Age is a significant risk factor for mild cognitive impairment (MCI) and Alzheimer's disease (AD) and identifying brain age patterns is critical for comprehending the normal aging and MCI/AD processes. Prior studies have widely established the univariate relationships between brain regions and age, while multivariate associations remain largely unexplored. Herein, various artificial intelligence (AI) models were used to perform brain age prediction using an MRI dataset (

BACKGROUND: High-grade gliomas (HGG) have a poor prognosis despite aggressive treatment. Severe, persistent lymphopenia occurring in HGG patients after concurrent chemoradiation is associated with worse survival. NT-I7, a long-acting interleukin-7 analog, has been shown to increase CD4 and CD8 counts in healthy, septic, and HIV-positive adults. This multi-institutional, NCI-funded dose-escalation trial is the first to evaluate NT-I7 safety and activity in HGG patients with severe treatment-related lymphopenia (TRL) and the effect of co-administered glucocorticoids.
METHODS: Eligible HGG patients had CD4 counts < 300 cells/mm
RESULTS: NT-I7 was well tolerated in all 12 patients (median age 64; median CD4 count 161 cells/mm³) before the …

BACKGROUND: Essential tremor (ET) is the most common movement disorder in adults, but its pathogenesis is incompletely understood. Deep brain stimulation of the ventral intermediate thalamic nucleus (VIM DBS) provides effective treatment for medically-refractory cases. We aimed to determine how pre-surgical resting-state functional connectivity (FC) in medically-refractory ET relates to VIM DBS clinical response.
METHODS: We analyzed resting-state FC MRI in 21 participants with medically-refractory ET who subsequently underwent VIM DBS and 34 matched controls. We applied rigorous quality assurance to minimize motion artifact. Whole-brain correlation matrices were computed across 300 cortical, subcortical, and cerebellar regions and compared across groups …

Histone post-translational modifications (PTMs) regulate gene expression patterns through epigenetic mechanisms. The five histone proteins (H1, H2A, H2B, H3, and H4) are extensively modified, with over 75 distinct modification types spanning more than 200 sites. Despite strong advances in mass spectrometry (MS)-based approaches, identification and quantification of modified histone peptides remains challenging because of factors, such as isobaric peptides, pseudo-isobaric PTMs, and low stoichiometry of certain marks. Here, we describe the development of a new high-throughput method to identify and quantify over 150 modified histone peptides by LC-MS. Fast gradient microflow liquid chromatography and variable window sequential windows acquisition of …

Copyright law is under attack. Scholars and activists have long argued that American copyright law is a shambolic mess—vague, unpredictable, and startlingly overbroad. But amidst the swirling chaos, one core principle has remained intact: the idea that copyright attaches only to intangible goods. In theory, copyright resides in an intangible work of authorship, not a physical artifact. It is the intellectual creation, rather than the material copy, that gives rise to copyright protection. Copyright law thus rests on a stark distinction between the intellectual property of authors and the personal property of consumers—in short, between the intangible work and the …

Filipinx Americans In U.S. Higher Education: A Mixed-Methods Critical Study Of Student Engagement Using The Culturally Engaging Campus Environments Model, Gerald Ariston Doton Corporal
Educational and Organizational Learning and Leadership Dissertations
Who are Filipinx American college students and what are their experiences in U.S. higher education? The academic literature emphasizes the importance of disaggregating empirical data on sub-populations within the Asian American and Pacific Islander racial diaspora to fully understand their experiences in a variety of contexts (Teranishi et al., 2015). In higher education administration, there is limited empirical data on Asian American and Pacific Islander college students and it is further limited when examining specific racial subgroups, such as Filipinx Americans (Museus & Maramba, 2011). This has implications for administrators in U.S. postsecondary institutions, particularly regarding their capacity to appropriately …

Heart failure (HF) affects 6 million people in the US, and type 2 diabetes (T2D) is a significant risk factor for HF. Since 2008, regulatory agencies have required cardiovascular safety trials for new T2D therapies, and these have consistently demonstrated the cardiovascular benefits of glucose-lowering drugs. Sodium-glucose cotransporter inhibitors (SGLTi) and glucagon-like peptide-1 (GLP-1) receptor agonists (RAs)/glucose-dependent insulinotropic polypeptide (GIP) reduce major adverse cardiovascular events and deaths, regardless of diabetes status, and improve cardiorespiratory fitness. SGLTis, such as dapagliflozin and empagliflozin, benefit patients with reduced and preserved ejection fraction, significantly reducing HF hospitalizations and cardiovascular deaths. The DAPA-HF and EMPEROR-Reduced …

Introduction: Previous literature has shown that clear cell renal cell carcinoma (ccRCC) is becoming a more prevalent diagnosis and that the incidence and mortality differ both regionally and racially. While the molecular profiles for ccRCC are studied regionally through biopsy and sequencing techniques, the genomic landscape and ccRCC diversity data are not well studied. We conducted a review of the known genomic data on 6 of the most clinically relevant DNA biomarkers in ccRCC: von Hippel-Lindau (vHL), Polybromo-1 (PBRM1), Breast Cancer Gene 1-Associated Protein 1 (BAP1), Histone-Lysine N-Methyltransferase Domain-Containing 2 (SETD2), Mammalian Target of Rapamycin (mTOR), and Lysine-Specific Demethylase 5C …

