T-Cell Redirecting Bispecific Antibodies: A Review Of A Novel Class Of Immuno-Oncology For Advanced Prostate Cancer, Julia Palecki, Amman Bhasin, Andrew Bernstein, Patrick Mille, William Tester, William Kelly, Kevin Zarrabi

Kimmel Cancer Center Faculty Papers

Novel T-cell immunotherapies such as bispecific T-cell engagers (BiTEs) are emerging as promising therapeutic strategies for prostate cancer. BiTEs are engineered bispecific antibodies containing two distinct binding domains that allow for concurrent binding to tumor-associated antigens (TAAs) as well as immune effector cells, thus promoting an immune response against cancer cells. Prostate cancer is rich in tumor associated antigens such as, but not limited to, PSMA, PSCA, hK2, and STEAP1 and there is strong biologic rationale for employment of T-cell redirecting BiTEs within the prostate cancer disease space. Early generation BiTE constructs employed in clinical study have demonstrated meaningful antitumor …

A Rising Tide That Lifts All Boats: Long-Term Effects Of The Alaska Permanent Fund Dividend On Poverty, Matthew Berman May 2024

A Rising Tide That Lifts All Boats: Long-Term Effects Of The Alaska Permanent Fund Dividend On Poverty, Matthew Berman

ISER Articles

Although not designed as a social program to redistribute income, the Alaska Permanent Fund Dividend (PFD) has been reducing poverty by providing equal annual payments to nearly all state residents for over 40 years. We examine direct effects of the PFD on Alaska poverty rates since 1990, using US Census and American Community Survey Public Use Microdata Sample records to adjust for under-reporting of children's PFD income in official statistics. We estimate that the PFD reduced the number of Alaskans with incomes below the US poverty threshold by 20%–40%. We measure only a small effect on income distribution: a 0.02 …

Efficacy Of Ropivacaine For Sub-Arachnoid Block In Patients With Recent History Of Scorpion Sting: A Case Series, Saurabh Trivedi, Hemendra Bhardwaj, Tapan K. Sahoo, Seema Gupta

Sultan Qaboos University Medical Journal

Failure of sub-arachnoid block (SAB), due to resistance to bupivacaine after a recent scorpion sting can lead to multiple block attempts and subsequent conversion to general anaesthesia. We report this case series of 10 patients with successful SAB with newly launched 0.75% hyperbaric ropivacaine, in patients with recent scorpion sting. Thus, intrathecal hyperbaric ropivacaine may be considered as the local anaesthetic agent of choice in patients with scorpion sting to prevent failure of SAB.

Faculty, Staff and Student Publications

Fetal alcohol spectrum disorders (FASDs) are leading causes of neurodevelopmental disability but cannot be diagnosed early in utero. Because several microRNAs (miRNAs) are implicated in other neurological and neurodevelopmental disorders, the effects of EtOH exposure on the expression of these miRNAs and their target genes and pathways were assessed. In women who drank alcohol (EtOH) during pregnancy and non-drinking controls, matched individually for fetal sex and gestational age, the levels of miRNAs in fetal brain-derived exosomes (FB-Es) isolated from the mothers’ serum correlated well with the contents of the corresponding fetal brain tissues obtained after voluntary pregnancy termination. In six …

Abscisic acid (ABA) plays a crucial role in promoting plant stress resistance and seed dormancy. However, how ABA regulates rice quality remains unclear. This study identifies a key transcription factor SLR1-like2 (SLRL2), which mediates the ABA-regulated amylose content (AC) of rice. Mechanistically, SLRL2 interacts with NF-YB1 to co-regulate Wx, a determinant of AC and rice quality. In contrast to SLR1, SLRL2 is ABA inducible but insensitive to GA. In addition, SLRL2 exhibits DNA-binding activity and directly regulates the expression of Wx, bHLH144 and MFT2. SLRL2 competes with NF-YC12 for interaction with NF-YB1. NF-YB1 also directly represses SLRL2 transcription. Genetic validation …

Glioblastoma Disrupts Cortical Network Activity At Multiple Spatial And Temporal Scales, Jochen Meyer, Kwanha Yu, Estefania Luna-Figueroa, Benjamin Deneen, Jeffrey Noebels

