Trauma diagnostics and management are major aims of research for implantable amperometric enzyme biosensor technology. Biosensors are capable of monitoring metabolic variables in a minimally invasive manner and have great potential to augment current wireless vital sign monitoring technologies in order to make a more robust physiologic status monitoring platform. The dual responsive Electrochemical Cell-on-a-Chip Microdisc Electrode Array (ECC MDEA 5037) is a recently developed electrochemical transducer for use in a wireless, implantable biosensor system for the continuous measurement of interstitial glucose and lactate. To expand the design envelope and supplement the materials library available to biomaterials scientists, the copper(I)-catalyzed azide-alkyne cycloaddition (CuCAAC) was explored as a route to design, synthesize and characterize bio-functional small-molecules, nanoparticles, and microfibers. In each engineered system, the use of click chemistry provided facile, bio-orthogonal control for materials synthesis; moreover, the results provided a methodology and more complete, fundamental understanding of the use of click chemistry as a tool for the synergy of biotechnology, polymer and materials science. It is estimated that this year more than 200,000 women in the United States will be diagnosed with breast cancer. Treatment for most occurrences of breast cancer will often include surgical removal of the tumorigenic tissue, resulting in a soft tissue defect within the subcutaneous tissue of the skin. Post-surgical reconstruction methods are often sought by patients to restore the aesthetic function of the breast via cellular or non-cellular methods; however, because of complications associated with currently used methods for breast reconstruction, researchers are investigating tissue engineering methods to produce viable autologous adipose tissue for breast reconstruction.

Despite over three decades of research in blood-material interactions, the biomaterials field has been unsuccessful in developing a truly non-thrombogenic biomaterial. This is due to an incomplete understanding of the factors underlying biomaterial-associated thrombosis, especially the mechanisms mediating the interactions of platelets with the adsorbed plasma protein layer(s) on the implant surface. The work presented here is motivated by the primary goal of delineating these mechanisms, and understanding the platelet receptors involved, as well as the domains/amino acid sequences they bind to in the protein molecules.

Spinal cord injuries cost the United States $20 billion per year, with an existing patient population of 256,000 growing by an estimated 12,000 each year. Current clinical therapies for spinal cord injury are limited to spinal immobilization and realignment via traction, surgery, administration of methylprednisolone sodium succinate (MPSS) within eight hours post-injury, and rehabilitation exercises. While these therapies are important and may minimize damage and restore limited function, there is a dire clinical need for treatments to address the growing population of chronically-injured patients.

Evaluation of retrieved joint arthroplasty bearings provides unique evidence related to the physiological environment in which bearing materials are expected to perform. This dissertation describes the development of novel spatial sensors and measurement strategies for standardized, quantitative assessments of arthroplasty bearings, including total knee replacements, unicompartmental knee replacements, and total hip replacements. The approach is to assess bearings that endured a finite duration of function in patients, with particular emphasis on expanding our understanding of the biomechanical conditions specific to bearing function and wear in the physiological environment. Several quantifiable parameters are identified that prove comparable to pre-clinical in vitro …
