RNA sequencing (RNA-seq) has recently been used in translational research settings to facilitate diagnoses of Mendelian disorders. A significant obstacle for clinical laboratories in adopting RNA-seq is the low or absent expression of a significant number of disease-associated genes/transcripts in clinically accessible samples. As this is especially problematic in neurological diseases, we developed a clinical diagnostic approach that enhanced the detection and evaluation of tissue-specific genes/transcripts through fibroblast-to-neuron cell transdifferentiation. The approach is designed specifically to suit clinical implementation, emphasizing simplicity, cost effectiveness, turnaround time, and reproducibility. The coppery titi monkey (Plecturocebus cupreus) is an emerging nonhuman primate model system for behavioral and neurobiological research. At the same time, the almost entire absence of genomic resources for the species has hampered insights into the genetic underpinnings of the phenotypic traits of interest. To facilitate future genotype-to-phenotype studies, we here present a high-quality, fully annotated de novo genome assembly for the species with chromosome-length scaffolds spanning the autosomes and chromosome X (scaffold N50 = 130.8 Mb), constructed using data obtained from several orthologous short- and long-read sequencing and scaffolding techniques. During the secretory phase of the menstrual cycle, endometrial fibroblast cells begin to change into large epithelial-like cells called decidual cells in a process called decidualization. This differentiation continues more broadly in the endometrium and forms the decidual tissue during early pregnancy. The cells undergoing decidualization as well as the resulting decidual cells, support successful implantation and placentation during early pregnancy. This study was carried out to identify new potentially important long non-coding RNA (lncRNA) genes that may play a role in human endometrial stromal fibroblast cells (hESF) undergoing decidualization in vitro, and several were found. Coagulation factor VIII (FVIII) and its carrier protein von Willebrand factor (VWF) are critical to coagulation and platelet aggregation. We leveraged whole-genome sequence data from the Trans-Omics for Precision Medicine (TOPMed) program along with TOPMed-based imputation of genotypes in additional samples to identify genetic associations with circulating FVIII and VWF levels in a single-variant meta-analysis, including up to 45 289 participants. Gene-based aggregate tests were implemented in TOPMed. We identified 3 candidate causal genes and tested their functional effect on FVIII release from human liver endothelial cells (HLECs) and VWF release from human umbilical vein endothelial cells. INTRODUCTION: This study examines a social-emotional learning intervention that teaches positive self-talk to high school students from underrepresented minority groups in science, technology, engineering, and math (STEM). The setting of this study is a place-based program that immerses students in hands-on STEM activities while located in Philadelphia’s life science hub. Before taking a science assessment, students will learn to say self-talk statements such as “I will give my best effort” and “I can achieve a high score.”

Urban settings face unprecedented environmental challenges that include legacy issues as well as emerging pollution problems (e.g., micro-plastics) that continually impact the public health of its communities. Determining the baseline abundances of potentially harmful contaminants in soil, sediment, air, and water is essential to protecting residents. This study examines anthropogenic pollution in street sediments collected from three neighborhoods located in northeast Terre Haute, Indiana. Using stereomicroscopy, SEM/EDS, pXRF analyses, and geospatial analysis (GIS), the goal of this work is to characterize the types of pollutants present and place important constraints on their potential sources.

