With the increasing prevalence of diabetes in the United States and worldwide, blood glucose monitoring must be accurate and reliable. Current enzymatic sensors have numerous disadvantages that make them unreliable and unfavorable among patients. Recent research in glucose affinity sensors correct some of the problems that enzymatic sensors experience. Dextran and concanavalin A are two of the more common components used in glucose affinity sensors. When these sensors were first explored, a model was derived to predict the response time of a glucose affinity sensor using concanavalin A and dextran. This dissertation describes the synthesis of photo-crosslinkable copolymers and their utilization for the fabrication and testing of tunable and responsive one-dimensional (1D) photonic multilayers. Photonic multilayers exhibit structural color due to the interference of incident light at layer interfaces, providing a convenient route towards optically responsive materials that do not rely on potentially light- or oxygen-sensitive chromophore-containing pigments and dyes. A fabrication technique based on sequential spin-coating and crosslinking of photo-crosslinkable polymers is used to assemble tunable and responsive photonic multilayers. A new fluorescence enhancement technical platform based on nanostructured aluminum oxide (NAO) has been discovered. Experiments reveal that this new platform can enhance the fluorescence signals up to two or three orders of magnitude compared to a traditional glass substrate platform. In order to realize a large-scale arrayed fluorescence biosensing platform for high throughput applications, a rapid and cost-effective process has been developed to fabricate the micropatterned NAO of a variety of shapes. An optical characterization technology, by introducing UV light irradiance, has been found for the first time to determine if the NAO has been formed from Al and …
