Mechanosensitive Ion Channels As Therapeutics Targets For Osteoarthritis, Alireza Savadipour
Mechanosensitive Ion Channels As Therapeutics Targets For Osteoarthritis, Alireza Savadipour
McKelvey School of Engineering Graduate Student Theses & Dissertations
Osteoarthritis (OA) is a common degenerative joint disease characterized by the degenerative changes in joint cartilage and underlying bone. It commonly occurs in weight-bearing joints like the knees, hips, and spine but can affect any joint in the body, resulting in symptoms such as pain, stiffness, and reduced range of motion. Post-traumatic osteoarthritis (PTOA) is a subtype of OA that occurs after a joint injury, such as a fracture, dislocation, or ligament tear, causing damage to the cartilage and other joint tissues leading to OA over time. PTOA accounts for a significant proportion of all OA cases. OA is a …
Why Teachers Stay: An Exploration Of Why Mid-Career Teachers Remain In Teaching, Jennifer K. Mccorvey
Why Teachers Stay: An Exploration Of Why Mid-Career Teachers Remain In Teaching, Jennifer K. Mccorvey
USF Tampa Graduate Theses and Dissertations
Much of the current literature on teacher retention centers around early-career teachers, with far less addressing mid-career teacher attrition. This research study explores the positive and negative experiences of five mid-career teachers, Toni, Sean, Susan, Debbie, and Jacqueline, explicitly focusing on the experiences that influenced their decisions to remain teaching. The participants were Teacher Leadership Academy alumni, a professional learning community in two separate Professional Development School programs. I used a qualitative case-study approach to examine the teachers’ experiences and Identity Theory as a lens through which to view the findings. Semi-structured interviews and participant-constructed timelines were the primary sources …

Trna Methyltransferases Mediate Acinetobacter Baumannii Pathobiology, Jenna Mcguffey
Arts & Sciences Graduate Student Theses and Dissertations
Bacterial pathogens commonly encounter hostile conditions, including disinfectants, antimicrobials, and the host immune system. To be successful, they must adapt through dynamic and complex regulatory mechanisms, such as post-transcriptional modifications on a variety of molecules, including tRNA. In recent years, this tRNA “epitranscriptome” has been increasingly appreciated as an under-researched mechanism for bacterial stress responses. These modifications can impact the translation efficiency of a cohort of stress response factors whose transcripts are enriched for the corresponding codons. However, the variety and roles of tRNA modification enzymes in human pathogens is still largely unknown. The Gram-negative pathogen Acinetobacter baumannii is currently …
T-Bet And Eomes Are Required For Mature Nk Cell Homeostasis And Function, Pamela Wong
T-Bet And Eomes Are Required For Mature Nk Cell Homeostasis And Function, Pamela Wong
Arts & Sciences Graduate Student Theses and Dissertations
Natural Killer (NK) cells are innate lymphocytes known for their role in recognizing and killing virus-infected or tumor cells without prior sensitization. NK cells express a variety of inhibitory and activating receptors that control their effector functions. Once activated by target cells, cytokines, or activating receptor ligation, NK cells are capable of potent IFN-γ secretion in addition to direct cytotoxicity mediated by perforin and granzymes. Because of their anti-tumor function, NK cells are currently utilized in promising immunotherapy approaches. NK cells develop in the bone marrow and arise from the common lymphoid progenitor, which also gives rise to T, B, …
Environmental Sustainability Of Small Wastewater Treatment Systems And Pathways For Improvement, Matthew James Thompson
Environmental Sustainability Of Small Wastewater Treatment Systems And Pathways For Improvement, Matthew James Thompson
Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–
Wastewater treatment protects society and the environment by reducing pollutants generated from communities prior to discharge into local water bodies. Small water resource recovery facilities (WRRFs) represent the majority of centralized wastewater treatment systems, are resource intensive, and often lack resources needed to evaluate sustainability improvements. The goal of this dissertation was to identify primary sources and drivers of environmental impacts in small WRRFs and exploration of pathways to improve their environmental sustainability.
Case study data collected from small WRRFs in Nebraska was analyzed using environmental life cycle assessment (LCA) to quantify the environmental impact of facilities and existing opportunities. …

Purdue University Libraries Open Access Publishing Fund
Herpesviruses are large DNA viruses that have long been used as powerful gene therapy tools. In recent years, the ability of herpesviruses to stimulate both innate and adaptive immune responses has led to their transition to various applications as vaccine vectors. This vaccinology branch is growing at an unprecedented and accelerated rate. To date, human herpesvirus-based vectors have been used in vaccines to combat a variety of infectious agents, including the Ebola virus, foot and mouth disease virus, and human immunodeficiency viruses. Additionally, these vectors are being tested as potential vaccines for cancer-associated antigens. Thanks to advances in recombinant DNA …