I explore in this paper the ways in which churches in the United States treat LGBTQIA+ issues and members in their congregations. To do so, I will focus on several case studies, including the Metropolitan Community Church, the Episcopal Church, the Roman Catholic Church, the Church of Jesus Christ of Latter-Day Saints, and the United Methodist Church. These examples range from socially affirming to conservative and from traditional to contemporary worship styles. White-light LED bulbs undergo degradation over time, leading to a rise in color temperature and a proportional increase in the emission of blue light from these bulbs. A frightening mucilage event occurred in the Sea of Marmara (SoM) from November 2020 to July 2021, leaving devastating effects on maritime industries, fishing activities and the benthic ecosystem. The mucilage led to intense fish mortalities, particularly affecting species like smelt, anchovy, and picarel, with severe consequences for benthic ecosystems and various marine species. A total of 134 Apodemus samples, whose genetic diagnoses had been previously conducted, were morphologically examined from 39 localities in Northern Anatolia, Thrace, Gökçeada, Bozcaada and the Marmara Island. The variation boundaries of dental variations in the distribution areas of five Apodemus species (Apodemus flavicollis, Apodemus witherbyi, Apodemus sylvaticus, Apodemus uralensis, Apodemus mystacinus) included in the research were determined. The defining morphological characters of Apodemus species, which have a complex taxonomic status, and their variations according to regions were identified using samples that had undergone genetic diagnoses. The Common wall lizard (Podarcis muralis) has the largest range among the genus Podarcis. Although there are a number of studies on its diet, data on regional level remains insufficient, particularly from a purely ecological perspective. We investigated 194 individual faecal samples of P. muralis obtained from two localities in western Bulgaria with the aim of clarifying the composition of the trophic spectrum of the species, as well as the intra-population characteristics in terms of age and sex. Sexual dimorphism has been established as one of the important evolutionary results in the process of sexual selection between males and females of different species. Distinct morphological features between males and females can also be seen in reptiles. Lizards of the genus Darevskia show sexual dimorphism, as Darevskia schaekeli was studied as an example. The morphological traits investigated between males and females of this species showed that head length, trunk length, length of forelimbs and hindlimbs as well as the number of femoral pores are more in males than in females. Background/Problem Statement: Acute musculoskeletal pain is one of the major chief complaints amongst Emergency Department (ED) visits each year. In the United States, approximately 41.9 million annual ED visits are musculoskeletal pain related (Fontánez, et al., 2021). Yet, data statistics demonstrate at least 40% of patients reported their pain remained unchanged during their presentation to an ED (Bussey et al., 2020).

This study examined the problems and challenges of social and academic integration in U.S. higher education institutions for Saudi Arabian and Mexican undergraduate international students. The purpose of this quantitative, cross-sectional, descriptive, and causal-comparative study was to examine the relationship among (a) demographic variables (i.e., age, gender, education, marital status, nationality, and previous academic performance), (b) academic integration, (c) social integration, and (d) intent to drop out among Saudi and Mexican students studying at U.S. colleges and universities. Osteosarcopenia is the duplicate diagnosis of osteoporosis, low bone mineral density, and sarcopenia, low muscle mass, co-existing together. The impending biodiversity crises demands urgent, effective action. The transboundary nature of many marine species at risk makes international law a necessary tool in this endeavour. The United Nations Environment Programme and its Regional Seas Programme consists of 18 individual progammes spanning the globe and bringing together 143 countries in regional collaborations. This research project evaluates potential effectiveness of four programmes within the Regional Seas Programme relative to each other on twelve elements looking at legal and institutional structure, as well as regional implementation. This action research study investigated the impact of open-note assessments on student learning as measured by the student’s overall score on unit assessments inside of a 9th grade Algebra 1 classroom. This study included 332 freshmen: an experimental group (open-note assessments) of 72 students and a control group of 260 students. Findings of this study did not show a statistically significant increase in the scores of students using open-note assessments. However, when looking at the different quartiles of test scores, the overwhelming evidence by the researcher revealed the benefits that open-note assessments had on student’s academic performance resulting in better …

Small RNAs play pivotal roles in post-transcriptional gene regulation across diverse phylum of protostomes. In this study, we investigate the functional significance of atypical miRNAs, mirtron miR-1017 in Drosophila. Through ectopic expression in neuronal cells, we demonstrate that miR-1017 extends lifespan by targeting its host transcript, acetylcholine receptor Dα2, and influencing its splicing. This novel trans-regulatory function suggests a mechanism for mirtron evolution, highlighting the interplay between splicing and post-transcriptional regulation. Additionally, we profile small RNA populations in the polychaete developmental model Capitella teleta, shedding light on the small RNA landscape in annelid worms. Our analysis reveals a rich …

The conventional methodology for sentiment analysis within large language models (LLMs) has predominantly drawn upon human emotional frameworks, incorporating physiological cues that are inherently absent in text-only communication. This research proposes a paradigm shift towards an emotionallyagnostic approach to sentiment analysis in LLMs, which concentrates on purely textual expressions of sentiment, circumventing the confounding effects of human physiological responses. The aim is to refine sentiment analysis algorithms to discern and generate emotionally congruent responses strictly from text-based cues.
