Exploring Structure-Property Relationships Of Gags To Tailor Ecm-Mimicking Hydrogels, Ralf Zimmermann, Carsten Werner, James Sterling
Business and Information Technology Faculty Research & Creative Works
Glycosaminoglycans (GAGs) are a class of linear polysaccharides that are ubiquitous in the extracellular matrix (ECM) and on cell surfaces. Due to their key role in development, homeostasis, pathogenesis, and regeneration, GAGs are increasingly used in the design of ECM-mimicking hydrogels to stimulate tissue formation and regenerative processes via specifically orchestrated cell-instructive signals. These applications first and foremost build on the ability of GAGs to effectively bind, protect, and release morphogens. The specificity and strength of morphogen-GAG interactions are largely governed by the number and spatial distribution of negatively charged sulfate groups carried by GAGs. Herein, we summarize a mean-field …

Advances in the study of glycosoaminoglycan biohydrogels, label-free electrokinetic analysis of soft-diffuse layers in contact with saline solutions, and elucidation of ion-specific behavior in many biochemical systems offer the opportunity to marry these principal features in a new mathematical model of the mucosal glycocalyx. The model is based on the electroquasistatic subset of Maxwell's equations in the form of the steady-state continuum Poisson–Boltzmann equation for electrostatics with explicit incorporation of pairwise binding of ions to fixed charged-groups in the hydrogel. The pairwise association is modeled using reversible bimolecular reactions via stoichiometric dissociation constants that represent the rule of matching water …

The ability to detect when a person is hiding important information has high value in many applications. A new class of systems, termed autonomous scientifically controlled screening systems (ASCSS), is designed to detect individuals' purposely hidden information about target topics of interest. ASCSS represents a systematic synthesis of structured interviewing, orienting theory, defensive response theory, non-invasive psychophysiological measurement, and behavioral measurement. To evaluate and enhance the design principles, an automated screening kiosk (ASK) system was constructed. The ASK system has been used in a physical security screening scenario in which participants constructed and attempted to smuggle a fake improvised explosive …

Discourse plays a central role in organizing vision and computerization movement perspectives on IT innovation diffusion. While we know that different actors within a community contribute to the discourse, we know relatively little about the roles different actors play in diffusing different types of IT innovations. Our study investigates vendor versus adopter roles in social media and big data diffusion. We conceptualize the difference between the two IT innovations in terms of their decentralizability, i.e., extent to which decision rights pertinent to adoption of an organizational innovation can be decentralized. Based on this concept, we hypothesized: (1) adopters would contribute …

The Impact Of Regulation Fd On The Information Environment: Evidence From The Stock Market Response To Stock Split Announcements, Lili Eng, Joohyung Ha, Sandeep Nabar
Business and Information Technology Faculty Research & Creative Works
This paper examines the information environment effects of regulation fair disclosure (Reg FD). We investigate the stock market response to stock splits in the pre- and post-regulation periods. We find that abnormal returns around split announcement are positive in both periods, but the magnitude of the returns is smaller in the post-FD period relative to the pre-FD period. The difference between the pre- and post-FD period abnormal returns persists even after we control for factors that may affect split announcement returns. We also find that the magnitude of the association between announcement returns, and the unexpected portion of the split …

Co-Space refers to interactive virtual environment modelled after the real world in terms of look-and-feel, functionalities and services. We have developed a 3D virtual world named Nan yang Technological University (NTU) Co-Space populated with virtual human characters. Three key requirements of realistic virtual humans in the virtual world have been identified, namely (1) autonomy: Agents can function on their own, (2) interactivity: Agents can interact naturally with players, and (3) personality: Agents can exhibit human traits and characteristics. Working towards these challenges, we propose a brain-inspired agent architecture that integrates goal-directed autonomy, natural language interaction and human-like personality. We conducted …

Researchers have continued to study green consumer demographics with mixed results. The relevant literature since 1998 (the last review) was summarized to determine what demographics seemed to impact environmental purchasing. The greatest consistency was found for gender with women more likely to exhibit green behaviors. This research article analyzed survey responses to determine the environmentally friendly behaviors of consumers and their relationships to demographics. Several demographics were found to be related to a number of specific environmentally friendly behaviors, more than in prior research. The authors concluded that using specific behaviors, in contrast to general statements or attitudes, may be …

