Liver fibrosis occurs due to injury or inflammation, which results in the excessive production of collagen and the formation of fibrotic scar tissue that impairs liver function. Despite the limited treatment options available, freshwater clams may hold promise in the treatment of liver fibrosis. In this study, we demonstrated the effects of ethanol extract of freshwater clam (FCE), ethyl acetate extract of FCE (EA-FCE), and trans-2-nonadecyl-4-(hydroxymethyl)-1,3-dioxolane (TNHD) on liver fibrosis induced by dimethylnitrosamine (DMN). Administration of FCE and TNHD alleviated liver injury, including tissue damage, necrosis, inflammation scores, fibrosis scores, serum enzymes, and triglyceride levels. The present research examined the levels of lead, cadmium, copper and zinc in water, sediments, and some vital organs of Solea solea fish from Lake Qaroun, Egypt, such as liver, gills, kidney, muscle and end of the alimentary. The contamination factor (CF) values of Pb, Cd, Cu, and Zn are low contamination which CF ˂ 1. Mycobacterium tuberculosis (Mtb)-specific γ9δ2 T cells secrete granzyme A (GzmA) protective against intracellular Mtb growth. However, GzmA-enzymatic activity is unnecessary for pathogen inhibition, and the mechanisms of GzmA-mediated protection remain unknown. We show that GzmA homodimerization is essential for opsonization of mycobacteria, altered uptake into human monocytes, and subsequent pathogen clearance within the phagolysosome. Although monomeric and homodimeric GzmA bind mycobacteria, only homodimers also bind cluster of differentiation 14 (CD14) and Toll-like receptor 4 (TLR4). Without access to surface-expressed CD14 and TLR4, GzmA fails to inhibit intracellular Mtb. Upregulation of Rab11FIP1 was associated with inhibitory activity. Furthermore, GzmA colocalized with …

The Austroasiatic (AA) languages are a large language family in Mainland Southeast and South Asia. Theoretical, methodological, and material constraints have limited research on the origin and dispersal of AA-speaking populations within historical-comparative linguistics. With the deepening of archaeological and genetic studies, interdisciplinary collaboration has become the key to solving this problem. Based on the latest achievements of linguistics, archaeology and molecular anthropology, we review the hypotheses and propose insights on the origin and dispersal of AA languages. Understanding gene variations in people living under extreme conditions has the potential of curing diseases caused by exposure to heat, cold, fatty diets, hypoxia, and pathogens. One candidate gene associated with heat resistance is ACE1, encoding angiotensin-converting enzyme 1. Associations have also been made between cold resistance or fatty diets and polymorphisms of several genes including ACTN3, encoding alpha-actinin-3, and CPTIA, encoding carnitine palmitoyltransferase 1A. A prominent role in resistance to hypoxia has been given for polymorphisms of EPAS1, encoding endothelial PAS domain protein 1, and EGLN1, encoding Egl-9 family hypoxia inducible factor 1. As the volume and diversity of Internet-of-Things (IoT) data continues to grow, traditional cloud-based processing methods face significant challenges, including latency, bandwidth constraints, and privacy concerns. Our research focuses on employing decision trees (DTs) as an intelligent filtering mechanism on the edge. Preliminary comparisons across four datasets revealed DTs are significantly more efficient than multilayer perceptrons (MLP), saving 97-98\% in area and power, leading to the selection of DT for our proposed architecture for lightweight IoT devices. Managing wildlife at airports poses a challenge for ensuring safety for aviation traffic. Airports, with their surrounding habitats and infrastructure, serve as attractants for avian species. Threats created by these species have led managers to develop management techniques in and around airports (DeVault et al 2013). One approach is the use of geographic information systems (GIS) for analyzing avian strike data and airport bird surveys. Spatial analysis of strike data identifies species of threat by categorizing strike occurrence, species sizes, phase of flight, and risk of damage. Unbiased stereology refers to a field of applied mathematics \cite{intro-to-stereology} focused on accurate (model and assumption-free) quantification of three-dimensional (3D) objects, typically based on their appearance in 2D sections (planes) through the objects. In the biological sciences, these techniques are widely used for making unbiased estimates of arbitrary-shaped (stochastic) objects such as stained cells, blood vessels, region volumes, etc., in tissue sections through a region of interest (ROI).

