Enzymatic Hydrolysis Of Biomass At High-Solids Loadings – A Review, Alicia A. Modenbach, Sue E. Nokes

Biosystems and Agricultural Engineering Faculty Publications

Enzymatic hydrolysis is the unit operation in the lignocellulose conversion process that utilizes enzymes to depolymerize lignocellulosic biomass. The saccharide components released are the feedstock for fermentation. When performed at high-solids loadings (≥ 15% solids, w/w), enzymatic hydrolysis potentially offers many advantages over conversions performed at low- or moderate-solids loadings, including increased sugar and ethanol concentrations and decreased capital and operating costs.

The goal of this review is to provide a consolidated source of information on studies using high-solids loadings in enzymatic hydrolysis. Included in this review is a brief discussion of the limitations, such as a lack of available …

The Use Of Co-Culturing In Solid Substrate Cultivation And Possible Solutions To Scientific Challenges, Wanying Yao, Sue E. Nokes Jul 2013

The Use Of Co-Culturing In Solid Substrate Cultivation And Possible Solutions To Scientific Challenges, Wanying Yao, Sue E. Nokes

Biosystems and Agricultural Engineering Faculty Publications

This perspective systematically summarizes the use of solid substrate co‐cultures in agriculture, food, plant, and industrial biotechnology applications. The summarization is organized by organism, i.e. fungus, bacteria, yeast and then co‐cultivation of either two or three organisms. Generally, in solid substrate co‐culture, the organisms synergistically penetrate and degrade the solid substrate, thereby increasing product yield and productivity over a monoculture. Efforts to increase co‐culture performance include optimizing process parameters (pH, temperature, moisture, and oxygen demand) and defining the acceptable types of substrate. Scientific challenges exist in understanding the interactions between microbial stains, such as viability, suite of products, and bio‐transformations. …

Horizontal Pressures In Cylindrical Metal Silos And Comparison With Different International Standards, José W. B. Do Nascimiento, José P. Lopes Neto, Michael D. Montross

Biosystems and Agricultural Engineering Faculty Publications

The focus of this research was to evaluate the horizontal pressures on a cylindrical metal silo of corrugated walls and flat bottom with 1.82m diameter and 5.4m high, and to compare the values with those obtained theoretically by the ISO 11697, EP 433 and AS 3774 standards. The silo was symmetrically filled and constant speed with wheat cv. soft red for two different height/diameter ratios (H/D) and was unloaded through three orifices with a diameter of 71.6mm, one concentric and two eccentrics. Horizontal pressures were measured on the walls of the silo at three positions using hydraulic type pressure cells. …

Influence Of Media Composition On The Growth Rate Of Chlorella Vulgaris And Scenedesmus Acutus Utilized For Co2 Mitigation, Czarena L. Crofcheck, Xinyi E, Aubrey Shea, Michael D. Montross, Mark Crocker, Rodney Andrews

Biosystems and Agricultural Engineering Faculty Publications

Atmospheric carbon dioxide levels have increased since the industrial revolution due to increasing combustion of fossil fuels. One possible CO2 mitigation strategy is the use of microalgae for mitigation of CO2. This paper focuses on the influence of media composition on the growth rate of two microalgae strains, Chlorella vulgaris and Scenedesmus actus. A KNO3 based medium was found to work better for Chlorella, while a urea based equivalent worked better for Scenedesmus. The urea based media investigated here resulted in growth similar to that found with previously reported KNO3 based media. …

The microbiological testing of foods is a well-established science. Due to the severity of foodborne pathogen illnesses, the widespread use and implementation of rapid detection methods in food testing labs is increasingly important. The first step for successful testing is sampling. Surfactants have been widely used in food microbiology, but there is not much, if any, published research about the use of fatty alcohols and chemical dispersants as aids in microbial separation and recovery. The microbial extraction efficiency of Escherichia coli K12 and Listeria innocua from three representative food matrices (hot dogs, spinach, and milk) was measured using chemical additives …

Designing Contour Weep Berms To Reduce Agricultural Nonpoint Source Pollution, Richard C. Warner, Carmen T. Agouridis, Ross L. Guffey Jan 2013

Designing Contour Weep Berms To Reduce Agricultural Nonpoint Source Pollution, Richard C. Warner, Carmen T. Agouridis, Ross L. Guffey

Biosystems and Agricultural Engineering Faculty Publications

Nonpoint source pollution (NPS) of surface waters is a significant issue in agricultural lands, and best management practices (BMPs) are often used to reduce these impacts. Since the effectiveness of a BMP depends on a large number of widely varying factors, it is important to continue to develop BMPs in order to provide designers with more tools to use to maximize NPS removal. The contour weep berm is a new structural BMP constructed out of earth and subsequently vegetated. It is a linear BMP that is used in combination with a down-gradient vegetated filter strip or forested riparian buffer. Preliminary …

Development Of A Semi-Automated Tobacco Stripping Machine Utilizing String Trimmers, Robert George Sperry, John H. Wilhoit, George A. Duncan Jan 2013

Development Of A Semi-Automated Tobacco Stripping Machine Utilizing String Trimmers, Robert George Sperry, John H. Wilhoit, George A. Duncan

Biosystems and Agricultural Engineering Faculty Publications

Conventional stripping of burley tobacco is labor intensive and typically requires 50 to 75 worker hours per acre (wkr•h/acre). The goal of the project was to reduce labor by optimizing leaf removal by string trimmer heads using combinations of strings lengths and motor speeds. In tests conducted on a single grade, all leaves outside the grade were removed by hand. Plants were run through the machine for string lengths of 13, 18, and 23 cm (5, 7, and 9 in.) and associated motor speeds which were monitored and recorded. Stripping efficiencies were calculated for each plant and collectively for each …

A Correlation-Based Optical Flowmeter For Enclosed Flows, Naiqian Zhang, Joseph S. Dvorak, Yali Zhang Jan 2013

A Correlation-Based Optical Flowmeter For Enclosed Flows, Naiqian Zhang, Joseph S. Dvorak, Yali Zhang

Biosystems and Agricultural Engineering Faculty Publications

A low-cost flowmeter would be very useful in a wide variety of monitoring situations. This article discusses the development of such a flowmeter based on optical components and its testing with water in an enclosed flow system. The sensor consisted of two sets of LEDs and phototransistors spaced 4 cm apart, monitoring the optical properties of the fluid at upstream and downstream locations, respectively. A small amount of dye was injected into the flow, which caused a change in the optical properties of the fluid at both locations. The time required for this change to move from the upstream to …

In 2006, a social marketing campaign was developed to increase the installation of rollover protective structures (ROPS) on unprotected New York tractors. Using data gathered from the program's hotline, the impact of price increases on farmers' interest in ROPS is examined. Pricing data were obtained for all rigid ROPS kits commercially available in the U.S. since 2006. These data were stratified into two groups of ROPS suppliers: (1) tractor manufacturers that sell ROPS for their own tractors, referred to in this study as original equipment manufacturers (OEMs), and (2) aftermarket (AM) ROPS suppliers. The trend in price increases was contrasted …

Atmospheric carbon dioxide levels have increased since the industrial revolution due to the increase in combustion of fossil fuels. One possible alternative strategy is the use of microalgae for CO2 capture and recycling. Major components in coal-derived flue gas that may accumulate and effect algae growth include both sulfur oxides and fly ash. However, in practical application, sulfur oxides will be converted quickly to the acid product (H2SO4) in the aerobic aqueous conditions of algae cultivation. In this article, the influence of elevated H2SO4 levels and the presence of coal-derived fly ash …