AlxGa1-xN/GaN high-electron-mobility transistor (HEMT) structures are key components in electronic devices operating at gigahertz or higher frequencies. In order to optimize such HEMT structures, understanding their electronic response at high frequencies and room temperature is required. Here, we present a study of the room temperature free charge carrier properties of the two-dimensional electron gas (2DEG) in HEMT structures with varying Al content in the AlxGa1-xN barrier layers between x = 0:07 and x = 0:42. We discuss and compare 2DEG sheet density, mobility, effective mass, sheet resistance, and scattering times, …

The movement ecology of raccoons varies widely across habitats with important implications for the management of zoonotic diseases such as rabies. However, the spatial ecology of raccoons remains poorly understood in many regions of the United States, particularly in the southeast. To better understand the spatial ecology of raccoons in the southeastern US, we investigated the role of sex, season, and habitat on monthly raccoon home range and core area sizes in three common rural habitats (bottomland hardwood, upland pine, and riparian forest) in South Carolina, USA. From 2018–2022, we obtained 264 monthly home ranges from 46 raccoons. Mean monthly …

Obsessive-compulsive disorder (OCD) is a debilitating psychiatric disorder. Worldwide, its prevalence is ~2% and its etiology is mostly unknown. Identifying biological factors contributing to OCD will elucidate underlying mechanisms and might contribute to improved treatment outcomes. Genomic studies of OCD are beginning to reveal long-sought risk loci, but >95% of the cases currently in analysis are of homogenous European ancestry. If not addressed, this Eurocentric bias will result in OCD genomic findings being more accurate for individuals of European ancestry than other ancestries, thereby contributing to health disparities in potential future applications of genomics. In this study protocol paper, we …

Reynolds-Averaged Navier-Stokes Simulations Of Nearshore Langmuir Circulation And The Formation Of Oil-Particle Aggregates, Juan J. Penaloza Gutierrez
USF Tampa Graduate Theses and Dissertations
Langmuir turbulence in the inner shelf coastal ocean is characterized by Langmuir circulation (LC) or cells that can span the full depth of unstratified water columns. A Reynolds-averaged Navier-Stokes (RANS) simulation strategy resolving full-depth LC coupled with an oil-particle aggregate (OPA) formation model is introduced. The RANS methodology is employed in two different cases. The first case corresponds to a shallow shelf zone of uniform depth, focusing on vertical mixing and transport of OPAs. The second case pertains to a surf-shelf transition zone with variable depth, comprising vertical and horizontal mixing of OPAs. In the latter case, a wave model …

Mycobacterium tuberculosis (Mtb) cultured axenically without detergent forms biofilm-like cords, a clinical identifier of virulence. In lung-on-chip (LoC) and mouse models, cords in alveolar cells contribute to suppression of innate immune signaling via nuclear compression. Thereafter, extracellular cords cause contact-dependent phagocyte death but grow intercellularly between epithelial cells. The absence of these mechanopathological mechanisms explains the greater proportion of alveolar lesions with increased immune infiltration and dissemination defects in cording-deficient Mtb infections. Compression of Mtb lipid monolayers induces a phase transition that enables mechanical energy storage. Agent-based simulations demonstrate that the increased energy storage capacity is sufficient for the formation …

Uncovering The Genetic Basis Underlying The Pseudomonas Syringae Pv. Tomato Strain Dc3000’S Response To Iaa And Its Role In Regulating Ptodc3000 Biology, Joshua Michael Brian Johnson
Arts & Sciences Graduate Student Theses and Dissertations
Indole-3-acetic acid (IAA) is a molecule ubiquitous to the environment and is known to be produced across all three domains of life. It is most well-known for its role as an important plant hormone (an auxin) and until recently, its roles in other organisms have not been explored. It is well established that bacteria, and pathogens in particular, need to sense and respond to environmental signals. One model organism for studying plant pathogen interactions that is known to both produce and respond to IAA is Pseudomonas syringae pv. tomato strain DC3000 (PtoDC3000). Many plant metabolites have been explored as potential …

The performance of the mid-wave infrared InGaAs/InAsSb nBn photodetector is investigated and its viability for space applications is assessed. Three structures are grown with unique absorber layer doping profiles via molecular beam epitaxy. Material and device characterizations are performed and analyzed to determine the effects of doping on fundamental material parameters and detector performance. Noise-equivalent irradiance is calculated to be a factor of 4x that of an ideal detector exhibiting Rule 07 dark current and 100% quantum efficiency, demonstrating high sensitivity. The structures are then irradiated with 63 MeV protons to evaluate the extent of performance degradation over the course …

