The Nagoya Protocol to the Convention on Biological Diversity (CBD) has threatened to impede access to genetic resources and related data for cross-border scientific research. In principle, every use of genetic resources would require a set of contracts under the CBD, in a “bilateral” regime. The related transaction costs could overwhelm many transnational research undertakings, affecting even public health responses to outbreaks and epidemics. However, the Nagoya Protocol also offers a unique opportunity to resolve this dilemma, despite struggles to define the meaning and coverage of “digital sequence information.” The coverage of genetic sequence data under the CBD remains controversial …

BACKGROUND: Metagenomic next-generation sequencing (mNGS) is increasingly being used for microbial detection in various infectious syndromes. However, data regarding the use of mNGS in solid organ transplant recipients (SOTR) are lacking.

OBJECTIVES: To describe and analyze real-world clinical impact of mNGS using plasma microbial cell-free DNA (mcfDNA) in SOTR.Design: Retrospectively reviewed all adult SOTR who underwent mNGS testing using plasma mcfDNA at Baylor St Luke's Medical Center from March 2017 to February 2023.

BACKGROUND: Metagenomic next-generation sequencing (mNGS) is a novel diagnostic tool increasingly used in the field of infectious diseases. Little guidance is available regarding its appropriate use in different patient populations and clinical syndromes. We aimed to review the clinical utility of mNGS in patients with a specific clinical syndrome and identify factors that may increase its utility.

METHODS: We retrospectively reviewed charts of 72 non-immunocompromised adults hospitalized with the clinical syndrome of 'fever of undetermined origin' and underwent mNGS testing. We report 9 patients with invasive Bartonella infections, including 5 with endocarditis, who were diagnosed with microbial cell-free DNA next-generation sequencing and Bartonella serology studies. Diagnosis with plasma mcfDNA NGS enabled a faster clinical and laboratory diagnosis in 8 patients. Prompt diagnosis impacted antibiotic management in all 9 patients.

INTRODUCTION: In response to the COVID-19 pandemic, we rapidly implemented a plasma coordination center, within two months, to support transfusion for two outpatient randomized controlled trials. The center design was based on an investigational drug services model and a Food and Drug Administration-compliant database to manage blood product inventory and trial safety.

METHODS: A core investigational team adapted a cloud-based platform to randomize patient assignments and track inventory distribution of control plasma and high-titer COVID-19 convalescent plasma of different blood groups from 29 donor collection centers directly to blood banks serving 26 transfusion sites.

BACKGROUND: Neuropathic adverse events occur frequently in linezolid-containing regimens, some of which remain irreversible after drug discontinuation.

OBJECTIVE: We aimed to identify and validate a host RNA-based biomarker that can predict linezolid-associated neuropathy before multidrug-resistant/rifampicin-resistant tuberculosis (MDR/RR-TB) treatment initiation and to identify genes and pathways that are associated with linezolid-associated neuropathy.

METHODS: Adult patients initiating MDR/RR-TB treatment including linezolid were prospectively enrolled in 3 independent cohorts in Germany. Mechanistic investigation of host-microbe interactions in the human gut are hindered by difficulty of co-culturing microbes with intestinal epithelial cells. On one hand the gut bacteria are a mix of facultative, aerotolerant or obligate anaerobes, while the intestinal epithelium requires oxygen for growth and function. Thus, a coculture system that can recreate these contrasting oxygen requirements is critical step towards our understanding microbial-host interactions in the human gut. Here, we demonstrate Intestinal Organoid Physoxic Coculture (IOPC) system, a simple and cost-effective method for coculturing anaerobic intestinal bacteria with human intestinal organoids (HIOs). In vitro models, such as primary cells and continuous cell lines routinely used for evaluating drug candidates, have limitations in their translational relevance to human diseases. Organotypic cultures are increasingly being used to assess therapeutics for various cancers and infectious diseases. Monitoring drug cytotoxicity in cell cultures is crucial in drug development, and several commercially available kits for cytotoxicity assessment offer distinct advantages and limitations. Cytomegalovirus (CMV) infection poses a significant threat to solid organ transplant (SOT) recipients and can lead to various complications and adverse outcomes. In an effort to prevent CMV infection, it is common to utilize prophylactic strategies, including antiviral medications such as valganciclovir, especially for high-risk patients. Risk factors for CMV infection in kidney transplant recipients (KTRs) include CMV mismatch between donor and recipient (i.e., donor positive, recipient negative), and intensity of immunosuppression, such as the use of T-cell depleting agents. Purpose: This study aimed to evaluate the risk of pediatric glioma in relation to multiple birth and parental characteristics as well as environmental exposures, in the overall California population and in racial and ethnic subgroups.

