Despite being classically reported as caused by mutations in solute carriers genes (SLC2A1), it has been recently shown that also mutations in ALDH7A1 can cause pyridoxine-dependent epilepsy (PDE). ALDH7A1 is a gene encoding for the antiquitin, an enzyme that catalyzes the nicotinamide adenine dinucleotide-dependent dehydrogenation of L-α-aminoadipic semialdehyde/L-Δ1-piperideine 6-carboxylate. It is a highly treatable disorder, but nevertheless it is still not certain when to consider this diagnosis and how to test for it. Punctuated Equilibrium Theory, as applied by Niles Eldredge and Stephen Jay Gould (1941-2002) to the study of species evolution, examined the infrequency of morphological change or stasis discontinuity.1 While groundbreaking in the field of Paleontology and the study of fossil change, the theory has been well-fitting to the decision sciences as introduced by Baumgartner and Jones2, in their 1993 and 2010 publications. Understanding the difference between signal and noise in a measurement is fundamental to analytical chemists. The typical instruments found in an undergraduate chemistry department can be used to teach this concept, as well as signal averaging, by giving students hands-on experience with measurements and data processing. In this study, we conducted high-throughput spatiotemporal analysis of primary cilia length and orientation across 22 mouse brain regions. We developed automated image analysis algorithms, which enabled us to examine over 10 million individual cilia, generating the largest spatiotemporal atlas of cilia. We found that cilia length and orientation display substantial variations across different brain regions and exhibit fluctuations over a 24-h period, with region-specific peaks during light-dark phases. Our analysis revealed unique orientation patterns of cilia, suggesting that cilia orientation within the brain is not random but follows specific patterns. The Kentucky Microbiome Dataset is a continuously updated geolocated dataset which describes the distribution and diversity of microorganisms across the Commonwealth of Kentucky. Each geolocated point represents a location reported in the peer-reviewed literature where microorganisms were sampled from soil, water, geologic substrates, or clinical settings. Environmental studies include only non-host-associated, non-clinical microorganisms sampled from natural environments. Clinical studies are mapped separately to highlight locations where investigations into host-associated or human health-related microbes have occurred.

[Objective] To explore the application of Polygonatum sibiricum powder in bread,enhancing its health benefits.[Methods] Polygonatum powder is added to wheat flour to study its effects on starch gelatinization,gluten structure,dough rheological properties,and bread baking quality.[Results]] The addition of Polygonatum powder increases the gelatinization temperature of the flour,while peak viscosity and retrogradation values decrease.The gluten yield in the dough is reduced.Changes in the secondary structure of gluten proteins are observed,with an increase in the content of random coil structures.Polygonatum sibiricum powder increases dough hardness,elasticity modulus,and viscosity modulus,but it reduces the dough's fermentation volume.The baking quality of the bread is affected,with the bread …

Antimicrobial resistance (AMR) is an escalating global health threat, with the greatest risk observed in low- to middle-income countries, particularly in the global south. The World Health Organization advocates for a One Health approach to address AMR, promoting collaboration across sectors, including in agriculture. This study aims to enhance understanding of antimicrobial use and stewardship in livestock within pastoralist communities in northern Kenya, where there is limited information. Current research suggests that children with Down Syndrome (DS) are at an increased risk for autism spectrum disorder (ASD) compared to the general population. Detecting ASD early in children with DS is crucial to providing access to additional treatment services and supports that may be necessary. Current radiobiological models focus primarily on nuclear DNA damage and fail to fully explain radiation-induced noncancerous health effects. This dissertation investigates whether non-nuclear subcellular structures contribute to radiation-induced biological damage through experimental biomarker analysis and computational microdosimetry. The central hypothesis posits that whole-body photon irradiation results in subcellular non-nuclear damage, specifically increased circulating mitochondrial DNA concentrations, that correlates with radiation dose and long-term noncancerous effects such as cardiovascular disease.

