Purpose: Therapeutic depletion of immunosuppressive regulatory T cells (Treg) may overcome resistance to cancer immunotherapies. RG6292 is an anti-CD25 antibody that preferentially depletes Tregs while preserving effector T-cell functions in preclinical models. The safety, pharmacokinetics, pharmacodynamics, and antitumor efficacy of selective Treg depletion by RG6292 administered as monotherapy or in combination with atezolizumab were evaluated in two phase I studies.
Patients and methods: Adult patients with advanced solid tumors were administered intravenous RG6292, given every 3 weeks alone (study 1: NCT04158583, n = 76) or with 1,200 mg atezolizumab every 3 weeks (study 2: NCT04642365, n = 49). …

Importance: Molecular techniques, including next-generation sequencing, genomic copy number profiling, fusion transcript detection, and genomic DNA methylation arrays, are now indispensable tools for the workup of central nervous system (CNS) tumors. Yet there remains a great deal of heterogeneity in using such biomarker testing across institutions and hospital systems. This is in large part because there is a persistent reluctance among third-party payers to cover molecular testing. The objective of this Review is to describe why comprehensive molecular biomarker testing is now required for the accurate diagnosis and grading and prognostication of CNS tumors and, in so doing, to justify …

Patients with recurrent high-grade glioma (rHGG) have a poor prognosis with median progression-free survival (PFS) ofheterogenous, suggesting a clinical need for prognostic models. Bayesian data analysis can exploit individual patient follow-up imaging studies to adaptively predict the risk of progression. We propose a novel sample size analysis for Bayesian individual dynamic predictions and demonstrate proof of principle. We coupled a nonlinear mixed effects tumor growth inhibition model with a survival model. Longitudinal tumor volumes and time-to-progression were simulated for 2000 in silico rHGG patients. Bayesian individual dynamic predictions of PFS curves were evaluated using area under the receiver operating characteristic …

This study develops a comprehensive system dynamics model to predict and manage African armyworm (Spodoptera exempta) outbreaks, a major threat to cereal crops across Africa. We applied system dynamics approache with its archetypes (causal loop diagram (CLD), reinforcing (R) and balancing (B)) to analyse the population dynamics of the pest. The VENSIM modelling platform (Ventana Systems Inc., DSS 8.2) was used to implement the models and carry out the simulations. The research integrates extensive data from 1980 to 2023, encompassing the African armyworm's life cycle stages, climatic variables, and intervention strategies, to simulate potential outbreak scenarios and evaluate the impacts …

Vascular stabilization is a mechanosensitive process, in part driven by blood flow. Here, we demonstrate the involvement of the mechanosensitive ion channel, Piezo1, in promoting arterial accumulation of vascular smooth muscle cells (vSMCs) during zebrafish development. Using a series of small molecule antagonists or agonists to temporally regulate Piezo1 activity, we identified a role for the Piezo1 channel in regulating klf2a, a blood flow responsive transcription factor, expression levels and altered targeting of vSMCs between arteries and veins. Increasing Piezo1 activity suppressed klf2a and increased vSMC association with the cardinal vein, while inhibition of Piezo1 activity increased klf2a levels and …

Explainable Neural Networks With Guarantees: A Sparse Estimation Approach, Antoine Ledent, Peng Liu
Research Collection School Of Computing and Information Systems
Balancing predictive power and interpretability has long been a challenging research area, particularly in powerful yet complex models like neural networks, where nonlinearity obstructs direct interpretation. This paper introduces a novel approach to constructing an explainable neural network that harmonizes predictiveness and explainability. Our model is designed as a linear combination of a sparse set of jointly learned features, each derived from a different trainable function applied to a single 1-dimensional input feature. Leveraging the ability to learn arbitrarily complex relationships, our neural network architecture enables automatic selection of a sparse set of important features, with the final prediction being …

Foundation models have emerged as critical components in a variety of artificial intelligence applications, and showcase significant success in natural language processing and several other domains. Meanwhile, the field of graph machine learning is witnessing a paradigm transition from shallow methods to more sophisticated deep learning approaches. The capabilities of foundation models in generalization and adaptation motivate graph machine learning researchers to discuss the potential of developing a new graph learning paradigm. This paradigm envisions models that are pre-trained on extensive graph data and can be adapted for various graph tasks. Despite this burgeoning interest, there is a noticeable lack …

Metacognition And The 5cs Of Positive Youth Development Programs: A Review Of Metacognitive Interventions, Omolola A. Odejimi, Leslie F. Clark
Journal of Youth Development
Positive youth development (PYD) programs have increased positive outcomes for adolescents, however research on the underlying change processes in these programs remains limited. The 5Cs (Competence, Confidence, Character, Caring and Connection) of PYD are a framework of desirable outcomes for youth programs. Change to metacognition is proposed as a mechanism by which PYD programming accomplishes these outcomes. This review describes findings from metacognitive interventions with PYD-relevant outcomes in adolescents aged 10-24. Fifteen electronic databases were searched using keyword phrases related to “metacognitive intervention” and “adolescents”. Seventeen studies met inclusion criteria. Included studies addressed 2 of the 5 goals of PYD …

