Peptide hydrogels are an important class of biomaterials that have gained significant attention in recent years. The stimuli responsive materials change their properties in response to pH, redox, temperature and enzymes and this property allowed us to design the materials with tunable properties based on their end application. This dissertation reports the de novo design, synthesis and characterization of short chain peptide sequences and optimizations of those in order to develop them into potential candidates in wound care and drug delivery applications. Access to electricity in underdeveloped communities is particularly crucial in improving the living standards of people and alleviating poverty by promoting sustainable development. This study deals with the exploration of incorporating wood-plastic composite (WPC) – specifically wood pulp (W) and high-density polyethylene (H) – into triboelectric nanogenerator (TENG) flooring system to convert mechanical energy to electrical energy, promoting sustainable energy, reducing greenhouse gas emissions, improving the quality of life, and serving as feasible flooring tiles. Filipino mothers traditionally serve as primary caregivers and, at times, as the main breadwinners for their families. With limited opportunities in the Philippines, many seek employment abroad, leaving their children behind. Data from the Philippine Statistics Authority indicated that female Overseas Filipino Workers (OFWs) often engage in elementary jobs, including domestic work and caregiving. The COVID-19 pandemic added a challenge in fulfilling their maternal needs since health and safety have become everyone’s priority. Herein, we present a protocol for culturing patient-derived organoids (PDOs) of cervical cancer that includes workflows for tumor biopsy/resection tissue and cytobrush-sampled cells. We describe steps for PDO culture initiation, including rinsing, gentle dissociation, Lymphoprep separation, and cell assessment, as well as seeding cells from surgical and cytobrush tissue digestion. We then provide guidance on PDO maintenance and passage and techniques for producing conditioned medium. Overall, this protocol serves as a valuable guide for establishing and maintaining cervical cancer PDOs. For complete details on the use and execution of this protocol, please refer to Colbert et al.

This research aims to 1) study the conceptual framework for high-efficiency learning management in the digital era, and 2) investigate the needs for developing high-efficiency learning management in the digital era. The study employs a mixed-methods research design divided into two phases. Modern imperial formations involve a violent reconfiguring of difference that is inseparable from the everyday. Modern-world archaeology as a project that studies interlocking vicissitudes of modernity globally has done much to elucidate the varied social orders and politics of knowledge through which such reshuffling occurs. Central to the ability of cells to both generate and mediate forces is the cell cytoskeleton, which forms the muscles and bones for cells. The Bruton tyrosine kinase inhibitor ibrutinib is an effective treatment for patients with chronic lymphocytic leukemia (CLL). While it rapidly reduces lymph node and spleen size, it initially increases the number of lymphocytes in the blood due to cell redistribution. A previously published mathematical model described and quantified those cell kinetics. Here, we propose an alternative mechanistic model that outperforms the previous model in 26 of 29 patients. Our model introduces constant subcompartments for healthy lymphocytes and benign tissue and treats spleen and lymph nodes as separate compartments. Clinical trials frequently include multiple end points that mature at different times. The initial report, typically based on the primary end point, may be published when key planned co-primary or secondary analyses are not yet available. Clonal hematopoiesis involves the clonal expansion of hematopoietic cells, potentially progressing into hematological malignancies. Here, we present a protocol for the development and characterization of two mouse models designed to simulate clonal hematopoiesis and acute myeloid leukemia. We describe steps for model generation, monitoring clonal expansion, harvesting, fixation, and staining of bone marrow and spleen tissues. We specifically focus on the visualization and analysis of p53 mutant clonal expansions, providing a comprehensive protocol for studying these phenomena in mouse models. Lignin is a complex, random, and heterogeneous biopolymer in lignocellulosic biomass. It has significant potential for conversion into valuable chemicals and fuels. However, its natural recalcitrant structure has made selective depolymerization challenging. This dissertation presents a detailed investigation into the mechanisms of lignin disassembly and structural evolution under various catalytic and solvent conditions. The research spans three key studies. Low back pain (LBP) is