The Supreme Court of the United States decision on race-based affirmative action policies in higher education has impacted equity, diversity and inclusion (EDI) programs in higher education institutions (Gonzales, 2023; Singh, 2023; Opie & Washington, 2023). This study explores the challenges higher education institutions face when communicating and implementing EDI programs. Almost two million Americans are incarcerated in state and federal prisons, juvenile corrections facilities, and local jails. While many studies have examined the impact of mass incarceration on the daily lives of wives, female partners, and children, little is known about support groups for families of incarcerated loved ones. Throughout 2021, I conducted virtual interviews with 13 respondents across the United States who created support groups for families of incarcerated individuals or are executive leaders in these organizations. El retamo espinoso ( Ulex europaeus L.), una planta europea altamente agresiva en Colombia, afecta los páramos y desplaza especies nativas, generando preocupación ecológica. Este estudio investiga su biología, a partir de un banco de semillas (113 semillas/m² a 5 cm), acumulación de biomasa (0,006732 g/día en 481 días), fenología en estado juvenil durante 378 días sin floración, y resultados preliminares del uso de herbicidas en los tallos. Estos datos iniciales son cruciales para desarrollar métodos de control, abordando disciplinas como ecología, fenología, hábitat, herbicidas, malezas, biodiversidad y bioseguridad.

This paper explores the potential impact of exercise timing on appetite hormones and metabolic outcomes, aiming to enhance understanding of the complex interplay between exercise, appetite regulation, and weight management. A randomized trial involving premenopausal women investigated the effects of morning versus evening exercise sessions over an eight-week intervention period. Participants underwent comprehensive screening and a supervised exercise regimen, with pre-and post-training laboratory sessions assessing body composition and appetite hormone levels. Statistical analyses revealed no significant differences in appetite hormone levels between morning and evening exercise groups, but notable correlations were observed between appetite hormones and body composition measures. The Precision Nitrogen Project (PNP) worked with more than 80 corn and winter wheat producers to inexpensively design and implement randomized, replicated field strip trials on whole commercial farm fields, and to provide site-specific testing of current nitrogen (N) technologies. This article proposes a conceptual framework and detailed procedure to select the N technology to be tested; design and implement field trials; generate, process, and manage field trial data; and automatically analyze, report, and share benefits from precision N technology. Tissue-resident macrophages (TRMs) are abundant immune cells within pre-metastatic sites, yet their functional contributions to metastasis remain incompletely understood. Here, we show that alveolar macrophages (AMs), the main TRMs of the lung, are susceptible to downregulation of the immune stimulatory transcription factor IRF8, impairing anti-metastatic activity in models of metastatic breast cancer. G-CSF is a key tumor-associated factor (TAF) that acts upon AMs to reduce IRF8 levels and facilitate metastasis. BACKGROUND: Immune-checkpoint inhibitors (ICIs) have showed unprecedent efficacy in the treatment of patients with advanced non-small cell lung cancer (NSCLC). However, not all patients manifest clinical benefit due to the lack of reliable predictive biomarkers. We showed preliminary data on the predictive role of the combination of radiomics and plasma extracellular vesicle (EV) PD-L1 to predict durable response to ICIs.

BACKGROUND. Vaccination is typically administered without regard to site of prior vaccination, but this factor may substantially affect downstream immune responses. METHODS. We assessed serological responses to initial COVID-19 vaccination in baseline seronegative adults who received second-dose boosters in the ipsilateral or contralateral arm relative to initial vaccination. We measured serum SARSCoV-2 spike–specific Ig, receptor-binding domain–specific (RBD-specific) IgG, SARS-CoV-2 nucleocapsid–specific IgG, and neutralizing antibody titers against SARS-CoV-2.D614G (early strain) and SARS-CoV-2.B.1.1.529 (Omicron) at approximately 0.6, 8, and 14 months after boosting. This project aims to investigate the DNA fragmentation produced by enzymatic bisulfite conversion compared to DNA fragmentation produced by traditional chemical bisulfite conversion techniques for the analysis of cytosine methylation in seminal cell-free DNA (cfDNA). Traditional chemical bisulfite (HSO3-) conversion methods are known to introduce biases and damage DNA, potentially compromising methylation analysis accuracy. In contrast, enzymatic conversion, utilizing biological molecules, is hypothesized to offer more precise visualization with reduced fragmentation (Lambert et al., 2019). While previous studies have analyzed methylation conversion efficiency in genomic DNA, this project specifically examines how these conversion techniques impact cfDNA.