Biodiversity—The Birds And The Bees And Healthy Grasslands, D. Borrenpohl, Pat D. Keyser
IGC Proceedings (1977-2023)
Losses in biodiversity abound in modern agroecosystems, and biodiversity loss hampers ecosystem function and primary productivity comparable to abiotic stress. One of the most endangered ecosystems in the world is North American tallgrass prairie, and native birds and pollinators that historically depended on tallgrass prairie are in precipitous decline. Fortunately, native warm-season grasses that dominated tallgrass prairie present a valuable opportunity for summer forage to beef producers in the Eastern United States—a time when coolseason grasses endure a lack in productivity called the “summer slump.” Tallgrass prairie was sustained by periodic disturbance from grazing and fire, and combining fire and …

The Soil Health Properties Of Native Grasslands, C. Bitler, Pat Keyser
IGC Proceedings (1977-2023)
Due to an increased interest in soil health and the role soils can have in carbon sequestration, native grasslands are getting heightened attention. Native grasslands are known for their deep top-soils that develop over time through the accumulation of soil organic matter. The deep and fibrous root systems that make up two-thirds of the biomass of native grasslands are the primary contributors to the soil organic carbon (SOC) content of grassland soils. Increased SOC content has a cascading effect on soil health metrics through increasing water infiltration and water holding capacity, supporting diverse and abundant soil microbial life, and improving …
Using Functional Ecology To Predict Restoration Success In Managed Grasslands, J. Bergman
Using Functional Ecology To Predict Restoration Success In Managed Grasslands, J. Bergman
IGC Proceedings (1977-2023)
Plants have developed remarkable functional variation over evolutionary time to cope with different environmental conditions. As a result, functional variation – measurable as functional traits above and below ground – can be found within but also among grassland communities today. The functional space covered by a community is thereby restricted by environmental factors while each single species within the community inhabits a specific niche with a certain breadth along different environmental gradients. Data from field sampling in European grasslands have shown, that species intraspecific functional variation can cover considerable amounts of possible functional spaces, hence holding the potential to establish …
Toxic Plants In Grasslands And Their Impact On Livestock, A. C. Barr
Toxic Plants In Grasslands And Their Impact On Livestock, A. C. Barr
IGC Proceedings (1977-2023)
Grasslands are critical infrastructure for most cow/calf operations. Quality and abundance of pasture grasses directly influence livestock rate of gain, thereby affecting livestock margins and production costs, which determine the profitability of stocker operations. Successful cattlemen control weeds to preserve healthy pasture but also to prevent plant poisonings that may reduce individual animal performance or cause fatalities. Integrated Toxic Plant Management (ITPM) programs teach ranchers to identify the potentially toxic plants, recognize symptoms of plant poisonings, and use appropriate strategies to control those plants. Toxic plants can produce a range of effects in cattle and other grazing species. Nightshades ( …

Response Of Reproductive Phenology To Soil Water Content Of Stipa Breviflora Under Warming And Nitrogen Application, L. Bai, H. Y. Ren, G. D. Han
IGC Proceedings (1977-2023)
Plant phenology is considered as the most direct and sensitive characteristic indicators of plant response to regional climate change. Based on the observation data of phenology and soil water content of Stipa breviflora in an experimental platform of warming and nitrogen application in Inner Mongolia desert steppe from 2009 to 2019. The results showed that warming and nitrogen application significantly advanced the reproductive phenological period (P < 0.001), and the interaction of warming and nitrogen application promoted the advance of reproductive phenological period (P < 0.05).

Evaluation Of Serum Melatonin Levels In Type 2 Diabetic Patients In Relation To Sleep Quality, Haidar K. Ibrahim, Zainab Hasan Hashim, Riyam Bassil Ali
Maaen Journal for Medical Sciences
Overview: The negative impact of diabetes mellitus on the quality of life is not limited to the clinical aspects, it also extends to the psychological and economic sides of their lives. Deteriorated sleep quality is one of the major factors that affect diabetics' health-related quality of life. This research is an attempt to track the pathophysiology of sleep disturbance in type 2 diabetic patients by evaluating serum melatonin levels as a key hormone that induces sleep. Methods: Eighty-eight relatively glycemic-controlled type 2 diabetic patients were recruited in a cross-sectional study to evaluate their quality of sleep. The study then tends …
