Trypanosomiasis, caused by protozoan parasites of the Trypanosoma genus, remains a significant health and veterinary burden in sub-Saharan Africa, with Human African Trypanosomiasis (HAT) and Animal African Trypanosomiasis (AAT) posing ongoing threats to underserved populations. Despite advances in therapeutics, the lack of sensitive and scalable diagnostic tools continues to limit early detection, effective disease management, and evaluation of emerging vaccine candidates. Current diagnostic methods such as light microscopy and serological tests frequently fail to detect low levels of parasitemia during the early stages of infection, thereby delaying intervention and increasing the risk of disease progression and transmission.

Recent research on human disease networks (HDNs) has revealed the importance of jointly analyzing multiple diseasesand their interconnections, yet many existing approaches focus on molecular-based data and do not fully capture non-molecular relationships. Building on the concept of the phenotypical HDN, we leverage Medicare inpatient claims data to construct a comprehensive disease co-occurrence network. In our framework, two diseases are deemed interconnected if their observed probability of co-occurrence significantly deviates from that expected under independence. We previously found that the levels of soluble amyloid precursor protein α (sAPPα) are partly regulated by acetylcholinesterase (AChE) in human non-small cell lung cancer cell lines A549 (p53 wild-type) and H1299 (p53-null). In this study, we hypothesize that sAPPα is partly regulated by leptin, a hormone classically recognized for its anorexigenic role in appetite regulation— yet recently shown to play a role in cancer cell signaling. It was observed that cells treated with leptin exhibited greater concentrations of sAPPα and lower concentrations of Aβ40/42 in the media compared to those of cells left untreated. Introduction: Advanced age is a primary risk factor for many chronic diseases and conditions; however, age-related immune dysregulation is not well understood. Animal models, particularly those that resemble human age-related physiological changes, are needed to better understand immunosenescence and to improve health outcomes. Here, we explore the utility of the olive baboon (Papio anubis) in studying age-related changes to the immune system and understanding mechanisms of immunosenescence.

Methods: We examined immune cell, inflammatory responses, cytokines, and cortisol levels using hematology and flow cytometry, mitogen stimulation, multiplex cytokine assay, and cortisol immunoassay.

Borderline personality disorder (BPD) is a severe and prevalent mental illness that is three times more likely to be diagnosed in women than men. BPD research has attempted to explain this gender disparity, with clinician gender bias emerging as a possible factor. Yet, no research to date has examined the role of social status in the overdiagnosis of women with BPD. There is merit in examining BPD through a class lens as low SES is a risk factor for BPD, poverty and income inequality disproportionately affect women, and low status is associated with stereotypical traits of femininity. This thesis examines the contemporary artist Patricia Piccinini and her posthuman hybrid creations within the context of a feminist ethic of care. Largely referring to scholarship by the political scientist Joan C. Tronto, I argue that Piccinini expands the definition of an ethic of care to include non-human entities as well as human ones. CRISPR-Cas12a is a rapidly evolving technology for improved gene editing in cells, as well as in vivo. The system, consisting of Cas12a nuclease and approximately 40-nt long guide RNA (gRNA), can efficiently recognize and cut genomic double-stranded DNA (dsDNA). Chemical modifications to the gRNA can potentially improve its dsDNA binding and enhance Cas12a-mediated nuclease activity. In this work, 5-methyl cytidine (m5C) and N6-methyl adenosine (m6A) were explored to improve the binding between gRNA and target DNA. Cancer cells often exhibit a Warburg-like metabolism, which includes increased fatty acid synthesis and storage that help them survive in hypoxic conditions. This shift is marked by the heightened synthesis and accumulation of fatty acids, acting as a survival mechanism in low-oxygen environments (Baumann et al., 2016). Interestingly, deep-diving whales may utilize a similar metabolic pathway to produce wax esters during their prolonged dives.

