Epigenetic Modifiers: Exploring The Roles Of Histone Methyltransferases And Demethylases In Cancer And Neurodegeneration, Lauren Reed, Janak Abraham, Shay Patel, Shilpa S Dhar

Faculty, Staff and Student Publications

Histone methyltransferases (HMTs) and histone demethylases (HDMs) are critical enzymes that regulate chromatin dynamics and gene expression through the addition and removal of methyl groups on histone proteins. HMTs, such as PRC2 and SETD2, are involved in the trimethylation of histone H3 at lysine 27 and lysine 36, influencing gene silencing and activation. Dysregulation of these enzymes often leads to abnormal gene expression and contributes to tumorigenesis. In contrast, HDMs including KDM7A and KDM2A reverse these methylation marks, and their dysfunction can drive disease progression. In cancer, the aberrant activity of specific HMTs and HDMs can lead to the silencing …

Accumulating evidence indicates that the gut microbiome influences cancer progression and therapy. We recently showed that progressive changes in gut microbial diversity and composition are closely coupled with tobacco-associated lung adenocarcinoma in a human-relevant mouse model. Furthermore, we demonstrated that the loss of the antimicrobial protein Lcn2 in these mice exacerbates protumor inflammatory phenotypes while further reducing microbial diversity. Yet, how gut microbiome alterations impinge on lung adenocarcinoma development remains poorly understood. In this study, we investigated the role of gut microbiome changes in lung adenocarcinoma development using fecal microbiota transfer and delineated a pathway by which gut microbiome alterations …

Intrinsically disordered protein regions (IDRs) are well established as contributors to intermolecular interactions and the formation of biomolecular condensates. In particular, RNA-binding proteins (RBPs) often harbor IDRs in addition to folded RNA-binding domains that contribute to RBP function. To understand the dynamic interactions of an IDR-RNA complex, we characterized the RNA-binding features of a small (68 residues), positively charged IDR-containing protein, Small ERDK-Rich Factor (SERF). At high concentrations, SERF and RNA undergo charge-driven associative phase separation to form a protein- and RNA-rich dense phase. A key advantage of this model system is that this threshold for demixing is sufficiently high …

School of Geography, Earth and Environmental Sciences

Soil erosion is a world-wide issue driven by land management and climate change. Research has focussed on soil loss rates from agricultural land. However, the loss of trace elements essential for soil and plant health, or potentially toxic elements that occur as impurities in fertilisers and manures, is poorly understood. This study reports on the loads and forms of copper, cadmium, manganese, nickel, selenium and zinc lost from three types of agricultural systems at Rothamsted Research’s North Wyke Farm Platform over five individual storm events. Loads reflected a combination of concentrations in the soil, annual additions from fertilisers, the ability …

Mechanical Degradation Of Plastics: An Implication For Agricultural Soil Health, Linkon Bhattacharjee

Electronic Theses and Dissertations Archive

The growing public awareness of plastic pollution and its harmful environmental impacts on public health and ecological safety has raised multiple questions regarding the rate of its accumulation, long-term fate, and transport within the environment. However, the extent of plastic release into the terrestrial environment, such as farmlands, remains inadequately understood, though it is expected to exceed that of the ocean. Therefore, this dissertation focuses on investigating the mechanical degradation of plastics that leads to the generation of microplastics through fragmentation in agricultural farmlands. This dissertation addresses several knowledge gaps regarding the influence of environmental conditions on the mechanical degradation …

On The Logistic Polynomials And Logistic Kernels With Applications In Machine Learning, Alexander Saint Jefferson

Electronic Theses and Dissertations Archive

Orthogonal polynomials have had vast historical applications in applied mathematics, particularly in differential equations and numerical analysis. In recent years, they have become instrumental as a natural basis for constructing kernel systems in machine learning (ML). In this thesis, we orthogonalize a set of polynomials, the Logistic Polynomials, first developed by George and Rousseau, using the Gram-Schmidt process. We show that this new Logistic Orthogonal Polynomial System (LOPS) satisfies the hypergeometric differential equation and we relate the system to the Legendre polynomials. The connection to the Legendre polynomials has motivated this study to explore applications of the LOPS in ML, …

Responding Through Listening: Promoting Listening Skills Through Lightning Talks And 2x2x2 Storytelling, Elizabeth L. Angeli

Journal of Response to Writing

Current scholarship in writing pedagogy and peer review focuses on how and what to write and say. What is often left unaddressed in these discussions are how we listen and how we teach our students to listen, making listening an often overlooked and understudied piece of the peer review and writing response process. Yet, the quality of the feedback we receive and offer is directly tied to how well we listen to what is said and written. To this fill this gap, this teaching article offers two activities, Lighting Talks and 2x2x2 Storytelling, that promote students’ listening skills so that …

The Trouble With Classifications, Aziz Z. Huq Dec 2024

The Trouble With Classifications, Aziz Z. Huq

Notre Dame Law Review

The Supreme Court relies increasingly on anticlassification rules to implement the Constitution’s various commands of evenhanded state treatment. These rules direct attention to whether an instance of a forbidden classification is present on the face of a challenged law. They contain two necessary steps. First, a court defines a general category of impermissible terms. Second, a court ascertains whether an instance of the category is found in enacted text—so triggering the familiar strict scrutiny analysis. So defined, anticlassification rules now dominate equal protection, free speech, “dormant” Commerce Clause, and even free exercise jurisprudence. The Roberts Court celebrates these doctrinal tests …

Comparing Metabolism And Hypoxia Tolerance Of Native And Non-Native Florida Freshwater Fishes In Climate Change Scenarios, Benjamin A. Share

All HCAS Student Capstones, Theses, and Dissertations

As climate change brings rising water temperatures and lower dissolved oxygen (DO) concentrations to freshwater bodies, it has become increasingly important to understand how this will affect ecosystems with established non-native or invasive species. Fishes of the family Cichlidae are the most pervasive non-native fishes in the Everglades and may increase their range further north under a changing climate. To better understand potential invasibility and competition with native fishes, the metabolism and hypoxia tolerance of non-native Blue Tilapia Oreochromis aureus was compared to native Bluegill Lepomis macrochirus when acclimated across a range of seasonal and projected temperatures. Intermittent-flow respirometry was …

Plants encounter considerable challenges due to environmental changes induced by anthropogenic activities or abrupt seasonal patterns. As sessile organisms, plants are continuously subjected to various abiotic stresses that adversely affect growth and productivity. Researchers have been studying for years to understand how plants develop resilience and respond to these environmental stressors. The application of phytohormones is widely recognized as a practical approach to mitigate the adverse effects of abiotic stresses on horticultural plants. Phytohormones play a crucial role in enhancing plant resilience by modulating key physiological functions such as seed germination, seedling growth, root formation, and photosynthetic efficiency. They also …
