In the United States, broccoli is a popular vegetable, important for its nutrition and economic contributions alike. However, its future production is at risk as broccoli prefers cooler climates, with an optimal temperature range of 15 - 23°C (59 - 73°F) for growth, especially in the weeks leading up to harvest. Temperatures above this threshold lead to failure to develop, damage, and unmarketability. There has also been an emphasis on bringing broccoli production back to the East coast from California, as climate events threatened Californian agriculture and local food has become popular, necessitating outside of season broccoli production during times …

Cellulose Synthase-Like Ds (CSLDs) are cellulose synthase known to participate in cell plate formation and cell wall remodeling. In the moss model, Physcomitrium patens, double csld2/6 knockout (KO) mutants were previously shown to have gametophore leaf phenotypes that are consistent with defects in cytokinesis, including irregular cell shape, cell separations, and incomplete midrib formation (Wu et al., 2023a). When these mutants were grown on ammonium-free medium to promote gametophore development, the severity of the phenotype was more extreme, including incompletely formed cell plates and multinucleated cells (A. This research study examines the challenges Black women encounter when in predominantly white, male academic and professional science settings, where they experience dual discrimination because of their race and gender. These biases perpetuate the negative stereotype that Black women are less competent than white men. This mixed methods study examines how Black women navigate their race, gender, and science identities within predominantly white institutions through their doctoral programs and professional science careers. The study implements Critical Race Mixed Methodology to combine survey data for a wide reach with interview data for detailed exploration. The study involved eight Black women science …

Navigating Stress In Higher Education Leadership: The Role Of Mindful Meditation For Women Administrators, Christine T. Harper

Women higher education administrators encounter distinct stressors which can significantly impact wellbeing and lead to burnout. These challenges include implicit bias, difficulty achieving work-life balance, and increasing institutional expectations. This study utilizes a Mixed Methods Action Research (MMAR) framework to assess stress levels, trait mindfulness, coping mechanisms, and the effectiveness of a structured mindfulness-based intervention. While the quantitative analysis revealed only a slight increase in trait mindfulness and no notable reduction in stress levels, the qualitative findings showcased meaningful personal benefits to the participants. They reported heightened awareness of stress triggers, improved emotion regulation, and an ability to apply the …

Magnetic heating mediated by magnetic iron oxide nanoparticles (MION) has several clinical applications including cancer thermal therapy, tissue thawing for organ preservation, and magnetogenetics. However, current models of magnetic heating, including the widely used linear response theory, inadequately predict MION heating efficiency. An experimental study covering alternating magnetic field (AMF) strengths and frequencies ranging from 3.98 – 27.85 kA/m and 103.6 – 984.1 kHz, respectively, with a range of MION core diameters 9.5- 30.3 nm was conducted to elucidate the relationships between these AMF parameters and MION sizes and the resulting heating efficiencies. Pandemic surveillance is a cornerstone in public health safety, as it enables identification of pathogens circulating within a community before they become widespread outbreaks. Wastewater based epidemiology has blossomed in recent years due to its success in the SARS-CoV-2 pandemic, but this form of public health surveillance has a global equity problem. A large majority of the wastewater-based surveillance work has been done in large metropolitan cities with access to advanced testing infrastructure, which poses a challenge as we consider where pandemics have historically emerged. Our goals were to carry out analyses of a multi-year δ¹⁸O and δ²H dataset of precipitation from central Kentucky and to analyze the source of precipitation for storm events from 2017 to 2022. δ¹⁸O and δ²H were measured for 244 weekly precipitation samples collected by the National Atmospheric Deposition Program at Mackville, Kentucky, USA. Modelling precipitation source was carried out using HYSPLIT model for 414 events. Results of δ¹⁸O and δ²H from central Kentucky show: consistent seasonality, including summer highs and winter lows for δ¹⁸O and δ²H; correlation with air temperature; and …

No Hope?: Queer Negativity, Afropessimism, And The Limits Of Futurity, Colton Barton

Since their inceptions, black and queer studies have sought to articulate the positions of “black” and “queer” within American social life—often converging on the goal of liberation. This thesis examines the intersection of two “negative” theoretical frameworks—Afropessimism and anti-social queer theory (or “queer negativity”)—to interrogate both the limits and generative potential of radical negativity. Though emerging from distinct intellectual traditions, these frameworks conceptualize blackness and queerness as constitutive negations—gaps within the dominant social order that render those so marked inassimilable to normative notions of humanity. While the convergences of these theories can be illuminating, their shared pessimism risks obscuring alternative …

