BACKGROUND: Transgenic (Tg) mice are widely used in biomedical research, and they are typically generated by injecting transgenic DNA cassettes into pronuclei of one-cell stage zygotes. Such animals often show unreliable expression of the transgenic DNA, one of the major reasons for which is random insertion of the transgenes. We previously developed a method called "pronuclear injection-based targeted transgenesis" (PITT), in which DNA constructs are directed to insert at pre-designated genomic loci. PITT was achieved by pre-installing so called landing pad sequences (such as heterotypic LoxP sites or attP sites) to create seed mice and then injecting Cre recombinase or …

Expanding Understandings Of Race In Postsecondary Language Classrooms: A Call For Multiraciality In Teacher Identity Research, Marcela Hebbard Jan 2024

Expanding Understandings Of Race In Postsecondary Language Classrooms: A Call For Multiraciality In Teacher Identity Research, Marcela Hebbard

Writing and Language Studies Faculty Publications

While issues of race in relation to teacher identity have been addressed in language education research, they have often been confined to special issues. Factors contributing to the “absent-present” nature of race include an imbalanced focus on intersectionality which tends to prioritize the teacher's linguistic identity over other social categories, such as race and the persistent dichotomy between the idealized native speaker and non-native speaker. To broaden the understandings of race in teacher identity research within postsecondary language classrooms, this chapter advocates for considering the notion of multiraciality. To support these arguments, results from a critical discourse analysis (CDA) of …

A Scoping Review Of Population Diversity In The Common Genomic Aberrations Of Clear Cell Renal Cell Carcinoma, Sean S. Kumar, Ninad Khandekar, Komal Dani, Saina R. Bhatt, Vinay Duddalwar, Anishka D'Souza

Department of Medicine Faculty Publications

Introduction: Previous literature has shown that clear cell renal cell carcinoma (ccRCC) is becoming a more prevalent diagnosis and that the incidence and mortality differ both regionally and racially. While the molecular profiles for ccRCC are studied regionally through biopsy and sequencing techniques, the genomic landscape and ccRCC diversity data are not well-studied. We conducted a review of the known genomic data on 6 of the most clinically relevant DNA biomarkers in ccRCC: Von Hippel-Landau (vHL), Polybromo-1 (PBRM1), Breast Cancer Gene 1-Associated Protein 1 (BAP1), Histone-Lysine N-Methyltransferase Domain-Containing 2 (SETD2), Mammalian Target of Rapamycin (mTOR), and Lysine-Specific Demethylase 5C (KDM5C). …

Minimally Invasive Therapies In The Treatment Of Arthropathy, Steven Bateman Jan 2024

Minimally Invasive Therapies In The Treatment Of Arthropathy, Steven Bateman

Physician Assistant Scholarly Project Papers

It is estimated that 27% of adults over 45 years of age suffer from osteoarthritis of the hip secondary to articular cartilage damage over the lifespan. Joint pain diminishes life quality, limits range of motion and overall activity, and frequently impairs basic ambulation. These functional deficits worsen the progression of comorbidities that are often referred to as lifestylerelated chronic disease as physical activity subsequently declines. This literature review explores the efficacy of minimally invasive therapies that precede and delay surgical intervention in adult patients experiencing functional limitations and pain in major joints. The study examines the effectiveness of various interventions, …

Probiotic Therapy For The Treatment And Prevention Of Bacterial Vaginosis, Chelsea Scheil Jan 2024

Probiotic Therapy For The Treatment And Prevention Of Bacterial Vaginosis, Chelsea Scheil

Physician Assistant Scholarly Project Papers

Bacterial vaginosis (BV) is a common presenting complaint in healthcare, and patients often experience recurrences at a frustrating rate. The mainstay of current treatment is antibiotic therapy, either via oral or vaginal route. When recurrences occur, stronger antibiotics are often employed. This current method of treatment does not address an underlying component that impacts infection recurrence and rate of recurrence- the patient’s baseline vaginal microbiome and the healthy bacteria that support it. To determine the efficacy and safety of probiotic therapy for the treatment of bacterial vaginosis, a literature review was completed using databases PubMed and Embase. The results of …

In this case study, protected and conserved areas practitioners working in the Chignecto Isthmus region of New Brunswick and Nova Scotia, Canada were interviewed assist in better understanding the …

