This open-label, randomized, phase 3 trial (NCT02577406) compared enasidenib, an oral IDH2 (isocitrate dehydrogenase 2) inhibitor, with conventional care regimens (CCRs) in patients aged ≥60 years with late-stage, mutant-IDH2 acute myeloid leukemia (AML) relapsed/refractory (R/R) to 2 or 3 prior AML-directed therapies. Patients were first preselected to a CCR (azacitidine, intermediate-dose cytarabine, low-dose cytarabine, or supportive care) and then randomized (1:1) to enasidenib 100 mg per day or CCR. The primary endpoint was overall survival (OS). Tlr5 Agonists Enhance Anti-Tumor Immunity And Overcome Resistance To Immune Checkpoint Therapy, Caleb Gonzalez, Sarah Williamson, Seth T Gammon, Sarah Glazer, Joon Haeng Rhee, David Piwnica-Worms

Primary and adaptive resistance to immune checkpoint therapies (ICT) represent a considerable obstacle to achieving enhanced overall survival. Innate immune activators have been actively pursued for their antitumor potential. Herein we report that a syngeneic 4T1 mammary carcinoma murine model for established highly-refractory triple negative breast cancer showed enhanced survival when treated intra-tumorally with either the TLR5 agonist flagellin or CBLB502, a flagellin derivative, in combination with antibodies targeting CTLA-4 and PD-1. Long-term survivor mice showed immunologic memory upon tumor re-challenge and a distinctive immune activating cytokine profile that engaged both innate and adaptive immunity. Antivirulence agents targeting endospore-transmitted Clostridioides difficile infections are urgently needed. C. difficile-specific DNA adenine methyltransferase (CamA) is required for efficient sporulation and affects persistence in the colon. The active site of CamA is conserved and closely resembles those of hundreds of related S-adenosyl-l-methionine (SAM)-dependent methyltransferases, which makes the design of selective inhibitors more challenging. We explored the solvent-exposed edge of the SAM adenosine moiety and systematically designed 42 analogs of adenosine carrying substituents at the C6-amino group (N6) of adenosine. The safety and efficacy of various pharmacotherapeutic regimens on refractory meningiomas have been the focus of investigations. We present a comprehensive review of the previous efforts and the current state of ongoing clinical trials. A PRISMA-compliant review of the MEDLINE and ClinicalTrial.gov databases of the National Library of Medicine were performed. The primary outcomes of interest for included articles were radiographic response, overall survival, progression-free survival, six-month progression-free survival, and adverse events. Overall, 34 completed trials and 27 ongoing clinical trials were eligible. Six-month progression-free survival was reported in 6-100% of patients in the completed studies. Patients with primary mitochondrial oxidative phosphorylation (OxPhos) defects present with fatigue and multi-system disorders, are often lean, and die prematurely, but the mechanistic basis for this clinical picture remains unclear. By integrating data from 17 cohorts of patients with mitochondrial diseases (n = 690) we find evidence that these disorders increase resting energy expenditure, a state termed hypermetabolism. We examine this phenomenon longitudinally in patient-derived fibroblasts from multiple donors. Genetically or pharmacologically disrupting OxPhos approximately doubles cellular energy expenditure. This cell-autonomous state of hypermetabolism occurs despite near-normal OxPhos coupling efficiency, excluding uncoupling as a general mechanism. Instead, hypermetabolism is associated …

The impact of zero-tolerance, exclusionary discipline, and the school-to-prison pipeline has become increasingly present in research in recent years. Studies centered around zero-tolerance, exclusion, and the school-to-prison pipeline have made the focus on such studies boys of color, specifically Black boys. These studies have contributed to the amount of awareness that schools, parents, and the public have about the effects of unfair punishment practices. However, Black girls have been invisible, criminalized in educational settings, and are at a great risk of encountering the criminal justice system. This study examines the history of environmental racism and unfair housing practices, and their connection to health and socioeconomic impacts. Detroit’s unfair housing practices date back to earlier racial covenants and redlining during the New Deal’s federal housing program. This study utilizes Geographic Information Systems (GIS) to investigate the relationship between toxin-producing sites and formerly redlined neighborhoods in Detroit, Michigan. Datasets from 2019 American Community Survey: 5-Year Data (2014-2019) and the Environmental Protection Agency 2017 Toxic Release Inventory (TRI) are interpreted using spatial analysis to identify the ratio of facilities between minority and non-minority populations. Previous research addressing phobia toward K-12 students in the LGBTQ+ community considers mental, emotional, physical, and spiritual effects. Students in the community have experienced progress in the past 20 years regarding equality, support, and empowerment from institutions, interpersonal relationships, and insecurities within themselves. In this study, I review several academic articles and textbooks which highlight the progress students have experienced regarding prejudice and backlash in K-12 schools in the United States. “Big events”, such as wars, economic crises, pandemics, or natural disasters, affect the risk environment in which people use drugs. While the impact of big events on injection risk behaviors and access to drug-treatment services is well documented, less is known about the effects of big events on drug markets. The study examined the impact of the audit expectation gap and auditors' statutory duty on financial reporting quality in Nigeria. Data for the study were obtained primarily through questionnaires administered to randomly selected 400 respondents comprising auditors, accountants, stockbrokers, and investors in various sectors of the Nigerian economy. Of the distributed questionnaires, 252 were received, representing a 63 per cent response rate. As a doctoral student, I was tasked to write a literature review for my dissertation, which focused on the experiences of Mexican/Mexican American/Xicanx undergraduate students at a predominantly white university in the Northeast. Honey bee colonies have perished at an alarming rate for over 15 years. Among the multitude of threats to bee health, virus infections and agrochemical exposure appear to be major drivers of colony loss. As virus infection is among the most damaging influences on bee health, we first investigated a putative target for antiviral immunity, termed KATP channels. We previously demonstrated that pharmacologically activating KATP channels induces the production of reactive oxygen species and increases survival during infection with Israeli Acute Paralysis Virus.
