Assessing Health Implications Of Monarch Butterfly Populations And Behaviors, Kira Goldman, Kelsey E. Fisher Dr. May 2024

Assessing Health Implications Of Monarch Butterfly Populations And Behaviors, Kira Goldman, Kelsey E. Fisher Dr.

Honors Scholar Theses

Pollinator populations have seen massive declines over the past decade due to anthropogenic activity and climate change posing serious threats to the health of humans and ecosystems (Ulyshen & Horn, 2023). As agents responsible for the production of about 35% of micronutrient rich food crops necessary for adequate nutrition to lead healthful lives and prevent nutrition associated diseases, pollinator decline is a public health concern (Aizen et al., 2009). Monarch butterflies (Danaus plexippus) serve as bio indicators for the health of pollinator populations because of their analogous habitat and food resources shared with many other pollinator species, including bees and …

Understanding The Role Of Minor Intron Splicing In Spermatogenesis, Jade Rosado, Kazumasa Takemoto, Rahul Kanadia May 2024

Understanding The Role Of Minor Intron Splicing In Spermatogenesis, Jade Rosado, Kazumasa Takemoto, Rahul Kanadia

Honors Scholar Theses

The minor spliceosome is composed of the unique small nuclear RNAs (snRNA), U11, U12, U4atac, and U6atac, and is necessary for the splicing of less than 0.5% of introns, termed minor introns. Minor intron containing genes (MIGs) regulate diverse processes, one of which is spermatogenesis. Specifically, 111 testis-specific MIGs have been identified, suggesting that minor splicing is necessary for spermatogenesis. To interrogate the role of minor splicing in spermatogenesis, we conditionally ablated the Rnu11 gene, which encodes for the U11 snRNA, in the developing testes via Stra8-iCre, creating a Rnu11Flx/Flx::Stra8-Cre+ mutant. We found, at postnatal …

Analysis Of Antibiotic Sensitivity Patterns In Pasteurellaceae Family Isolates: A Microbial And Molecular Investigation, Jillian Barron

Honors Scholar Theses

Bacteria in the Pasteurellaceae family are known to cause disease in both human and animal species. Like all species of bacteria, the pathogens in the Pasteurellaceae family evolve and acquire antimicrobial-resistant traits. Antimicrobial resistance of bacteria is a significant public health concern, as treatment of resistant pathogens becomes more difficult, if not impossible, with current medical capabilities. In this project, phenotypic and genotypic analyses were performed on Pasteurellaceae family isolates sourced from the Connecticut Veterinary Medical Diagnostic Laboratory. Phenotypic antibiotic sensitivity patterns of each isolate were assessed against a panel of antibiotics through a Kirby-Bauer disk diffusion test. Disk diffusion …

The Association Between Aerobic Exercise And Changes In Hdl Quality Induced By Egg Consumption – A Post-Hoc Analysis Of A Randomized, Controlled, Crossover Dietary Intervention Study, Rachael Woodruff, Catherine J. Andersen

Honors Scholar Theses

Abstract

Objective: Daily intake of whole eggs and aerobic exercise have independently been shown to increase high-density lipoprotein cholesterol (HDL-C) and measures of HDL function, suggesting cardioprotective effects of these lifestyle interventions. However, it is unknown whether exercise volume can synergistically or differentially impact diet responses and egg effects on HDL. Therefore, we evaluated whether exercise volume was associated with distinct effects of consuming different egg-based diets on measures of HDL size, function, and composition.

Methods: Twenty-six healthy men and women completed a 16-week randomized, controlled, crossover intervention study which aimed to investigate the effects of egg-free, egg-white, and whole-egg …

Archaeal Diversity In The Anna's Hummingbird Microbiome, Lauren E. Chance May 2024

Archaeal Diversity In The Anna's Hummingbird Microbiome, Lauren E. Chance

Honors Scholar Theses

The microbial communities that are present in and on vertebrates are collectively called the microbiome. The composition of a microbiome is dependent upon the host, the environment, and evolution. There has been extensive research on the bacterial composition of host-associated microbiomes, however, there has been much less work on the archaeal composition of host-associated microbiomes. Archaea have previously been assumed to primarily exist in extreme environments, but this may not be true and has been influenced by their generally low abundance and methodological difficulties in detection. It is possible they are consistent members of diverse host-associated microbiomes.

Archaea-specific PCR primers …

Investigating Effects Of Co-Culturing With Vibrio Fischeri On Secondary Metabolite Production Of Hawaiian Bobtail Squid Symbionts, Mariam A. Zedan May 2024

Investigating Effects Of Co-Culturing With Vibrio Fischeri On Secondary Metabolite Production Of Hawaiian Bobtail Squid Symbionts, Mariam A. Zedan

Honors Scholar Theses

Microbes are a fruitful source of natural product drug discovery since many of their secondary metabolites show defensive activity and have unique structures. However, extracting and elucidating the structure of those compounds can be challenging in vitro due to particular environmental and nutritional requirements of some bacteria. Those requirements may include proximity to other organisms that the microbes have symbiotic relationships with. Under those conditions, the organism may activate silent biosynthetic gene clusters (BGCs) responsible for production of certain secondary metabolites not typically expressed otherwise. One method of BGC induction is co-culturing with other microbes since this mimics the natural …

