Assessing The Effects Of Dasatinib On Mesenchymal Stem/Stromal Cells, David P. Heinrichs, Vitali Maldonado, I Kade Karisma Gita Ardana, Ryan M. Porter, Rebekah M. Samsonraj

Biomedical Engineering Faculty Publications and Presentations

Introduction

Progressive aging, or senescence, of mesenchymal stem/stromal cells (MSCs) is a major obstacle faced when trying to culture potent stem cells for use in therapy. Senescent cells are irreversibly nondividing cells that cease performing critical functional effects. Elimination of senescent cells using biochemical means, such as the use of senolytic drugs like dasatinib, may be useful in retaining the viable and proliferating populations of the cells.

Methods

An in vitro approach was used to investigate the effect of dasatinib on phenotypic, genotypic, and immunomodulatory functionality of osteogenic and adipogenic differentiated MSCs. Replicative senescence was achieved through multiple sub-culturing in …

Electric Interrow Control Of Lupine Plants Does Not Adversely Affect The Neighboring Non-Target Lupine Plants, Catherine Borger, Miranda J. Slaven Oct 2024

Electric Interrow Control Of Lupine Plants Does Not Adversely Affect The Neighboring Non-Target Lupine Plants, Catherine Borger, Miranda J. Slaven

Journal articles

Interrow weed control is used in a wide range of crops, traditionally applied via physical cultivation or banded herbicide application. However, these methods may result in crop damage, development of herbicide resistance, or off-target environmental impacts. Electric interrow weed control presents an alternative, although its potential impact on crop yield requires further investigation. One of the modes of action of electric weed control is the continuous electrode–plant contact method, which passes a current through the weed and into the roots. As the current passes into the roots, it can potentially disperse through the soil to neighboring root systems. Such off-target …

Background: Although the physiological stress response is well established, self-reported stress has received less attention. Some validated measures assess task-related stress via self-report in English. However, no such measure exists in Germany. Aims: To address this research gap, we translated and validated the Short Stress State Questionnaire in German (SSSQ-G). It assesses stress-related affective states (e.g., anger) and appraisals (e.g., feelings of control). We expected to replicate the factor structure of the English SSSQ in the SSSQ-G. Methods: We included the SSSQ-G in five studies that induced stress either via the gold standard task for acute stress …

Dynamic Dysregulation Of Retrotransposons In Neurodegenerative Diseases At The Single-Cell Level\, Wankun Deng, Citu Citu, Andi Liu, Zhongming Zhao

Faculty, Staff and Student Publications

Retrotransposable elements (RTEs) are common mobile genetic elements comprising ∼42% of the human genome. RTEs play critical roles in gene regulation and function, but how they are specifically involved in complex diseases is largely unknown. Here, we investigate the cellular heterogeneity of RTEs using 12 single-cell transcriptome profiles covering three neurodegenerative diseases, Alzheimer's disease (AD), Parkinson's disease, and multiple sclerosis. We identify cell type marker RTEs in neurons, astrocytes, oligodendrocytes, and oligodendrocyte precursor cells that are related to these diseases. The differential expression analysis reveals the landscape of dysregulated RTE expression, especially L1s, in excitatory neurons of multiple neurodegenerative diseases. …

We examined the zoographics, or the characteristics of nonhuman animal subjects, reported for the entirety of the Journal of the Experimental Analysis of Behavior (JEAB) through the most recent complete year (1958–2023). Animal sex in particular was evaluated to determine whether the lack of inclusion of female subjects in other disciplines extends to JEAB. Through systematic coding of all nonhuman empirical articles, we found consistent underreporting of most zoographics and a disproportionate use of male subjects relative to female subjects. Additionally, animal sex was commonly unreported and the inclusion of both male and female subjects was sparse. …

Pseudomonas Aeruginosa Aggregate Formation In The Cystic Fibrosis Airway, Alexa D. Gannon Oct 2024

Pseudomonas Aeruginosa Aggregate Formation In The Cystic Fibrosis Airway, Alexa D. Gannon

USF Tampa Graduate Theses and Dissertations

Bacterial aggregates are observed in both natural and artificial environments. In the context of disease, aggregates have been isolated from both chronic and acute infections. Pseudomonas aeruginosa (Pa) aggregates contribute significantly to chronic infections, particularly in the lungs of people with cystic fibrosis (CF). Unlike the large biofilm structures observed in vitro, Pa in CF sputum forms smaller aggregates (~10-1000 cells), and the mechanisms behind their formation remain underexplored. This study aims to identify genes essential and unique to Pa aggregate formation in a synthetic CF sputum media (SCFM2). We cultured Pa strain PAO1 in SCFM2 and LB, both with …