the number one musculoskeletal cause of disability with a lifetime prevalence of up to 85%. The causes of LBP are multifactorial with potential contributions from factors such as tissue injury and degeneration as well as social economic and psychological factors. Attenuation compensation (AC) is beneficial for visual interpretation and serves as a prerequisite for quantification tasks in single-photon emission computed tomography (SPECT) imaging. However, conventional AC methods typically rely on a separate X-ray CT component, leading to multiple challenges. In this context, studies have shown that scatter-window projection data contains information to estimate the attenuation distribution. Influenza A virus (IAV) poses substantial epidemiological burdens around the world, with an estimated annual global mortality rate in the hundreds of thousands. Its rapid rate of mutation gives it a propensity for causing seasonal epidemics and occasional pandemics. As an airborne virus, IAV is most commonly transmitted through droplets and aerosols. Antibodies play a crucial role in the immune defense against IAV infection. Low back pain (LBP) is the number one musculoskeletal cause of disability with a lifetime prevalence of up to 85%. The causes of LBP are multifactorial with potential contributions from factors such as tissue injury and degeneration as well as social economic and psychological factors. Intervertebral disc (IVD) degeneration is associated with 26-40% of LBP cases and in a 2015 meta-analysis, it was estimated that a total 403 million people suffer from symptomatic IVD degeneration. The discrete element method (DEM) is a widely used numerical approach for simulating and analyzing the behavior of granular materials in various applications. This research focuses on the iron ore beneficiation process, addressing the clumping issue in cohesive iron ores from existing crushing processes through a novel double-layered disperser design. The significance of this study lies in its potential to enhance the efficiency of the existing crushing phase and improve the linkage to subsequent purification stages that target smaller-sized iron ore pellets, thereby enhancing the overall efficiency of the ore beneficiation process and achieving significant energy savings. Lignin is a complex, random, and heterogeneous biopolymer in lignocellulosic biomass. It has significant potential for conversion into valuable chemicals and fuels. However, its natural recalcitrant structure has made selective depolymerization challenging. This dissertation presents a detailed investigation into the mechanisms of lignin disassembly and structural evolution under various catalytic and solvent conditions. The research spans three key studies. In Chapter 2, operando magic-angle spinning (MAS) solid-state nuclear magnetic resonance (NMR) spectroscopy was employed to study reaction pathways and kinetics of lignin model polymers during catalytic hydrogenolysis. Attenuation compensation (AC) is beneficial for visual interpretation and serves as a prerequisite for quantification tasks in single-photon emission computed tomography (SPECT) imaging. However, conventional AC methods typically rely on a separate X-ray CT component, leading to multiple challenges. In this context, studies have shown that scatter-window projection data contains information to estimate the attenuation distribution. Furthermore, large amounts of SPECT emission data in both photopeak and scatter-energy windows and corresponding CT scans are available. Economic choices entail computing the values of different options and then making choice by comparing values. Work in primates and rodents has shown that these mental processes rely on the OFC. Neurophysiology studies identified in OFC distinct groups of neurons encoding the value of individual options, the binary choice outcome, and the chosen value. These variables capture both the input and the output of the decision process, suggesting a decision circuit existing in OFC. The landscape of spatial biology research has transformed drastically in recent years allowing a comprehensive understanding of cellular organization and molecular mechanisms in various biological context. This thesis presents three complementary studies that advance our understanding of spatial organization in normal tissue biology and tumor evolution, utilizing cutting-edge spatial multi-omics approaches. The first study presents a comprehensive investigation of kidney sexual dimorphism across the lifespan, integrating six distinct technological platforms: single-nucleus transcriptomics, epigenomics, Visium spatial transcriptomics, Xenium single-cell resolution spatial transcriptomics, conventional immunofluorescence imaging, and co-detection by indexing (CODEX) multiplexed protein imaging. Social support signifies a system that provides for family, relatives, and friends or is derived from external sources such as social services or health promotion networks and can reduce depression and increase well-being.
