The De-Domestication Of Ornithopus Sativus Brot. To Develop Cultivars With Physical Dormancy (Hardseed), Robert J. Harrison, Bradley J. Nutt, Ronald J. Yates, Belinda F. Hackney, John G. Howieson

Grain and Other Field Crops Research Articles

Ornithopus sativus Brot. (French serradella) is a forage legume that is well adapted to acidic coarse textured soils (sands) which are characterized by poor nutrition and an inability to retain water. During the process of domestication of O. sativus, there was an unintentional loss of seed physical dormancy (PY) thus compromising its self-regeneration after a cropping interval. Through mass screening of seed, we identified for the first time that heritable sources of PY exist in three populations of O. sativus. This rare genetic material was then incorporated into suitable genetic backgrounds of differing maturity through targeted hybridization. We demonstrated …

We report the development of an experimentally-validated computational fluid dynamics (CFD) model for simulation of an anhydrous HCl electrolyzer. The experimental data from 3 membrane variants was used to provide kinetic and membrane parameters for the model. The model not only accurately simulates overall electrolyzer performance, but it also provides key insights into the transport phenomena within the electrolyzer. The model allows simulation of experimental parameters like high HCl flowrates and increased cell pressure that pose a high safety risk to researchers. The model shows hotspots in the temperature distribution that will need to be addressed by flow field modification …

Saline lakes were under pressure of high EC, …

Develop And Optimize Clinical System Of Focused Ultrasound-Enabled Liquid Biopsy (Sonobiopsy) For Brain Cancer Diagnosis, Lu Xu May 2024

Develop And Optimize Clinical System Of Focused Ultrasound-Enabled Liquid Biopsy (Sonobiopsy) For Brain Cancer Diagnosis, Lu Xu

McKelvey School of Engineering Graduate Student Theses & Dissertations

Brain cancer is a devastating disease characterized by the abnormal growth of cells within the brain. It disrupts neurological function, carries a grim prognosis, and significantly decreases the quality of life. Glioblastoma (GBM), the most common malignant primary brain tumor in adults, is particularly aggressive, with less than 5% of patients surviving for five years. The conventional treatment portfolio, including radiation therapy, chemotherapy, and surgical resections, is less effective for GBM than other cancer diseases due to its high spatial heterogeneity and adaptability to treatment. This suggests that an accurate and comprehensive diagnosis of the molecular subtypes is critical for …

Engineering Macrophages For Self-Regulated, Localized Delivery Of Anti-Inflammatory Biologics, Molly Klimak

McKelvey School of Engineering Graduate Student Theses & Dissertations

Immunoengineering continues to revolutionize healthcare, generating new approaches for treating previously intractable diseases. In chronic inflammatory diseases such as rheumatoid arthritis (RA), a chronic autoimmune disease characterized by joint inflammation and destruction, anti-cytokine treatments have previously been at that forefront of therapeutic innovation. However, while many of the existing anti-cytokine treatments are successful for a subset of patients, these treatments can also pose severe adverse events and off-target effects from systemic administration of anti-inflammatory drugs at constant high dosages or a lack of disease-specific targets. These limitations have motivated the development of new treatment strategies that are well-controlled, precise, safe, …

Improving And Modeling Heterogeneous Streaming Computation, Clayton Faber May 2024

Improving And Modeling Heterogeneous Streaming Computation, Clayton Faber

McKelvey School of Engineering Graduate Student Theses & Dissertations

Data streaming algorithms are a class of problems that deal with moving data through a system while being processed. When implementing these types of algorithms a developer will spend time tuning an implementation for deployment, but if they are using heterogeneous architectures or when nodes of computation are physically separate from one another performance may be lost to unforeseen data movement complications. In this work we aim to alleviate some of these pain points through a combination of programming advice and mathematical models. One of the pain points often unseen and underappreciated by developers is a type of data streaming …

We sought to define the cooccurring mutational profile of FLT3-ITD-positive (ITDpos) acute myeloid leukemia (AML) in pediatric and young adult patients and to define the prognostic impact of cooperating mutations. We identified 464 patients with FLT3-ITD mutations treated on Children's Oncology Group trials with available sequencing and outcome data. Overall survival, event-free survival (EFS), and relapse risk were determined according to the presence of cooccurring risk stratifying mutations. Among the cohort, 79% of patients had cooccurring alterations across 239 different genes that were altered through mutations or fusions. Evaluation of the prognostic impact of the cooccurring mutations demonstrated that patients …

Spatial transcriptomics (ST) assays represent a revolution in how the architecture of tissues is studied by allowing for the exploration of cells in their spatial context. A common element in the analysis is delineating tissue domains or "niches" followed by detecting differentially expressed genes to infer the biological identity of the tissue domains or cell types. However, many studies approach differential expression analysis by using statistical approaches often applied in the analysis of non-spatial scRNA data (e.g., two-sample t-tests, Wilcoxon's rank sum test), hence neglecting the spatial dependency observed in ST data. In this study, we show that applying linear …

Abnormal Morphology And Synaptogenic Signaling In Astrocytes Following Prenatal Opioid Exposure, Ethan B. Niebergall, Daron Weekley, Anna Mazur, Nathan A. Olszewski, Kayla M. Deschepper, N. Radant, Aishwarya S. Vijay

Biomedical Sciences

In recent decades, there has been a dramatic rise in the rates of children being born after in utero exposure to drugs of abuse, particularly opioids. Opioids have been shown to have detrimental effects on neurons and glia in the central nervous system (CNS), but the impact of prenatal opioid exposure (POE) on still-developing synaptic circuitry is largely unknown. Astrocytes exert a powerful influence on synaptic development, secreting factors to either promote or inhibit synapse formation and neuronal maturation in the developing CNS. Here, we investigated the effects of the partial µ-opioid receptor agonist buprenorphine on astrocyte synaptogenic signaling and …

Tree Recovery By Dynamic Programming, Gustavo Alberto Gratacos May 2024

Tree Recovery By Dynamic Programming, Gustavo Alberto Gratacos

McKelvey School of Engineering Graduate Student Theses & Dissertations

Tree-like structures are common, naturally occurring objects that are of interest to many fields of study, such as plant science and biomedicine. Analysis of these structures is typically based on skeletons extracted from captured data, which often contain spurious segments or cycles that need to be removed. We propose a dynamic programming algorithm which seeks to recover directed trees from these noisy skeletons. Our method recovers trees by removing edges and duplicating nodes while adhering to edge-label constraints. Our algorithm proceeds by iteratively merging graph nodes, such that the solution on the original graph can be obtained from those on …

MV140 is an inactivated whole-cell bacterial mucosal vaccine with proven clinical efficacy against recurrent urinary tract infections (UTIs). These infections are primarily caused by uropathogenic E. coli (UPEC) strains, which are unique in their virulence factors and remarkably diverse. MV140 contains a non-UPEC strain, suggesting that it may induce an immune response against different UPEC-induced UTIs in patients. To verify this, we experimentally evaluated the cellular and humoral responses to UTI89, a prototypical UPEC strain, in mice vaccinated with MV140, as well as the degree of protection achieved in a UPEC UTI89 model of acute cystitis. The results show that …
