Wnt proteins are lipid-modified morphogens fundamental in development and disease. During Wnt biogenesis, the G-protein-coupled receptor (GPCR)-like transporter Wntless (WLS) escorts lipidated Wnts from the endoplasmic reticulum to the plasma membrane, then transfers them to extracellular carriers, forming active and soluble morphogen-carrier complexes. To dissect the mechanisms involved, we solve cryo-EM structures of Wnt-bound WLS and unliganded WLS, and perform structure-guided functional experiments. Wnts engage WLS via three conserved hairpins, which are all required for Wnt trafficking to the cell surface and carrier-mediated secretion. Background: Ophthalmic diseases are a significant public health concern globally and in Uzbekistan, where approximately 1 million individuals are affected.
Objectives: The objectives are to conduct an assortment analysis of ophthalmic medicines registered in Uzbekistan (2010–2024), to identify trends in drug registration, composition, dosage forms, and manufacturing sources.
Materials and Methods: Based on State Register data, medicines classified under Anatomical Therapeutic Chemical S01 were analyzed using marketing and qualimetric methods, focusing on the International Nonproprietary Names, dosage forms, manufacturers, and countries of origin.

The monitoring of electrical power quality requires advanced analysis techniques capable of accurately detecting and localizing non-stationary disturbances. This work proposes a structured and adaptive optimization of the Stockwell Transform (ST), referred to as Two-Power Optimization (TPO). A multiparametric Gaussian window governed by four parameters (m, p, k, r), is introduced, and a genetic algorithm is applied independently to each signal to optimize these parameters. The optimization process adapts the analysis scale to the local characteristics of the signal while preserving the physical consistency of the window function. Chromium (Cr+6)  toxicity is a significant environmental stress factor that severely hampers plant growth and productivity. This study aimed to evaluate whether the combined application of phosphorus fertilizer and melatonin could alleviate chromium induced stress in maize  (Zea mays L.).  A foliar spray of melatonin (50 µM) was applied alongside two phosphorus sources—phosphoric acid (PA) and diammonium phosphate (DAP), each at 60% of the recommended rate. Two maize cultivars (FH-1046 and YH-1898) were grown under chromium stress (0, 15, and 30 mg kg–1) to assess treatment effects. Developing resistant chickpea cultivars is essential for managing Fusarium wilt caused by Fusarium oxysporum, a major pathogen responsible for substatial yield losses. In this study, we conducted RNA-seq–based transcriptome analysis to characterize gene expression changes in resistant and susceptible genotypes of Cicer arietinum L. and C. reticulatum following inoculation with Fusarium oxysporum f. sp. ciceris race 0. Susceptible genotypes exhibited markedly higher numbers of differentially expressed genes compared with resistant genotypes. Defense-related genes were induced in all plants upon infection; however, resistant genotypes specifically upregulated genes functioning in the apoplast or extracellular region while downregulating intracellular processes—a pattern not observed …

The aim of the present research was to evaluate, for the first time, the effect of ozonated water treatments on the postharvest quality of capia pepper (Capsicum annuum L. ‘Kaptan’). The peppers were harvested at the optimum time and transported to the laboratory immediately. Uniform and quality fruit was selected and precooled. AFP, a cancer biomarker, is monitored during fetal development and in the serum of pregnant women. Sensitive and rapid monitoring of AFP is crucial for effective management. In this study, screen-printed carbon electrodes (SPCEs) modified with multi-walled carbon nanotubes (MWCNT-COOH), polyAzur A (PAA), which was prepared in a deep eutectic solution (DES), and gold nanoparticles (AuNP) were fabricated. Nonsense suppression rescues functional protein from mRNAs containing in-frame premature termination codons (PTCs). This approach employs small molecules that promote insertion of tRNAs at a PTC during translation (called “readthrough”), enabling translation to continue past the PTC to generate a full-length, functional protein. Some aminoglycosides such as G418 promote readthrough; however, their long-term use is thwarted by toxicity and modest potency. ELX-02, a novel synthetic aminoglycoside, was developed to have both enhanced readthrough activity and reduced toxicity in mammalian cells compared to traditional aminoglycosides. This thesis analyzed two media texts about former Olympic gymnast Aly Raisman and how these texts constructed her identity during and after the Larry Nassar scandal. I analyzed these texts through the lenses of whiteness, vulnerability, and carceral feminism. Looking at how the media acts as a disciplinary apparatus in this case, I argue that women athletes’ bodies are portrayed as having a vulnerability to abuse that only the "good" patriarchy can address. I seek to show that the media use depictions of women’s vulnerability and need for patriarchal help to remind women athletes that, in cases of abuse, who …

Violations of consent and sexual assault occurred frequently within the genre of romance popular across medieval England and France during the late Middle Ages. While many of the characters who experienced sexual assault in these pieces of literature were women, “Subjection to Chivalry” investigates two masculine characters whose consent is violated within their respective romances. As populations age and chronic illness increasingly characterizes the final stages of life in the United States, end-of-life doulas (EOLDs) have emerged as non-medical practitioners who support dying people and their families through education, companionship, advocacy, and ritualized care. Drawing on a thematic analysis of 18 semi-structured interviews with practicing EOLDs across the United States, this thesis examines how EOLDs conceptualize a “good death,” navigate emotionally intensive work, and sustain themselves in an unregulated and often marginal professional landscape. Erwinia amylovora is the causal agent of fire blight, which is known as one of the most destructive diseases of apple. The use of fire blight-resistant cultivars/rootstocks is one of the most effective methods available to manage this disease. We previously selected 199 local apple cultivars that showed no clearly visible symptoms of fire blight on the plants from the coastline of the eastern Black Sea Region in Türkiye. Background/aim: Polyunsaturated fatty acids (PUFAs) and the hepatic cholinergic axis are critical regulators of lipid metabolism and inflammatory signaling; however, their time-dependent interplay in hepatocytes remains incompletely understood. Clarifying how n-6 and n-3 PUFAs interact with cholinergic pathways may provide mechanistic insight into metabolic dysfunction-associated steatotic liver disease (MASLD).
