Lesbian, Gay, Bisexual, Transgender, Queer/Questioning, and More (LGBTQ+) individuals are part of a diverse and complex community that has unique considerations regarding physical and mental health. While there is ongoing health equity research regarding LGBTQ+ individuals, the focus is often on health disparities. A strengths-based approach to helping LGBTQ+ individuals through research and other avenues may be beneficial. Kitchen (1991–96) was the first major work of art by American artist Liza Lou. It is an installation of a life-size kitchen in which every surface and object is covered in glass beads. Kitchen, which Lou refers to as a monument to women’s unsung domestic labor, received considerable critical attention and drew the artist into ongoing conversations around the relationship between “fine art” and “craft” and about the role of feminism in the art of the 1990s. In contemporary interviews, however, Lou expressed discomfort with the label of “craft” as well as with feminist art that she considered “didactic.” …

Nitrogen oxides (NOx = NO + NO₂) are trace gases that play a critical role in the atmosphere by influencing air quality, oxidation chemistry, and the deposition of fixed nitrogen. Historically, NOx emissions have been primarily linked to anthropogenic sources such as fossil fuel combustion, industrial activities, and agriculture. However, as successful legislation has significantly reduced these emissions, the relative importance of natural NOx sources has become an increasing concern. The ubiquitous presence of nanoparticles (NPs) and nanoplastics (NPPs) and their effects on environmental and human health is a significant concern. They are used in every aspect of our lives, and while they offer many benefits, there are concerns about their impacts on ecosystems and human health. The primary aim of this dissertation is to quantify NP and NPP transformations in the presence of algae and determine how those transformations influence bioavailability. The transport of phosphorus (P) and nitrogen (N) from sites containing varying quantities of legacy nutrients was evaluated in this investigation. The data that were examined were collected during four previously reported field hydrologic studies performed in eastern Nebraska. Additional flow was introduced to the top of the rainfall simulation plots to replicate runoff conditions occurring along a hillslope. It was observed that both P and N delivery were influenced by runoff rate on sites where beef cattle manure or inorganic fertilizer had been applied. Objectives: To carry out a quantitative assessment of the therapeutic effect of Chinese herbal medicine (CHM), with Rehmannia glutinosa as the main ingredient, in treating perimenopausal syndrome (PMS) in breast cancer patients. Methods: Randomized controlled trials (RCTs) comparing CHM with Rehmannia glutinosa as the principal ingredient and Western medicine in treating PMS among breast cancer patients were retrieved from multiple databases. ABSTRACT Objectives: To evaluate dupilumab’s outcomes in chronic rhinosinusitis with nasal polyps (CRSwNP) cases. Methods: A retrospective analysis was carried out at Dr. Sulaiman Al Habib Hospitals, Riyadh, Saudi Arabia on 145 adult CRSwNP patients treated with dupilumab between January 2020 and December 2023 following the European position paper on rhinosinusitis and nasal polyps 2020 guidelines. Primary outcomes included changes in Lund-Mackay computed tomography (CT) scores, immunoglobulin E levels, and visual analog scale (VAS) smell score over +12 months. Results: Significant improvements were noted post-treatment. Lund-Mackay CT scores decreased from 12.5 to 5.5 ( p <0.001), and VAS smell scores dropped from 10.0 to 0.0 ( p <0.001). Immunoglobulin E levels reduced from 250.5 to 63.5 ( p <0.001). After treatment, 93.3% of patients avoided sinus surgery, 78.6% reported improved asthma status, and 60% used dupilumab as monotherapy. Conclusion: This study supports dupilumab’s effectiveness in treating severe CRSwNP, showing significant improvements in CT imaging and overall medical condition over 12 months. Dupilumab reduced systemic corticosteroid use and rescue surgeries. While promising, further trials are needed to validate its use as monotherapy. Identifying those who gain most from biologic therapy is crucial.