Care To Criminalization: How Adults With Lived Crossover Kid Experience Percieve Their Experiences In The Child Protection System And The Criminal Justice System, Clarissa Kurzawski Jan 2024

Care To Criminalization: How Adults With Lived Crossover Kid Experience Percieve Their Experiences In The Child Protection System And The Criminal Justice System, Clarissa Kurzawski

Theses and Dissertations (Comprehensive)

Crossover kids are youth who were/are in the child protection system and cross over into the juvenile/criminal justice system. Children who have been in the child protection system are 52% more likely to end up in the criminal justice system (Baidawi, 2020; Bateman, 2021; Turpel-Lafond, 2009). This creates a concerning trajectory for youth who are in the child protection system, through no fault of their own, because criminal justice involvement can follow them into adulthood. Children are entering a system of care that is criminalizing them and moving them into a system of custody. There has been scholarship using extant …

However, vouchered specimens could be at risk if staining techniques are destructive. For instance, the pH of unbuffered Lugol's iodine (I

Examining How Non-Antifungal Medications Impact Host-Fungal Interactions, Parker Ryan Reitler Jan 2024

Examining How Non-Antifungal Medications Impact Host-Fungal Interactions, Parker Ryan Reitler

Theses and Dissertations (ETD)

Invasive fungal infections (IFIs) are estimated to kill 1.5 million people each year with mortality rates remaining high (around 30-70%), despite the availability of three major classes of antifungal drugs. Furthermore, millions more suffer debilitating infections of the mucosal membranes or destructive subcutaneous mycoses. One of the most common IFIs is invasive candidiasis, mostly commonly caused by Candida albicans infections, with mortality rates of approximately 30%. In addition, the population of susceptible individuals and the incidences of both mucosal and systemic mycoses continue to rise due to the widespread use of broad-spectrum antibacterial agents, cancer chemotherapies, and the immune suppressive …

Molecular Determinants Of Trp Channel Gating, Briar J. Bell Jan 2024

Molecular Determinants Of Trp Channel Gating, Briar J. Bell

Theses and Dissertations (ETD)

Transient Receptor Potential (TRP) ion channels are a family of proteins that contribute to a myriad of physiological functions. They have been extensively studied since their discovery 35 years ago; however, there are still many questions regarding the mechanisms by which they function. This family of proteins is characterized by a generally shared structure: an intracellular N terminus featuring ankyrin repeats, six transmembrane helices including the pore domain, a re-entrant loop and TRP helix, and an intracellular C terminus with varying structure. Each of these regions has been implicated in the regular function of TRPs, while mutations that alter trafficking …

Characterizing The Cellular Response To Astrovirus Infection In The Central Nervous System, Amy E. Davis

Theses and Dissertations (ETD)

Central nervous system (CNS) diseases caused by infectious agents pose significant health challenges, often leading to severe neurological dysfunction with high morbidity and mortality rates. Astroviruses (AstVs), traditionally known for causing gastroenteritis, have recently been identified as neuroinvasive pathogens capable of inducing neurological diseases. This study explores the cellular and molecular mechanisms by which AstV, specifically non-classical VA1 and classical HAstV1 genotypes, induce CNS pathology in human neurons and astrocytes. Utilizing primary human neurons and astrocytes from various brain regions, this research investigates the cytotoxic effects, apoptotic pathways, and inflammatory responses triggered by these viruses. This study reveals that VA1 …

Ftsk Regulates The Assembly Of The Division Machinery In The Ftsz-Less Chlamydia Trachomatis, Mckenna Katherine Harpring Jan 2024

Ftsk Regulates The Assembly Of The Division Machinery In The Ftsz-Less Chlamydia Trachomatis, Mckenna Katherine Harpring

Theses and Dissertations (ETD)

In evolving to obligate intracellular dependence, Chlamydia trachomatis serovar L2 (Ct), has eliminated several gene products essential for cell division in most other bacteria, including FtsZ, the central coordinator of divisome assembly. In the absence of FtsZ, we show that divisome assembly in Ct is initiated by FtsK, a chromosomal translocase. Chlamydial FtsK forms discrete foci at the septum and at the base of the progenitor mother cell, and our data indicate that FtsK foci at the base of the mother cell mark the location of nascent divisome complexes that form at the site where a daughter cell will emerge …