Objectives: This study aimed to assess the incidence of breast cancer in young women over a ten-year period and examine its association with clinical characteristics, risk factors, treatment modalities, and survival outcomes. Methods: A retrospective analysis of the Breast Cancer Registry at our institution was conducted. Of the 2,238 women diagnosed with breast cancer between 2012 and 2021, 535 (23.9%) were aged 40 years or younger at the time of diagnosis. Introduction: This study evaluated and compared orthodontic forces from different archwire designs and materials used to correct a deep curve of Spee. Methods: A three-dimensional orthodontic force tester (OFT) was used to measure forces and moments exerted by archwires on brackets from central incisors to first molars. Two setups were tested: one with a full dentition from central incisor to first molar, and another simulating bicuspid extractions with the first premolar removed. Dentoform models representing a deep curve of Spee were created using cone-beam computed tomography (CBCT) data. [Objective] To investigate the effect of complexes produced by protein -polyphenol interaction on the bioavailability of polyphenols and proteins.[Methods] Rutin -casein complexes are prepared using rutin and casein as raw materials.Their physicochemical and structural properties are analyzed.[Results]] The optimal preparation conditions for the rutin -casein complexes are determined as follows:mass ratio of rutin to casein =1.89∶1.00,reaction time of 5.94 h,pH 8.5,and temperature of 50 ℃.Under such conditions,the binding degree is 40.23%.Compared with casein,the complexes exhibit a decrease in emulsifying activity index (8.47 m2/g) and emulsifying stability (9.39%).The solubility increases by 5.11%,surface hydrophobicity is significantly enhanced,and antioxidant activity markedly improves (P<0.05).Scanning electron microscopy reveals that the rutin -casein complexes display a lamellar structure with smooth surfaces,which may contribute to its functional properties.Infrared spectroscopy demonstrates peak broadening and redshifts in the amide I and II bands,and fluorescence intensity (including synchronous fluorescence ) decreases due to fluorescence quenching.It indicates that rutin alters the secondary structure and conformational arrangement of casein.[Conclusion] Compared with the uncomplexed casein,the rutin -casein complexes are enhanced in solubility,surface hydrophobicity,and antioxidant activity.引用格式:张楠,刘丽莉,杨乐,等.芦丁—酪蛋白复合物的制备及特性研究 [J].食品与机械,2025,41(7):9-17.C itation:ZHANG Nan,LIU Lili,YANG Le,et al.Preparation and properties of rutin-casein complexes [J].Food & Machinery,2025,41(7):9-17.

Lung cancer remains the leading cause of cancer-related death globally, with metastasis driven by circulating tumor cells (CTCs)-particularly clusters-being a major treatment challenge. Despite their critical role, the biological differences between single CTCs and CTC clusters remain unclear. Here, we comprehensively compared their behavioral, transcriptomic, and proteomic profiles in lung cancer models. Compared with single cells, CTC clusters present enhanced metastatic potential, greater survival in the bloodstream and increased resistance to microenvironment. Any failure in frontonasal development can lead to malformations at the middle facial region, such as frontonasal dysplasia, midfacial clefts, and hyper/hypotelorism. Various environmental factors influence morphogenesis through epigenetic regulations, including the action of noncoding microRNAs (miRNAs). However, it remains unclear how miRNAs are involved in the frontonasal development. In our analysis of publicly available miRNA-seq and RNA-seq datasets, we found that miR-28a-5p, miR-302a-3p, miR-302b-3p, and miR-302d-3p were differentially expressed in the frontonasal process during embryonic days 10.5 to 13.5 (E10.5–E13.5) in mice. Background: Despite the global public health threat posed by carbapenem-resistant Enterobacter spp, clinical and molecular epidemiological studies on international isolates remain scarce. Historically, the taxonomy of Enterobacter has been challenging, limiting our understanding of the clinical characteristics and outcomes of carbapenemase-producing Enterobacter spp infections.
Methods: Hospitalized patients enrolled in the CRACKLE-2 study (ClinicalTrials.gov, NCT03646227) from 2016 to 2018 with cultures positive for carbapenemase-producing Enterobacter spp were included. Clinical and microbiologic data were collected from health records. Whole genome sequencing was performed, and the population structures of selected predominant clones were analyzed.

Malaria is a vector-borne disease and is a major global public health threat caused by the genus Plasmodium. Of the five species that infected humans, Plasmodium vivax and Plasmodium falciparum contribute the highest rates of morbidity and mortality. Research efforts to develop antimalarial drugs and effective vaccines is imperative to the prevention of future cases. Plasmodium has three reproductive stages: sporogonic, liver, and blood. Respiratory syncytial virus (RSV) is a significant cause of morbidity and mortality in high-risk populations. Although prophylactic options are available, there are no effective oral therapeutics for RSV infection. Obeldesivir (ODV) is an orally bioavailable prodrug of the nucleoside analog GS-441524, which is converted intracellularly to its active nucleoside triphosphate and inhibits the RSV RNA polymerase. Here we report the potent antiviral activity of ODV against geographically and temporally diverse RSV A and B clinical isolates (EC50: 0.20–0.66 μM). [Objective] To establish a method for determining the total vitamin B12 content in infant formula milk powder based on the indirect detection of α-ribazole,a degradation product and nucleoside component of vitamin B12.[Methods] After protein denaturation and sugar removal,the sample underwent acid hydrolysis and enzymatic dephosphorylation to release α-ribazole,which was then purified using a boronate affinity column.Quantitative analysis was performed using hydrophilic interaction liquid chromatography with fluorescence detection (HILIC -FLD ).[Results]] Alpha -ribazole showed good linearity in the range of 1.00~10.0 μg/L,with a correlation coefficient (R²) of 0.999 1.The spiked recovery ranged from 72.5% to 78.3%,with relative standard deviations (RSDs ) …