Personalized cancer vaccines (PCVs) can generate circulating immune responses against predicted neoantigens1-6. However, whether such responses can target cancer driver mutations, lead to immune recognition of a patient's tumour and result in clinical activity are largely unknown. These questions are of particular interest for patients who have tumours with a low mutational burden. Here we conducted a phase I trial (ClinicalTrials.gov identifier NCT02950766) to test a neoantigen-targeting PCV in patients with high-risk, fully resected clear cell renal cell carcinoma (RCC; stage III or IV) with or without ipilimumab administered adjacent to the vaccine. At a median follow-up of 40.2 …

Aim: Pre-degradation baseline conditions (references) provide crucial context for restoration actions. Here, we compare vegetation structure and its driving processes across the main pre-agricultural references discussed for temperate Europe: the Last Interglacial and the early-mid Holocene—before and after the arrival of Homo sapiens, respectively. Location: Temperate Europe. Time Period: The first ~4000–6000 years, excluding the initial early-successional phases, of the Last Interglacial (PAAZ III) and Holocene (8700–5700 BP). Major Taxa Studied: Plants. Methods: We use large datasets of pollen-based vegetation reconstructions (REVEALS) to compare open vegetation, light woodland and closed forest between the two periods. We use Random Forest modelling …

Estimating thresholds to distinguish between good and degraded ecosystem states is key for assessing and managing marine environments. Numerous methods are used to estimate thresholds; however, there is no standardized framework to evaluate their accuracy and reliability, which reduces the consistency and transparency of thresholds estimated for 'good' status. Statistical robustness of four methods was evaluated by varying stochastic noise, sample size, and shape of the pressure-state relationship of simulated indicator data. Range of natural variation and statistically detectable change methods, which quantify natural variation in undisturbed reference conditions, reliably estimated status thresholds for noisy, small datasets, but thresholds were …
Quantum Control And Simulation Using Hamiltonian Engineering In Solid-State Nmr, Linta Joseph
Quantum Control And Simulation Using Hamiltonian Engineering In Solid-State Nmr, Linta Joseph
Dartmouth College Ph.D Dissertations
Lattices of dipolar coupled nuclear spins in natural crystals are large, interacting quantum systems -- ideal platforms to simulate non-equilibrium many-body dynamics. Using the magnetic resonance toolkit, which includes Dynamic Nuclear Polarization (DNP), Hamiltonian engineering, and multiple-quantum Nuclear Magnetic Resonance (NMR) experiments, we study aspects of coherent control, manipulation, and readout of the complex dynamics of the spin system in NMR quantum simulation.
First, applying Hamiltonian engineering sequences, we control the system evolution. Specifically, we use a combination of numerical simulations and NMR experiments on adamantane to evaluate and compare the performance of several known sequences that aim to suppress …

Tick-borne diseases are a growing concern globally. Understanding the factors that influence tick ecology and people’s understanding of that ecology is crucial for developing effective tick management and disease prevention strategies. In this work, I investigated fundamental questions about the ecology of a primary vector of medically important pathogens in the United States, the blacklegged tick Ixodes scapularis Say, and identified common misconceptions about ticks and tick-borne diseases on social media. First, I examined the effects of temperature and moisture stress on the movement of adult female I. scapularis in laboratory experiments. Using a Drosophila Activity Monitor, ticks were exposed …

Learning verbs is an important part of learning one's native language. Prior studies have shown that children younger than 5 years can have difficulty in learning and extending new verbs. The current study extended these studies by showing children multiple events that can be compared during learning, including Japanese- and English-speaking children. In the study, 2-, 3-, and 4-year-olds saw two similar events and then one varied (progressive alignment) or three varied (low alignable) events in a learning phase before test, and this was repeated for four sets. Children were asked to extend these novel verbs in easy (non-cross-mapping) and …

Optimizing Biofungicide Use In Managing Winegrape Powdery Mildew On The Central Coast Of California, Edgar G. Godoy Monterroso
Master's Theses
Grapevine powdery mildew caused by the fungal pathogen Erysiphe necator is a persistent threat to wine grape production in California. While synthetic fungicides remain the primary management strategy, increasing concerns over pesticide resistance, environmental impact, and regulatory restrictions have driven interest in biofungicides as a sustainable alternative. However, their efficacy is inconsistent, requiring research into optimized application strategies, integration with synthetic fungicides, and their effects on disease suppression, grape yield, and fruit quality.

In Parafamily, Chen and Mulligan have managed to cover vast terrain, to catalogue important developments in law, to canvass insights from generations of thinkers, and to propose an ambitious yet pragmatic approach to the laws that govern human relationships, directly and indirectly. They do all this in a relatively short article composed of unusually readable prose. Quite a feat.
The focus of the piece is Chen and Mulligan’s contribution to the field of legal scholarship dedicated to relationships that go beyond the traditional nuclear family. As their title reveals, the authors contribute a new term and concept to this …

Evaluating Dry Air Personnel Decontamination Of Methyl Salicylate As A Chemical Agent Surrogate In Extreme Cold Environments To Reduce Airborne Risks Using A Manikin, Lance E. Campbell
Theses and Dissertations
This research investigated a mobile air shower as an alternative to water washing for personnel chemical decontamination in Arctic environments, where traditional methods like disrobing and rinsing are impractical. Interest in Arctic operations has grown with recent U.S. military focus on strategic advancements by Russia and China, yet research on air shower effectiveness for chemical decontamination remains limited. This study examined whether a commercial off-the-shelf air shower could effectively decontaminate methyl salicylate (MES), a surrogate for chemical warfare blister agents, from a manikin outfitted in military cold-weather gear. Researchers used a ppbRAE 3000 photo-ionization detector to measure MES concentrations following …
Molecular Scale Investigations Of Interactions Between Pfas And Concrete, Eric E. Keeler
Molecular Scale Investigations Of Interactions Between Pfas And Concrete, Eric E. Keeler
Theses and Dissertations
Per- and polyfluoroalkyl substances (PFAS), widely referred to as “forever chemicals,” exhibit high environmental persistence and potential health risks due to their robust carbon-fluorine bonds. These substances are prevalent in aqueous film-forming foams (AFFF), used in industrial and military applications, and are known to contaminate environmental surfaces, including concrete. This study aims to characterize the molecular-level interaction energies of six PFAS species—PFOA, PFOS, PFHxS, PFHxA, 6:2 FTS, and PFBS—with calcium silicate, a key component of concrete, using density functional theory (DFT) calculations. Change in Gibbs free energy (ΔG) was determined for each of the interactions, revealing negative ΔG values for …
Improving Zero Shot Learning By Linking Multi-Label Cnns With Llms, Michael A. Wegner
Improving Zero Shot Learning By Linking Multi-Label Cnns With Llms, Michael A. Wegner
Theses and Dissertations
Classifying previously unseen objects poses a significant challenge for traditional computer vision algorithms, which rely on extensive labeled training data. Zero-shot reasoning offers a way to overcome this limitation. This research explores a novel method for image recognition using the Animals with Attributes 2 (AWA2) dataset as a proof of concept. A multi-label ResNet50 model predicts core attributes like color, ear shape, or number of limbs. Those attributes then feed into ChatGPT which leverages its extensive knowledge base to classify the animal based on the provided attributes. This novel approach skips the need to train on every possible class. Instead, …
Discovering Design Requirements For Next Generation Arctic Tension Fabric Shelters, Mark W. Mcveigh
Discovering Design Requirements For Next Generation Arctic Tension Fabric Shelters, Mark W. Mcveigh
Theses and Dissertations
Military operations to remote Arctic regions require large-span temporary shelters to house tactical aircraft and provide heated maintained spaces. However, the current System-50 Large Area Maintenance Shelters used by the United States Air Force and Department of Defense are inadequate for Arctic deployments. These shelters lack durability against extreme subzero temperatures, heavy snow accumulation, and high wind speeds. They also fail to address critical Arctic-specific design challenges including permafrost protection, foundation disruption caused by frost heaves, and efficient heating as a result of their inadequate thermal resistance properties. This research analyzed 20 years of climatological data from 8,399 weather observation …
Atmospheric Water Generation: Bacterial And Inorganic Chemical Quality, Garrett E. Stanley
Atmospheric Water Generation: Bacterial And Inorganic Chemical Quality, Garrett E. Stanley
Theses and Dissertations
This study investigated the quality of untreated water collected from a vapor compression-based atmospheric water generator (AWG) operated outdoors in Dayton, Ohio. The concentrations of bacterial and chemical constituents were determined. The concentration of culturable bacteria was between 200 and 2300 CFU/mL, significantly higher than the reference levels described in recreational and drinking water quality guidance. Three discrete bacteria phenotypes were visually identified; two were yellow in color, rod-shaped and gram-negative while the third was milky white, circular, and gram-positive. Chemical analysis on 15 randomly selected samples revealed the presence of Barium (average = 0.16 mg/L), Magnesium (0.08 = mg/L), …
Early-Phase Cost Estimation For Department Of Defense Construction Projects Via Artificial Neural Networks: Implications And Policy Recommendations For Artificial Intelligence Integration, Kobe T. Thompson
Theses and Dissertations
The Department of Defense lost over 500 million dollars between 2016 and 2024, partially due to poor early cost estimates resulting in cost overruns. practice for cost estimation relied on parametric techniques that incorporate historical data, subject matter experts in cost estimating, and predictive software applications. The main motivation for this study was to assess the viability of artificial neural networks as a means of providing a more accurate cost estimate in the early design phases of a construction project. The dataset initially contained approximately 48,000 data points from a database of various Air Force projects, including maintenance, repair, minor …