The Characterization Of Vig-1’S Expression Patterns In Ipnv-Infected, Or Dsrna-Treated, Rainbow Trout Cells, Samantha Lum

Theses and Dissertations (Comprehensive)

In Canada, annual trout production is valued at over $60M with approximately 10,000 tonnes of trout produced per year. Rainbow trout (Oncorhynchus mykiss) are one of the most commonly farmed fresh-water fish in Canada. The high-density farming practices involved in aquaculture increase rainbow trout susceptibility to aquatic viruses such as infectious pancreatic necrosis virus (IPNV). The innate antiviral immune response in rainbow trout is poorly characterized, and the functions of numerous interferon stimulated genes (ISG) in rainbow trout remain unclear. One such gene, virus-induced gene 1 (vig-1) has been poorly studied; however, the human orthologue for vig-1 …

Comparing Ionoregulation And Modes Of Nitrogen Excretion Across The Life Cycle Of Parasitic And Non-Parasitic Lamprey Species, Shannon Davidson Jan 2024

Comparing Ionoregulation And Modes Of Nitrogen Excretion Across The Life Cycle Of Parasitic And Non-Parasitic Lamprey Species, Shannon Davidson

Theses and Dissertations (Comprehensive)

Abstract

Ionoregulation and nitrogenous waste (N-waste) metabolism in fishes has been extensively documented in teleosts and elasmobranchs, but less so in lampreys (Petromizontiformes). All lamprey species undergo metamorphosis which drastically changes their physiology and modes of feeding. However, differences in ionoregulatory mechanisms due to anadromous versus non anadromous life history strategies emerge following metamorphosis in different lamprey species. Furthermore, some species, such as anadromous juvenile sea lamprey (Petromyzon marinus), are parasitic and migrate to seawater (SW) to ingest blood protein from host fishes, while juveniles of freshwater (FW), non-parasitic species, such as the American brook lamprey …

Mainstream Media Portrayal Of Banishment And Nation-Imposed Punishment, Keely Ormond Jan 2024

Mainstream Media Portrayal Of Banishment And Nation-Imposed Punishment, Keely Ormond

Theses and Dissertations (Comprehensive)

“In a traditional village, we wouldn’t have a teepee with no door on it and throw somebody in there. We wouldn’t cast them out, because banishment meant death. What we had to do was restore relationships” – Ryan Beardy (Thorpe, 2022).

The following project examines the representation of Indigenous traditions, customs, and issues in Canadian mainstream media. Specifically, this project is interested in the portrayal of banishment as an Indigenous practice in Canadian mainstream news outlets. This project is based on an interpretive paradigm informed by grounded theory and concepts of media framing, postcolonialism, settler colonialism and restorative justice. Nineteen …

Theses and Dissertations (Comprehensive)

Pauline Oliveros, renowned lesbian composer, left a legacy of experimental music and groundbreaking ideas about music and sound, both as maker and listener. She is best known for her concept of Deep Listening, the active engagement of listening to sound and being open to all possibilities, being receptive to the resonant currents emanating from the sounds, to go beyond hearing and to listen, to Deeply Listen, to the sounds, to experience fully those sounds. Deep Listening was inspired by an experience Oliveros had in a cistern in 1988, where beneath the earth she truly experienced listening and the sounds present …

Incorporating Effects-Based Approaches Into Environmental Impact Assessment To Improve Post-Development Monitoring, Carolyn J M Brown Jan 2024

Incorporating Effects-Based Approaches Into Environmental Impact Assessment To Improve Post-Development Monitoring, Carolyn J M Brown

Theses and Dissertations (Comprehensive)

Over the last 50 years, improvements in design of industrial facilities have significantly reduced environmental impacts. But impacts still occur and monitoring programs are the main mechanism to inform when modification/implementation of mitigation is needed. Informed decisions require adequate baseline (pre-development) data to predict impacts based on the development’s design and to understand when the post-development environment has changed. An adaptive monitoring plan provides an effective way to evaluate monitoring results and allow for proactive responses to environmental change before impacts become difficult or challenging to reverse. Unfortunately, baseline data gathered during an environmental impact assessment (EIA) is often inadequate …