Non-tuberculosis mycobacteria (NTM) represent a diverse repertoire of species, most of which are ubiquitous in the environment. Some NTM species, such as Mycobacterium abscessus, can cause life-threatening infections in people with underlying medical conditions. The clinical incidence of such infections is on the rise, posing an emerging threat to public health. M. abscessus is characterized by intrinsic and acquired antibiotic resistance, complicating treatment strategies. The transcription factor p63 is critical for development of the epidermis and other epithelial-related structures. Mutations in the DNA-binding domain of p63 lead to a range of human developmental disorders characterized by symptoms such as ectrodactyly, painful skin dysplasia, and cleft lip and palate. The TP63 gene is also amplified in squamous cell carcinomas, contributing to tumor pathogenesis. While the importance of p63 in human health is well established, the mechanisms underlying its control of an essential gene network remain unclear. Puberty is a postnatal developmental process required to initiate key sexual maturation events that are necessary for the propagation of species. Signals to initiate these events first begin with the Gonadotropin releasing hormone-1 (GnRH-1) neurons within the hypothalamus. The hormone GnRH-1 released by these neurons is crucial for puberty, fertility and reproductive processes by establishing the hypothalamic-pituitary-gonadal (HPG) axis, regulating release of sex hormones (Progesterone, Estrogen, Testosterone). Early-life stress is a risk factor for increased susceptibility to stress- and trauma-related disorders, including Posttraumatic Stress Disorder (PTSD). PTSD is characterized by exposure to trauma that results in exaggerated and extinction-resistant fear responses that persist for over a month. Understanding how the severity of early-life stressors in animal models impacts adult behavior, cognition, and underlying neurobiology could aid in the development of more effective PTSD treatments.

Background: People living with diagnosed HIV (PLWDH) have a higher chance of co-infections and co-morbidities than the general population. When SARS-CoV-2 was first reported in China in December 2019 and later declared a pandemic, it was speculated that PLWDH might be more prone to SARS-CoV-2 infection and severe disease outcomes given their increased vulnerability to other respiratory infections like influenza, pneumococcal pneumonia and tuberculosis. This dissertation examines whether and how experiences of grandparenthood—including becoming a grandparent, providing grandchild care, living with grandchildren, and exchanging intergenerational support—affect the well-being of middle-aged and older adults in China. Drawing on data from the China Health and Retirement Longitudinal Study (CHARLS, 2011–2018), a nationally representative survey of Chinese adults aged 45 and older, the study employs Ordinary Least Squares (OLS) regression and random effects models to investigate the associations between multigenerational family relationships and depressive symptoms among this population.

Grounded in developmental psychobiology, this research aimed to elucidate how birthing people’s physiological regulation during pregnancy may shape the early emergence of socioemotional development in early infancy. The present study examined how birthing people’s prenatal parasympathetic nervous system (PNS) activity, specifically baseline high-frequency heart rate variability (HF-HRV) and HF-HRV withdrawal, and postnatal sensitivity contributes to infant temperamental reactivity at six months of age. Zika virus (ZIKV) remains a significant public health concern due to major gaps in our understanding of its infection mechanisms and immune evasion strategies. Despite its link to severe complications such as microcephaly and Guillain-Barré Syndrome (GBS), no vaccine or targeted antiviral therapies exist. Compounding this concern is the expanding habitat of ZIKV-carrying mosquito vectors, driven by climate change, which increases the risk of future outbreaks in previously unaffected regions.

Controlled research shows that cognitive-behavioral therapy (CBT) is an effective treatment for insomnia, yet clinician experiences with applying CBT in routine settings has received limited attention. This study replicated and extended upon the Two-Way Bridge initiative (Goldfried et al., 2014), which aimed to establish a dialogue between researchers and clinicians on the identification of variables that promote and interfere with the effectiveness of evidence-based therapies. This survey study asked clinicians to identify variables they have encountered that make it challenging to implement CBT and related interventions for insomnia.
