Myotonic dystrophy type 1 (DM1) is a multisystemic disorder caused by the expression of expanded CUG (CUGexp) repeat RNA from the myotonic dystrophy protein kinase (DMPK) gene. This CUGexp repeat RNA’s gain-of-function mechanism leads to the sequestration of muscleblind-like (MBNL) proteins, resulting in widespread alternative splicing dysregulation across tissues. Despite significant research, there are currently no approved therapeutics targeting the underlying causes of DM1. This dissertation explores the development and optimization of a novel class of small molecules, Modified Polycyclic Compounds (MPCs), designed to address splicing dysregulation in DM1. Mercury (Hg) exposure from environmental sources and consumer products presents a significant public health concern. The toxicity of methylmercury (MeHg) differs considerably from inorganic mercury (iHg), such that their separation and quantification in whole blood is helpful in identifying sources and possible pathways of exposure. Speciation methods provide a more detailed picture regarding human exposure to toxic mercury species and their effects on human health. This dissertation utilizes the lens of Social Reproduction Theory to examine the consequences of viewing race and racism and other systems of oppression like Patriarchy as separate or additive autonomous ontologies. Intersectionality Theory posits oppressions as independent systems that “add” or “intersect” onto oppressed subjects in particular circumstances. On the other hand, responding to Intersectionality Theory, Social Reproduction Theory understands systems of oppression as mutually constitutive social relations. Social Reproduction Theory therefore allows for a more thorough understanding of the relationship between racism and other systems of oppression and their own relationship to capitalism. Electrocoagulation (EC) with a zinc anode demonstrated promising results to remove perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) from an aqueous solution. However, the energy requirement for EC is usually very high due to water electrolysis or aeration. The second chapter of this dissertation aims to reduce energy consumption using an air-cathode in EC (ACEC) to supply oxygen electron acceptor without aeration for attenuating PFOA/PFOS in this new configuration. For the high PFOA concentration (0.25 mM), ACEC with 45 min of the reaction time exhibited an excellent PFOA removal (99.8 ± 0.3% removal) comparable to an EC with aeration (EC-aeration, …

Background. Implementation science aims to identify methods that support the adoption of evidence-based practices (EBPs), including measurement-based care (MBC) and MBC-informed patient-therapist matching. Examining clinician attitudes can elucidate potential barriers to implementing EBPs, and attitudinal differences may exist across groups of therapists. In the context of a patient-therapist matching implementation project, this multi-method study investigated the relationship between clinician characteristics, attitudes toward EBPs/MBC, and implementation climate. Method. A sample of N = 72 therapists completed measures of demographic and clinical characteristics, attitudes toward EBPs, perceptions of implementation climate, and attitudes toward monitoring and feedback. Gamification has emerged over the past ~10-12 years as a popular design approach to problems of motivation in online or blended learning, with a particular emphasis on distinguishing between extrinsic motivation driven by points, grades, class rank, etc, vs. intrinsic motivation driven by personal values and interest in the curriculum. The egg's quality is crucial for the healthy development of the next generation, making oogenesis a tightly regulated process with multiple benchmarks. To investigate the intricacies of oogenesis, we utilized the Drosophila oogenesis system as our primary model. This choice was motivated by several factors. American alligators (Alligator mississippiensis) have received little scientific attention in the context of human-driven land use change. The goal of my study is to investigate how land use change may be affecting the spatial ecology of alligator nesting in Florida. Areas directly impacted by humans are growing rapidly in Florida, yet the impacts of this land use change on alligator populations and reproduction are largely unknown. Unlike mammals, shark endoskeletons are composed of cartilage, which is less stiff compared to bone. Despite this structural disadvantage, sharks have risen to become apex predators, with reported maximum posterior bite force estimates approximating 5914 N in bull sharks. Shark jaws are estimated to undergo high cycle loading within the organism’s lifecycle without evidence of failure. Fatigue resistance may relate to mineral distribution within a component of the endoskeletal system – tesserae. Tesserae are thin, roughly hexagonal tiles composed of calcium phosphate hydroxyapatite (HA) crystals and grow by accretion to eventually surround the hyaline cartilage core. This thesis investigates the effectiveness of microbially-induced calcium carbonate precipitation (MICP) using the recently introduced bioslurry treatment method via surface percolation. To simulate realistic water forcing conditions, a piston-style erosion rate testing apparatus, specifically the Florida Department of Transportation Sediment Erosion Rate Flume, was used. Initial tests conducted at a high shear stress of 42 Pa revealed that a combination of 15% bioslurry and 85% cementation solution exhibited the strongest erosion resistance when compared to other ratios between bioslurry and cementation solution. Asphondylia borrichiae, like other gall-forming gnats, are obligate symbionts to the endophytic fungi (EF) they deposit when laying eggs. Asphondylia spp. and EF are important biocontrol agents, yet details surrounding their diversity, life cycles, and host interactions remain poorly understood. Galls and leaf-stem tissues from two host plants were surveyed to identify and compare EF diversity between the host tissues and species of A. borrichiae. Identification of 114 isolates was carried out by sequencing the internal transcribed spacer (ITS 1/4 primers). The Nourish to Flourish study aimed to decrease the prevalence of obesity among food insecure children in Jacksonville, FL. Specifically, the program focused on nutrition education and access to healthy foods. The study consisted of a 4-week nutrition education series emphasizing the major determinants of healthy weight known as 5-2-1-0! Freshwater harmful algal blooms (HABs) are a global water quality issue. Algae, especially cyanobacteria, are integrally associated with complex microbial assemblages, which cycle nutrients, provide microenvironments, breakdown potentially harmful molecules, etc. However, these same microbial consortia are competitors or even inhibitors to algae in these systems. One potential biological control is Rhodospirillum rubrum, a Gram- proteobacterium. These involve reordering scrambled sequences of numbers into a sequential arrangement known as the identity permutation.
