Sperm cryopreservation is a basic element of assisted reproductive technology, yet it often compromises cell viability through thermal and osmotic stress. The cellular response involves HSP90, a stress marker and ZPII, which is essential for sperm-oocyte interaction and preventing polyspermy. In addition, this modified cryoprotectant combines glycerol and raffinose is expected to reduce damage and increase spermatozoa's recovery rate after cryopreservation. To address these challenges, we investigated the expression levels of HSP90 and ZPII to assess the protective capacity of modified cryoprotectant formulations. Groundwater-obligate taxa (stygobionts) exhibit variable degrees of habitat specialization, ranging from those restricted to single micro-habitats to generalists occurring in a broad range of habitat types. The karstic Edwards Aquifer is the most biodiverse and well-studied groundwater habitat in the continental United States, but some Edwards Aquifer-associated species also occupy the hyporheic zone of rivers downstream from karst. Langerhans cell histiocytosis (LCH) is a rare malignancy marked by clonal proliferation of Langerhans cells, with BRAF V600E mutations identified in over 50% of the cases. BRAF inhibitors (BRAFi), such as dabrafenib, have shown efficacy in treating BRAF V600E-mutant LCH. However, BRAFi therapy is associated with cutaneous adverse effects, including the development of new melanocytic nevi, verrucae, and keratinocyte carcinoma (KC). This case report describes a 78-year-old woman with BRAF V600E-mutant LCH who developed multiple dermatologic side effects following dabrafenib salvage therapy. Background and Objectives: Vamorolone demonstrated similar efficacy for Duchenne muscular dystrophy (DMD) compared with prednisone in a 24-week exploratory analysis and may reduce key side effects compared with classic corticosteroids. In this study, we compare the efficacy and anthropometric effect of vamorolone 6 mg/kg/d with prednisone 0.75 mg/kg/d and deflazacort 0.9 mg/kg/d in steroid-naïve boys aged 4 to < 7 years using data from 2 trials. Methods: VISION-DMD was a phase 2b, 48-week randomized, double-blind trial assessing vamorolone 2 and 6 mg/kg/d vs placebo and prednisone 0.75 mg/kg/d to 24 weeks. Finding the Optimum Steroid Regimen for DMD (FOR-DMD) was a double-blind, parallel-group randomized trial comparing daily prednisone 0.75 mg/kg/d, daily deflazacort 0.9 mg/kg/d, and intermittent prednisone 0.75 mg/kg (10/10 days on/off). Entropy balancing generated weighted data for mixed model for repeated measures analyses that compared VISION-DMD vamorolone 6 mg/kg/d efficacy and anthropometric outcomes with FOR-DMD prednisone and deflazacort outcomes at 6 and 12 months. Inclusion criteria were boys with genetically confirmed DMD, age 4 to < 7 years, ambulatory, and able to complete a time-to-stand (TTSTAND) assessment in < 10 seconds at baseline. Results: All interventions showed motor improvements relative to baseline at 6 and 12 months. Using the weighted cohorts and at 12 months, vamorolone 6 mg/kg/d (n = 28) had similar changes in TTSTAND velocity, the primary motor endpoint component in both trials, vs daily prednisone (n = 50) or deflazacort (n = 55; mean baseline age 5.42 years all groups), but with CIs overlapping minimal clinically important thresholds of 0.023 rises per second (TTSTAND velocity least squares mean [LSM] difference [95% CI]: vamorolone 6 mg/kg/d vs prednisone 0.75 mg/kg/d, 0.004 rises per second [−0.025 to 0.032] and vs deflazacort 0.9 mg/kg/d, 0.001 rises per second [−0.027 to 0.028]). Body mass index (BMI) Z-scores increased in all groups, greatest with vamorolone (12-month LSM difference [95% CI]: vamorolone vs prednisone 0.57 [0.24–0.90]; vamorolone vs deflazacort 0.29 [0.03–0.56]), whereas growth slowdown occurred in prednisone- and deflazacort-treated boys but not vamorolone (12-month LSM difference in height Z-scores [95% CI]: vamorolone vs prednisone 0.57 [0.24–0.90]; vamorolone vs deflazacort 0.72 [0.53–0.91]). Discussion: Vamorolone demonstrated numerically similar TTSTAND velocity changes to prednisone and deflazacort at 1 year; however, interpretations of differences are limited by 95% CIs crossing minimally important difference thresholds. Further evidence of the growth-protective effect of vamorolone was observed; however, all treatments increased BMI. Vamorolone provides a linear growth-protective classic corticosteroid alternative.

