Glioblastoma Multiforme (GBM) is an aggressive cancer with patients surviving 13 months on average after diagnosis. The PI3K-PKB/AKT pathway is conserved from flies to mammals. The PI3K signaling pathway aids in the growth and survival of tumor cells. Forkhead box transcription factors subclass O (FOXO1, FOXO3, and FOXO4) act as tumor suppressors in some contexts. In GBM, our group and others showed that FOXO transcription factors drive stem gene expression to promote growth and survival. We disrupted the FOXO4 gene in U87MG cells using CRISPR Cas9 genome editing and performed RNA-seq to identify targets of this …

The application of environmental DNA (eDNA) has improved the ability of humans to detect the existence and geographical distribution of aquatic organisms. The present study highlights the development and application of eDNA techniques to detect rough-footed mud turtles and yellow mud turtles in different areas in Texas. Both mud turtle species were successfully detected in lakes/resacas using direct eDNA sampling techniques. Moreover, passive eDNA sampling techniques were conducted using different membrane filters to detect mud turtle species. The findings indicated that only the mixed cellulose esters (MCE) filter membranes effectively identified eDNA of yellow mud turtles in the fall, but …

Combination antiretroviral therapy (cART) is widely used to treat HIV infection, but it fails to eradicate viral reservoirs, such as microglia in the brain, leading to HIV-associated neurological disorders (HAND). Advances in nanomedicine have enabled the development of targeted drug delivery systems, and nanodiamonds (NDs) and fluorescent nanodiamonds (FNDs) have emerged as a potential solution due to their biocompatibility, non-toxic nature, and efficiency as drug carriers. As global energy demands have risen in the past decades, various species of microalgae have been chosen and used as sources of lipids for third-generation biofuel. I selected two species of green microalgae, Bracteacoccus sp. and Tetrastrum sp., from isolates from South Texas to determine their potential as sources of biofuel. For comparison, Chlamydomonas reinhardtii was used as a model organism. Affectionate behaviors have biological and physiological implications for humans. Affectionate communication encompasses behaviors that convey affection to the senders and receivers. The purpose of this qualitative study was to investigate the role of culture and cultural traditions in the expression of affectionate behaviors among members of the Nigerian Yoruba family. Two focus groups were conducted in which participants (18-39 years of age) identified the affectionate behaviors they use in their families. The objective of this research is to establish a fundamental approach to tuning PID (Proportional-Integral-Derivative) parameters for a simulated quadrotor drone. Implementing a PID controller for autonomous flight provides a straightforward and efficient method for monitoring and correcting robotic movement based on the robot's current state. However, applying a PID approach to a quadrotor's flight controller poses challenges, such as assigning multiple parameters to control an inherently under-actuated system. This includes the need to find optimal parameter values that reduce the likelihood of large overshoots and lengthy adjustment times. This thesis delves into the security of stochastic pheromone-based foraging algorithms within swarm robotic systems, a subset of foraging algorithms distinguished by their reliance on probabilistic decision-making mechanisms inspired by the natural world. Such algorithms face vulnerabilities in stigmergic communication that threaten to disrupt swarm operations. This research investigates these vulnerabilities, presenting two distinct contributions.

This interpretive phenomenological study addresses the underrepresentation of Latinas in higher education leadership roles by focusing on the lived experiences of Latina poets who have served as higher education leaders. Data, collected through semi-structured participant interviews and participant writing samples, was analyzed through a standard method of thematic analysis. Latin American and U.S. theories of decoloniality and Yosso’s community cultural capital guided interpretation of participant experiences. Five themes emerged from the analysis: border negation, foundational values, othered leadership, importance of community, and writing as necessity. The Generalized Poisson distribution is useful in modeling epidemiological processes as a branching stochastic processes problem. Our goal is to construct accurate and reliable estimators for the reproduction number (R0) (i.e., the number of secondary infections), particularly in the context of disease outbreaks modeled by a Galton-Watson process. Towards this goal, we construct the classical Bayes estimator, the Maximum Likelihood estimator, and the Empirical Bayes (EB) estimator under the Square Error Loss function in Chapter II. Soil organic carbon (SOC) plays a vital role in the global carbon cycle and aids in climate change mitigation. However, deforestation and intensive agricultural practices threaten topsoil and carbon (C) pools through soil disturbances releasing vast amounts of carbon dioxide (CO2) by enhancing the decomposition of organic materials. Degraded soils typically lack stable aggregates, nutrient and water retention, and overall low fertility. This paper is about leveraging cultural ways of knowing such as ethnobotany in order to increase STEM engagement in Chicanx populations. The conceptual framework utilized in this study combines social justice, Chicana feminist epistemology, land-based pedagogy, critical place-based pedagogy, pedagogical praxis, pluralism and ecological sustainability to achieve this end. Four pre-service Chicana science teachers provided testimonio from their elder assignment and engaged in two focus groups in order to identify relevant themes that impact Chicana engagement in STEM. There has been a growing interest in organometallic chemistry to utilize late Earth-abundant metals towards challenging molecular functionalization of abundant feedstocks to value-added products. There is precedent of utilizing cooperativity to try and address some of the shortcomings of traditional mononuclear complexes by integrating metal-metal and or metal-ligand cooperativity to facilitate multi-electron transformations. The utility of a redox-noninnocent scaffold allows for the metal centers to be stable over a range of oxidation states, using the ligand to store electrons. Limited research exists about the experiences of nurses’ caring for patients during the COVID-19 pandemic post hoc. To understand nurses’ realities of caring for patients with COVID-19, I aimed to understand the lived experience of nurses caring for patients with COVID-19 in the United States. A qualitative design with a hermeneutic phenomenological methodology was used. Sixteen participants were recruited via purposive sampling, augmented with snowball sampling. Opioid use disorder (OUD) impacts 5.6 million people in the US. Buprenorphine (BUP) is a commonly prescribed opioid medication used to treat OUD, including in pregnant women. However, opioid use during pregnancy is associated with poorer infant outcomes including reduced fetal growth, neurodevelopmental deficits, and neonatal opioid withdrawal syndrome (NOWS). Recent clinical data suggests that providing mothers with a lower dose of BUP may result in fewer negative outcomes in infants. Here, a preclinical rodent model of low-dose perinatal BUP exposure was used to study offspring health outcomes in the neonate, juvenile, and adolescent offspring. Urbanization is an intensive form of habitat disturbance associated with detrimental effects on biodiversity. However, few studies have investigated its effects on the number, identity and structure of species interactions while considering seasonal fluctuations in communities. Avian seed dispersal is a vital ecosystem service, and the interplay of urbanization and seasonality may impact seed dispersal in ways not predicted by either factor alone. Land cover conversion has resulted in severe habitat fragmentation, degradation, and loss in bottomland hardwood (BLH) forests throughout the Lower Mississippi Alluvial Valley. BLH forests in southern Illinois are home to diverse wildlife communities, many of which are of conservation concern and utilize BLH habitat year-round or during migration and/or breeding. Yersinia pestis causes the human disease known as plague. A key manifestation of plague is a delayed inflammatory response. Because this delay in inflammation is required for virulence, I was interested in defining the molecular mechanisms used by Y. pestis to evade immune recognition. Eicosanoids are produced early during infection and necessary to initiate a rapid inflammatory response. Despite the importance of these lipids in mediating inflammation, the role of eicosanoids during plague has not been previously investigated. The central goal of my dissertation is to better understand undergraduate biology students’ perceptions of climate change specifically using a localized lens. Research relating to climate change perceptions and understanding is common in the K-12 setting; however, there have been far fewer studies that use undergraduate student populations.