Faculty, Staff and Students Publications

The emergence of glioblastoma in cortical tissue initiates early and persistent neural hyperexcitability with signs ranging from mild cognitive impairment to convulsive seizures. The influence of peritumoral synaptic density, expansion dynamics, and spatial contours of excess glutamate upon higher order neuronal network modularity is unknown. We combined cellular and widefield imaging of calcium and glutamate fluorescent reporters in two glioblastoma mouse models with distinct synaptic microenvironments and infiltration profiles. Functional metrics of neural ensembles are dysregulated during tumor invasion depending on the stage of malignant progression and tumor cell proximity. Neural activity is differentially modulated during periods of accelerated and …

School of Nursing and Midwifery

Many coastal and rural areas in Britain are deprived. This regional aspect multiplies the effect of digital exclusion for older people. Younger people in these regions can also be digitally excluded. Codesigning digital technologies, using the natural and heritage resources of such regions would address this, but is rarely done. We present our methods and preliminary results, half-way through a 30-month project to develop novel technologies in extended reality, underwater telepresence, digital social games, and artificial intelligence voice interfaces and to use these assets to tackle digital exclusion. We are taking an intergenerational approach, working with 35 partner organizations, some …

The epithelial cell lining of the oviduct plays an important role in oocyte pickup, sperm migration, preimplantation embryo development, and embryo transport. The oviduct epithelial cell layer comprises ciliated and nonciliated secretory cells. The ciliary function has been shown to support gamete and embryo movement in the oviduct, yet secretory cell function has not been well characterized. Therefore, our goal was to generate a secretory cell-specific Cre recombinase mouse model to study the role of the oviductal secretory cells. A knock-in mouse model, Ovgp1Cre:eGFP, was created by expressing Cre from the endogenous Ovgp1 (oviductal glycoprotein 1) locus, with enhanced green …

Bagging Improves The Performance Of Deep Learning-Based Semantic Segmentation With Limited Labeled Images: A Case Study Of Crop Segmentation For High-Throughput Plant Phenotyping, Yinglun Zhan, Yuzhen Zhou, Geng Bai, Yufeng Ge

Department of Statistics: Faculty Publications

Advancements in imaging, computer vision, and automation have revolutionized various fields, including field-based high-throughput plant phenotyping (FHTPP). This integration allows for the rapid and accurate measurement of plant traits. Deep Convolutional Neural Networks (DCNNs) have emerged as a powerful tool in FHTPP, particularly in crop segmentation—identifying crops from the background—crucial for trait analysis. However, the effectiveness of DCNNs often hinges on the availability of large, labeled datasets, which poses a challenge due to the high cost of labeling. In this study, a deep learning with bagging approach is introduced to enhance crop segmentation using high-resolution RGB images, tested on the …

Carbon Sequestration In Peatlands: Using Environmental Proxies To Understand The Impact Of A Changing Climate On Global Carbon Storage, Sarah Schuller, Don Sullivan May 2024

Carbon Sequestration In Peatlands: Using Environmental Proxies To Understand The Impact Of A Changing Climate On Global Carbon Storage, Sarah Schuller, Don Sullivan

DU Undergraduate Research Journal Archive

Peatlands are a type of terrestrial wetland ecosystem in which consistently waterlogged conditions prevent the decomposition of organic matter. These conditions allow for sequestered carbon in plant matter to remain stored in the soil to such a great degree that peatlands store more carbon than all other vegetation types in the world combined. By inhibiting decomposition, the composition of water-rich peat soil remains representative of the environmental conditions during the period in which the peat was formed. The collected peat samples can then be utilized as environmental proxies to determine historical temperature, moisture, and carbon content and extrapolated to predict …

Caprin1 And Fmr1 Genetically Interact To Regulate The Development Of The Larval Drosophila Neuromuscular Junction, Seraphina Loukas, Scott Barbee

DU Undergraduate Research Journal Archive

Fragile X Syndrome (FXS) is the most prevalent inherited neurodevelopmental disorder and the most common single-gene cause of autism (Richter and Zhao 2021). FXS occurs due to the loss of the Fmr1 gene, and its respective protein, the Fragile X Messenger Ribonucleoprotein (FMRP). FMRP is an RNA-binding protein (RBP) with notable functions in synaptic development.  Given that cellular processes often entail the collaborative actions of multiple proteins acting as binding partners to regulate mRNA metabolism, identifying FMRP's associates is essential for comprehending FXS mechanisms. Caprin1 was identified as a high-confidence interactor via its co-immunoprecipitation with FMRP in an IP/LC experiment …

