Mutations affecting Clostridioides difficile flagellin (FliC) have been shown to be hypervirulent in animal models and display increased toxin production and alterations in central metabolism. The regulation of flagellin levels in bacteria is governed by a tripartite regulatory network involving fliC, fliW, and csrA, which creates a feedback system to regulate flagella production. Through genomic analysis of C. difficile clade 5 strains (non-motile), we identified they have jettisoned many of the genes required for flagellum biosynthesis yet retain the major flagellin gene fliC and regulatory gene fliW. Humans are altering biological communities on local to global scales, driving profound changes in biodiversity and ecosystem functioning. However, significant gaps and biases in biodiversity data hinder our understanding of ecosystem interactions, making it challenging to develop effective, data-driven policies to protect remaining biodiversity. Bats, the second most species-rich groups of mammals, have a global distribution, and perform a broad array of vital ecosystem functions. Their diversity peaks in the tropics, where they often constitute more than 50% of mammal species diversity. Fibroblast growth factor 21 (FGF21) is an endocrine FGF that plays a vital role in regulating essential metabolic pathways. FGF21 increases glucose uptake by cells, promotes fatty acid oxidation, reduces blood glucose levels, and alleviates metabolic diseases. However, detailed studies on its stability and biophysical characteristics have not been reported. Herein, we present the overexpression, biophysical characterization, and metabolic activity of a soluble recombinant FGF21 (rFGF21). The far-UV circular dichroism spectra of rFGF21 show a negative trough at 215 nm, indicating that the protein's backbone predominantly adopts a 13 sheet conformation. rFGF21 shows intrinsic tyrosine fluorescence at 305 nm. Objectives: Factors associated with peripheral arthritis and enthesitis, especially Achilles tendonitis and plantar fasciitis, were examined in a longitudinal cohort of 1075 patients with ankylosing spondylitis (AS) (also known as radiographic axial spondyloarthritis).
Methods: Patients were derived from the Prospective Study of Outcomes in Ankylosing Spondylitis cohort. Disease activity and functional indices, as well as physical examination and medications used, were measured at every study visit. Univariable and multivariable analyses of the association of peripheral arthritis and enthesitis with clinical, sociodemographic factors were performed. Human leucocyte antigen (HLA)-B alleles were analysed by single-stranded conformational polymorphism analysis.

Breaking Binaries: Gender Non-Conforming Lesbians And The Undercurrent Of Biopower On Dating Apps, Lily M. Connolly

This study examines how gender nonconforming lesbians experience and navigate algorithmic constraints, commodification, and self-expression on dating apps. Drawing on Michel Foucault’s concept of biopower, this study analyzes focus group discussions to argue that dating apps encode and enforce traditional gender norms. However, rather than acting solely as sites of constraint, dating apps also provide spaces where gender nonconforming lesbians creatively resist these limitations and subvert platform design to assert their politicized queer identities. This research highlights how biopower operates within digital dating platforms while also illuminating the subversive strategies users employ to reclaim agency. Exercise technique, defined as the controlled execution of bodily movements to ensure an exercise effectively targets specific muscle groups while minimizing the risk of injury, is a resistance training (RT) variable frequently highlighted as critical to successful RT program outcomes, with proper technique suggested to play a role in maximizing muscle development. Heterogeneous Wireless Sensor Networks (WSNs) involve nodes with varying capabilities, such as different energy levels, sensing ranges, and computational abilities, which enable them to execute different tasks professionally. Tuberculosis (TB) remains a significant public health challenge worldwide. Currently, Bacillus Calmette-Guerin (BCG) is the only vaccine available for TB prophylaxis. However, the efficacy of the BCG vaccine against adult pulmonary TB is considered inconsistent. This condition encourages researchers to look for more effective options, such as subunit vaccines. This condition requires the development of a more effective subunit vaccine to protect active TB in productive and adult ages. Ras El-‘Amud, Jerusalem: Faunal Remains From The Intermediate Bronze And Middle Bronze Age Settlement, Nimrod Marom
Qadum: Journal of Excavation Reports from Israel
The small faunal assemblage from Ras el-‘Amud provides insights into the economic strategies of a rural settlement during a period of urban growth. A shift from a self-sufficient goat and pig economy to a specialized sheep-focused one is observed, likely driven by urban demand. Analysis of caprine skeletal remains suggests consumption patterns and potential ritualistic practices. The Middle Bronze IIA village exhibits a decrease in butchery waste, possibly due to changes in spatial organization or waste disposal practices. The assemblage offers valuable insights into the economic and social dynamics of rural settlements during the Middle Bronze Age.