Objectives: To determine the prevalence of functional disability among older people attending primary healthcare centers in Bisha, Saudi Arabia. Methods: The study used a cross-sectional design and gathered data from older adults (60 years and older) attending primary healthcare centers in Bisha. Data was collected using structured interviews based on the Katz Index of Independence in activities of daily living (ADL). Ordinal logistic regression models identified major ADL limiting variables. Objectives: To evaluate the effect of radial extracorporeal shockwave therapy (rESWT) combined with evidence-based physical therapy (EBPT) on adhesive capsulitis (AC). Methods: A total of 30 participants with AC were randomly assigned to receive either rESWT combined with EBPT (experimental group) or sham rESWT combined with EBPT (control group) over a 6-week intervention period. The primary outcomes included pain (assessed using the visual analogue scale) and disability (disabilities of the arm, shoulder, and hand questionnaire). Patients admitted to a pediatric intensive care unit (PICU) need individualized nutrition support that is tailored to their particular disease severity, nutritional status, and therapeutic interventions. We aim to evaluate how calories and proteins are provided during the first seven days of hospitalization for children in critical condition with organ dysfunction (OD). A single-center retrospective cohort study of children aged 2–18 years, mechanically ventilated > 48 h, and admitted > 7 days to a PICU from 2016 to 2017 was carried out. Background and objectives: Emerging evidence suggests that immune dysregulation plays a pivotal role in triggering cryptogenic new-onset refractory status epilepticus (c-NORSE), prompting a consensus on early initiation of immunotherapy. However, despite similar timing of administration, responses to immunotherapies have been varied and unpredictable, suggesting the presence of heterogeneous underlying mechanisms. The aim of this study was to identify distinct inflammatory response subtypes in patients with c-NORSE by analyzing their cytokine profiles. Insights into underlying mechanisms were sought to understand the pathophysiology and guide personalized therapies to improve patient outcomes.

Cells undergo dramatic morphological changes during embryogenesis, yet how these changes affect the formation of ordered tissues remains elusive. Here, we show that a phase transition leading to the formation of a nematic liquid crystal state during gastrulation in the development of embryos of fish, frogs, and fruit flies occurs by a common mechanism despite substantial differences between these evolutionarily distant animals. Importantly, nematic order forms early before any discernible changes in the shapes of cells. Purpose: Immune checkpoint inhibitors (ICI) have limited efficacy in pleural mesothelioma. We investigated the role of Dickkopf WNT signaling pathway inhibitor 3 (DKK3) in overcoming treatment resistance.

Patients and methods: We performed preclinical studies to elucidate DKK3's role in ICI-resistant mouse mesothelioma. Based on these findings, we conducted a single-arm, phase II clinical trial of a combination of Ad-SGE-DKK3 and nivolumab for chemotherapy-refractory epithelioid pleural mesothelioma, with the objective response rate as the primary outcome.

Background: Epigenetic regulatory genes (epiRG) are pivotal in the epigenetic regulation of the human genome, primarily through DNA and histone modifications. These genes are frequently mutated in human cancers, particularly bladder cancer (BC). However, the functional impact of epiRG mutations on patient outcomes remains poorly understood.

Methods: In this study, we developed gene signatures for the most frequent genomic aberrations of epiRG using The Cancer Genome Atlas Bladder Carcinoma (TCGA-BLCA) dataset and validated these signatures with independent tumor expression profiles for prognostic relevance. Synthetic biology has enabled the development of biosensors to detect intestinal inflammation, yet few target the clinically validated biomarker of intestinal inflammation calprotectin with both diagnostic and therapeutic capabilities. Here, an optimized calprotectin biosensor is presented that leverages a zinc uptake regulator (Zur) controlled promoter coupled with a memory circuit to detect and record intestinal inflammation in vivo. The level of biosensor activation strongly correlates with calprotectin levels in the colon of two independent mouse models of colitis. In 2025, the National Institute of Diabetes and Digestive and Kidney Diseases (NIDDK) at the National Institutes of Health celebrates 75 years of leadership in diabetes research. The NIDDK serves people of the U.S. affected by or at risk for many chronic diseases, including diabetes and other endocrine, metabolic, and digestive disorders, by funding innovative research to develop better treatment and prevention and a cure for these conditions. Dystonia is a movement disorder with varied clinical features and diverse etiologies. Here we present a revision of the 2013 consensus definition and classification of dystonia in light of subsequent publications and experience with its application during the last decade. A panel of movement disorder specialists with expertise in dystonia reviewed the original document and proposed some revision. There was broad consensus to retain the definition of dystonia with only minor clarifications to the wording. Background: Anemia is a common complication of inflammatory bowel disease (IBD), impacting patient quality of life and clinical outcomes. This study examines anemia management practices and clinical outcomes in a national cohort of veterans with IBD.