Lake Tanganyika is a large, ancient freshwater body with exceptional biodiversity that is located in the East African Rift System. Lake Tanganyika’s size and longevity make its sedimentary record useful for understanding environmental changes that cross spatial and temporal boundaries. Lake Tanganyika is known to be susceptible to global warming given its tropical location. Over 12 million people living within Lake Tanganyika’s watershed rely on fishing for their income and nutrition. Understanding the long-term responses of ecosystems to climate variability is crucial for predicting and mitigating future changes in the Afrotropics, a region where a significant portion of the population is highly vulnerable to extreme climatic events. Lake Tanganyika, the largest rift lake in East Africa, offers a unique sedimentary archive that can be used to investigate the limnological changes, hydroclimatic shifts, and vegetation dynamics across various timescales. Karst aquifers represent some of the world’s most productive yet vulnerable groundwater systems, supplying freshwater to approximately 10% of the global population across 15% of Earth’s ice-free land surface. Their distinctive hydrological characteristics— including rapid recharge, preferential flow paths, and limited natural filtration— create unique vulnerabilities to both climate variability and contamination that pose significant challenges for sustainable water resource management. This dissertation presents a comprehensive multi-scale analysis of karst aquifer vulnerability through three interconnected studies that examine climate change impacts, microbial contamination dynamics, and contaminant transport processes across diverse geographic and hydrogeologic settings.

What does it mean to “eat right?” The notion is, of course, culturally, historically, nationally, politically, ethnically, and class determined. For the purposes of this project, however, I am particularly interested in changing notions of what it means to “eat right” within the post-World War II to the early new millennium American context, as such notions are represented within the prescriptions of popular culture. Epigenetic clocks are algorithms that use epigenetic biomarkers of aging to calculate an organism’s ‘biological age.’ Through discursive analysis of commercially available epigenetic clock marketing materials, scientific articles, and interviews with actors in the longevity biotechnology industry, this study examines the commercialization of epigenetic clocks in the form of consumer test-kits. The study argues that longevity biotechnology companies leverage the malleable and fluid qualities of epigenetic markers of biological age to extend the domains of profitable therapies. Media consumption reflects aspects of identity while also assisting in identity formation (Koller, 2008; Zemmels, 2012; Wu, 2006). This thesis examines the phenomenon of lesbians who engage with men-loving-men (MLM) media. The practice seems odd due to the contrast of lesbians desiring women for their social, platonic, romantic, and sexual relationships, but this thesis finds that repeated consumption of media that display men and their desires can index lesbianism. This practice helps lesbians construct their identities through three processes of indexicality: indexical disjuncture, indexical bleaching, and indexical denial (Barrett & Hall, 2024). “A True And Broad Sympathy:” Thomas Hughes, Medievalism, And Social Justice In The Nineteenth Century, Sarah E. Stapleton

Thomas Hughes, author of Tom Brown’s Schooldays (1857), used medievalism to help solve social problems of the Nineteenth Century. This contravenes the traditional view of medievalism, especially Anglo-Saxonism, since others have used it to justify the subjugation of people of lower social status, namely the poor in England, enslaved African Americans in the United States, and Indians under British rule.
While the use of medievalism is veiled in Hughes’ work, the subtext of Tom Brown’s Schooldays demonstrated Hughes’ Anglo-Saxonism. Tom Brown and the intertextual material surrounding his narrative exhibit the characteristics of Anglo-Saxonism through cultural primitivism, the concept of allodial, …

Protein Arginine Methyltransferase 5 (PRMT5) is a member of PRMT family that controls both intrinsic and extrinsic oncogenic processes [1]. Protein arginine methyltransferase 5 (PRMT5) is a type II methyltransferase that exerts widespread influence over gene expression, splicing, DNA repair, and immune regulation through symmetric dimethylation of arginine residues on histone and non-histone proteins [2]. The addition of methyl groups induces gene silencing through producing dictatorial histone marks like H2AR3ne2s, H3R8me2s and H4R3mes [3], which act as epigenetic repressive marks on gene promoters. Prior studies indicated Borrelia burgdorferi SpoVG protein was regulated, SpoVG binds specifically to a number of nucleic acids, and SpoVG binds to the non-variable section of the major virulence factor vlsE. Although a direct transcriptional activator or inhibitor was not elucidated, it is shown that SpoVG is regulated based on cell culture density independent of the growth rate of B. burgdorferi, indicating that SpoVG regulation is outside the scope of the alternative sigma factor RpoS. Using a dCas9 CRISPRi construct to target SpoVG resulted in protein expression trending towards upregulation of the BosR-RpoS regulon, and a marked decrease …

Psychedelics are rapidly garnering research attention for their ability to successfully treat a variety of psychiatric diseases after one or a handful of treatments in recent clinical research. Among the conditions treated is substance use disorder (SUD), which afflicted 48.5 million (16.7%) Americans aged 12 and older in 2023 (NSDUH). As clinical trial results continue to show lasting positive effects of psychedelic psychotherapy, there remains an enormous gap in knowledge regarding how these drugs cause lasting changes in the brain to achieve these therapeutic effects. Efforts to improve treatment of spinal cord injury (SCI) often use animals to develop effective therapeutics, intending to improve the survival and the quality of life of persons with SCI. Over half of all SCIs in the United States occur at the cervical level and often cause locomotor deficits and life-threatening breathing dysfunction. Thus, my dissertation is focused on this important patient population.