Ionizing radiation (IR) poses a dual challenge in medicine; while essential for cancer therapy, it inflicts collateral damage to normal tissues, particularly the gastrointestinal (GI) tract. High-dose IR triggers acute radiation syndrome (ARS), characterized by crypt stem cell depletion, mucosal barrier disruption, inflammation, and potential progression to fibrosis and secondary malignancy. Emerging evidence identifies the epithelial kinase doublecortin-like kinase 1 (DCLK1)—highly expressed in GI tuft cells and cancer stem-like cells—as a master regulator of post-IR responses. DCLK1 integrates DNA repair (via p53/ATM), and survival signaling (via NF-κB, TGF-β, and MAPK) to promote epithelial regeneration, yet these same mechanisms contribute to …

Quantifying Links Between Phytoplankton Physiology And The Environment, Gabrielle M. Armin
Quantifying Links Between Phytoplankton Physiology And The Environment, Gabrielle M. Armin
Open Access Dissertations
Phytoplankton are microscopic, primary producers in aquatic ecosystems worldwide. They are key links in global biogeochemical cycles, converting inorganic compounds into bioavailable nutrients which fuel fisheries. Through photosynthesis, phytoplankton play an essential role in the carbon cycle and marine biological pump, drawing carbon from the atmosphere, which is eventually sequestered in the oceans, regulating our Earth’s climate. Due to their influential role in large scale processes, there have been many efforts to characterize and quantify phytoplankton impacts to ecosystem health and global nutrient cycles. Often, phytoplankton physiology has been highly simplified in global or ecosystem models, limiting their representation to …

The latest edition of Global Cancer Statistics released by the American Cancer Society reported approximately 20 million newly diagnosed cancer cases worldwide in 2022 with 9.7 million deaths. This ranks cancer as the second leading cause of death worldwide. For the majority of cancer patients, the standard treatment approach is chemotherapy and/or radiation therapy, and while this has been successful in some patients, a lack of specificity to cancer cells causes severe side-effects when these agents damage healthy cells. Despite extensive efforts that have gone into developing targeted cancer therapeutics that block functions specific to cancer, they have not been …

Electroencephalography (EEG) and functional near-infrared spectroscopy (fNIRS) are complementary noninvasive neuroimaging modalities that have contributed greatly to our understanding of the mechanisms of auditory processing. As an emerging technology, fNIRS has produced promising insights into the mechanisms of auditory processing, although considerable work remains to characterize hemodynamic responses in auditory tasks. In addition, the complementary nature of EEG and fNIRS and the relationships between the signals they record have been underexplored. In particular, nonlinear interactions between these signals remain under-characterized, despite the potential benefit to future multimodal neuroimaging studies. A challenge in understanding these interactions is the influence of systemic …
Light, Symbiosis, And The Biology Of Corals, Taylor R. Lindsay
Light, Symbiosis, And The Biology Of Corals, Taylor R. Lindsay
Open Access Dissertations
Light is critical for the survival of most shallow water scleractinian corals. Corals are mixotrophs, acquiring energy via both feeding on water column plankton and photosynthesis via a symbiotic relationship with dinoflagellates living within their tissues. Corals rely heavily on photosynthetically derived metabolites through this symbiotic relationship, intrinsically linking their biology to light availability. Corals are known to modulate their biology to maximize light capture. However, there is a limited understanding of the interaction between morphology, physiology, and energetic strategy, as well as how light may affect these relationships. In Chapter 1, we quantified the morphology, physiology and energetic …

Development Of A Scituate Reservoir Mitigation Model For Assessing Flood Control During Extreme Scenarios, Supria Paul
Open Access Dissertations
Effective reservoir management is essential for mitigating downstream flooding, especially with the increasing frequency and intensity of extreme weather events impacting major drinking water reservoirs such as the Scituate Reservoir in Rhode Island. These reservoirs provide water for large populations but can be overwhelmed by intense precipitation, flooding and land cover changes. Climate change is expected to increase precipitation in the study area located in New England, highlighting the need for improved reservoir systems, especially given the aging dams and higher risk of failures. Additionally, land cover changes, like the widespread defoliation caused by the Gypsy Moth infestations, affect water …
Development And Characterization Of Polysaccharide-Based Controlled Drug Delivery Platforms, Amanda Pepler
Development And Characterization Of Polysaccharide-Based Controlled Drug Delivery Platforms, Amanda Pepler
Open Access Dissertations
Drug delivery research focuses on overcoming challenges in patient treatment. Most therapeutics are delivered systemically through oral or intravenous routes. However, these administration methods are often inefficient due to therapeutic hydrophobicity, poor bioavailability, and liver metabolic clearance, necessitating frequent dosing. When treatment plans require frequent dosing, patients experience significant fluctuations in therapeutic concentration in the bloodstream, which can increase adverse side effects. These factors contribute to low patient compliance. Therefore, the core focus of drug delivery research is to develop new administration routes for therapeutics. Drug delivery systems (DDS) that can target therapeutics to a localized region offer advantages over …

