Legume-grass mixtures may be a useful alternative to nitrogen-fertilized grass monocultures, but pasture and animal responses have not been assessed for pastures grazed by horses in Florida. This 2-yr study compared pasture and horse responses of continuously stocked, mixed pastures of rhizoma peanut (RP, Arachis glabrata Benth) and bahiagrass (BG, Paspalum notatum Flüggé) receiving 30 kg nitrogen (N)/ha (RP-BG) compared with BG pastures fertilized with 120 kg/N ha (BG-N) or with no N (BG-No N). Legumes and forbs contain bioactives or plant secondary compounds (PSC) with potential to enhance animal health through their antibiotic, antioxidant and immunomodulatory properties that are evident even at small dietary concentrations. In turn, ruminants can regulate their ingestion of PSC through behavioral mechanisms that allow for the efficient achievement of homeostasis. High concentrations of PSC lead to food avoidance, whereas lower content of PSC in the diet achieved through regulatory mechanisms of ingestion could promote medicinal and/or prophylactic effects in the animal and concomitant health benefits to milk and meat products. Spring grass availability has a major impact on dry matter intake (DMI) of dairy cows during early lactation. The objective of this study was to investigate the effects of opening farm cover (OFC) on DMI during early lactation. A twelve week experiment, which was split into two 6 week periods (Period 1 (Week 1 – 6) and period 2 (Week 7 – 12), was conducted over a two year period. The hindgut microbiome plays an essential role in horses consuming forage-based diets high in fiber, such as pasture. The equine hindgut harbors a large microbial community that ferments dietary fiber and produces by-products which provide a substantial portion of daily energy requirements. Despite the importance in nutrition and health, research evaluating the hindgut microbiome of grazing horses is relatively limited. Rotational grazing management offers potential advantages for economic and environmental sustainability, as well as for horse health. Rotational grazing has the potential to increase pasture yields and available forage for grazing, and therefore reduce supplemental feed costs. A rotational grazing management strategy can also prevent or mitigate negative effects of overgrazing, with implications for pasture productivity and the environment. Overgrazing can reduce vegetative cover over time, increasing the potential for erosion and nutrient run-off. Overgrazing may also result in long-term decreased forage stand persistence and weed invasion, necessitating more frequent pasture renovation. All of this can negatively impact the horse’s …

Nationally, the lack of improvement in literacy scores continues to baffle experts. Instructional leaders from math, science, and social studies in one district knew the value of leveraging their subject areas to support literacy achievement in secondary students. Starting with an engaging STEM-related novel, the group of curriculum experts developed meaningful literacy connected tasks for their teacher and teacher leader participants. Energetic Value Estimates Of Wild And Domesticated Food Items Consumed By Congolese Bayaka Foragers, Lucie Benoit, Sheina Lew-Levy, Yann R. Ouamba, Alyssa N. Crittenden

We present a dataset that systematically estimates the energetic value of wild and domesticated food resources collected and consumed by Congolese BaYaka foragers. This dataset is unique in that it accounts for everyday practices of food processing and consumption. Reported values of inedible portions, moisture content, and estimated energetic values (kcal/g) were extracted from field research and the published food chemistry and nutrition literature. These data are central to understanding how nutritional needs are met in rainforest environments and will allow for current and future tracking of diet composition in this and similar ecologies. Introduction: Cannabis use is increasing among pregnant people, and cannabidiol (CBD), a constituent of cannabis, is often perceived as ‘‘natural’’ and ‘‘safe’’ as it is non-intoxicating. In utero, cannabis exposure is associated with negative health outcomes, including fetal growth restriction (FGR). The placenta supplies oxygen and nutrients to the fetus, and alterations in placental development can lead to FGR. Copper−cystine-based high aspect ratio structures (CuHARS) possess exceptional physical and chemical properties and exhibit remarkable biodegradability in human physiological conditions. Extensive testing has confirmed the biocompatibility and biodegradability of CuHARS under diverse biological conditions, making them a viable source of essential Cu2+. These ions are vital for catalyzing the production of nitric oxide (NO) from the decomposition of Snitrosothiols (RSNOs) found in human blood. The ability of CuHARS to act as a Cu2+ donor under specific concentrations has been demonstrated in this study, resulting in the generation of elevated levels of NO. A large convection-cloud chamber has the potential to produce drizzle-sized droplets, thus offering a new opportunity to investigate aerosol-cloud-drizzle interactions at a fundamental level under controlled environmental conditions. One key measurement requirement is the development of methods to detect the low-concentration drizzle drops in such a large cloud chamber. In particular, remote sensing methods may overcome some limitations of in situ methods. Here, the potential of an ultrahigh-resolution radar to detect the radar return signal of a small drizzle droplet against the cloud droplet background signal is investigated. Neurons of the basal forebrain nucleus basalis and posterior substantia innominata (NBM/SIp) comprise the major source of cholinergic input to the basolateral amygdala (BLA). Using a genetically-encoded acetylcholine (ACh) sensor in mice, we demonstrate that BLA-projecting cholinergic neurons can 'learn' the association between a naïve tone and a foot shock (training) and release ACh in the BLA in response to the conditioned tone 24h later (recall). In the NBM/SIp cholinergic neurons express the immediate early gene, Fos following both training and memory recall. Oligopeptides of l-lysine have the potential for applications in various scientific and technical areas. The number of residues in polycationic compounds such as oligolysine is also reported to have an effect on its biological properties. Hence, there is a necessity for developing efficient oligolysine synthesis methods where the oligopeptide dispersity can be tailored, along with optimum yield values. The ability of proteases to reverse their proteolytic activity to synthesize peptides has been reported in the literature. Brazil is a leading grain producer and exporter, producing on average 6.6 million tons (Mt) of wheat per year from 2013 to 2023. However, to meet its average annual domestic consumption of 11.1 Mt over the same period, the country continued to rely on imports of 4.5 Mt of wheat each year. This dependency on imports leads to high wheat prices in Brazil compared to the global average.

