Optimized Nano-Bilosomes Loaded Ocular Insert As A Promising Approach For Boosting Voriconazole Ocular Bioavailability: In-Vitro, Microbiological, And In-Vivo Assessments, Eman Gomaa

The eye possesses numerous physiological barriers that significantly limit the absorption of administered drugs, as only < 5 % get through. These barriers restrict drug bioavailability, often necessitating multiple dosing, which can lead to poor patient compliance. The study goal is to efficiently incorporate Voriconazole (VRC), a wide-spectrum antifungal candidate with a short half-life, into stable, biocompatible, non-irritating bile salt nanoparticles (Bilosomes, BLOs) to enhance its ocular delivery. Bilosomes were prepared by the ethanol injection method. Experimental (33) Box–Behnken Design (BBD) was employed; the amount of bile salt, cholesterol, and span-60 were chosen as independent variables, while entrapment efficiency (EE%), cumulative VRC release (Q8h), particle size (PS), zeta potential (ZP), and polydispersity index (PDI) were the dependent variables. The formula of optimization showed EE% (59.04 ± 1.88 %), cumulative release after 8 h (81.35 ± 0.69 %), PS (222.53 ± 2.04 nm), ZP (−72.53 ± 1.75 mV), and PDI (0.225 ± 0.03). It was chemically characterized using DSC and FTIR. Its spherical arrangement was confirmed using TEM. For ophthalmic application, the optimized formula was included in ocular inserts to extend ocular residence time while minimizing dosing frequency. The optimal VRC-loaded bilosomal ocular insert met the acceptable physicochemical criteria, was sterilized using UV radiation for conducting microbiological assay, in-vivo study, and pharmacokinetic analysis. For safety evaluation, an eye irritation study and histopathological examination were conducted. The results confirmed its effectiveness as evidenced by a significantly larger inhibition zone and 8.39-fold enhancement in VRC bioavailability compared to its suspension. These findings support the potential of bilosomes-loaded ocular insert as an effective ocular delivery of VRC.

This monograph explores three main strategies of vocal exotification in Anglophone film: representations of accented speech, inclusion of constructed languages, and non-subtitling of snippets of other languages. Using Stuart Hall’s encoding/decoding framework, I approach these practices as rhetorical devices that both construct and are constructed by the complexity of our social landscapes. Portrayals of accented speech, constructed languages, and subtitling practices form a gradient of abstraction, where the denotative meaning of words gradually loses importance to the sonic characteristics of the voice. Connectivity has become integral to various application domains, with the automotive sector as a prime example. Advances in electronics, computing, and telecommunications have driven the evolution of the connected car ecosystem, transforming it into a data-rich environment that enhances road safety, efficiency, and overall mobility. However, the success of this ecosystem depends on seamless, reliable, and resilient communications. We identify key challenges that may affect communications in the connected car environment and discuss solutions that enhance resiliency and robustness. Finally, we propose a multi-layered network architecture that will enhance communication resilience in the connected car ecosystem.

This thesis examines the crisis that developed after US president John Quincy Adams signed, then disavowed (allegedly over moral concerns), the fraudulent 1825 Treaty of Indian Springs with the Creek Indian Nation. The thesis probes Adams' motives during the Treaty of Indian Springs crisis, and seeks to connect the crisis to the evolution of US federal Indian policy in the 1820s, prior to the mass Indian removal of the 1830s.

The following review examines how youth exposure to sexual images online is conceptualized and measured as abusive within epidemiological research on online child sexual abuse (OCSA). As digital environments reshape sexual socialization, scholars debate whether and when exposure to sexual content constitutes abuse. To address this issue, this study conducts a qualitative content analysis of 27 survey items drawn from 20 empirical studies of youth online experiences. Survey questions about sexual image exposure were coded across four dimensions: volition, personalization, image or text, and explicitness. Findings reveal significant definitional inconsistency. Balance deficits following repeated mild traumatic brain injuries are well documented, yet research examining the delayed outcomes specifically in college-aged women remains limited. The present study investigated whether women with a history of two or more mTBIs exhibit lasting balance impairments compared to women with zero or one prior mTBI, tested at least six months after their most recent injury. The capital city of Concord, New Hampshire, has been served by Concord Area Transit (CAT) since 1989. Despite continued population growth and economic development within the city, CAT’s fixed route bus system has remained unaltered since 2011. The purpose of this project is to rework the CAT system to provide service that better aligns with Concord resident’s current needs. A public survey was conducted to gather data regarding Concord resident’s needs, habits, and perception of the existing system, which was used in the development of the revised system. The Iberian ribbed newt Pleurodeles waltl can regenerate the lens by transdifferentiating iris pigmented epithelial (IPE) cells into lens epithelial cells (LECs), but the mechanism by which this occurs remains largely uncharacterized. We aim to mechanistically show an early and critical role for FGF signaling by using small molecules to inhibit FGF receptors and observe the histological and intracellular effects. Two inhibitors were found to completely stall lens regeneration. Cell cycle analysis revealed decreased DNA synthesis and uniformly low but unchanged entry to mitosis under signaling inhibition by staining for EdU incorporation and phospho-histone H3 (pH3), respectively. Substance use disorder (SUD) is a major public health concern characterized by compulsive substance use despite adverse consequences. While contemporary models emphasize biological, psychological, and environmental contributors, stress- and reward-related systems are often examined in isolation. This review integrates these systems within an evolutionary mismatch framework to examine how chronic stress exposure and repeated substance use increase vulnerability to addiction. Stress and reward systems are evolutionarily conserved but may become chronically dysregulated under modern conditions characterized by persistent psychosocial stressors and highly potent rewarding stimuli.
