We adopted a proteomic and follow-through approach to investigate how mechanical overload (MOV) potentially affects novel targets in skeletal muscle, and how a perturbation in this response could potentially affect the adaptive response. First, we determined that 10 wk of resistance training in 15 college-aged females increased sarcolemmal-associated protein content (+10.1%, P < 0.05). Sarcolemmal protein isolates were then queried using mass spectrometry-based proteomics, ∼10% (38/387) of proteins putatively associated with the sarcolemma or extracellular matrix (ECM) were upregulated (>1.5-fold, P < 0.05), and one target (intermediate filament vimentin; VIM) warranted further investigation due to its correlation to myofiber hypertrophy (r = 0.652, P = 0.009). VIM expression was then examined in 4-mo-old C57BL/6J mice following 10 and 20 days of plantaris MOV via synergist ablation. Relative to Sham (control) mice, VIM mRNA and protein content was significantly higher in MOV mice, and immunohistochemistry indicated that VIM predominantly resided in the ECM. MOV experiments were replicated in Pax7-DTA (satellite cell depleted) mice, which reduced VIM in the ECM by ∼74%. A third MOV experiment was performed in C57BL/6 mice intramuscularly injected with either AAV9-scrambled (control) or AAV9-VIM-shRNA. Although VIM-shRNA mice possessed lower VIM in the ECM (∼45%), plantaris masses in response to MOV were similar between groups. However, VIMshRNA mice possessed smaller and more centrally nucleated MyHCemb-positive fibers in response to MOV. In summary, skeletal muscle VIM appears to be enriched in the ECM following MOV, satellite cells may regulate its expression, and a disruption in expression during MOV leads to an excessive regenerative phenotype.

This study investigates the complex relationship between cellular senescence, aging, and pulmonary fibrosis through a comprehensive analysis of bulk RNA-seq data from patients with idiopathic pulmonary fibrosis (IPF), chronic obstructive pulmonary disease (COPD), and scleroderma. Using differential expression (DE) and age-associated differential expression (AADE) analyses, we revealed a paradoxical pattern: senescence-related gene sets were significantly downregulated in disease comparisons but progressively upregulated with aging within these same diseases. Gene set enrichment analysis (GSEA) revealed that gene sets associated with DNA damage/telomere stress and oxidative stress were among the most enriched senescence inducers in fibrotic disease states, while cellular senescence and …

Over time, viruses have evolved a myriad of mechanisms to gain an advantage in the virus-host battle, such as inhibiting host mRNA export through RNA Export Factor 1 (Rae1) and Nucleoporin 98 (Nup98). This strategy employed by the Vesicular Stomatitis Virus (VSV) M protein, Kaposi’s Sarcoma-Associated Herpesvirus (KSHV) ORF10, and SARS-CoV-1/2 ORF6 disrupts antiviral production. Despite their evolutionary distance, these viral proteins share a key methionine flanked by acidic residues that competitively bind to the ssRNA binding pocket of Rae1. Adipocyte differentiation plays an important role in bone remodeling due to secretory factors that can directly modulate osteoblast and osteoclast, thus affecting overall bone mass and skeletal integrity. Excessive adipocyte differentiation within the bone marrow microenvironment can lead to decreased bone mass, eventually causing osteoporosis. Background: There is a significant delay between symptom onset and diagnosis of childhood asthma, but the impact of this delay on asthma outcomes has not been well understood.

Objectives: We sought to study the association of delayed diagnosis of asthma with asthma exacerbations (AEs) in children.

Methods: Using the Mayo Clinic birth cohort, we identified children with a diagnosis of asthma from electronic health records. We defined onset date as the date when subjects first met predetermined asthma criteria ascertained by an electronic health records-based natural language processing algorithm. Students are increasingly expected to conduct research and scholarship. This paper explores podcast creation as a medium for students to deliver new knowledge and the elements of teaching and learning that support this outcome. Through focus groups, students at a large Midwestern university expressed the importance of contributing to the information landscape. The prospect of their podcasts contributing knowledge to academic communities held more interest to them than creating a podcast for an assignment. This paper addresses how librarians and libraries can support the information literacy standards and technical aspects of students creating new knowledge through a podcast assignment.

