Vegetation along roadsides is important to prevent soil erosion, provide habitat, and filter water running off the road. Vegetation close to the pavement along highways in Nebraska does not readily establish and persist. It is thought that the sodium and bulk density are the driving factors behind the lack of vegetation. After a construction project the shoulder is seeded into the compacted soil, and during winter salts can accumulate in the soil because of deicing agents being used. Wildflowers are crucial in the ecological function of the low-input roadside plant communities in terms of water andnutrient cycling, nutrient inputs such as nitrogen, total plant canopy cover, stand longevity, and provision of habitat for numerous small animals. Variable rate irrigation (VRI) may improve center pivot irrigation management, including deficit irrigation. A remote-sensing-based evapotranspiration model was implemented with Landsat imagery to manage irrigations for a VRI equipped center pivot irrigated field located in West-Central Nebraska planted to maize in 2017 and soybean in 2018. In 2017, the study included VRI using the model, and uniform irrigation using neutron attenuation for full irrigation with no intended water stress (VRI-Full and Uniform-Full treatments, respectively). Mid-late season weeds are those that escape the early season herbicide applications and those that emerge late in the season. They might not affect the crop yield, but if uncontrolled, will produce a large number of seeds causing problems in the subsequent years. In this study, high-resolution aerial imagery of mid-season weeds in soybean fields was captured using an unmanned aerial vehicle (UAV) and the performance of two different automated weed detection approaches – patch-based classification and object detection was studied for site-specific weed management. With the increasing meat demand and awareness of sustainability, concerns have been raised regarding the sustainability of beef production and processing. However, scarce data and inadequate sustainability assessment frameworks for the U.S. beef processing industry limit the ability to develop new technologies and policies comprehensively without shifting sustainability burdens. Distillery wastewater treatment is a great ecological problem, for example, India produces 2.7 billion liters of alcohol that results in 40 billion liters of wastewater. However, this material can be seen as a resource since 11 million cubic meters of biogas at 60% methane could be produced in addition to cleaning the water. The primary goal of this dissertation was to leverage the capabilities of remote sensing technology for capturing detailed spatial information at different spatial resolutions to monitor agricultural crops and generate accurate input datasets for water resources models. Individuals with different genotypes are impacted differently by exposure to the same environment. In this technology review, the process of turning an average biogas into methanol will be presented. The purpose is to find a better method of transporting the energy contained within biogas so that it may be used in industry as a value added product. The first step is removal of hydrogen sulfide, a corrosive chemical that can breakdown mechanical parts. The second step is carbon capture and conversion of the biogas into hydrogen. The final step is the hydrogenation of carbon dioxide into methanol using a copper catalyst and an alcohol co-catalyst.

DNAzymes have been widely explored owing to their excellent catalytic activity in a broad range of applications, notably in sensing and biomedical devices. These newly discovered applications have built high hopes for designing novel catalytic DNAzymes. However, the selection of efficient DNAzymes is a challenging process but one that is of crucial importance. Initially, systemic evolution of ligands by exponential enrichment (SELEX) was a labor-intensive and time-consuming process, but recent advances have accelerated the automated generation of DNAzyme molecules. Sorghum (Sorghum bicolor [L.] Moench) is the fifth most productive cereal crop worldwide with some hybrids having high biomass yield traits making it promising for sustainable, economical biofuel production. To maximize biofuel feedstock yields, a more complete understanding of metabolic responses to low nitrogen (N) will be useful for incorporation in crop improvement efforts. Small RNAs (sRNAs) remain an understudied class of regulatory molecules in bacteria in general and in Gram-positive bacteria in particular. In the major human pathogen Staphylococcus aureus, hundreds of sRNAs have been identified; however, only a few have been characterized in detail. In this study, we investigate the role of the sRNA Teg41 in S. aureus virulence. We demonstrate that Teg41, an sRNA divergently transcribed from the locus that encodes the cytolytic alpha phenolsoluble modulin (αPSM) peptides, plays a critical role in αPSM production. Fungi are successful eukaryotes of wide distribution. They are known as rich producers of secondary metabolites, especially terpenoids, which are important for fungi-environment interactions. Horizontal gene transfer (HGT) is an important mechanism contributing to genetic innovation of fungi. However, it remains unclear whether HGT has played a role in creating the enormous chemical diversity of fungal terpenoids. Here we report that fungi have acquired terpene synthase genes (TPSs), which encode pivotal enzymes for terpenoid biosynthesis, from bacteria through HGT. Natural astaxanthin, a high-value carotenoid that is currently extracted mainly from marine organisms, was extracted from engineered camelina seed using ethanol-modified supercritical carbon dioxide (SC-CO2) for the first time, and compared with hexane and accelerated solvent extraction using hexane and ethanol. Response surface methodology (RSM) based on central composite rotatable design was employed to investigate the effect of pressure (30–45 MPa), temperature (40–60 °C), and ethanol concentration (10–35%, w/w). RSM-optimized conditions (41.6 MPa, 36.6 °C and 42.0% ethanol concentration) predicted the astaxanthin concentration as 437 μg/g oil, whereas the actual concentration was 421 ± 14 μg/g oil. Longifolene is a naturally occurring tricyclic sesquiterpene widely used in many different fields. Up to now, this valuable terpene was mainly manufactured from the high-boiling fraction of certain pine resins. Microbial production can be a promising alternative to the extraction from natural plant sources. Here, we present the metabolic engineering strategy to assemble biosynthetic pathway for longifolene production in Escherichia coli. E. coli was rendered to produce longifolene by heterologously expressing a codon optimized longifolene synthase from Picea abies. Hyperinsulinemia affects 72% of Fanconi anemia (FA) patients and an additional 25% experience lowered glucose tolerance or frank diabetes. The underlying molecular mechanisms contributing to the dysfunction of FA pancreas β cells is unknown. Therefore, we sought to evaluate the functional role of FANCA, the most commonly mutated gene in FA, in glucosestimulated insulin secretion (GSIS). This study reveals that FANCA or FANCB knockdown impairs GSIS in human pancreas β cell line EndoC-βH3.