Rearranging any given permutation into the identity permutation with the minimum number of specific operations related to strips gives rise to various combinatorial optimization scenarios. Zoos and aquariums utilize research-based management to ensure animals remain in a positive welfare standing. While vital, this research is taxa-biased towards mammals, with few studies observing other species that are frequently observed in captivity, such as elasmobranchs, or sharks and rays. Often maintained in exhibits that allow direct guest interaction, or touch pools, elasmobranchs have been shown to respond to these exposures through behavior changes in activity and social interactions. This study focuses on the development of an adaptive ride-on toy specifically designed for a 2.5-year-old child with spastic diplegic cerebral palsy. As a feasibility study, the primary objective of this innovative device is to enhance motor function, foster autonomy, and improve the overall quality of life for the child. Florida's newest legislation leaves many Florida queer teachers feeling marginalized and unprotected in the classroom (Block, 2022). The “Parental Rights in Education” Bill (FL HB 1557 and FL HB 1069), infamously renamed “Don’t Say Gay”, was created ostensibly to help protect parental rights and choices in education. The new law prohibits the discussion of gender identity and sexual orientation in kindergarten through twelfth grade classrooms. “Don’t Say Gay” provides vague guidelines for queer teachers on approaching topics like sexual orientation and gender identity. Recently, there has been increasing use of baited remote underwater video (BRUV) surveys for assessing shark and fish populations in marine ecosystems. This approach can be useful because it may provide a non-destructive, non-extractive, and cost-effective alternative to traditional fishing-based methods. This paper discusses the treatment of Florida beach sand by surface percolating Bioslurry, a microbially-induced calcite precipitation (MICP) variant that was developed and refined in recent years. Researchers examined how variations in pore volume and surface area affected the cemented depth of soil treated via surface percolation. Results suggested that while the pore volume is an important factor that governs cementation in soil treated via surface percolation, it does not appear to be the only controlling factor and treatment volume per unit surface area is another important factor to consider when treating soil with Bioslurry using surface percolation. This chapter explores how practices of digital kinning and digital homing enacted through smartphones shape the experiences of Chinese older adults in Perth, Australia. Transnational grandparent carers must balance the risks of diminished social networks with the benefits of providing intergenerational care. Some form strong friendships in Perth, but many yearn for a leisured life with their friends in China. Findings illustrate the central role of digital migration practices in transnational families and friendships, not only sustaining distant support networks, but also building new communities in host settings. Critical Belief Analysis for Security Studies is a companion volume to Security Analysis: A Critical-Thinking Approach. Critical Belief Analysis (CBA) is an emerging analytic technique for describing, explaining, and predicting decisions and actions of agents (actors, believers, decision-makers). Understanding the sources of primary production supporting marine food webs is important for conserving critical habitats. However, the importance of allochthonous versus autochthonous production is often unclear. We identify the sources of primary production supporting three species of predatory fish (Lutjanus carponotatus, Lutjanus fulviflamma, and Lethrinus punctulatus) in the Dampier Archipelago, Western Australia. The proportions of mangrove, seagrass, macroalgae, and suspended particulate organic matter (SPOM) assimilated by fish of different life-history stages and from different habitats were determined using stable isotopes and Bayesian mixing models. How does Australia's latest Indigenous defeat relate to Environmental Education? The answer is direct complicity. This paper begins with the premise that the failure of Australia's 2023 referendum on "The Voice to Parliament"is directly connected with education. The chapter builds on the proposition that local and Indigenous public knowledge could - and should - be the heart of environmental education. The humoral response is complex and involves multiple cellular populations and signaling pathways. Bacterial and viral infections, as well as immunization regimens, can trigger this type of response, promoting the formation of microanatomical cellular structures called germinal centers (GCs). GCs formed in secondary lymphoid organs support the differentiation of high-affinity plasma cells and memory B cells. There is growing evidence that the quality of the humoral response is influenced by genetic variants. N-Nitrosamines, carcinogenic compounds present in dietary and environmental sources and formed endogenously, are believed to be linked with the presence of nitrate and nitrite, both within dietary sources and after intake. To fully evaluate this potential threat to human health, an accurate analytical method to measure N-nitrosamines in biological matrices is necessary. We report a simple, fast, selective mass spectrometry method to detect N-nitrosamines in human urine. Background: Chronic lymphocytic leukaemia (CLL) typically presents with asymptomatic, early-stage disease that is monitored until disease progression (‘treatment-naïve’ CLL). The objective of this pilot study was to assess the feasibility and preliminary safety of an exercise program in treatment-naïve CLL. We also sought to preliminarily assess the impact of the exercise program on disease activity, as it has been proposed that exercise training may reduce disease outgrowth in treatment-naïve CLL.