A Systems Theoretic View Of Speculative Realism, Martin Zwick May 2024

A Systems Theoretic View Of Speculative Realism, Martin Zwick

Complex Systems Faculty Publications and Presentations

Recent developments in Continental philosophy have included the emergence of a school of “speculative realism,” which rejects the human-centered orientation that has long dominated Continental thought. Proponents of speculative realism differ on several issues, but many agree on the need for an object-oriented ontology. Some speculative realists identify realism with materialism, while others accord equal reality to objects that are non-material, even fictional. Several thinkers retain a focus on difference, a well-established theme in Continental thought. This paper looks at speculative realism from the perspective of systems theory. Many of the tenets of speculative realism have long been features of …

DU Undergraduate Research Journal Archive

Past research has investigated the impact of gender and gender biases on hiring decisions for cisgender individuals. However, there is a lack of research on how perceptions of gender diverse individuals impact hiring decisions. The current work explores how a job applicant’s gender pronouns may impact the likelihood of the applicant getting hired. We also investigated whether this likelihood varied depending on the job description—specifically whether it included content related to gender stereotypes (e.g., “nurturing” versus “leadership capabilities” as a desired trait in job applicants). Ninety-six participants were randomly assigned to view an application package including she/her, he/him, or they/them …

Examination Of Practices Employed In Correctional Facilities Concerning Transgender Women, Nivu Srinivasan May 2024

Examination Of Practices Employed In Correctional Facilities Concerning Transgender Women, Nivu Srinivasan

Student Theses

This thesis investigates how traditional gender norms may influence correctional facility regulations, particularly those affecting transgender women. Traditional correctional facilities operate relying on these norms that marginalize those who do not conform, compounding issues like limited access to gender-affirming healthcare and greater risks of assault and isolation. The study investigates how these norms influence incarceration policy, which frequently overlooks the unique requirements of transgender inmates. Transgender women confront significant challenges in correctional settings, including limited healthcare access, increased vulnerability to physical and psychological damage, and placement in facilities that do not reflect their gender identity.

Virtual Confessions: Examining The Clergy Privilege’S Extension To Artificially Intelligent Religious Robots, Samuel N. Dick May 2024

Virtual Confessions: Examining The Clergy Privilege’S Extension To Artificially Intelligent Religious Robots, Samuel N. Dick

The University of Cincinnati Intellectual Property and Computer Law Journal

Artificial Intelligence (AI) is fundamentally changing the world. AI’s rapid development is driving its integration into every industry, including those traditionally untouched by technology—such as religion. Today, faith groups in America and globally, are integrating AI-driven robots in roles traditionally held by human priests, clergy, or pastors. AI robots have begun giving sermons, conducting funerals/weddings, providing spiritual counseling, and conducting the sacrament of confession. Some faith groups have gone further claiming the worship of AI as an independent religion, and have received § 501(c)(3) tax-exempt status as a church. Whether thoughts of sacrileges, inevitability, or a science-fiction novel emerge, AI’s …

Estimating Increased Transient Water Storage With Increases In Beaver Dam Activity, Konrad Hafen, Joseph M. Wheaton, Brett B. Roper, Philip Bailey, William W. Macfarlane, Bethany T. Neilson, Christopher J. Tennant

Watershed Sciences Faculty Publications

Dam building by beaver (Castor spp.) slows water movement through montane valleys, increasing transient water storage and the diversity of residence times. In some cases, water storage created by beaver dam construction is correlated to changes in streamflow magnitude and timing. However, the total amount of additional surface and groundwater storage that beaver dams may create (and, thus, their maximum potential impact on streamflow) has not been contextualized in the water balance of larger river basins. We estimate the potential transient water storage increases that could be created at 5, 25, 50, and 100% of maximum modeled beaver dam capacity …

An Evaluation Of Sonderegger Pine Morphology, Kelsey Denise Shoemaker May 2024

An Evaluation Of Sonderegger Pine Morphology, Kelsey Denise Shoemaker

Master's Theses

Sonderegger pine (Pinus x sondereggeri H.H. Chapm.), the naturally occurring hybrid of longleaf (Pinus palustris Mill.) and loblolly pine (Pinus taeda L.), is considered a producer of poor-quality wood due to observations of stem forking and a high number of large-diameter branches at maturity. Because landowners generally decline planting Sonderegger pine seedlings, nursery workers cull putative hybrids from longleaf pine seedling stock during lifting and processing. However, over the past 100 years, very few studies have quantified stem quality of Sonderegger pine. To gain a better understanding of the early growth and development of Sonderegger pine, evaluations of height, diameter …

