Genetic And Mechanistic Dissection Of Missing-Self Killing By Natural Killer Cells, Jeesang Yoon

Natural killer (NK) cells eliminate transformed and infected cells through activating receptor-mediated cytotoxicity and through recognition of cells that have lost major histocompatibility complex class I (MHC-I) expression, a process termed missing-self recognition. Although missing-self recognition is a central principle of NK-cell biology, the mechanisms that execute missing-self killing remain incompletely understood. In this dissertation, we used complementary genetic and functional approaches to investigate the molecular basis of missing-self killing. We first implemented a CRISPR–Cas9 ribonucleoprotein electroporation platform for efficient genetic manipulation of primary murine NK cells, providing a versatile approach for functional studies of NK-cell biology. Maintenance of protein homeostasis is essential for cellular function and depends on the coordinated regulation of protein synthesis and degradation. The major intracellular pathways responsible for protein degradation are the ubiquitin-proteasome system (UPS) and autophagy. Disruption of these processes has been implicated in a broad range of diseases, including neuromuscular disorders. X-linked myopathy with excessive autophagy (XMEA) is a rare vacuolar myopathy caused by mutations in VMA21, which encodes an assembly chaperone required for proper assembly of the vacuolar H⁺-ATPase (V-ATPase), the primary proton pump responsible for acidifying intracellular compartments, including lysosomes. Novae are thermonuclear explosions on the surface of the white dwarf in a close binary system. They are multi-wavelength transients emitting across the electromagnetic spectrum from radio to gamma-rays. This thesis primarily focuses on the X-ray emission from one particular nova, V1674 Her. V1674 Her (Nova Her 2021) is known for its ultra-fast decline time of ��2 ∼ 1 day. This under normal circumstances implies massive white dwarf potentially approaching the Chandrasekhar limit. The recent cyberattacks targeting municipal water facilities across multiple U.S. states serve as another reminder that America’s critical infrastructure remains under constant attack. According to the FBI, Environmental Protection Agency (EPA), and the Cybersecurity and Infrastructure Security Agency (CISA), attackers successfully compromised operational technology (OT) devices, including programmable logic controllers (PLCs), by exploiting Internet-connected systems, changing administrator credentials, and disrupting normal operations. Fortunately, local operators responded quickly by isolating affected systems, restoring backups, and preventing what could have become a significant public health crisis.

While these incidents focused on water treatment facilities, they represent a much larger national security issue. …

This thesis demonstrates how machine learning techniques can solve computationally challenging problems across diverse areas of physics, from high-energy astrophysics to condensed matter systems, accelerating traditional computation. My first contribution addresses the computational expense of Monte Carlo calculations for radiative processes in relativistic plasmas. I develop a neural network sampling method that enables fast sampling from an arbitrary probability density, and demonstrate the method on inverse Compton scattering, achieving a speedup of up to an order of magnitude beyond standard methods. My second contribution addresses the structure and radiation of neutron star magnetospheres. I use physics-informed neural networks to model …

Background: Bipolar disorder (BD) features episodic shifts among mania, hypomania, depression, mixed states, and euthymia. Timely detection of mood transitions is difficult due to infrequent clinical touchpoints. Digital health technologies, including wearables and smartphones, offer a unique opportunity to passively and continuously monitor behavior and physiology that could reflect underlying mood dynamics in real-world settings.

Background/aim: Epilepsy is a chronic neurological disorder that significantly affects psychosocial functioning, self-perception, and quality of life. Emerging evidence suggests that posttraumatic stress (PTS) symptoms may be more prevalent in individuals with epilepsy than previously recognized, particularly following seizure-related experiences perceived as traumatic. The aim of this study was to evaluate short- and long-term PTS levels in individuals with newly diagnosed epilepsy and examine their associations with anxiety, depression, early maladaptive schemas, and quality of life.

Background/aim: Thoracic trauma accounts for approximately 25% of all trauma-related deaths and remains a significant public health challenge. Early identification of high-risk patients is essential for improving clinical outcomes. Background/aim: To assess the histopathological effects of three preservation solutions (70% ethanol, povidone-iodine, and ciprofloxacin) on human septal cartilage obtained during septoplasty and to compare their preservation characteristics for potential future graft applications. Optimal preservation of septal cartilage is clinically important because well-preserved grafts may reduce the need for secondary cartilage harvesting and the associated donor-site morbidity while facilitating reconstructive procedures during revision surgery.

Background/aim: To evaluate the clinical, demographic, and operational characteristics of patients with infectious diseases transported by helicopter ambulance and to examine differences in the transfer patterns, flight durations, and diagnostic categories.

Materials and methods: Registry data of patients with infectious diseases transported by helicopter ambulance were analyzed. Demographic characteristics, diagnoses, referring and receiving hospital types, intraprovincial and interprovincial transfer status, flight durations, and temporal distribution of transfers (hours, days, and months) were evaluated. Flight duration was calculated in minutes and compared between intraprovincial and interprovincial transfers.

Salinity stress impairs plant growth, nutrient homeostasis, and metabolism, particularly in arid ecosystems. Phyla nodiflora, a ground cover used for erosion control, shows preliminary salinity tolerance, but the mechanistic basis of its long-term adaptation and the potential for enhancement via potassium humate (KH) were unknown. Plant growth, key biochemical attributes, and nutrient uptake were assessed to elucidate how KH modulates physiological performance under increasing salinity. Plum (Prunus domestica L.) is a nutritionally valuable fruit with significant commercial importance, yet consumer acceptance is often constrained by inconsistent fruit quality. This study investigated the effects of cultivar, training system, and harvest stage on the chemical composition of plum fruit, focusing on polyphenols, organic acids, sugars, and sugar alcohols. With the rapid advancement of agricultural modernization and the increasing demand for agricultural products, inefficient logistics distribution has become a major bottleneck in rural supply chains. This study addresses the capacitated vehicle routing problem (CVRP) in agricultural logistics. A genetic algorithm (GA)-based optimization model was proposed to enhance distribution efficiency. The model integrates critical agricultural characteristics, including multidistribution center networks, seasonal delivery schedules, and regional road infrastructure constraints, to minimize both transportation distance and operational costs. The carob tree (Ceratonia siliqua L.) is a drought-tolerant species native to the Mediterranean Basin. It has been cultivated for centuries for its highly nutritious, edible pods. Genetic diversity is one of the key requirements for the effective management and utilization of plant genetic resources. In this study, we evaluated the genetic diversity and population structure of 169 seminatural carob individuals from Lebanon, Spain, and Morocco using nine expressed sequence tag-simple sequence repeat (EST-SSR) markers. Background: Discharge delay in resource-constrained Intensive Care Units (ICUs) remains a concerning and persistent challenge, resulting in delayed ICU admissions and after-hours discharges. These delays are associated with adverse patient outcomes and system inefficiency. While centralised patient flow centres and other strategies have been implemented to address these issues, their impact on ICU discharge delays remains unexplored.
