Resveratrol (RES) is a natural polyphenol compound with a wide range of health-promoting activities, including protective effects against colon cancer and renal disease. However, the premise of these benefits has been dampened since RES shows a poor oral bioavailability due to its rapid and extensive biotransformation after oral consumption. The paradox (low bioavailability but high bioactivity) warrants further investigations to determine the contribution of RES metabolites to the health benefits associated with RES. We identified 11 metabolites of RES in mice with high-resolution HPLC-MS/MS, then quantified two major metabolites - dihydro-resveratrol (DHR) and lunularin (LUN). Recent studies reported the positive association between exposure to insecticides and increased risks of obesity and type 2 diabetes, which is closely associated with non- alcoholic fatty liver disease (NAFLD). However, the effect of insecticide exposure on NAFLD remains unknown. Therefore, this project investigated the roles of insecticide exposure in the development of NAFLD using HepG2 hepatocytes. First, 12 insecticides were tested for their effects on triglycerides (TG) accumulation and interactions with palmitic acid (PA), which induces steatosis. Among tested insecticides, permethrin and ivermectin significantly altered TG accumulation and interacted with PA. Further mechanistic studies showed that permethrin promoted lipogenesis …

Consequently, the desire to find a replacement for these antibiotics has garnered interest in both farmers and consumers. The ideal substance would display antimicrobial activity without promoting bacterial resistance and would still function as a growth promoter in animals. The Food Safety Modernization Act (FSMA) has a regulation stating that there can be no detectable generic Escherichia coli in agricultural water. The objective of this study was to optimize the use of peracetic acid (PAA) as a mitigating strategy in postharvest processing. This project began by investigating the impact of storage temperatures on stability of a commercially available PAA sanitizer. Regression analysis showed a quadratic relationship in the concentration of PAA over 12 weeks when storing the product at 4, 20 and 37°C. There is lack, however, of an effective method that can monitor and characterize pesticide behaviors including penetration, persistence and translocation. Herein, we developed a novel method for real-time and in situ monitoring of pesticide behaviors in fresh produce based on surface-enhanced Raman scattering (SERS) mapping method. The food that we consume contain many dietary compounds which are biologically active. In this thesis we will discuss the biological effects of dietary compounds and the mechanisms behind their activities.

First, we studied on the anti-metastatic effects of curcumin, a dietary compound derived from turmeric, through lymphangiogenesis inhibition. Curcumin inhibited vascular endothelial growth factor-C (VEGF-C)-induced lymphangiogenesis in vivo and in vitro. Curcumin inhibited lymphangiogenesis, in part through suppression of proliferation, cell cycle progression and migration of lymphatic endothelial cells. Nobiletin (NBT) is a unique flavonoid mainly found in citrus fruits, and has been reported to inhibit colon carcinogenesis in multiple rodent models. However, the direct molecular targets of NBT are unknown. Heat shock cognate 70 protein (HSC70) contributes to cancer cell survival and resistance to chemotherapies, thereby the inhibition of HSC70 is a promising strategy in cancer chemoprevention. Using affinity chromatography, proteomics analysis and computer modeling, we demonstrated that NBT bound to HSC70 at its ATP-binding site and specifically inhibited its ATPase activity. Phospholipids can regenerate oxidized tocopherols and help delay lipid oxidation. The impact of emulsifier type, tocopherol homologue and phospholipid head group on tocopherol-phospholipid interaction was investigated in this study.

Three µmol tocopherol/kg emulsion and 15.0µmol/kg emulsion of PE or PS were dissolved in oil and emulsions were prepared. Tween 20 or bovine serum albumin(BSA) was used as emulsifier and the continuous phase contained 10mM imidazole/acetate buffer at pH 7. Microbial cross-contamination remains an on-going challenge in the food sector despite implemented sanitation programs. Antimicrobial coatings with inherent self-sanitizing properties have been explored to enhance current cleaning practice and support food safety. Prior work has demonstrated successful incorporation of dual antimicrobial characters, cationic polymers and N-halamines, into one coating system. In addition to the rechargeable nature of N-halamines, the coating was reported to exhibit biocidal effects due to the inherently antimicrobial cationic moieties and the chlorinated N-halamines.
