In his article "Theories of Opiate Addiction in the Early Works of Burroughs and Trocchi" Richard English discusses William S. Burroughs's and Alexander Trocchi's representations of opiate addiction with special reference to their early writings. English examines the concept of homo heroin that can be attributed to Burroughs and lists and expounds its qualities. Among these are: immorality, criminality, mono-objectuality, self- and other-indifference, and, most importantly, the radical physical transformation into a new species, which Burroughs extends in Naked Lunch. Maladaptive reactivity to stress is linked to improper decision making, impulsivity, and discounting of delayed rewards. Chronic unpredictable stress alters dopaminergic function and re-shapes dopaminergic circuits in key areas involved in decision making, and impairs prefrontal-cortex dependent response inhibition and working memory. Glial-derived neurotrophic factor (GDNF) is essential for regulating dopamine release in the basal ganglia and the survival of dopaminergic neurons, and GDNF-deficient mice are considered an animal model for aging-related Parkinsonism. Background: The provision of smoking cessation support in Australian drug and alcohol treatment services is sub-optimal. This study examines the cost-effectiveness of an organisational change intervention to reduce smoking amongst clients attending drug and alcohol treatment services. Methods/design: A cluster-randomised controlled trial will be conducted with drug and alcohol treatment centres as the unit of randomisation. Biochemically verified (carbon monoxide by breath analysis) client 7-day-point prevalence of smoking cessation at 6 weeks will be the primary outcome measure. Since the sample size of a typical neuroimaging study lacks sufficient statistical power to explore unknown genomic associations with brain phenotypes, several international genetic imaging consortia have been organized in recent years to pool data across sites. The challenges and achievements of these consortia are considered here with the goal of leveraging these resources to study addiction. The authors of this review have joined together to form an Addiction working group within the framework of the ENIGMA project, a meta-analytic approach to multisite genetic imaging data.