Mutational Landscape Of Noncoding Ultraconserved Elements In Human Cancers: Functional Relevance And Clinical Utility, Recep Bayraktar

Dissertations and Theses (Open Access)

The mutational landscape of phylogenetically ultraconserved elements (UCEs), especially those in noncoding DNA regions (ncUCEs), and their functional and clinical relevance in cancers remain poorly characterized. In this thesis, we perform a systematic analysis of whole-genome sequencing (WGS) data from the International Cancer Genome Consortium (ICGC) and in-house targeted sequencing of 13,736 UCEs and demonstrate that ncUCE somatic alterations are pervasive. Using a genome-wide multiplexed CRISPR knockout screen in colorectal cancer (CRC) cell lines, we show that the loss of several mutated ncUCEs impacts cancer cell proliferation. In-depth functional studies in vitro and in vivo further reveal that UCE_11311 is …

Chemistry Faculty Publications and Presentations

Fluorescent gold nanoclusters (AuNCs) have shown promise as metal ion sensors. Further research into surface ligands is crucial for developing sensors that are both selective and sensitive. Here, we designed simple tripeptides to form fluorescent AuNCs, capitalizing on tyrosine’s reduction capability under alkaline conditions. We investigated tyrosine’s role in both forming AuNCs and sensing metal ions. Two tripeptides, tyrosine–cysteine–tyrosine (YCY) and serine–cysteine–tyrosine (SCY), were used to form AuNCs. YCY peptides produced AuNCs with blue and red fluorescence, while SCY peptides produced blue-emitting AuNCs. The blue fluorescence of YCY- and SCY-AuNCs was selectively quenched by Fe3+ and Cu2+, whereas red-emitting YCY-AuNC …

A Fish Out Of Water: An Examination Of Self-Identified Introverted Business Students' Perceptions Of Their Leadership Ability, Katie J. Jolicoeur

Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–

Current American cultural and societal ideals favor personalities that are outgoing, positive, and talkative in nature, due to the rise of the Culture of Personality (Wilmot, 2019; Kim, 2012; Cain, 2013). Beginning with the industrial revolution and expanding rapidly midcentury, parents began to view children who are quiet as less favorable than those who are more gregarious, thus children and parents began forced socialization at an earlier age (Wilmot, 2019; Cain, 2013). Very little, if any, research has been conducted to understand the impact that the cultural ideal of favoring extraversion has upon undergraduate students seeking a degree in business, …

Artificial Intelligence (AI) has advanced rapidly in the past two decades. Internet of Things (IoT) technology has advanced rapidly during the last decade. Merging these two technologies has immense potential in several industries, including agriculture.

We have identified several research gaps in utilizing IoT technology in agriculture. One problem was the digital divide between rural, unconnected, or limited connected areas and urban areas for utilizing images for decision-making, which has advanced with the growth of AI. Another area for improvement was the farmers' demotivation to use in-situ soil moisture sensors for irrigation decision-making due to inherited installation difficulties. As Nebraska …

Vibratory Noise Impacts The Sensory Ecology Of A Funnel-Weaving Spider, Brandi June Pessman May 2024

Vibratory Noise Impacts The Sensory Ecology Of A Funnel-Weaving Spider, Brandi June Pessman

Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–

Animals experience their world through diverse sensory systems, leveraging information from prey, predators, and potential mates to inform behavioral decisions. Environmental noise disrupts information via spectral or amplitude masking, degradation, or distortion, potentially leading to costly errors in responses. Critically, urbanization has accelerated the proliferation of novel noise sources, triggering significant shifts in communication, foraging, and predator-prey dynamics among urban wildlife. While vertebrate studies have illuminated urban animals’ flexibility to anthropogenic acoustic noise, a comprehensive understanding requires broader investigations across diverse animal taxa and sensory modalities. To address these gaps, my dissertation explored the impacts of anthropogenic substrate-borne (vibratory) noise …

Sex education in schools is a pivotal yet contentious aspect of adolescent development, with enduring impacts on individuals' sexual exploration and liberation. However, the types of sex education offered have varied over time, often disseminating misinformation, and reinforcing traditional gender norms. Particularly for women, sex education can perpetuate harmful stereotypes, limit sexual agency, and contribute to disparities in sexual and reproductive health. This qualitative study aims to address the gap of intersecting sex education and sexual identity by examining the relationship between women's sexual liberation and exploration in adulthood and the sex education they received. Drawing from a phenomenological approach, …

The Sexual Trauma Of The Female Body: Violence Against Women In Contemporary Spanish And Latin American Cultures, Lara Armenteros Garrido May 2024

The Sexual Trauma Of The Female Body: Violence Against Women In Contemporary Spanish And Latin American Cultures, Lara Armenteros Garrido

Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–

This dissertation focuses on the idea that women’s bodies have a sexual stigma attached to them as a consequence of different forms of heteropatriarcal violence inflicted on them. From violence in public spaces, such as street harassment or the sexual exploitation of women, to the intimacy of the relationship, like intimate partner violence or the orgasm gap, it becomes increasingly difficult for women to escape violence. Through the analysis of films, documentaries, narratives, and the testimonies of women from Spain and Latin America, I argue there is still a lack of adequate sexual and emotional knowledge to help complete agency …