Methods: Leveraging a population-based linkage of statewide birth records and cancer registry data in California, we conducted a case-control study that included 3,244 cases who were born in California during 1982-2015 and diagnosed with first primary glioma at the age of 0-19 years during 1988-2015, and 162,200 controls who were born in California and frequency-matched to cases on year of birth. For this study, DNA classifier dataset was extracted from UCI repository of machine learning databases. Food allergies have been an increasing concern worldwide with very little information on preventative measures and treatment. They can have long lasting and life-threatening consequences with a wide range of symptoms from hives to anaphylactic shock. Long recommended feeding guidelines, vitamin and mineral deficiencies, and gastrointestinal alterations are thought to play a role in the increase in food allergies in infants. Early introduction of food, immunotherapy, and a properly supported gut microbiome have been recommended to decrease the likelihood of developing a food allergy and desensitizing the immune system.

Sand tigers are frequently observed at shipwrecks and other artificial reef habitats off North Carolina (USA), but data about occupancy, movement ecology, and site fidelity are lacking. Spot A Shark USA researchers used Wildbook© photoidentification software to spot map sand tigers in images provided by recreational SCUBA divers, or collected from remotely operated vehicles, and an offshore live-streaming camera. We uniquely identified 1837 sand tigers, 101 of which were resighted on more than one date between 2005–2021. News organizations and journalists are important and influential actors in environmental politics. Their reporting on social and environmental issues often follows issue-attention cycles (IACs) that emphasize drama and problematization to maintain public interest. This study examined nearly 13 years of news coverage to understand the media discourse about the Critically Endangered North Atlantic right whale (NARW) and its conservation. Content analysis and critical discourse analysis were used to analyze 356 texts published in six large US newspapers between January 1, 2010 and March 15, 2023. This study investigates the practices and impact of School Self-Evaluation/Internal School Quality Assurance (SSE/ISQA) on academic achievement in secondary schools within the Mara region of Tanzania. Employing a qualitative approach and case study design, 41 participants were purposefully sampled, including school administrators, teachers, and students. Data collection involved semi-structured interviews, focus group discussions, and document analysis, with thematic research applied for data interpretation. Key findings revealed that specific SSE practices, including teacher-student collaboration, follow-up on teaching and learning, and the provision of examinations, positively influence academic achievement. In 1966, in response to growing fears that men would attempt to pose as women to win medals in elite competition, the International Olympic Committee (IOC) began gender testing all female athletes through various visual and gynecological evaluations. These tests were quickly deemed to be too invasive and were replaced by other chemical and genetic tests as scientific capabilities advanced. The deposition offers a singular opportunity to handcuff the deponent to an irreversible script. Consequently, both the attorney taking the deposition and defending counsel must prepare for and conduct the deposition with equal if not greater care than the trial.

Traditionally, lawyers have used the deposition to discover facts relating to the legal elements and the credibility, perception, and recollection of the witness. Urban trees contribute to decarbonisation. However, the removal of trees may release the stored carbon within them. In Hong Kong, Acacia confusa and Leucaena leucocephala are being removed, but with unknown carbon impacts. This industry-academia-government collaborative research aimed to: (1) report the amount of carbon stored inside A. confusa and L. leucocephala harvested from slopes; (2) estimate the carbon storage of a tree using dendrometric measurements; and (3) explain the variation in carbon content percentage of urban trees with respect to tree species, part of the tree, and position within the part. 10 A. confusa and 10 L. leucocephala were …
