SMARCB1 loss has long been observed in many solid tumors. However, there is a need to elucidate targetable pathways driving growth and metastasis in SMARCB1-deficient tumors. Here, we demonstrate that SMARCB1 deficiency, defined as genomic SMARCB1 copy number loss associated with reduced mRNA, drives disease progression in patients with bladder cancer by engaging STAT3. SMARCB1 loss increases the chromatin accessibility of the STAT3 locus in vitro. Orthotopically implanted SMARCB1 knockout (KO) cell lines exhibit increased tumor growth and metastasis. SMARCB1-deficient tumors show an increased IL6/JAK/STAT3 signaling axis in in vivo models and patients. Furthermore, a pSTAT3 selective inhibitor, TTI-101, reduces …

Double-Blinded Randomized Control Trial Investigating The Efficacy Of Probiotic Mouth Rinse In Enhancing Oral Health, Sudeep C B Prof. Dr., Sunil P M Prof. Dr.
Annual Research Symposium
The efficacy of probiotic mouth rinse in enhancing oral health was investigated through a double-blinded randomized controlled trial involving 45 children aged 12 to 15 over a two-week period. Comparing probiotic mouth rinse, chlorhexidine mouth rinse, and a control group, plaque and gingival accumulation were assessed using established indices. Results indicated both probiotic and chlorhexidine groups exhibited reduced plaque compared to the control, with the probiotic group showing superior reduction in gingival inflammation. These findings suggest the potential of probiotic mouth rinse in improving oral hygiene and overall oral health.

Investigating Optimal Laboratory Growth Conditions Of Gracilibacillus Halotolerans In Media Supplemented With Salt, Isaac Young
Annual Research Symposium
As interest continues to grow in the field of persister cells and their morphology, there arises an ever-evolving desire to further understand specific strains of bacteria that exemplify the qualities of seemingly anomalous survival regardless of anti-bacterial treatment. In the case of the Gracilibacillus halotolerans, a halotolerant extremophile extracted from the Great Salt Lake with known persistent characteristics, uncovering its optimal growth conditions was essential for future investigations. Identifying the optimal salinity for the growth of G. halotolerans will allow us to standardize our growth methods, uncover several mechanisms of saline tolerance, and add to future investigations of persistence with …

Evidence Of Prostate Cancer Progression In Transgender Women After Hormone Replacement Therapy - Scoping Review Protocol, Emily Nelson
Annual Research Symposium
This scoping review aims to determine the extent and quality of evidence regarding prostate cancer progression outcomes in transgender women who have undergone hormone replacement therapy. Notably, transgender females undergoing hormone replacement therapy as a component of the gender affirmation treatment commonly have their prostate, rendering them vulnerable to developing prostate cancer. There is no current agreement about the likelihood of transgender women developing prostate cancer or the impact of hormone replacement therapy on oncogenic molecular pathways. Unfortunately, due to a lack of scientific data and inadequate trust and awareness in the medical community, transgender women are often not provided …

Role Of Mumio In Periodontal Wound Healing–An In Vitro Study On Human Pdl Cells, Jagadish Hosmani, Abdullah Alqarni, Abdullah Alqahtani, Nasser Alhamid
Annual Research Symposium
Mumio, used in ancient medicine for almost 4000 years, is the gold standard of traditional medicine. Mumio, a tar-like compound made from rock dissolved minerals and degraded organic matter, takes a century to form. Dibenzo-alpha-pyrones, humic acid, and fulvic acid are important in Mumio, which includes 60–80% humus, benzoic acid, fatty acids, ichthyol, ellagic acid, resin, triterpenes, sterol, aromatic carboxylic acids, bioactive 3,4-benzokoumarins, amino acids, phenol Active chemicals improve Mumio's antioxidant effects. Despite its anti-ulcerogenic, anti-inflammatory, antioxidant, immunomodulatory, memory-enhancing, and anxiolytic characteristics, mumio has been studied rarely on oral tissues. The present study examined the cytotoxic, antioxidant, anti-inflammatory, and reparative …
Interplay Of Periodontal Bacterial Metabolites In The Progression Of Coronary Artery Disease: A System Biological Approach, Janakiraman V
Interplay Of Periodontal Bacterial Metabolites In The Progression Of Coronary Artery Disease: A System Biological Approach, Janakiraman V
Annual Research Symposium
Purpose:
The purpose of this study is to investigate the intricate relationship between periodontal disease (PD) and coronary artery disease (CAD), as evidenced by epidemiological associations. Metalloproteinase inhibitor (TIMP1) plays a pivotal role in cellular signaling, differentiation, cell death, and migration by binding to target metalloproteinases, forming complexes with other molecules (collagenases) to inactivate them. However, the expression of TIMP1 is reduced in both PD and CAD, leading to an upregulation of other metalloproteinases. This research explores the hypothesis that metabolites released from (Porphyromonas gingivalis), a prevalent bacterium in atherosclerotic patients, may inhibit TIMP1, thereby influencing CAD progression. …