Divalent cations, particularly calcium (Ca2+) and magnesium (Mg2+), are essential for maintaining chromosomal architecture, yet their roles in plants remain poorly characterized. This study investigated the effects of Ca2+ and Mg2+ on the chromosomal structure of wheat (Triticum aestivum, 2n = 6x = 42), a widely used model in plant cytogenetics. Root-tip chromosomes were isolated enzymatically and treated with BAPTA (a Ca2+-specific chelator), EDTA (a broad cation chelator), and buffer solutions containing either 5 mM Mg2+ (XBE5) or no Mg2+ (XBE). Oxytropis is a taxonomically challenging genus within the tribe Galegeae of the family Fabaceae. Despite its systematic importance, the seed morphology of Oxytropis has been poorly investigated. In the present study, we examined the seed morphology of 18 Oxytropis species representing 10 sections from the Pakistan, with the aim of evaluating the diagnostic value of seed traits using both light and scanning electron microscopy. The current investigation revealed two distinct seed shapes, namely cordiform and prolonged reniform, across the studied taxa. Documenting species, particularly those that are threatened or endangered, is fundamental to biodiversity conservation. However, many taxa remain scientifically unrecognized, being either undiscovered or unpublished. While some plants can be easily recognized as new species, other identifications are obscured by morphological, taxonomic, and nomenclatural complexity. Detecting cryptic or “hidden” species is therefore essential to understanding biodiversity and plant evolution. Our revisionary studies on Salvia demonstrate that the morphologically complex Salvia caespitosa is not a single species but instead comprises at least four distinct species, three with narrow distributions. KMT2A-rearranged (KMT2A-r) acute leukemias are especially prevalent in the pediatric population. KMT2A-fusion proteins drive leukemogenic gene expression through an interaction with a chromatin complex that includes the scaffold protein menin, giving rise to aggressive acute leukemias. RAS pathway mutations are also common in pediatric leukemia. In a cohort of 1750 patients enrolled on Children's Oncology Group (COG) trials, we identified RAS pathway mutations in 43% of acute myeloid leukemia (AML) cases. The presence of RAS pathway mutations in KMT2A-r AML was associated with a lower complete remission rate, poor event-free survival and overall survival (OS), and early relapses. Given the …

Breast cancer prevalence and incidence are increasing globally every year. Medication resistance and recurrence are more likely in breast cancer cells that express CD44+/CD24–. C.nutans leaves are a traditional medicinal herb with numerous pharmacological benefits, including antioxidant and antiproliferation properties, although the activity against breast cancer stem cells is still unclear. Salinity stress is one of the major abiotic factors that significantly impact plant growth. In this regard, this work investigated a way to alleviate the adverse impacts by simultaneously spraying methyl jasmonate and titanium nanoparticles on lettuce (Lactuca sativa L.) plants, which are grown under induced salinity conditions. Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by synovial inflammation and autoantibody production. This study aimed to assess serum levels of TNF-α , IL-17, and IL-13 in RA patients and to explore their association with disease severity and autoantibody status in comparison to healthy controls. The study involved 67 RA patients and 20 healthy individuals. Patients were initially classified based on autoantibody status (RF-positive vs. RF-negative and anti-CCP-positive vs. anti-CCP-negative), and further categorized into low, moderate, and high disease activity groups. Teachers are the foundational component of the educational system, yet many enter the classroom without adequate knowledge of dyslexia -- a specific learning disability affecting up to 17.5% of the student population. Despite growing legislative mandates requiring teacher training on dyslexia, recent research confirms that misconceptions and knowledge gaps persist among both preservice and inservice teachers, and the question of which professional development models most effectively address those gaps remains largely unanswered. (Scorpiones: Caraboctonidae) is newly recorded as a successfully intercepted non-native invasive species in Bermuda, based on examination of a desiccated and damaged immature female specimen. A second record of an intercepted Centruroides gracilis (Latreille, 1804) (Scorpiones: Buthidae) is reported based on examination of an adult male.

[Objective] To investigate the influence of three extraction methods (alkaline, enzymatic, and acidic) on the emulsification characteristics of corn fiber gum (CFG). [Methods] Corn bran was used as the raw material, and CFG was extracted using sodium hydroxide, xylanase, and formic–acetic acid–water solutions. The changes in particle size, zeta potential, rheological properties, creaming index, appearance, and microstructure of CFG emulsions were measured. [Results] All three CFG emulsions exhibited shear-thinning behavior, with viscosities decreasing as the shear rate increased. [Objective] To establish a model for the rapid quantification of sorghum starch based on near-infrared spectroscopy and machine learning algorithms. [Methods] Brewing sorghum was used as the raw material. Near-infrared diffuse reflectance spectra were collected using a near-infrared spectrometer. The Isolation Forest algorithm was employed to remove outlier samples and optimize the dataset. Subsequently, multiple preprocessing methods and regression models were compared using various modeling software to analyze and establish a quantitative model for sorghum starch. [Methods] With ultrasonic temperature, power, time, as well as solid–liquid ratio and ethanol volume fraction as experimental factors, the response surface method was employed to optimize the ultrasonic extraction process of polyphenols from C. anisum-olens kernel. [Objective] To prepare Flammulina velutipes crude polysaccharide (FVP) and evaluate its structural characteristics and immunomodulatory activity. [Methods] FVP was prepared from Flammulina velutipes using hot-water extraction and ethanol precipitation, and its structural characteristics were characterized by combining Fourier transform infrared spectroscopy (FTIR) and high-performance size-exclusion chromatography–multi-angle laser light scattering–refractive index detection (HPSEC–MALLS–RI). The immunomodulatory activity of FVP was evaluated using RAW 264.7 macrophages and a cyclophosphamide-induced immunosuppressed mouse model. [Objective] To investigate the effects of probiotic fermentation conditions and drying methods on the quality of fermented sea buckthorn leaf products. [Methods] Lactiplantibacillus plantarum is used for fermentation. The fermentation process of sea buckthorn leaves is optimized through single-factor experiments combined with response surface methodology. Fermented sea buckthorn leaf extract is prepared under the optimal fermentation conditions and subsequently processed into freeze-dried powder by direct freeze-drying and spray freeze-drying, with or without low-voltage electrostatic field assistance.