KRAS mutations drive oncogenic alterations in numerous cancers, particularly in human pancreatic ductal adenocarcinoma (PDAC). About 93% of PDACs have KRAS mutations, with G12D (∼42% of cases) and G12V (∼32% of cases) being the most common. The recent approval of sotorasib (AMG510), a small-molecule, covalent, and selective KRASG12C inhibitor, for treating patients with non-small cell lung cancer represents a breakthrough in KRAS targeted therapy. However, there is a need to develop other much-needed KRAS-mutant inhibitors for PDAC therapy. Stimulator of interferon genes (STING) is an immune adaptor protein that senses cyclic GMP-AMP (cGAMP) in response to self or microbial cytosolic DNA as a danger signal. STING is ubiquitously expressed in diverse cell populations including cancer cells with distinct cellular functions such as activation of type I interferons, autophagy induction, or triggering apoptosis. The TP53 tumor suppressor gene is mutated early in most of the patients with triple-negative breast cancer (TNBC). The most frequent TP53 alterations are missense mutations that contribute to tumor aggressiveness. Here, we used an autochthonous somatic TNBC mouse model, in which mutant p53 can be toggled on and off genetically while leaving the tumor microenvironment intact and wild-type for p53 to identify physiological dependencies on mutant p53. In TNBCs that develop in this model, deletion of two different hotspot p53R172H and p53R245W mutants triggers ferroptosis in vivo, a cell death mechanism involving iron-dependent lipid peroxidation. Pathological consequences of circadian misalignment, such as shift work, show considerable individual differences, but the lack of mechanistic understanding hinders precision prevention to prevent and mitigate disease symptoms. Here, we employed an integrative approach involving physiological, transcriptional, and histological phenotypes to examine inter-individual differences in pre-symptomatic pathological progression, preceding irreversible disease onset, in wild-type mice exposed to chronic jet-lag (CJL). We observed that CJL markedly increased the prevalence of hepatic steatosis with pronounced inter-individual differences. Stratification of individual mice based on CJL-induced hepatic transcriptomic signature, validated by histopathological analysis, pinpoints dysregulation of lipid metabolism. In recent years, there has been increased interest in incorporation of backfilling into dose-escalation clinical trials, which involves concurrently assigning patients to doses that have been previously cleared for safety by the dose-escalation design. Backfilling generates additional information on safety, tolerability, and preliminary activity on a range of doses below the maximum tolerated dose (MTD), which is relevant for selection of the recommended phase II dose and dose optimization. Purpose: The neutralizing peptibody trebananib prevents angiopoietin-1 and angiopoietin-2 from binding with Tie2 receptors, inhibiting angiogenesis and proliferation. Trebananib was combined with paclitaxel±trastuzumab in the I-SPY2 breast cancer trial.

Patients and methods: I-SPY2, a phase II neoadjuvant trial, adaptively randomizes patients with high-risk, early-stage breast cancer to one of several experimental therapies or control based on receptor subtypes as defined by hormone receptor (HR) and HER2 status and MammaPrint risk (MP1, MP2). The primary endpoint is pathologic complete response (pCR). A therapy "graduates" if/when it achieves 85% Bayesian probability of success in a phase III trial within a given subtype. …

Early-stage lung adenocarcinoma (LUAD) patients remain at substantial risk for recurrence and disease-related death, highlighting the unmet need of biomarkers for the assessment and identification of those in an early stage who would likely benefit from adjuvant chemotherapy. To identify circulating miRNAs useful for predicting recurrence in early-stage LUAD, we performed miRNA microarray analysis with pools of pretreatment plasma samples from patients with stage I LUAD who developed recurrence or remained recurrence-free during the follow-up period. PURPOSE: Camonsertib is a highly selective and potent inhibitor of ataxia telangiectasia and Rad3-related (ATR) kinase. Dose-dependent anemia is a class-related on-target adverse event often requiring dose modifications. Individual patient risk factors for the development of significant anemia complicate the selection of a "one-size-fits-all" ATR inhibitor (ATRi) dose and schedule, possibly leading to suboptimal therapeutic doses in patients at low risk of anemia. We evaluated whether early predictors of anemia could be identified to ultimately inform a personalized dose-modification approach.

Although the number of Nigerian immigrants to the United States has increased in the past few years, few studies have explored their mental health experiences. This study explored the lived experience of Nigerian immigrants living in the United States (US). Belief perseverance theory was utilized as a lens of analysis to understand if prior experiences, perceptions, and/or stigma can influence help-seeking behavior and expectations through face-to-face, semi-structured, and audiotaped interviews. The study explored a sample of 10 Nigerian immigrants lived experiences, understanding, perceptions, and stigma of mental illness and whether or not they seek psychological help.
