The Role And Therapeutic Possibilities Of Pelp1 In Hepatocellular Carcinoma (Hcc) Cells, Marian Hernandez Fernandez
The Role And Therapeutic Possibilities Of Pelp1 In Hepatocellular Carcinoma (Hcc) Cells, Marian Hernandez Fernandez
Honors Program Theses and Research Projects
Hepatocellular carcinoma (HCC) is the most rapidly increasing cause of cancer mortality in the USA and ranks as the fourth leading cause of cancer-related deaths worldwide. Moreover, advanced HCC has a bleak prognosis, with a median survival rate of approximately 10 months. Latinos in South Texas exhibit the greatest incidence of liver cancer nationwide. Proline-, glutamic acid-, and leucine-rich protein 1 (PELP1) is an oncogene associated with the advancement of several cancers; however, its function in hepatocellular carcinoma (HCC) remains unclear. Analysis of publicly accessible cancer databases revealed elevated expression of PELP1 oncogene in HCC relative to the control group. …

Physics-Informed Transfer Learning For Process Control, Samuel Arce Munoz
Physics-Informed Transfer Learning For Process Control, Samuel Arce Munoz
Theses and Dissertations
In the realm of process control, managing complex systems with limited prior knowledge presents significant challenges, particularly in environments where traditional mechanistic models are either unavailable or computationally prohibitive. This work explores the integration of deep transfer learning with system identification and Model Predictive Control (MPC) to develop control strategies that are both data-efficient and computationally streamlined. Initially, Long Short-Term Memory (LSTM) networks are employed to create approximate MPC controllers by leveraging transfer learning from a source system to a target system, demonstrating that transfer learning can achieve comparable performance to traditional MPC methods with reduced training data. Building upon …

Direct reciprocity is a wide-spread mechanism for the evolution of cooperation. In repeated interactions, players can condition their behavior on previous outcomes. A well-known approach is given by reactive strategies, which respond to the coplayer's previous move. Here, we extend reactive strategies to longer memories. A reactive-n strategy takes into account the sequence of the last n moves of the coplayer. A reactive-n counting strategy responds to how often the coplayer cooperated during the last n rounds. We derive an algorithm to identify the partner strategies within these strategy sets. Partner strategies are those that ensure mutual cooperation without exploitation. …

Staphylococcus aureus is a highly significant pathogen with several well studied and defined virulence factors. However, the metabolic pathways that are required to facilitate infection are not well described. Previous data have documented that S. aureus requires glucose catabolism during initial stages of infection. Therefore, certain nutrients whose biosynthetic pathway is under carbon catabolite repression and CcpA, including arginine, must be acquired from the host. However, even though S. aureus encodes pathways to synthesize arginine, biosynthesis of arginine is repressed even in the absence of glucose. Why is S. aureus a functional arginine auxotroph? This review discusses recently described regulatory …

Bacterial membranes are complex and dynamic, arising from an array of evolutionary pressures. One enzyme that alters membrane compositions through covalent lipid modification is MprF. We recently identified that Streptococcus agalactiae MprF synthesizes lysyl-phosphatidylglycerol (Lys-PG) from anionic PG, and a novel cationic lipid, lysyl-glucosyldiacylglycerol (Lys-Glc-DAG), from neutral glycolipid Glc-DAG. This unexpected result prompted us to investigate whether Lys-Glc-DAG occurs in other MprF-containing bacteria, and whether other novel MprF products exist. Here, we studied protein sequence features determining MprF substrate specificity. First, pairwise analyses identified several streptococcal MprFs synthesizing LysGlc-DAG. Second, a restricted Boltzmann machine-guided approach led us to discover an …