Regional food insecurity caused by the inability to use conventional farming techniques year-round in cold climates increases the need for access to quality, locally sourced, and affordable foods year-round. Shifting from a soil-based system to aeroponics can increase yields, as aeroponic systems are not weather-reliant and can operate year-round. Aeroponics, a subgroup of hydroponics, induces growth by misting nutrient-rich solutions onto the plant roots. These systems use space and water more efficiently and rely less on fertilizers and pesticides but are vulnerable to errors in nutrient application, power loss, and rapid pathogen spread. Spinal cord injury (SCI) disrupts white matter, causing periaxonal swelling, axonal degeneration, and impaired function. To examine vascular edema’s role, we developed a 3Dprinted dual-compartment imaging chamber to mimic and manipulate interstitial and vascular fluid dynamics in real time. We hypothesized that hypertonic saline (HTS) in the "vascular" chamber would osmotically shift fluid from the periaxonal space and preserve myelinated fibers after SCI. Utilizing aquaculture wastewater as microalgae growth medium is a potential opportunity for nutrient recycling and industrial symbiosis, but more research is required to understand how best to design and scale-up a dual-purpose facility. This study investigates the growth of Coelastrella sp. in aquaculture wastewater compared to industry-standard growth medium BG-11 and modified tap water as controls. The STAT2 gene encodes a protein called Signal Transducer and Activator of Transcription 2 and is an excellent target for genetic manipulation due to its essential role in type I and III interferon signaling pathways11. A member of the STAT family of transcription factors, this protein is directly involved in mediating cellular responses to various cytokines and growth factors, particularly those related to the innate immune response11. Impact Of Altering Gut Microbiota With Multinutrient Supplementation On Neuropsychiatric Symptoms In Attention Deficit/Hyperactivity Disorder, Benjamin Hinkle

Attention-Deficit/Hyperactivity Disorder (ADHD) is a prevalent neuropsychiatric condition affecting children, often characterized by inattention, hyperactivity, and impulsivity. Recent evidence suggests a potential link between gut microbiota composition and neuropsychiatric disorders, including ADHD, through the gut-brain axis (GBA). A literature review of studies published between 2017 and 2024 explores the efficacy of multinutrient interventions on influencing gut microbiota to treat the core symptoms of ADHD in children ages 6-12 years old. Findings indicate that multinutrient supplementation may modulate gut microbiota and subsequently impact neuropsychiatric symptoms, offering a promising adjunctive treatment for ADHD. According to the American Heart Association, there are approximately 1.4 million people who are hospitalized and 700,000 deaths attributed to acute coronary syndrome on a yearly basis in the United States. It is standard therapy for patients to receive a drug eluting stent and dual antiplatelet therapy to prevent occlusion of the stent and other coronary arteries. Dual antiplatelet therapy typically consists of aspirin and a P2Y12 inhibitor. Dual antiplatelet leads to an increased risk of bleeding. The incidence of nonalcoholic steatohepatitis (NASH) is continually rising and has now emerged as one of the leading causes of cirrhosis requiring liver transplantation (Maurice et al., 2018). Until recently, the management of this disease has been limited to lifestyle modification and treatment of comorbid risk factors. Obesity and type 2 diabetes are interconnected global health challenges, contributing significantly to morbidity, mortality, and healthcare costs. This literature review compares the efficacy, safety, and applicability of glucagon-like peptide-1 (GLP-1) receptor agonists and bariatric surgery as treatment options. A literature search with defined inclusion and exclusion criteria was conducted using PubMed and EBSCOhost from May to July 2024, resulting in 14 randomized controlled trials for evaluation. This literature review supports the efficacy of both GLP-1 receptor agonists, particularly semaglutide, and bariatric surgery as effective interventions for weight loss and glycemic control in patients with obesity and type 2 diabetes. …