Sulforaphane Pre-Treatment Improves Cytoprotection Against Opportunistic Pathogens, Robert Darby, Nathan Clark
Sulforaphane Pre-Treatment Improves Cytoprotection Against Opportunistic Pathogens, Robert Darby, Nathan Clark
Annual Research Symposium
Alcohol is associated with increased mortality and morbidity globally. Pulmonary infections with opportunistic pathogens can occur in healthy humans; however, binge alcohol intoxication (≥ 0.08% BAC) is a major risk factor. We have previously shown that a single dose of alcohol comparable to binge alcohol intoxication increases infection by reducing alveolar macrophage function in vivo. The aim of this study was to 1) test the therapeutic potential of the phytonutrient sulforaphane (SFN) given as a pre-treatment, and 2) test the alcohol-induced effects on phagocytic function in murine and human macrophages in vitro.

Conservation is increasingly focused on preventing losses in species’ populations before they occur. Tracking changes in demographic parameters that can impact a population’s resilience in response to drivers of global change can support early conservation efforts. We assessed trends in population productivity (late summer juveniles per 100 females) relative to drivers of global change in 40 pronghorn (Antilocapra americana) herds across sagebrush (Artemisia spp.) steppe in Wyoming. Pronghorn are an iconic rangeland species that have been exposed to increasing levels of anthropogenic, climatic, and land-use change. Using data collected across the state of Wyoming, we (1) assessed …

The rapid growth of cultural evolutionary science, its expansion into numerous fields, its use of diverse methods, and several conceptual problems have outpaced corollary developments in theory and philosophy of science. This has led to concern, exemplified in results from a recent survey conducted with members of the Cultural Evolution Society, that the field lacks ‘knowledge synthesis’, is poorly supported by ‘theory’, has an ambiguous relation to biological evolution and uses key terms (e.g. ‘culture’, ‘social learning’, ‘cumulative culture’) in ways that hamper operationalization in models, experiments and field studies. Although numerous review papers in the field represent and categorize …

The present research is primarily focused on investigating the characteristics of environmentally persistent free radicals (EPFRs) generated from commonly recognized aromatic precursors, namely, 1,2-dichlorobenzene (DCB) and 2- monochlorophenol (MCP), within controlled laboratory conditions at a temperature of 230 °C, termed as DCB230 and MCP230 EPFRs, respectively. An intriguing observation has emerged during the creation of EPFRs from MCP and DCB utilizing a catalyst 5% CuO/SiO2, which was prepared through various methods. A previously proposed mechanism, advanced by Dellinger and colleagues (a conventional model), postulated a positive correlation between the degree of hydroxylation on the catalyst’s surface (higher hydroxylated, HH and …
Impairment Of The Glial Phagolysosomal System Drives Prion-Like Propagation Of Huntington’S Disease, Graham Davis
Impairment Of The Glial Phagolysosomal System Drives Prion-Like Propagation Of Huntington’S Disease, Graham Davis
Theses and Dissertations
The ability of glia to tightly regulate neuronal health and homeostasis in the CNS is conserved across species. Yet, despite the ability to degrade protein aggregates, glia are vulnerable to the accumulation of neurotoxic amyloid aggregates during neurodegenerative disease progress, and even exacerbate their spread. A developing narrative highlights glia as a double-edged sword in neurodegenerative diseases: initially capable of dynamically responding to amyloid aggregate-ladened dying neurons but also capable of inducing chronic inflammation and creating seeding-competent amyloid oligomers. Thus, uncovering the mechanisms that allow glia to control aggregate deposition while preventing the neurotoxic effects and seed generation is vital …
Misgivings In Measuring Happiness, Sudhanva Char
Misgivings In Measuring Happiness, Sudhanva Char
International Review of Business and Economics
According to a resolution of the UN General Assembly (Resolution 66/281), March 20th is observed annually as International Day of Happiness. A nation’s overall success is measured by people’s happiness, the litmus test. The World Happiness Report (WHR) states there is consensus about measuring happiness, whereas, happiness is idiosyncratic and its connotation differs from culture to culture, language to language, and even person to person. Personal ‘space’ in all spheres matters, and so do democracy or dictatorship, all factors leading to mismeasures of happiness scores. And so, there are paradoxes in happiness rankings in WHR. Economists have yet to …