Despite very high rates of suicidal thoughts and behaviors (STB) in autistic adults, the key psychosocial drivers of this phenomenon remain unknown. To investigate, we examined how lifetime stressor exposure and severity, which have been found to predict STB in non-autistic populations, related to STB in a multinational dataset of 226 autistic adults from the United Kingdom and Australia (67% female; Mage = 41.8, SD = 13.6, range = 19–73 years old). Results revealed that autistic men and women differ with respect to the count, severity, and type of stressors they experienced over the life course. Whereas autistic men …
African Women Custodians: Teaching, Toponymy And Tacit Reverie In Heritage Education, Tokie Laotan-Brown
African Women Custodians: Teaching, Toponymy And Tacit Reverie In Heritage Education, Tokie Laotan-Brown
Wagadu: A Journal of Transnational Women's & Gender Studies
This paper investigates the critical role of women in African cultures as guardians of cultural heritage. It explores their central function in transmitting traditional ecological knowledge and practices across generations through established pedagogical methods, toponymic knowledge systems (place-names within cultural landscapes), and the cultivation of tacit knowledge. The paper emphasizes the significance of women's contributions to environmental education, often overlooked or undervalued within Western scientific paradigms. This marginalization restricts access to resources and participation in decision-making processes. It argues for the integration of traditional knowledge systems, informed by women's expertise, into formal education and pedagogy. This inclusion serves to validate …

Theses and Dissertations
University students face a variety of mental health challenges, including increasingly high rates of anxiety. As demand for college counseling services among student populations rises, universities have an increasing need for effective and resource-efficient methods of intervention that support student mental health on their campuses. One area in need of research is that of using university classes themselves as a form of preventive intervention. If effective, this kind of preventive measure might be a particularly attractive method of reducing incidence of mental illness on campus and decreasing demand on university therapy resources. This study involves students at a private university …

Adeno-associated virus (AAV) gene therapy for neurological disease has gained traction due to stunning advances in capsid evolution for CNS targeting. With AAV brain delivery now in focus, conventional improvements in viral expression vectors offer a complementary route for optimizing gene delivery. We previously introduced a novel AAV gene therapy to slow amyloid aggregation in the brain based on neuronal release of an Aβ sequence variant that inhibited fibrilization of wild-type Aβ. Here we explore three coding elements of the virally delivered DNA plasmid in an effort to maximize the production of therapeutic peptide in the brain. We demonstrate that …

Childhood exposure to social disadvantage is a major risk factor for psychiatric disorders and poor developmental, educational, and occupational outcomes, presumably because adverse exposures alter the neurodevelopmental processes that contribute to risk trajectories. Yet, given the limited social mobility in the United States and other countries, childhood social disadvantage is frequently preceded by maternal social disadvantage during pregnancy, potentially altering fetal brain development during a period of high neuroplasticity through hormonal, microbiome, epigenetic, and immune factors that cross the placenta and fetal blood-brain barrier. The current study examines prenatal social disadvantage to determine whether these exposures in utero are associated …

Angiogenin (Ang), an endoribonuclease belonging to the RNase A superfamily, cleaves the anticodon-loops of tRNAs to produce tRNA-half molecules. Although previous studies have demonstrated the involvement of Ang in the pathobiology of neurodegenerative disorders, the characterization of Ang-generated tRNA halves in neuronal cells remains limited. This is partly due to the technical limitations of standard RNA-seq methods, which cannot capture Ang-generated RNAs containing a 2',3'-cyclic phosphate (cP). In this report, we established an Ang-treatment model using SH-SY5Y, a human neuroblastoma cell line, and demonstrated Ang-dependent accumulation of tRNA halves. By performing cP-RNA-seq, which selectively captures cP-containing RNAs, we identified Ang-generated …

Infectious diseases and autoimmune diseases, such as multiple sclerosis (MS) and type 1 diabetes (T1D), have traditionally been seen as distinct, with separate etiologies and courses of disease. Recent research has shown that infection and autoimmune disease are more closely woven together than previously thought; certain pathogens like Epstein-Barr virus and M. tuberculosis have been shown to cause long-term disruption to the immune system, for example (Bordignon et al 2011, #1) (Zhang et al 2023, #2) (Bjornevik et al 2023, #3). Conversely, a distinct pattern has emerged in developed countries where as rates of infection fall, rates of autoimmune disease …

BACKGROUND: A goal of mucosal human immunodeficiency virus type 1 (HIV-1) vaccines is to generate mucosal plasma cells producing polymeric IgA (pIgA)-neutralizing antibodies at sites of viral entry. However, vaccine immunogens capable of eliciting IgA neutralizing antibodies (nAbs) that recognize tier 2 viral isolates have not yet been identified. METHODS: To determine if stabilized native-like HIV-1 envelope (Env) trimers could generate IgA nAbs, we purified total IgA and IgG from the banked sera of six rhesus macaques that had been found in a previous study to develop serum nAbs after subcutaneous immunization with BG505.664 SOSIP and 3M-052 adjuvant, which is …
