. ARDS/ALI is both difficult to study and treat as it is not in itself a specific pathology but rather a syndrome consisting of many pathologies that vary case by case. It is, however, consistently characterized by an explosive acute inflammatory response in the lung parenchyma leading to hypoxia. Although time has seen to an increase in the understanding of ARDS/ALI, the mortality rate remains in the range of 30-50%. For these …
Diagnosis Of Melanoma Disease State From Patient Blood Samples Using Photoacoustic Flow Cytometry, Jacob Salvatore
Diagnosis Of Melanoma Disease State From Patient Blood Samples Using Photoacoustic Flow Cytometry, Jacob Salvatore
Electronic Theses and Dissertations
Approximately 1 in 50 Americans develop melanoma in their lifetime. Early detection of melanoma is pivotal to the survival of the patient, with a 99% survival rate for 5 or more years after an early diagnosis. Metastasis, or the spread of cancer, increases the chances of cancerous growth in other parts of the body. In this study, we propose the use of photoacoustic flow cytometry as a diagnostic of patient disease state, by closely monitoring the amount of circulating melanoma cells (CMCs) in a patient’s blood before and after chemotherapy treatments. Using this patient data, we were able to make …
Detecting A Heterogenous Sample Of Pigmented Melanoma Cell Lines Using Photoacoustic Flow Cytometry, Margaret Cappellano
Detecting A Heterogenous Sample Of Pigmented Melanoma Cell Lines Using Photoacoustic Flow Cytometry, Margaret Cappellano
Electronic Theses and Dissertations
Metastatic melanoma is the deadliest form of skin cancer, which is in part, attributed to its rapid aggression and lack of response to typical treatment methods. There are far too often cases where a lymph node biopsy does not detect the severity of the cancer, which in turn causes a lack of diagnosis until a mass can be visually detected on a scan, such as a PET, CT, or MRI. Once visible on a scan, the cancer is too progressive for successful treatment. To avoid this, we investigated how a blood sample can be used to negate a missed diagnosis, …
Growth Of Perkinus Marinus By Bioreactor Fermentation, Caitlin Murphy
Growth Of Perkinus Marinus By Bioreactor Fermentation, Caitlin Murphy
Electronic Theses and Dissertations
Perkinsus marinus is a marine protozoan responsible for “Dermo” disease in the eastern oyster species, Crassostrea virginica. P. marinus has been detected along the Atlantic Coast of the United States and Mexico. In laboratory studies, many parasites need to be maintained in vivo, which complicates the study of the organism. However, In vitro cell culture for P. marinus at a scale of 1 mL was established in 1993 by several groups with the 1995 optimization (Gauthier & Vasta, 1993, 1995) considered the gold standard for small scale growth of this species. In addition to its importance as a …

Electronic Theses and Dissertations
A healthy gait is often taken for granted when walking. However, if a stroke, spinal cord injury (SCI), or traumatic event occurs the ability to walk may be lost. In order to relearn how to walk, gait rehabilitation is required. Including arm swing in gait rehabilitation has been shown to help in this process. This thesis presents two tasks to investigate the mechanics of arm swing and ways to provide assistance to induce arm swing in gait rehabilitation.
The firsts task completed was a study on the effects of forearm movement during gait. Twelve healthy subjects walked under three conditions …

Isomotive Dielectrophoresis For Enhanced Analyses Of Cell Subpopulations., Mohamed Zakarya Rashed
Electronic Theses and Dissertations
As the relentless dream of creating a true lab-on-a-chip device is closer to realization than ever before, which will be enabled through efficient and reliable sample characterization systems. Dielectrophoresis (DEP) is a term used to describe the motion of dielectric particles/ cells, by means of a non-uniform electric field (AC or DC). Cells of different dielectric properties (i.e., size, interior properties, and membrane properties) will act differently under the influence of dielectrophoretic force. Therefore, DEP can be used as a powerful, robust, and flexible tool for cellular manipulation, separation, characterization, and patterning. However, most recent DEP applications focus on …
Nanocellulose Conduits For Enhanced Regeneration Of Peripheral Nerve Injuries, Nicklaus R. Carter
Nanocellulose Conduits For Enhanced Regeneration Of Peripheral Nerve Injuries, Nicklaus R. Carter
Electronic Theses and Dissertations
Peripheral neuropathy is estimated to afflict 20 million people in the United States. Most cases of neuropathy result from physical injuries and trauma arising from automobile accidents and war. Peripheral nerves have the intrinsic ability to regenerate over time, bridging the injury gap. However native regeneration is limited to a distance of only a few millimeters. Current methods utilized to assist in the regeneration of peripheral nerves over distances exceeding those amenable to native repair include nerve autografts and allografts, and implantation of conduits. Nerve autografts are regarded as the most effective method but require a second surgical site to …

Simulation Of Fracture Strength Improvements Of A Human Proximal Femur Using Finite Element Analysis., Waleed Ebraheem Alim
Electronic Theses and Dissertations
The most common hip fracture in the elderly occurs as a result of a fall to the side with impact over the greater trochanter resulting in a fracture of the proximal femur. The fracture usually involves the femoral neck or the intertrochanteric region. It has recently been determined that the fracture crack of a hip fracture typically initiates on the superior-lateral cortex of the femoral neck and then propagates across the femoral neck, resulting in a complete fracture. The strength of the superior-lateral cortex of the femoral neck is likely determined by the combined properties of the generally thin cortex …

Electronic Theses and Dissertations
T-cell transformation is an ever-expanding treatment for several types of cancer, with a potential to be adapted to other disorders in which the immune system plays a key role in the pathophysiology. Currently, all FDA approved chimeric antigen receptor (CAR) T-cell cancer therapies rely on transformation via viral transduction. However, viral transduction is plagued by poor consistency and the potential to create adverse immune reactions when T-cells are reintroduced into a patient. Other transformation methods are being explored, with an alternative called acoustofluidic sonoporation showing promise. In these procedures, cells are passed through a channel, of the millimeter scale, while …

Rational Design Of Advanced Functional Materials For Electrochemical Devices, Shun Lu
Electronic Theses and Dissertations
In recent years, there has been a fast-growing trend in developing urea (CO(NH2)2) as a substitute H2 carrier in energy conversion due to its high energy density, nontoxicity, stability, and nonflammability. Urea, a byproduct in the metabolism of proteins and a frequent contaminant in wastewater, is an abundant compound that has demonstrated favorable characteristics as a hydrogen-rich fuel source with 6.7 wt % gravimetric hydrogen content. Also, there is 2-2.5 wt % urea from mammal urine; therefore, 0.5 million ton of additional fuels will be produced per year just from human urine (240 million ton each year). Electrochemical oxidation has …
