Polyriboinosinic acid-polyribocytidylic acid (Poly I:C) serves as a synthetic mimic of viral double-stranded dsRNA, capable of inducing apoptosis in numerous cancer cells. Despite its potential, therapeutic benefits, the application of Poly I:C has been hindered by concerns regarding toxicity, stability, enzymatic degradation, and undue immune stimulation, leading to autoimmune disorders. To address these challenges, encapsulation of antitumor drugs within delivery systems such as cationic liposomes is often employed to enhance their efficacy while minimizing dosages. Digital microfluidics in combination with emulsion microfluidics are crucial building blocks of droplet-based microfluidics, which are prevalent in a wide variety of industrial and biomedical applications, including polymer processing, food production, drug delivery, inkjet printing, and cell-based assays. The manipulation of magnetic microparticles or microdroplets in microscale fluid environments is one of the most critical processes in the systems and platforms based on microfluidic technology. "In this dissertation, two dimensional and three dimensional, transient CFD simulations are conducted to investigate the active pumping and mixing in microfluidics driven by Electromagnetic/Lorentz force. Shallow disk/ring cylindrical microfluidic cell and shallow cuboid microfluidic cell with electrodes deposited on the bottom surface are modelled for mixing and pumping purposes respectively. By applying voltage across specific pair of electrodes, an ionic current is established in the weak conductive liquid present in the cell. Heterogeneity in responses of cells to a stimulus, such as a pathogen or allergen, can potentially play an important role in deciding the fate of the responding cell population and the overall systemic response. Measuring heterogeneous responses requires tools capable of interrogating individual cells. Cell signaling studies commonly do not have single-cell resolution because of the limitations of techniques used such as Westerns, ELISAs, mass spectrometry, and DNA microarrays. Due to the high flexibility and versatility in the process development of microfluidic devices, an epoxybased, high-aspect ratio photoresist SU-8 is now attracting attention as the main structuring material of the fluidic channels. Manipulation of the surface properties of SU-8 channel wall is critical to attach functional films such as enzyme-immobilized layers or biocompatible layers. We describe a new in situ patterning method to form a hydrogel film on SU-8 by a photografted polymerization procedure. The hydrophobic surface of SU-8 is modified using a surface bound initiator HCPK (1-hydroxycyclohexyl phenyl ketone). The objective of this work is to develop a new microsensor system that can monitor dissolved oxygen and hydration environment at the plant root zone. A miniaturized plant growth system is prepared including the root zone layer, either a porous ceramic tube or porous ceramic wafer on which the plant is grown, and an underlying fluidic channel to deliver nutrients and water to the root zone. We demonstrate the feasibility of using a flexible microsensor array for dissolved oxygen detection, and a four-electrode impedance microelectrode for wetness detection on the surface of a porous tube nutrient delivery system. A new dissolved oxygen monitoring microsystem is proposed to achieve in situ intelligent self-calibration by using an electrochemically actuated fluidic system. The electrochemical actuation, based on water electrolysis, plays two critical roles in the proposed microsystem. First, the electrochemically generated gases serve as the calibrants for the in situ 2-point calibration/diagnosis procedure of the microsensor in a chip. Secondly, the electrochemical generation and collapse of gas bubbles provide the driving force of the bidirectional fluidic manipulation for sampling and dispensing of the sample solution.
