We annotated three newly discovered bacteriophages. TukTuk and Shamu were isolated on the host Microbacterium folorium and Megatron06 on Mycobacterium smegmatis. Based on based on gene-content similarity (GCS) of 35% or higher to sequenced bacteriophages present in the Actinobacteriophage database, phagesDB, TukTuk was assigned to cluster EB, Shamu to cluster EA, and Megatron06 to Cluster H1. Here we report a summary of our annotation findings along with one in-depth analysis of an aspect of our annotation for each phage.

Cardiovascular disease, chronic kidney disease, diabetes, and various mental health conditions have been shown to be associated with gut microbiota alterations that may contribute to the development and pathogenesis of those diseases. Cardiovascular disease, inflammatory bowel disease and diabetes have been associated with a lack of butyrate- producing bacteria which are responsible for lowering blood pressure, limiting atherosclerosis, abdominal pain, and lowering insulin resistance. Medications for individuals with schizophrenia may decrease bacteria with anti-inflammatory properties. Potential research avenues may be directed at investigating the di@erences between gut microbiomes between men and women to cultivate unique and targeted treatment and prevention …

Bacteriophages are viruses that infect bacteria and can be used in phage therapy to combat bacterial infections resistant to traditional antibiotics. One current limitation of phage therapy is the lack of understanding about phage genetics and biochemistry. We can learn more about bacteriophages by identifying patterns in their protein structure. In our experiments, we immunized BALB/c mice using whole phage or ~71>kDa proteins extracted from Sbash and Island3. We tail-bled the mice to collect antibodies raised against these phages, then used ELISA assays to detect anti-phage protein antibodies in mouse serum. Perineal tearing is one of the most common injuries that can be sustained during vaginal delivery, it can be physically and mentally debilitating for the mother to endure. Preventing this injury can lead to an easier healing process for the mother, allowing her to have a more impactful postpartum journey. There is debate as to whether warm compresses, perineal massage, and alternative birthing positions decrease the incidence of perineal tearing. This review of literature focuses on how these interventions work and if they are effective at decreasing the risk for perineal tearing during vaginal delivery.

Sbash is a temperate bacteriophage, which was isolated on the host, Mycobacterium smegmatis mc2 155 from soil collected in South Africa. It is classified as cluster I and sub-cluster I2. Its genome consists of 55,832 base pairs and 89 protein-coding genes of which only 25 genes were assigned a function by bioinformatic analysis. We are using a genetic screen to uncover the functions of phage genes for which function is unknown. Background: In Taiwan, there were 7,038 reports of attempted suicide among people aged 24 or younger in 2018. Anxiety disorders are the most common mental illness in the U.S., affecting 40 million adults in the United States age 18 and older.

Purpose: In this study, we compared United States (US) and Taiwanese college students’ spiritual well-being, psychological well-being, and suicidal ideation.

Methods: A total of 661 college students (481 from the US and 180 from Taiwan) were recruited to complete the survey study. This was a cross-sectional comparative research design. SPSS 28.0 was used to do the data analysis. Chi-square …

