Development And Application Of Small-Molecule Probes Targeting Ghrelin O-Acyltransferase, Jacob Moose

Ghrelin is a 28-amino acid peptide hormone that has wide-ranging physiological effects, including on metabolism, adipogenesis, insulin regulation, and addiction. Ghrelin offers an intriguing option for treatment of pathophysiologies related to these effects, as it requires a unique post-translational octanoyl modification to bind and activate its cognate receptor, growth hormone secretagogue 1a (GHSR). The enzyme responsible for this modification, ghrelin O-acyltransferase (GOAT) therefore offers the ability to modulate ghrelin signaling through inhibition of the maturation pathway. Specialized cancer treatments have the potential to exploit glutamine dependence to increase patient survival rates. Glutamine diagnostics capable of tracking a patient’s response to treatment would enable a personalized treatment dosage to optimize the tradeoff between treatment success and dangerous side effects. Current clinical glutamine testing requires sophisticated and expensive lab-based tests, which are not broadly available on a frequent, individualized basis. To address the need for a low-cost, portable glutamine diagnostic, this work engineers a cell-free glutamine biosensor to overcome assay background and signal-to-noise limitations evident in previously reported studies. Agrarian movements around the world use agroecology to build sovereignty and steward dynamic ecosystems. Research has shown that agroecological farmers steward more resilient crops, more resilient soil biomes, and greater biodiversity than conventional agriculture. The proceeding dissertation is a collection of three articles exploring the phenomenon of women’s wings in rebel groups. The articles ask different questions and utilize diverse methodological tools. The three abstracts are below. Paper 1 exposes the patterns of women’s wings in rebel groups through a large-N analysis of 372 rebel groups existing between 1946 and 2015. Studying variation in individual behavior can elucidate drivers related to factors like age, social context, and the environment. Differences in behavior with age are a product of survival needs and developmental differences as animals physiologically mature and modify their behaviors in response to experience. In this dissertation, representation, participation, and conflict are examined in state-mandated environmental justice councils. A novel dataset is constructed from council meeting minutes to explore patterns across these key concepts at the individual-level, meeting-level, and council level over time. The first essay examines how meeting-level factors – such as participant diversity, attributes of the meeting, and context – associate with meeting-level participation measured as different types of communication. This essay builds on participation research in collaborative governance, exploring participation through the lens of communication. This research examines the effect of the right to legislative representation for people with disabilities in the fields of Political Science and Law through a combination of doctrinal analysis and interviews with 12 individuals with disabilities. Climate Justice Before the Anthropocene examines British and Irish Romantic literature through the lens of climate. The tail end of the Little Ice Age and two volcanic eruptions halfway around the world contributed to severe and unpredictable climate patterns in the British Isles between 1790 and 1820 that exacerbated existing social inequities and economic distresses. This mixed methods study explored how elementary teachers reported using a commercially published instructional unit to plan and deliver science instruction in their elementary classrooms. Of particular focus is what elements of the program teachers eliminated during planning and instruction, what outside materials teachers added to instruction, what modifications they made to the materials, and their rationales for these changes to the prescribed program. Engaging with contemporary Latinx and Indigenous border literatures, films, and other cultural productions, my dissertation considers the tensions between Latinx migration and Indigenous sovereignty, claiming that their uneasy relation demands sustained critical and political attention. While Latinx cross-border migration and Indigenous peoples’ sovereignty struggles share important connections in the context of a colonial racial capitalist system of oppression, they cannot be easily collapsed into each other. Osvlado Golijov’s The Dreams and Prayers of Isaac the Blind, scored for klezmer clarinet and string quartet, remains one of his most influential works since its premier in 1994. Inspiration for the work came from the writings of Rabbi Isaac the Blind, a prominent 13th century figurehead in the branch of Jewish mysticism known as Kabbalah. All-atom molecular dynamics (MD) simulations are essential for examining the structural and dynamic aspects of proteins at high resolution, complementing experiments limited by resolution, size, and complexity. Advancements in algorithms and hardware have enhanced the role of computational tools in scientific research, allowing for more accurate simulations of proteins with standard residues. This dissertation explores the philosophical and socioeconomic context of late imperial Chinese pharmacology and medical therapies from the sixteenth to the eighteenth centuries. It examines the rise of human-derived drugs (renyao) in the sixteenth century and their subsequent decline in the late seventeenth and eighteenth centuries, influenced by a complex interplay of changes in social mores, urban tastes, commercial publishing, and philosophical ideologies. Treating biofilm infections on medical implants is a significant healthcare challenge, often requiring device removal and potentially leading to amputations. This study investigates the efficacy of biocompatible peptoids as a novel strategy to combat these infections. This study employs atomistic simulations and small-angle X-ray scattering to investigate the self-assembly behavior of four peptoid sequences (TM1, TM4, TM6, TM22). Peptoids forming supramolecular assemblies display enhanced complexation with polyanionic microgels, promising for antimicrobial coatings on implantable devices. Atomistic simulations and experimental data analysis reveal a strong link between peptoid sequence, hydrophobicity, and propensity for self-assembly. The methodology that was used for this study was constructivist grounded theory. My first chapter, “‘Ugly Monstrous Shapes’: Reading Signs of Alterity,” looks at presentations of “othered” female bodies in Book I of Edmund Spenser’s The Faerie Queene. This chapter draws our attention to various old, deformed, or otherwise dysfunctional bodies. In doing so, it not only emphasizes the necessity for reading but also listening to the “ugly monstrous shapes, that elsewhere no man may” find.