Impacts Of Multiple Coastal Stressors Across Life-History Stages In The Eastern Oyster, Amaelia Zyck
Open Access Dissertations
Urbanized estuaries are characterized as a complex of biotic and abiotic stressors, which currently challenge marine life and are expected to intensify and become increasingly unpredictable under the ongoing impacts of climate change. The persistence of coastal species that inhabit these stressful environments will ultimately depend on their ability to adapt. Many of these species have complex life cycles, featuring distinct morphological and physiological developmental stages that can exhibit unique responses to environmental pressures. However, since all stages share the same genome, selective pressures acting on one stage can have cascading effects throughout the life cycle. The larval stage, being …

Novel Methodology And Proof-Of-Concept For Proton Ct Using Spot Profile Rather Than Bragg Peak Location, Brett Albert Ruben
Open Access Dissertations
Proton radiation therapy offers a highly precise method for cancer treatment, theoretically providing superior dose deposition characteristics compared to conventional X-ray radiation therapy. Its steep and finite dose fall-off region allows for superior tumor coverage while reducing, though not eliminating, dose to surrounding healthy tissue. However, its full potential is limited by range uncertainties, particularly those arising from the reliance on X-ray-based imaging methods called computed tomography (CT) for proton-based treatment. CT image Hounsfield Units (HU) must be converted to stopping power ratios (SPR) as these ratios are the clinically relevant value for accurately planning particle interactions during proton therapy. …

Intersections Of Inattention: A Qualitative Inquiry Into The Experiences Of Multiply Marginalized Students In Community College Settings, Jana N. Knibb
Open Access Dissertations
Educational research has examined the impact of expanded access to higher education for historically marginalized groups. However, research on the experiences of students who are both racially minoritized and dis/abled is limited. Findings suggest that, due to their intersectional identities, multiply marginalized dis/abled students (MMDS) navigate policies and practices related to racist ableism, which further marginalize them and create additional barriers in their higher education experiences. Expanding this research requires theoretical frameworks that account for students' identities, experiences, and barriers in higher educational spaces and contexts. This qualitative study, grounded in Dis/ability Studies and Critical Race Theory in Education (DisCrit), …

Open Access Dissertations
This dissertation investigates how twenty-first-century prose fiction navigates the ecological and temporal dimensions of planetary crisis. In response to accelerating pressures - climate change, ecological collapse, and the intensification of capitalist modernity - literary studies has turned increasingly to the planetary as a conceptual frame. Yet this shift has often outpaced the development of adequate critical vocabularies and narrative methodologies. By drawing on ecocriticism, narrative theory, phenomenology, and related fields, Textual Sediments proposes a new framework for reading what it terms the planetary novel: a sedimentary genre characterized by temporal layering, formal hybridity, and a recursive engagement with scale. Quantifying Mixotrophy Potential As A Function Of Environmental And Biological Drivers In Coastal And Estuarine Plankton Communities, Erica Ewton
Open Access Dissertations
The discovery of mixotrophic plankton has bridged gaps in nutrient cycling budgets and altered the understanding of food webs and energy transfer. Mixotrophic plankton are capable of simultaneous or sequential photosynthesis and heterotrophy, providing multiple avenues of nutrition and thus reducing limitations that affect specialists. These energy dense cells provide increased biomass for higher trophic levels, which may increase organism sizes throughout the food web. Additionally, they may be physically denser, which alters biogeochemical cycling including carbon sequestration. The work presented here uses LysoTracker Green, an acidotropic stain that attaches to phagocytic vacuoles within potentially phagotrophic small (< 20 µm diameter) phytoplankton communities (‘mixoplankton’), to better understand their abundance, relative abundance, and growth rates in relation to the matrix of natural conditions they inhabit. Understanding the prevalence and drivers of mixoplankton is vital to better parameterize biogeochemical cycling and energy transfer in the ocean and for models that predict the fate of future oceans. In the first study, I compared the abundance of mixoplankton in the Northeast U.S. Shelf (NES) and Coastal California Current System (CCS) during two cruises in the NES in summer 2021 and one cruise in the CCS in summer 2022. These two contrasting coastal systems span the breadth of environmental conditions observed in temperate marine ecosystems and thus provided an opportunity to examine indicators of mixoplankton across temperate coastal regions. I discovered higher mixoplankton abundance in the NES than CCS yet similar population sizes of nanoeukaryotic photosynthesizers, resulting in higher relative abundance of mixoplankton in the NES than CCS. Across both systems, I found increased mixoplankton abundance associated with decreasing temperature and combined nitrate + nitrite concentrations (N+N), and increasing light intensity and Synechococcus abundance …