Competitive Language Use: A Linguistic Analysis Of Speech And Debate, Gina Iberri-Shea
Studies in Debate and Oratory
This study explores linguistic variation in competitive debate and prepared speech. A corpus of competitive public speech was collected and the situational characteristics of two contexts were analyzed. The linguistic characteristics of texts in each category were calculated along five dimensions of variation found across texts in English, and placed among other types of texts. Results show that three of the dimensions were able to distinguish between the two registers. The results indicate a clear direction for future work and implications for English for Academic Purposes instruction.

Historical critical approaches to the Bible that reexamine doctrines of inerrancy are unheard of, dismissed, or viewed as a threat throughout Christian faith communities in Sumner County, TN. This underutilizes a credible method of interpretation for Christians who have only been exposed to views that tightly hold biblical inerrancy. The lack of awareness and disregard for aspects of biblical criticism in local churches by leaders and congregation members in Sumner County, TN, is multifaceted.

Progesterone receptors are classified into nuclear and membrane-bound receptor families. Previous unbiased proteomic studies indicate a potential association between cardiac diseases and the progesterone receptor membrane-bound component-2 (PGRMC2); however, the role of PGRMC2 in the heart remains unknown. In this study, we use a heart-specific knockout (KO) mouse model (MyH6•Pgrmc2flox/flox) in which the Pgrmc2 gene was selectively deleted in cardiomyocytes. Here we show that PGRMC2 serves as a mediator of steroid hormones for rapid calcium signaling in cardiomyocytes to maintain cardiac contraction, sufficient stroke volume, and adequate cardiac output by regulating the cardiac pressure-volume relationship. Purpose of Review: Findings from the Global Aging & Geriatric Experiments in Bipolar Disorder (GAGE-BD) project, including sociodemographic and clinical information from older age bipolar disorder (OABD) and healthy participants around the globe (approximately N = 5000) were reviewed. Data was collected in multiple waves to create a large integrated dataset. Recent Findings: BD does not seem to fade with age. BD subtype and early/late onset did not show significant differences in daily functioning. Physical comorbidities were more frequent in OABD compared with controls. Women with OABD had an earlier age at onset and more psychiatric hospitalizations. Summary: GAGE-BD is …

Current work suggests that basic psychological needs are related to higher intrinsic motivation, which in turn, can promote more positive academic outcomes. However, few studies have examined how perceptions around one’s abilities in science, engineering, technology, and math (STEM) are related to intrinsic motivation and what role needs satisfaction plays in this association. This study aims to explore the everyday life and health information behavior of people who inject drugs (PWID) in Bangladesh, as well as the effectiveness of HIV and STI informational programs in promoting safe injecting and sexual behavior. The study received 97 completed survey responses and conducted two focus group discussions (FGDs) with 13 PWID. Using de Certeau's everyday life practices framework, the study aims to understand the tactical information strategies of PWID in the context of a developing country where access to government services and trust in authorities may be significantly limited among marginalized populations. Changing ocean temperatures are already causing declines in populations of marine organisms. Predicting the capacity of organisms to adjust to the pressures posed by climate change is a topic of much current research effort, particularly for species we farm or harvest. Arginine has been a mainstay in biological formulation development for decades. To date, the way arginine modulates protein stability has been widely studied and debated. Here, we employed a hydrophobic polymer to decouple hydrophobic effects from other interactions relevant to protein folding. While existing hypotheses for the effects of arginine can generally be categorized as either direct or indirect, our results indicate that direct and indirect mechanisms of arginine co-exist and oppose each other. At low concentrations, arginine was observed to stabilize hydrophobic polymer folding via a sidechain-dominated direct mechanism, while at high concentrations, arginine stabilized polymer folding via a …

