Leaf extracts are known to be rich in phytoconstituent biomolecules, making them a valuable source of medicinal compounds. They also serve as both capping and reducing agents in nanoparticle fabrication. A reaction between CuSO4.5H2O aqueous solution and the Indigofera tinctoria leaf extract results in the formation of stable copper nanoparticles. Phytochemical screening of the Indigofera tinctoria leaf extract revealed the presence of various compounds including carbohydrates, terpenoids, phenol, tannins, flavonoids, saponins, and glycosides in the sample. Xanthone and its derivatives are well known for their broad biological activities. This research aims to investigate the anticancer and antimalarial activities of synthesized xanthenol and xanthene compounds and to elucidate their mechanisms of action through molecular docking. The xanthenol compound was obtained through xanthone reduction with sodium triacetoxyborohydride, and xanthene synthesis was studied through three different reaction conditions: uncatalyzed and catalyzed by Lewis acid or Bronsted acid. The reduction reaction produced xanthenol in 30.50% yield, whereas the three reaction conditions produced xanthene and xanthone in 39.35–75.48% yield. Quercetin is an important phytochemical that belongs to the flavonoid group of plant pigments. It has potential benefits as an antioxidant, antiviral, and anti-inflammatory agent. However, the utilization of quercetin in aquaculture is not optimal. Hence, this study conducted a literature review to analyze the potential benefits of quercetin in aquaculture, especially for fish and prawn. Results showed that quercetin can be isolated from herbs using several methods, including sonication, maceration, and fractionation. Its amount in plants can be potentially determined using high-performance liquid chromatography and thin-layer chromatography. Infant nutrition and immune protection require the consumption of human milk, with macrophages mainly contributing to immune defense and infant development. This study was performed to investigate the correlation between macrophage cell numbers in human milk and various maternal factors. Thirteen human milk samples were collected from lactating mothers and were evaluated for macrophage cell numbers using a hemacytometer. The recorded questionnaire containing several questions, including age, multivitamin consumption, coronavirus disease 2019 vaccination status, allergy history, and lactation duration, was used to determine the maternal factors. Pearson’s correlation and Mann-Whitney test was employed in statistical analysis, with p-values < 0.05 considered significant. The macrophage cell number reached 54.236 ± 7.456 cells per mL. The use of soil microbes is one of the most effective methods for degrading pesticide residue in soil. Microbes can accelerate the degradation of persistent pollutants including chlorpyrifos in soil. This research aims to find soil bacteria with the greatest potential to degrade chlorpyrifos residue in tropical inceptisol soil and determine its degradation index (DI). The soil used in this experiment was collected from agricultural fields in the districts of Demak, Central Java Province, Indonesia. Acute kidney injury (AKI) strongly upregulates the transcription factor Foxm1 in the proximal tubule in vivo, and Foxm1 drives epithelial proliferation in vitro. Here, we report that deletion of Foxm1 either with a nephron-specific Cre driver or by inducible global deletion reduced proximal tubule proliferation after ischemic injury in vivo. Foxm1 deletion led to increased AKI to chronic kidney disease transition, with enhanced fibrosis and ongoing tubule injury 6 weeks after injury. We report ERK mediated FOXM1 induction downstream of the EGFR in primary proximal tubule cells. We defined FOXM1 genomic binding sites by cleavage under targets and release using …

Tumor cell plasticity contributes to intratumoral heterogeneity and therapy resistance. Through cell plasticity, some lung adenocarcinoma (LUAD) cells transform into neuroendocrine (NE) tumor cells. However, the mechanisms of NE cell plasticity remain unclear. CRACD (capping protein inhibiting regulator of actin dynamics), a capping protein inhibitor, is frequently inactivated in cancers. CRACD knockout (KO) is sufficient to de-repress NE-related gene expression in the pulmonary epithelium and LUAD cells. In LUAD mouse models, Cracd KO increases intratumoral heterogeneity with NE gene expression. Single-cell transcriptomic analysis showed that Cracd KO-induced NE cell plasticity is associated with cell de-differentiation and stemness-related pathway activation. The …

The B cell receptor (BCR) signals together with a multi-component co-receptor complex to initiate B cell activation in response to antigen binding. Here, we take advantage of peroxidase-catalyzed proximity labeling combined with quantitative mass spectrometry to track co-receptor signaling dynamics in Raji cells from 10 s to 2 h after BCR stimulation. This approach enables tracking of 2,814 proximity-labeled proteins and 1,394 phosphosites and provides an unbiased and quantitative molecular map of proteins recruited to the vicinity of CD19, the signaling subunit of the co-receptor complex. The present work focuses on the synthesis of a vanadium nitride (VN)/carbon nanocomposite material via the thermal decomposition of vanadyl phthalocyanine (VOPC). The morphology and chemical structure of the synthesized compounds were characterized using scanning electron microscopy (SEM), transmission electron microscopy (TEM), energy dispersive spectroscopy (EDS), Fourier transformed infrared spectroscopy (FTIR), X-ray diffraction (XRD), and X-ray photoemission spectroscopy (XPS). The successful syntheses of the VOPC and non-metalated phthalocyanine (H2PC) precursors were confirmed using FTIR and XRD. The VN particles present a needle-like morphology in the VN synthesized by the sol-gel method. The morphology of the VN/C composite material exhibited small …

