Proteomics And Machine Learning For Pulmonary Embolism Risk With Protein Markers, Yaa Amankwah Awuah

This thesis investigates protein markers linked to pulmonary embolism risk using proteomics and statistical methods, employing unsupervised and supervised machine learning techniques. The research analyzes existing datasets, identifies significant features, and observes gender differences through MANOVA. Principal Component Analysis reduces variables from 378 to 59, and Random Forest achieves 70% accuracy. These findings contribute to our understanding of pulmonary embolism and may lead to diagnostic biomarkers. MANOVA reveals significant gender differences, and applying proteomics holds promise for clinical practice and research.

Preterm birth is the number one cause of death in neonates, accounting for 35% of neonatal mortality. The preterm birth rate in the U.S. in 2021 was 10.5%, disproportionally affected by race and ethnicity. Obese women have an increased risk of preterm pregnancy, and if delivered before 37 weeks of gestation, the offspring have higher rates of complications that extend from the neonatal period into life-long metabolic and immune adverse outcomes. In the early months after delivery, preterm infants have higher rates of adverse health outcomes than term infants, including infections, extrauterine growth restrictions, respiratory, metabolic, and neurological complications, necrotizing …

Spider silk and hagfish intermediate filaments show the potential to be advantageously utilized in textile and biomedical industries. Bee silk is a sparsely studied biopolymer that has shown potential to be added to these other impressive biopolymers. Osmia lignaria, is a solitary bee native to the western United States. Opioid use disorder (OUD) affects over two million Americans, and over 42,000 Americans perished of opioid overdoses in 2016 (Volkow et al., 2019). During the initial years of the opioid crisis, overdose deaths were primarily attributed to prescription analgesics, heroin, and synthetic opioids. However, it was illicit prescription opioid painkiller use that ranked highest in terms of fatalities (Volkow et al., 2019). The capacity of SARS-CoV-2 to evolve poses challenges to conventional prevention and treatment options such as vaccination and monoclonal antibodies, as they rely on viral receptor binding domain (RBD) sequences from previous strains. Additionally, animal CoVs, especially those of the SARS family, are now appreciated as a constant pandemic threat. We present here a new antiviral approach featuring inhalation delivery of a recombinant viral trap composed of ten copies of angiotensin-converting enzyme 2 (ACE2) fused to the IgM Fc. The increasing complexity of artificial intelligence models has given rise to extensive work toward understanding the inner workings of neural networks. Much of that work, however, has focused on manipulating input data feeding the network to assess their affects on network output or pruning model components after the often-extensive time-consuming training. It is postulated in this study that understanding of neural network can benefit from model structure simplification. This dissertation explored the effect of Gagne's nine events of instruction on student academic achievement and satisfaction during an online English class for French speakers. A group of 30 Facebook users from French-speaking countries in North and Sub-Saharan Africa participated in the research. They were divided randomly into two groups: one receiving traditional instruction through an asynchronous online lecture with PowerPoint presentation, and the other using instruction utilizing Gagne's nine events of instruction integrated with an LMS designed for this purpose. The regulation of reproductive physiology in animals is mediated by the Hypothalamus-Pituitary-Gonad (HPG) axis. In seasonal breeders, a conserved reciprocal switching of deiodinase enzymes alters thyroid hormone signaling, which controls HPG activity. This study tested the hypothesis that neural regulation of the HPG axis as described in seasonal systems is conserved and repurposed to respond to other cues in aseasonal species. Using water deprivation and nest restriction in the opportunistic Zebra Finch, we were able to create a breeding group and non-breeding group, confirmed by egg laying and changes in ovarian anatomy. Shallow, river-influenced coastal sediments are important for global carbon storage and nutrient cycling and provide a habitat for diverse communities of invertebrates (infauna). Elevated bed shear stress from extreme storms can resuspend, transport, and deposit sediments, disrupting the cohesive structure of muds, and sorting and depositing sand eroded from beaches. These physical disruptions can also resuspend or smother infauna, decreasing abundances and changing community structure. Infaunal activities such as burrowing, tube construction, and feeding can impact sediment structure and stability. In many rural, low-income communities such as the Black Belt of Alabama, there is a lack of wastewater infrastructure and solutions leaving homeowners non-compliant to state and federal law and with potential health risks on their property. New regulations have allowed for onsite, surface discharge for single family homes that are not connected to a centralized sewer or large flow development. These new regulations also contain strict disinfection criteria for the treated wastewater that will be discharged. Alexandra Vozick Hans was born to Canadian parents, who, upon moving to New York, lived in the Amalgamated Housing Cooperative, where she grew up. She remained in the Bronx until she graduated college in 1966. After college, she moved to Boston and became a social worker.