Investigating The Role Of The Mammalian Dsrnai Pathway In Preventing Infection By Respiratory Syncytial Virus And Human Coronavirus, Iliana Portelli Jan 2024

Investigating The Role Of The Mammalian Dsrnai Pathway In Preventing Infection By Respiratory Syncytial Virus And Human Coronavirus, Iliana Portelli

Theses and Dissertations (Comprehensive)

dsRNA interference (dsRNAi) is an antiviral innate immune response that uses long dsRNA (>40bp) to inhibit gene expression through the RNA interference (RNAi) pathway. dsRNAi is an important antiviral defense mechanism in plants, invertebrates, and fungi. However, its use as an antiviral mechanism in vertebrates is not well understood. Evidence proving the significance of dsRNAi in vertebrate cells does exist, but there is still a lack of research in this area, particularly with the use of long dsRNA. Respiratory Syncytial Virus (RSV) and human coronaviruses (HCoVs) are two single stranded RNA (ssRNA) viruses that infect airway cells. This study …

Social Interaction, Social Cognition, And Personality In Gartersnakes And Ball Pythons, Morgan Skinner Jan 2024

Social Interaction, Social Cognition, And Personality In Gartersnakes And Ball Pythons, Morgan Skinner

Theses and Dissertations (Comprehensive)

Snakes are often considered non-social animals. However, this is mostly an untested assumption and the limited research that has been done has shown that snakes interact socially when given the opportunity. Here, the complexity of snake social interactions was explored through a social cognition and personality lens. Group and individual behavioural assays were used to explore flexibility and consistency in social interactions. The results provide evidence that elucidate the function, development, and evolution of these behaviours in snakes. In the first experimental chapter, chapter 2, social network analysis was used to provide a detailed description of aggregation behaviour in 4 …

The Role Of Care Webs In Sustaining Engagement And Well-Being Among Marginalized Emerging Adults Involved In Climate Justice Activism, Vincent Boisclair Jan 2024

The Role Of Care Webs In Sustaining Engagement And Well-Being Among Marginalized Emerging Adults Involved In Climate Justice Activism, Vincent Boisclair

Theses and Dissertations (Comprehensive)

The settler-caused climate crisis poses severe global threats, driving action from youth and emerging adults. Prior to COVID-19 lockdowns, youth-led climate organizations were highly active, but social distancing has hindered youth engagement and exacerbated psychological distress among marginalized emerging adult climate justice activists. Previous research indicates that well-being and social participation are interrelated. Guided by the critical transformative paradigm, this qualitative research examines the effects of care webs on the well-being and sustained engagement of marginalized emerging adults in climate justice activism. Care webs, informed by disability justice, are de-professionalized, non-hierarchical, reciprocal, and anti-oppressive systems of care. Participants were asked …

Reducing Prescription Drug Prices: How Competition Can Make Medications Affordable For Patients - Testimony Of Rachel E. Sachs, Rachel Sachs

Scholarship@WashULaw

In this testimony, I will explain how existing law both keeps branded drug prices high but has also enabled the development of lower-cost generic and biosimilar competition for branded prescription drugs and biological products. This competition can be used to promote access to affordable prescription drugs, benefiting not only patients but also our public payers. However, I will also explain the ways in which existing legislative and regulatory efforts have not always succeeded in promoting competition and will offer a path forward for this Committee to examine reforms that not only encourage the approval of lower-cost products but also ensure …

The Relationship Between Soil Test Phosphorus And Soil Biology - A Natural Management Of Soil Phosphorus Through Conservation Practices, Clarence Frederick Winter Jan 2024

The Relationship Between Soil Test Phosphorus And Soil Biology - A Natural Management Of Soil Phosphorus Through Conservation Practices, Clarence Frederick Winter

Electronic Theses and Dissertations

Phosphorus (P) management is an ongoing challenge in modern agriculture. Due to the importance of P in agriculture, P fertilizer additions are made throughout the U.S. to avoid nutrient-depleted soil. However, the application of P is problematic, as an excess of P in an agricultural system elevates the risk of environmental degradation, and current synthetic P sources are a fleeting resource. Additionally, a majority of P found in agricultural soils are insoluble, in the form of secondary and primary P minerals, making this large pool of P unavailable to plants. Fortunately, soil fungi organisms, such as arbuscular mycorrhizal fungi (AMF) …