TRAAK, TREK-1, and TREK-2 are mechanosensitive two-pore domain K+ (K2P) channels that contribute to action potential propagation, sensory transduction, and muscle contraction. While structural and functional studies have led to models that explain their mechanosensitivity, we lack a quantitative understanding of channel activation by membrane tension. Here, we define the tension response of mechanosensitive K2Ps using patch-clamp recording and imaging. All are low-threshold mechanosensitive channels (T10%/50% 0.6-2.7 / 4.4-6.4 mN/m) with distinct response profiles. TRAAK is most sensitive, TREK-1 intermediate, and TREK-2 least sensitive. Cannabis use disorder (CUD) is increasingly prevalent in the United States, but there is no effective pharmacological means to treat it. The endocannabinoid (eCB) system has emerged as a candidate therapeutic target demonstrating some evidence of efficacy in treating CUD. However, clinical trials evaluating eCB-modulating therapeutics have historically undervalued individual differences that could contribute to variation in treatment outcome (e.g. sex, comorbid psychiatric illness). Conifer forest resilience may be threatened by increasing wildfire activity and compound disturbances in western North America. Fire refugia enhance forest resilience, yet may decline over time due to delayed mortality—a process that remains poorly understood at landscape and regional scales. To address this uncertainty, we used high-resolution satellite imagery (5-m pixel) to map and quantify delayed mortality of conifer tree cover between 1 and 5 years postfire, across 30 large wildfires that burned within three montane ecoregions in the western United States. Literature on traumatic events, particularly those of a sexual nature, suggests that these events have an impact on one’s wellbeing and ability to parent. However, limited literature centers a women’s experience, with little attention focused on identity formation, the role of mothering, and healing as an outcome (Babcock et al., 2015; Clifford et al., 2020. Type 1 diabetes (T1D) is a life-threatening autoimmune disease where the immune system destroys the insulin producing beta-cells in the pancreas. There is no cure for the disease, and the current treatment involves life-long, daily injection of insulin. An ideal therapy for T1D will be the suppression of autoimmunity and increased functional beta-cell mass. The immune regulatory cytokine IL10 can suppress inflammation in various autoimmune diseases. On the other hand, several growth factors and peptide hormones are known to promote an increase in beta-cell mass, through neogenesis and beta-cell proliferation. Background: Stemming from poverty and systemic racism, Black youth are disproportionately represented in hospital-based violence intervention programs (HVIPs) due to greater violence exposure. HVIPs are a critical intervention that have been shown to reduce rates of reinjury in urban hospitals and trauma centers across the United States; however, they are plagued by low enrollment and engagement rates. Few studies have examined factors related to engagement, particularly among Black youth. Methods: Guided by Trauma Theory and Critical Race Theory, this study uses a retrospective cohort design. While we recognize the prognostic importance of clinicopathological measures and circulating tumor DNA (ctDNA), the independent contribution of quantitative image markers to prognosis in non-small cell lung cancer (NSCLC) remains underexplored. In our multi-institutional study of 394 NSCLC patients, we utilize pre-treatment computed tomography (CT) and 18F-fluorodeoxyglucose positron emission tomography (FDG-PET) to establish a habitat imaging framework for assessing regional heterogeneity within individual tumors. This framework identifies three PET/CT subtypes, which maintain prognostic value after adjusting for clinicopathologic risk factors including tumor volume. Juvenile idiopathic arthritis (JIA) is a complex rheumatic disease encompassing several clinically defined subtypes of varying severity. The etiology of JIA remains largely unknown, but genome-wide association studies (GWASs) have identified up to 22 genes associated with JIA susceptibility, including a well-established association with HLA-DRB1. Continued investigation of heritable risk factors has been hindered by disease heterogeneity and low disease prevalence. In this study, we utilized shared genomic segments (SGS) analysis on whole-genome sequencing of 40 cases from 12 multi-generational pedigrees significantly enriched for JIA. Functional medicine is a personalized and holistic approach to treating chronic disease. In this article, I build upon posthumanist literature by examing how functional medicine practitioners are decentering and destabilizing what it means to be human. Functional medicine discourse on the holobiont, which considers the human as an assemblage of different microbial species, reframes the “humananimal” (see Nayar 2018) as the “humicrobe.” I engage Gaia 2.0 (see Lenton and Latour 2018) when analyzing the interconnectivity, interdependence, and mutualism of all life. Gut inflammation involves contributions from immune and non-immune cells, whose interactions are shaped by the spatial organization of the healthy gut and its remodeling during inflammation. The crosstalk between fibroblasts and immune cells is an important axis in this process, but our understanding has been challenged by incomplete cell-type definition and biogeography. To address this challenge, we used multiplexed error-robust fluorescence in situ hybridization (MERFISH) to profile the expression of 940 genes in 1.35 million cells imaged across the onset and recovery from a mouse colitis model. Seminoles, Soldiers, And Settlers: Identity And Power On The Florida Frontier, Jean Louise Lammie

This project uses an historical archaeological approach to analyze the processes of cultural negotiation between disparate social groups living in a remote borderland in the early to mid-nineteenth centuries. Florida, first colonized by the Spanish in the 1500s was passed to the United States in 1821 as part of the young nation’s nascent Manifest Destiny. The Seminoles experienced multiple colonial conflicts, dealt with new diseases, and selected aspects of European culture to incorporate into their daily practices of lifeways.