Reports Of Autosomal Recessive Disease And Consanguineous Mating Within The Human Population, Johnathon L. Schluter

Master's Theses

It is anecdotally evident when investigating published reports of autosomal recessive disease that a substantial number of cases are the result of related (consanguineous) mating. This research seeks to quantify the percent of manuscripts describing autosomal recessive diseases published between 2000 and 2020 in which consanguineous mating is indicated. We analyzed 602 peer-reviewed manuscripts to identify the percentage of cases presented in which consanguineous mating was indicated, the underlying genes (novel gene or new mutation) and geographical region. These papers were accessed through a specific set of parameters on the free access PubMed Central (PMC) database. A total of 552 …

Effect Of Sex On The Accumulation Of Hepatic Mercury In Mallards Wintering In Louisiana, Latorya E. Mason May 2024

Effect Of Sex On The Accumulation Of Hepatic Mercury In Mallards Wintering In Louisiana, Latorya E. Mason

Master's Theses

The increase in mercury production through anthropogenic activities has led to a global crisis of mercury pollution that poses a significant threat to the health of both humans and wildlife. Aquatic ecosystems are especially at risk due to the creation of methylmercury, a toxic form of mercury, by sulfate-reducing bacteria in anoxic sediments and soils. Methylmercury enters the food chain where it bioaccumulates in individual organisms and then biomagnified with increasing trophic level. Aquatic birds are useful bioindicators of many pollutants, including mercury, due to their abundance and position in the food chain. Previous studies have primarily focused on mercury …

Chronic High-Salt Diet Activates Tumor-Initiating Stem Cells Leading To Breast Cancer Proliferation, Lisa Tucker, Umer Ali, Deborah A. Lannigan, Jeffrey C. Rathmell, Venkataswarup Tiriveedhi

Biology Faculty Research

Several chronic inflammatory diseases have been linked to high-salt (HS) diets. Chronic inflammation is an established causative hallmark of cancer. However, a direct role of HS diets in tumorigenesis is yet to be defined. Previous orthotopic murine breast tumor studies have shown that short-term HS diets caused inhibition of tumor growth through the activation of cytotoxic adaptive immune responses. However, there have been experimental challenges in developing a viable chronic HS-diet-based murine tumor model. To address this, we have developed a novel chronic HS diet tumor model through the sequential passaging of tumor cells in mice under HS dietary conditions. …

Comparison Of Vivitrol And Suboxone In Terms Of Lowering Relapse Among Opioid Addicts, Nicolle Tourdot

Walden Dissertations and Doctoral Studies

Drug overdoses are increasing, and the heroin epidemic is becoming more evident. There are ways to get these individuals’ help. Medically assisted treatments (MATs) involve use of medications alongside counseling and therapies to treat individuals with a substance abuse disorder. This subject is important to the human services administration field, more specifically those who focus on addiction. As the epidemic of opioid use continues, so does the need for treatment. Without that treatment, the problem will get worse and inevitably have negatives effects on the entire world. Those effects involve crime, increasing costs to the public, overall health, and child …

Phase Interface Dynamics And Heat Transfer Mechanisms In Evaporating Droplet And Pool Boiling Processes, Md Tanbin Hasan Mondal May 2024

Phase Interface Dynamics And Heat Transfer Mechanisms In Evaporating Droplet And Pool Boiling Processes, Md Tanbin Hasan Mondal

Doctoral Dissertations

Despite the significant importance and widespread use of phase-change cooling techniques, there are still fundamental questions about the microscopic processes that govern the heat transfer mechanisms. In order to gain a better understanding of the underlying physics involved, it is essential to have information at the microscale regarding the surface temperature distribution with time as well as the location and speed of the moving contact line (MCL). A comprehensive understanding of heat transfer mechanisms and phase-interface behavior during phase-change cooling is crucial for improving heat transfer models, optimizing surface engineering, and maximizing overall effectiveness. Firstly, this dissertation presents a capacitance-based …