Three related analyses were performed to try to understand the immediate effects of Medicaid expansion on hospital systems in Kentucky. Medicaid expansion led to a large, sudden increase in the number of individuals eligible for various health care needs. This sudden increase in demand for health services lead to initial hypotheses regarding hospitals’ ability to handle this demand. The first analysis examined hospital average length of stay (LOS) using a linear mixed-effects model. Length of stay was modeled longitudinally between 2013 and 2015 to determine if significant changes in the slope of LOS could be detected after expansion (2014). Separate …

Protein-protein interactions are critical in cellular signaling pathways. This project investigates binding interactions between SHP-2, a tyrosine phosphatase, and DDX3, an RNA helicase. There is a specific focus on the impact that gain-of-function SHP-2 mutations may impact binding to DDX3 via conformational change. Using immunoprecipitation protocols as well as western blotting we were able to provide insights into SHP-2 molecular mechanisms and subsequent binding to DDX3.

Maternal environmental exposures and inflammation are known to impact fetal development in utero as well as increase the risk for chronic diseases later in life. Therefore, it is essential that we understand how the maternal environment alters placental structure and immune function.
Therefore, we isolated fetal (chorionic villous) tissues from the placentas of healthy pregnant participants (control) and those with inflammatory conditions, including SARS-COV-2 infection, pregravid obesity, and opioid use disorder. Environmental Surveillance (ES) has emerged as a powerful tool to monitor the emergence and persistence of a pathogen within a facility or community using wastewater/water analysis. Over the past four years, implementation of ES has experienced an explosive acceleration, driven by efforts to monitor the spread of SARS-CoV-2, which has made ES a routinely used public health tool in many locations. Process-induced residual stress (RS) plays a critical role in determining the structural integrity, fatigue life, corrosion resistance, and dimensional stability of manufactured components. Tensile residual stress can be harmful to the machined workpiece by lowering fatigue strength, while compressive residual stress helps to prevent such issues. Therefore, accurate prediction and control of machining-induced residual stresses are essential for optimizing manufacturing processes and enhancing component performance. Precise fluid transport through microscale geometries can help optimize electrochemical and biochemical processes by maximizing the mass transfer rate of reactants. By introducing intricate structural features inside microchannels, it is possible to manipulate the fluid motion and direct the flow towards any surface of interest. In this study, we have demonstrated that rationally designed fluid pathways can improve the performance of electrochemical and biochemical systems.

This dissertation investigates the hemodynamics of pulsating blood flow in curved vessels through direct numerical simulations (DNS), with the overarching aim of uncovering fluid dynamic mechanisms linked to the onset and progression of aortic dissection in humans. The work is structured in two parts: (i) fundamental studies in idealized geometries and (ii) preliminary investigations in patient-specific anatomies. In the first part, curved pipe models representing the aortic arch are used to isolate the effects of pulsation frequency, amplitude ratio, and curvature ratio on transitional and turbulent flow dynamics. Cytoreductive surgery and heated intraperitoneal chemotherapy (HIPEC) is a treatment used for primary peritoneal malignancy or metastases, including colon cancer. We aimed to explore the effects of hyperthermia and/or mitomycin C (MMC) on mismatch repair (MMR) proteins and heat shock proteins (hsps) and the potential for post-HIPEC treatment with pembrolizumab treatment. Colon cancer cells were treated with hyperthermia and/or MMC for 90 minutes. Subcellular fractionation and immunofluorescent imaging were performed to identify protein abundance and localization of MSH2, MLH1, hsp70, and hsp27. Ovarian cancer typically presents at an advanced stage, has a poor prognosis, and is a leading cause of cancer-related deaths in women. Extracellular vesicles (EVs) are cell-derived membrane-bound nanoparticles that function in specific cell-to-cell communication and are under development as novel drug delivery vehicles and modulators of the tumor microenvironment. Artificial cell-derived vesicles (ACDVs) derived from M1 macrophages are able to repolarize M2 macrophages to the M1 phenotype and target tumor cells in in vitro studies.