Maternal grouping dynamics involve trade-offs between: (1) infant protection from predation (predation hypothesis), (2) infant protection from male harassment (infant safety hypothesis), and (3) reducing scramble competition for prey resources (foraging-type scramble competition hypothesis). Using unmanned aerial vehicles, we assessed grouping dynamics in common dolphin (Delphinus delphis) groups containing mother-calf pairs to address these hypotheses. We analyzed social aspects and structural group elements using generalized additive mixed models and modeled group formation using multinomial generalized estimating equations. Calf proportion was higher in very compact groups and in groups of 10–20 individuals but decreased in larger groups. The frequency of socio-sexual …

Organometallic drug development is still in its early stage, but recent studies show that organometallics having iron as the central atom have the possibility of becoming good drug candidates because iron is an important micro-nutrient, and it is compatible with many biological systems, including the human body. Being an eco-friendly Lewis acid, iron can accept the lone pair of electrons from imino(sp2)-nitrogen, and the resultant iron-imine complexes with iron as a central atom have the possibility of interacting with several proteins and enzymes in humans. Iron-imine complexes have demonstrated significant potential with anticancer, bactericidal, fungicidal, and other medicinal activities in …

The melting cryosphere adds heterogeneity to the abiotic and biotic characteristics of many high latitude and montane rivers. However, climate change threatens the cryosphere's persistence in many regions. While existing research has explored the impacts of cryospheric loss on the diversity and structure of freshwater communities, implications for functional traits of communities, such as production of aquatic invertebrates, remain unresolved. Here, we quantified aquatic invertebrate community structure and secondary production in southeast Alaska (USA) streams that represent a meltwater to non-meltwater gradient, including streams fed primarily by: (1) glacier-melt, (2) snowmelt, (3) rainfall, and (4) a combination of these sources. …

Stroke results in varying levels of motor and sensory disability that have been linked to the neurodegeneration and neuroinflammation that occur in the infarct and peri-infarct regions within the brain. Specifically, previous research has identified a key role of the corticospinal tract in motor dysfunction and motor recovery post-stroke. Of note, neuroimaging studies have utilized magnetic resonance imaging (MRI) of the brain to describe the timeline of neurodegeneration of the corticospinal tract in tandem with motor function following a stroke. However, research has suggested that alternate motor pathways may also underlie disease progression and the degree of functional recovery post-stroke. …

Elucidating Neuroinflammation In Multiple Sclerosis By Network Analysis, Nora C. Welsh
Dartmouth College Ph.D Dissertations
Multiple sclerosis (MS) is a heterogeneous disease, differing on many variables, including disease course, sex, and overall activity. Key characteristics of the disease encompass demyelination, axonal damage, neuronal loss, glial cell activation, and the infiltration of peripheral immune cells. Molecular proxies of these functions are secreted proteins, including cytokines and immunoglobulins, which, in the central nervous system (CNS), can be secreted into the cerebrospinal fluid (CSF). A detailed analysis of these secreted proteins can offer insights into the evolving immunological and neurodegenerative features as the disease progresses. To understand the dynamic biological processes involved in MS, I used network analysis …

Traditional methods have relied on video examination by investigators, which is labor-intensive and susceptible to bias. To address these challenges, research efforts have focused on computational methods and image-processing algorithms for automated behavioral classification. Two primary approaches have emerged: marker-and markerless-based tracking systems. In this study, we showcase the utility of “Augmented Reality University of Cordoba” (ArUco) markers as a marker-based tracking approach for assessing rat engagement during a nose-poking go/no-go behavioral task. …
