Hdac5 Enzymatic Activity Regulates Scn4b Expression To Control Relapse-Like Cocaine Seeking, Daniel J. Wood Apr 2025

Hdac5 Enzymatic Activity Regulates Scn4b Expression To Control Relapse-Like Cocaine Seeking, Daniel J. Wood

MUSC Theses and Dissertations

Substance Use Disorder (SUD) is a relapsing disorder characterized by repeated seeking and use of psychoactive substances despite negative consequences to the individual. Epigenetic changes within the medium spiny neurons (MSNs) of the nucleus accumbens (NAc) are thought to underly the lasting propensity to relapse in SUD. The epigenetic enzyme histone deacetylase 5 (HDAC5) in NAc MSNs limits the formation of reward-cue associations in rodent models of drug seeking, but not sucrose seeking, leading to decreased relapse-like behavior. However, whether HDAC5 reduces drug seeking behavior through its own deacetylase activity or through the recruitment of co-repressors is unknown. Furthermore, these …

What Is A Nation? Colonial Legacies, Identity, And Reclaiming In Métisse Autobiographical Narratives – A Case Study Of Sugar And Slate And Garçon Manqué, Thaïs Maëlle Autin

Foreign Languages & Literatures ETDs

This thesis examines the question of national belonging in autobiographical narratives of second-generation immigrant métisse women. I study the similar aesthetic manner in which individuals with different regional and cultural influences grapple with concepts of identity, gender, racism, exile, and negotiation, and the way in which the authors anchor themselves in a broader familial and national heritage. I argue that such an enterprise can be understood as part of the alternative heritage wave, based on Andrew Higson’s theory. The texts put in question traditional understandings of nation. Doing so opens the possibility of engaging in a dialogue that expands the …

Bio-Inspired Highest Lift-To-Drag-Ratio Fin Shape And Angle For Maximum Surfboard Stability: Flow Around Fish Fins, Megan S. Macneill, Brian Barkdoll

Michigan Tech Publications

Wave surfing is a multi-billion dollar industry involving both maneuverability and speed, yet little research has been performed regarding the highest lift-to-drag-ratio fin shape for these competing qualities. Numerical modeling and laboratory experiments were performed here to identify a bio-inspired fin shape that maximized lateral stability and minimized drag forces, in order to increase surfing maneuverability. Nine fins based on dorsal fins of real fish were tested. Both the CFD and laboratory experiments confirmed that the fin of the same shape as that of the Short-Finned Pilot Whale at an angle of attack of 10° had the greatest lift-to-drag ratios. …

Mhuireach, Susan Collins, Leslie Dietz, Patrick Finn Horve, Aurélie Laguerre, Dale Northcutt, Jason Stenson, Kevin Van Den Wymelenberg, Elliott Gall, Mark Fretz

Mechanical and Materials Engineering Faculty Publications and Presentations

IntroductionHumans have used wood as a construction material throughout history. Currently, mass timber products, such as cross-laminated timber (CLT), are becoming more popular as a structural material, since they are renewable and have a lower carbon footprint than concrete or steel. Nonetheless, some building types, such as healthcare, veterinary, and food manufacturing, avoid using structural mass timber due to concerns about microbial growth in the event of wetting. One solution is to use protective coatings on mass timber products to increase moisture resistance, although the coatings themselves may generate concerns about volatile organic compound (VOC) emissions. Natural uncoated wood also …

The Association Of Sleep Quality And Depression On Having Three Vs. All Five Diagnostic Criteria Of Metabolic Syndrome, Nina Pollard

Walden Dissertations and Doctoral Studies

Metabolic syndrome is increasing in prevalence in the United States. To be diagnosed, three of the five diagnostic criteria must be met. There is a higher risk of metabolic syndrome due to lifestyle and social factors, with increased criteria occurring as an individual has unmet social determinants of health. This quantitative quasi-experimental/quasi-comparative study examined the association between education level, with sleep quality and depression as moderators, on a diagnosis of metabolic syndrome with all five criteria versus three criteria using a binary logistic regression while controlling for age, race, gender, income, employment, and marital status. Bronfenbrenner’s social ecological theory served …

Structural and functional alterations in the brain's reward circuitry are present in cocaine use disorder (CocUD), but their molecular underpinnings remain unclear. To investigate these mechanisms, we performed single-nuclei multiome profiling on postmortem caudate nucleus tissue from six individuals with CocUD and eight controls. We profiled 30,030 nuclei, identifying 13 cell types including D1- and D2-medium spiny neurons (MSNs) and glial cells. We observed 1485 differentially regulated genes and 10,342 differentially accessible peaks, with alterations in MSNs and astrocytes related to neurotransmitter activity and synapse organization. Gene regulatory network analysis identified transcription factors including ZEB1 as exhibiting distinct CocUD-specific subclusters, …

Methicillin-resistant Staphylococcus aureus (MRSA) is a major public health menace. The global spread of MRSA is characterized by successive waves of epidemic clones dominating specific geographical regions. The acquisition of genes encoding resistance to heavy metals (HMRGs) is thought to be a key feature in the geographic divergence of MRSA. However, the cause-effect relationship between the presence of HMRGs and the divergence of MRSA clones remains to be clarified. In this study, we assessed the role that HMRGs may have played in the evolutionary divergence of the MRSA ST5-SCCmecI lineage in Latin America. We conducted a genomic characterization …

Pilrα Sustains Microglial Capacity To Process Myelin In A Mouse Model Of Demyelination, Kent Lin

Arts & Sciences Graduate Student Theses and Dissertations

Alzheimer's disease (AD) is characterized by amyloid-β (Aβ) plaques, neurofibrillary tangles, and neurodegeneration. Microglia, the resident immune cells of the central nervous system (CNS), play a crucial role in AD pathogenesis by containing the spreading of Aβ plaques. Genetic studies have associated a variant of the receptor Paired Immunoglobulin-like Type 2 Receptor Alpha (PILRα) with reduced risk for AD. PILRα is a receptor found on myeloid cells, such as microglia, that binds to endogenous cell surface glycoproteins and sends inhibitory signals within the cell. Additionally, it serves as an entry receptor for Herpes Simplex Virus 1. To elucidate the role …

Jurnal Kultivasi

Tea plants (Camellia sinensis) are important commodities with high economic value, but their production is greatly affected by environmental stresses such as drought and extreme temperatures. Global warming in recent years has led to extreme weather events and an increase in the earth's temperature, which also causes drought. Drought stress is one of the most significant abiotic factors affecting crop productivity. However, despite significant progress, there remains a notable gap in research, particularly the lack of integrated studies that combine morphological, physiological, and biochemical indicators for early and precise detection of drought stress in tea plants. To identify and mitigate …