Hans and her family were socialists, and Hans recalls tensions between the socialists and the communists in the Amalgamated. She also recalls tensions over the issue of integration in the building: the Amalgamated was not integrated, and residents complained about the lack of integration. Protein-targeted degradation is an emerging and promising therapeutic approach. The specificity of degradation and the maintenance of cellular homeostasis are determined by the interactions between E3 ubiquitin ligase and degradation signals, known as degrons. The human genome encodes over 600 E3 ligases; however, only a small number of targeted degron instances have been identified so far. In this study, we introduced DegronMD, an open knowledgebase designed for the investigation of degrons, their associated dysfunctional events, and drug responses. Cervical degenerative disc diseases such as myelopathy and radiculopathy often require conventional treatments like artificial cervical disc replacement or anterior cervical discectomy and fusion (ACDF). When designing a medical device, like the stand-alone cage, there are many design inputs to consider. Background: Opioid prescribing in the United States is decreasing, however, the opioid epidemic is continuing at an uncontrollable rate. Available data show a significant number of opioid deaths, primarily associated with illicit fentanyl use. It is interesting to also note that the data show no clear correlation between opioid prescribing (either number of prescriptions or morphine milligram equivalent [MME] per capita), opioid hospitalizations, and deaths. In the last few years, single cell and single nucleus RNA-sequencing technologies have been leveraged in exercised adult skeletal muscle to reveal complex cell type-specific regulation of muscle plasticity. Expanding -omics technology at single cell resolution, complimented by advanced computational approaches, promise to rapidly accelerate our understanding of how exercise confers its numerous beneficial effects, paving the way for cell type-specific and targeted therapeutics to increase exercise responses during aging and in the face of chronic disease.

OBJECTIVE: We assessed whether biological age, measured by the epigenetic clock GrimAge, mediates the association of objective and subjective neighborhood disadvantage with incident HF among Black persons.

METHODS: Participants were 1448 self-reported Black adults (mean age (standard deviation, SD) = 64.3 (5.5)) dually enrolled in two community-based cohorts in Jackson, Mississippi, the ARIC and JHS cohorts, who were free of HF as of January 1, 2000. Incident HF events leading to hospitalization through December 31, 2017, were classified using ICD-9 discharge codes of HF. OBJECTIVE: The alignment between environmental stimuli (e.g., dark, light) and behavior cycles (e.g., rest, activity) is an essential feature of the circadian timing system, a key contributor to metabolic health. However, no previous studies have investigated light-activity alignment in relation to glycemic control in human populations.

RESEARCH DESIGN AND METHODS: The analysis included ∼7,000 adults (aged 20-80 years) from the National Health and Nutrition Examination Survey (NHANES) (2011-2014) with actigraphy-measured, multiday, 24-h activity and light data. The microbiome's role in shaping host phenotypes is a critical area of investigation, with implications for ecology, evolution, and host health. Dynamic plant-microbe interactions are influenced by factors like soil type, environment, and genotype. Understanding their impact on microbial communities is key for tailored plant benefits. An artificial ecosystem selection experiment was done for eight generations with Arabidopsis thaliana Ler and Cvi. This revealed distinct microbial communities shaped by genotypes and biomass treatments. Initially, environment dominated, but over time, genotype and biomass gained influence, explaining ~40% of the variation. This process includes the combination of 2-photon polymerization (2PP) and masked-stereolithography (MSLA). Education plays an important role in preparing current and future generations for the challenges ahead caused by climate change. The purpose of this dissertation was to examine the educational influence textbooks and undergraduate instructors have on their students. Three studies were carried out to explore these influences on climate change instruction. The first study looked at the extent, type, and location of climate change topics across 24 undergraduate general chemistry textbooks. Soybean seeding rate (SR) is selected according to planting date, maturity group, soil properties, and yield goal. Even though one SR is applied in most fields, site-specific adjustments with variable-rate seeding (VRS) technology can help optimize production. The project goal was to determine if VRS could be beneficial to soybean producers in Arkansas. The project objectives were to i) assess planter performance and soybean yield response to five seeding rate treatments, ii) characterize the drivers of yield variability, and iii) create a-posteriori prescription maps for VRS.