Methods: A retrospective cohort study of 89 687 veterans with IBD from 2000 to 2017 was conducted using Veterans Health Administration data. We assessed anemia screening, iron store assessments, and iron therapy. Primary outcomes included anemia resolution within 12 months. Secondary outcomes included IBD-related hospitalizations, steroid use, and surgery within five years. Multivariate logistic regression models assessed associations between patient characteristics and clinical outcomes. …

Chimeric antigen receptor T cell (CAR-T) therapy is an effective treatment for relapsed-refractory large B-cell lymphoma (LBCL). However, toxicities, particularly cytokine release syndrome (CRS) and immune effector cell-associated neurotoxicity syndrome (ICANS), remain significant concerns. Analyze temporal trends, risk factors, and associations between these toxicities and their severity. In this registry study by the Center for International Blood and Marrow Transplant Research, we studied CRS and ICANS in 1916 LBCL patients treated with commercial CAR-T therapies (axicabtagene ciloleucel 74.9%, tisagenlecleucel 25.1%) between 2018 and 2020. Metastasis is the predominant cause of cancer mortality, primarily driven by complex tumor-host interactions within specialized metastatic niches. Recent advances in single-cell technologies have provided unprecedented insights into metastatic niche formation, evolution and function, including how primary tumors precondition distant organs for metastases and how disseminated tumor cells dynamically interact with host cells to modulate their environments. Integrated single-cell studies across multiple cancer types have also revealed divergent and convergent metastatic adaptation strategies. Purpose: There is evidence that statins might improve the outcome of patients with breast cancer. The role of statins in patients with early HER2-positive breast cancer is unknown. Therefore, we explored the association between statin use and survival outcomes in early HER2-positive breast cancer patients in the phase III APHINITY trial (adjuvant pertuzumab/trastuzumab).

Methods: All patients (intent-to-treat population, n = 4804) were included (6.2 years median follow-up database). Mitochondrial membrane dynamics control the shape, number, and distribution of mitochondria and regulate energy production and cell health. In a screen for yeast mutants with increased levels of templated insertions (~10-1000 bp) in the nuclear genome, we identified mitochondrial fusion deficient mutants (mgm1Δ, ugo1Δ, fzo1Δ). We found that fusion mutants activate the iron regulon, have decreased iron-sulfur clusters (ISCs), and increased DNA damage, suggesting a role of iron homeostasis in preventing insertions. Consistently, a secondary screen found mutants affecting iron-sulfur cluster production (yfh1Δ, grx5Δ), vacuolar iron storage (ccc1Δ) or general iron homeostasis (aft1Δ) to exhibit high insertion levels. Carbamoyl phosphate synthetase 1 (CPS1) deficiency is a rare metabolic disorder that, in neonatal onset, is typically characterized by severe life-threatening and neurologically injuring hyperammonemic episodes with high unmet patient need. Patients that retain limited enzyme activity may present later in life with less severe hyperammonemia. CPS1 drives the first step in the urea cycle, the pathway terrestrial mammals utilize to metabolize nitrogen. In order to probe the effect of hyperammonemia on the developing nervous system and explore new therapies, a murine Cps1 exon 3-4 mutant was previously generated. Introduction: Validation of the primary cognitive composite and baseline cognitive characteristics are presented for the US-Study-to-Protect-Brain-Health-Through-Lifestyle-Intervention-to-Reduce-Risk (US POINTER).

Methods: US POINTER is a multicenter, randomized clinical trial of two lifestyle interventions testing cognitive benefit in older adults without significant cognitive impairment but at-risk for decline due to well-established factors. Cognition is measured using a global cognitive composite (US POINTER modified Neuropsychological Test Battery-PmNTB).

Objective: To examine rates of postpartum hemorrhagic (PPH) morbidity among patients who did and did not have immediate skin-to-skin contact (SSC).

Methods: This study was a retrospective cohort of all non-anomalous, term singleton vaginal births at a Level IV center over 2 years. Exclusion criteria included COVID-19. Immediate SSC was defined as at least 60 min of direct contact initiated between parturient and neonate within 10 min of birth. The primary outcome was a composite of maternal morbidity related to PPH compared among those with and without immediate SSC.