Neural tube defects (NTDs), including anencephaly and spina bifida, contribute significantly to neonatal morbidity and mortality worldwide. Despite the widespread implementation of folic acid (FA) supplementation, its ineffectiveness in certain genetic populations, alongside the persistent prevalence of NTDs, underscores the need for alternative preventative strategies. This study investigates the potential of isopropyl isothiocyanate (IPI), a compound derived from Moringa oleifera (M. oleifera), in mitigating NTDs through the modulation of oxidative stress pathways. In a previous report, keratinocytes were shown to share their gene expression profile with surrounding Langerhans cells (LCs), influencing LC biology. Here, we investigated whether transferred material could substitute for lost gene products in cells subjected to Cre/Lox conditional gene deletion. We found that in human Langerin-Cre mice, epidermal LCs and CD11b+CD103+ mesenteric DCs overcome gene deletion if the deleted gene was expressed by neighboring cells. The mechanism of material transfer differed from traditional antigen uptake routes, relying on calcium and PI3K, being susceptible to polyguanylic acid inhibition, and remaining unaffected by inflammation. BACKGROUND: Increased inflammation caused by SARS-CoV-2 infection can lead to severe coronavirus disease 2019 (COVID-19) and long-term disease manifestations. The mechanisms of this variable long-term immune activation are poorly defined. One feature of this increased inflammation is elevated levels of proinflammatory cytokines and chemokines. Autoantibodies targeting immune factors such as cytokines, as well as the viral host cell receptor, angiotensin-converting enzyme 2 (ACE2), have been observed after SARS-CoV-2 infection. Autoantibodies to immune factors and ACE2 could interfere with normal immune regulation and lead to increased inflammation, severe COVID-19, and long-term complications.

Transgender and nonbinary youth are under legislative and political siege as the latest victims in our nation’s culture wars. They are acutely aware of the hostility towards their existence and best interests, damaging their often already precarious well-being. There is a concerning risk they will associate biased and antagonistic lawmakers with our entire legal system, including legal service providers. Fear of encountering discrimination and bias leads targeted individuals to avoid accessing services. I fear that means too many among this generation transgender and nonbinary youth may avoid addressing their legal health needs as they age.

Purpose: The lungs are the most common site of metastasis for patients with soft tissue sarcoma. SABR is commonly employed to treat lung metastases among select patients with sarcoma with limited disease burden. We sought to evaluate outcomes and patterns of failure among patients with sarcoma treated with SABR for their lung metastases.
Methods and materials: We performed a retrospective review of patients treated at a tertiary cancer center between 2006 and 2020. Purpose: Increased glucocorticoid receptor (GR) signaling is a proposed compensatory mechanism of resistance to androgen receptor (AR) inhibition in metastatic castration-resistant prostate cancer (mCRPC). ORIC-101 is a potent and selective orally-bioavailable GR antagonist.
Patients and methods: Safety, pharmacokinetic/pharmacodynamic, and antitumor activity of ORIC-101 in combination with enzalutamide were studied in patients with mCRPC progressing on enzalutamide. ORIC-101 doses ranging from 80 to 240 mg once daily were tested in combination with enzalutamide 160 mg once daily. Background: Follicular lymphoma (FL) and marginal zone lymphoma (MZL) are indolent non-Hodgkin lymphomas (iNHL). Median survival for iNHL is approximately 20 years. Because standard treatments are not curative, patients often receive multiple lines of therapy with associated toxicity-rationally designed, combination therapies with curative potential are needed. The immunomodulatory drug lenalidomide was evaluated in combination with rituximab for the frontline treatment of FL in the phase 3 RELEVANCE study. Ibrutinib, an oral Bruton tyrosine kinase inhibitor, is active in NHL and was evaluated in combination with lenalidomide, rituximab, and ibrutinib (IRR) in a phase 1 study.