Plum curculio is a brown weevil (beetle with a snout) native to eastern North America, where it is a major pest of pome and stone fruits. It was detected in Box Elder County, Utah, in the early 1980s, and this population remains the only known infestation in western North America, where it is occasionally found in residential and wild fruit trees. Plum curculio is a quarantine pest in western North America (U.S. and Canada), and fruit grown in infested counties is restricted from being exported. Thus, it is a threat to Utah’s fruit industry and requires ongoing monitoring and management …

Underrepresented populations are often excluded from genomic studies owing in part to a lack of resources supporting their analyses. The 1000 Genomes Project (1kGP) and Human Genome Diversity Project (HGDP), which have recently been sequenced to high coverage, are valuable genomic resources because of the global diversity they capture and their open data sharing policies. Here, we harmonized a high-quality set of 4094 whole genomes from 80 populations in the HGDP and 1kGP with data from the Genome Aggregation Database (gnomAD) and identified over 153 million high-quality SNVs, indels, and SVs. Scenarios once relegated to science fiction movies and novels are now potentially within the grasp of bioengineering. The world’s oceans and waterways contain untold amounts of munitions, whether dumped or fired, and the sometimes-harsh underwater environment can make their clearance difficult. Yet we are approaching a time when we will have to face the reality of removing them to continue enjoying the planet’s most abundant natural resource. Much of the ammunition in areas from the Baltic Sea to the rivers of Cambodia to the Gulf of Mexico has been immersed for decades, subject to a variety of underwater influences. Human sirtuin-2 (SIRT2) has emerged as an attractive drug target for a variety of diseases. The enzyme is a deacylase that can remove chemically different acyl modifications from protein lysine residues. Here, we developed a high-throughput screen based on a homogeneous time-resolved fluorescence (HTRF) binding assay to identify inhibitors of SIRT2's demyristoylase activity, which is uncommon among many ligands that only affect its deacetylase activity. From a test screen of 9600 compounds, we identified a small molecule that inhibited SIRT2's deacetylase activity (IC50 = 7 μM) as well as its demyristoylase activity (IC50 = 37 μM). It has become increasingly apparent that even mature forests are susceptible to plant invasions. However, invasive plants are often more abundant in younger forest stands. It is difficult to disentangle possible mechanisms that would explain this pattern due to the scarcity of long-term studies in successional forests. Several mechanisms have been proposed to explain patterns of invasions as forests age, including biotic resistance, window of opportunity, historical legacies, and invasion debt. We explored patterns and potential mechanisms of plant invasions over 70 years in a regenerating forest with different land-use histories in the Bolleswood Natural Area, Connecticut, USA. Insects are the most diverse animal taxon on Earth and play a key role in ecosystem functioning. However, they are often neglected by ecological surveys owing to the difficulties involved in monitoring this small and hyper-diverse taxon. With technological advances in biomonitoring and analytical methods, these shortcomings may finally be addressed. Here, we performed passive acoustic monitoring at 141 sites (eight habitats) to investigate insect acoustic activity in the Viruá National Park, Brazil. Ephrin-B-EphB signaling can promote pain through ligand-receptor interactions between peripheral cells, like immune cells expressing ephrin-Bs, and EphB receptors expressed by DRG neurons. Previous studies have shown increased ephrin-B2 expression in peripheral tissues like synovium of rheumatoid and osteoarthritis patients, indicating the clinical significance of this signaling. The primary goal of this study was to understand how ephrin-B2 acts on mouse and human DRG neurons, which express EphB receptors, to promote pain and nociceptor plasticity. Social media data provide unprecedented access to discussions of active, naturalistic, and often real-time cannabis use in an era of cannabis policy liberalization. The aim of this study was to explore psychological and environmental correlates of cannabis effects by applying computational social science approaches to a large dataset of unprompted reports of naturalistic cannabis use with corresponding self-reported numerical ratings of subjective highness. This article examines genealogical ethics in the digital age. At a time when more resources for research are available digitally than ever previously, digital media also pose challenges for the large-scale dissemination of false or misleading information as well as the incautious presentation of more careful research that then might be misconstrued by some. This article first reviews the literature about the development of academic genealogy and professional ethical standards. It then provides a series of case studies, each of which examines particular situations in which ethical questions have arisen about the presentation of research findings. This article argues for …