Automated Human Screening For Detecting Concealed Knowledge, Nathan W. Twyman
Business and Information Technology Faculty Research & Creative Works
Screening individuals for concealed knowledge has traditionally been the purview of professional interrogators investigating a crime. But the ability to detect when a person is hiding important information would be of high value to many other fields and functions. This dissertation proposes design principles for and reports on an implementation and empirical evaluation of a non-invasive, automated system for human screening. The screening system design (termed an automated screening kiosk or ASK) is patterned after a standard interviewing method called the Concealed Information Test (CIT), which is built on theories explaining psychophysiological and behavioral effects of human orienting and defensive …

Two mechanical lysis devices have been developed as compact, robust components to provide rapid sample preparation for nucleic acid diagnostic systems. One such component, known as the Micro Bead-Beater™ (μBB™, BBTM, Claremont BioSolutions, Upland, CA), is a compact device that is capable of ultrarapid lysis (>90% lysis in 30 s) of micro volumes (< 80 μL) of Bacillus spores in a continuous-flow format or in a disposable single-tube format. The μBB is also capable of processing much larger volumes of solutions containing spores or vegetative cells using a continuous-flow mode. A second mechanical lysis device designed as a disposable component is the microfluidic bead blender, which uses a small electric motor to spin vanes within the bead-laden solution. DNA quantification results using dsDNA-binding fluorescence dyes and real-time PCR are presented, comparing the lysis of Bacillus subtilis spores using the μBB™ with other well-known lysis techniques. Nanoscale imaging results obtained using scanning electron microscopy and transmission electron microscopy on B. subtilis spores lyzed using the μBB™ are also presented. © 2009, Society for Laboratory Automation and Screening. Moment Analysis Of Near-Equilibrium Binding Interactions During Electrophoresis, Yousef Daneshbod, James D. Sterling, Ali Nadim
Business and Information Technology Faculty Research & Creative Works
The electrophoretic transport of three chemically reacting species, two of which can bind reversibly to form the third, is analyzed mathematically. The species are assumed to move horizontally through a long channel with different electrophoretic mobilities and diffusion coefficients. By considering small perturbations of the system about equilibrium or when one of the two binding species is much more abundant than the other, the governing advection-reaction- diffusion equations can be linearized and studied via the method of moments. The result is a set of coupled ordinary differential equations for the moments that can be solved analytically. Analysis of the long-time …

Electrowetting Droplet Microfluidics On A Single Planar Surface, Christopher G. Cooney, Chao Yi Chen, Michael R. Emerling, Ali Nadim, James D. Sterling
Business and Information Technology Faculty Research & Creative Works
Electrowetting refers to an electrostatically induced reduction in the contact angle of an electrically conductive liquid droplet on a surface. Most designs ground the droplet by either sandwiching the droplet with a grounding plate on top or by inserting a wire into the droplet. Washizu and others have developed systems capable of generating droplet motion without a top plate while allowing the droplet potential to float. In contrast to these designs, we demonstrate an electrowetting system in which the droplet can be electrically grounded from below using thin conductive lines on top of the dielectric layer. This alternative method of …

By using high concentrations of buffer, electroosmotic flow within uncoated channels of a microfluidic chip was minimized, allowing the free solution electrophoretic separation of DNA. More importantly, because of the ability to efficiently dissipate heat within these channels, field strengths as high as 600 V/cm could be applied with minimal Joule heating (< 2 °C). As a result of the higher field strengths, separations within an 8-cm-long channel were achieved within a few minutes. However, when the electrophoretic separation of single-stranded DNA (ssDNA) less than 22 bases in length was performed, containing the fluorophore Texas Red as an end label, more than the expected single peak was observed at this high electric field. On the other hand, the free solution electrophoresis of a double-stranded DNA (dsDNA) consisting of a random sequence did exhibit the expected single peak. The appearance of these multiple peaks for ssDNA is shown to be dependent upon the base content and sequence of the ssDNA as well as on the chemical structure of the fluorophore used to tag the DNA for detection. Specifically, the peaks can be attributed to different secondary structures that result either from hydrophobic interactions between the DNA bases and an uncharged fluorescent dye or from G-quadruplexes within guanine-rich strands.
