A Microfluidic Assay For Single Cell Bacterial Adhesion Studies Under Shear Stress, Amanda Trusiak

The study of bacterial adhesion to host cells is important in understanding bacterial pathogenesis and developing new therapeutic approaches. Here, we studied bacterial adhesion under shear stress using a novel microfluidic method. Specifically, the adhesion of a uropathogenic E. coli strain (FimHOn, ATCC 700928/CFT073) to mannose-modified substrates was studied under flow conditions. The FimHOn E. coli strain expresses FimH which is a mannose-specific adhesin found on the fimbriae that binds to glycoproteins on the epithelium. Fecal indicator bacteria (FIB), including E. coli, are the number one cause of water quality impairments in the United States according to the USEPA. FIB are used as a predictor to identify the possible presence of pathogens in waterbodies. E. coli is a useful indicator of gastrointestinal (GI) related illnesses from contact with fresh water. Fecal indicator bacteria, including E. coli, are the leading cause of water quality impairments within assessed waters in the United States. The source of E. coli includes WWTP, leaking sewers, animal manure, wildlife, livestock, and stream bed sediment. Storm events contribute to bacteria loading within waters through wash-in of land sources of bacteria and resuspension of bacteria within sediments. Permeable pavements systems and tree boxes are a common type of Green Infrastructure (GI) Stormwater Control Measures (SCMs) that are often used for mitigating the stormwater runoff. In this study two permeable pavement systems and a tree box installed along parking lanes of an urban street in Louisville, KY, were investigated to evaluate their performance on improving stormwater runoff quality. The objective of the study was to compare the trihalomethanes (THMs) produced from ferrate with hypochlorite and to determine how different the THM production would be for a given degree of disinfection (3 log reduction in Heterotrophic Plate Count (HPC)). Different water samples were collected from Lake Claire, Atlantic Ocean, and secondary effluent from an advanced wastewater treatment plant. THM formation was determined using a standard assay over 7 days at room temperature. In addition samples were tested for Total Coliform Escherichia coli (TC/E.coli), and heterotrophic bacteria using HPC by spreadplating on R2A agar. Dissolved organic carbon (DOC) was measured …