Differences between normal tissues and invading tumors that allow tumor targeting while saving normal tissue are much sought after. Here we show that scarcity of VDAC2, and the consequent lack of Bak recruitment to mitochondria, renders hepatocyte mitochondria resistant to permeabilization by truncated Bid (tBid), a Bcl-2 Homology 3 (BH3)-only, Bcl-2 family protein. Increased VDAC2 and Bak is found in most human liver cancers and mitochondria from tumors and hepatic cancer cell lines exhibit VDAC2- and Bak-dependent tBid sensitivity. Background: Although electronic health record nursing summaries aim to provide a concise overview of patient data, they often fall short of meeting nurses' information needs, leading to underutilization. This gap arises from a lack of involvement of nurses in the design of health information technologies.
Objective: The purpose of this exploratory co-design case study was to solicit insights from nurses regarding nursing summary design considerations, including key information types and the preferred design prototype.

Members of the Pol A family of DNA polymerases, found across all domains of life, utilize various strategies for DNA strand separation during replication. In higher eukaryotes, mitochondrial DNA polymerase γ relies on the replicative helicase TWINKLE, whereas the yeast ortholog, Mip1, can unwind DNA independently. Using Mip1 as a model, we present a series of high-resolution cryo-EM structures that capture the process of DNA strand displacement. Our data reveal previously unidentified structural elements that facilitate the unwinding of the downstream DNA duplex. Parkinson's disease (PD) starts decades before symptoms appear, usually in the later decades of life, when age-related changes are occurring. To identify molecular changes early in the disease course and distinguish PD pathologies from aging, we generated Drosophila expressing alpha-synuclein (αSyn) in neurons and performed longitudinal bulk transcriptomics and proteomics on brains at six time points across the lifespan and compared the data to healthy control flies as well as human post-mortem brain datasets. Disease-modifying therapies are standard of care (SOC) for sickle cell disease (SCD), but hematopoietic cell transplantation (HCT) has curative potential. We compared outcomes prospectively through 2 years after biologic assignment to a donor or no donor (SOC) arm based on the availability of an HLA-matched sibling or unrelated donor (BMT CTN 1503). A donor search was commenced after eligibility confirmation. The primary end point was a comparison of survival between the treatment arms 2 years after biologic assignment. Power calculations required 60 participants in the donor arm and 140 in the no donor arm to determine if early transplant-related mortality …

Persistent organic pollutants (POPs) are lipophilic environmental contaminants accumulated in the adipose tissue. Weight loss interventions, such as bariatric surgery, can mobilize POPs from adipose tissue into the bloodstream. We hypothesized that this mobilization could contribute to increases in blood pressure among 57 adolescents with severe obesity undergoing bariatric surgery. POPs and metabolic features were measured from visceral adipose tissue collected during surgery using gas and liquid chromatography, coupled with high-resolution mass spectrometry. Structure-forming foundation species facilitate consumers by providing habitat and refugia. In return, consumers can benefit foundation species by reducing top-down pressures and increasing the supply of nutrients. Consumer-mediated nutrient dynamics (CND) fuel the growth of autotrophic foundation species and generate more habitat for consumers, forming reciprocal feedbacks. Such feedbacks are threatened when foundation species are lost to disturbances, yet testing these interactions requires long-term studies, which are rare. Here, we experimentally evaluated how disturbance to giant kelp, a marine foundation species, affects (1) CND of the forest animal community and (2) nutrient feedbacks that help sustain forest primary production during …
