Amygdala Npy+ Interneurons Are Activated By Aversive Stim-Uli, Patric Perez
Amygdala Npy+ Interneurons Are Activated By Aversive Stim-Uli, Patric Perez
All ETDs from UAB
Neuropeptide Y (NPY) serves as a critical endogenous anxiolytic and stress-buffering molecule, yet the temporal and spatial dynamics of NPY+ cells during fear learning re-main unknown. Temporal dynamics can be measured with in vivo calcium imaging tech-niques, which enable real-time monitoring of cell-type-specific neural activity in freely behaving animals. Snapshots of neuronal activation can be measured using c-Fos im-munohistochemistry, thereby providing excellent spatial resolution. Here, I employ cut-ting-edge approaches to characterize the temporal and spatial activation patterns of NPY+ cells in amygdala, a key fear-regulating brain region negatively impacted by aversive events. I first used in vivo fiber photometry, together …
Understanding Psychotic Disorders Using Normative Modeling-Derived Individual-Level Brain Structural Abnormalities, Natalie Remiszewski
Understanding Psychotic Disorders Using Normative Modeling-Derived Individual-Level Brain Structural Abnormalities, Natalie Remiszewski
All ETDs from UAB
Psychosis is an heterogenous condition that occurs in many psychiatric disorders. It has a variable symptom presentation and recent initiatives have suggested that investigating the underlying neurobiology of psychiatric disorders rather than their symptomatology would provide a more accurate picture for diagnosis, prognosis, and treatment. Grey matter has been implicated in the pathophysiology of psychosis and psychotic disorders, though significant natural variation due to age and sex have impeded the distinction of pathologically relevant grey matter abnormalities in psychosis. Additionally, case-control approaches to exploring grey matter abnormalities in psychotic disorders often obscure subtle variation in individuals. Here, we utilize a …
Standardizing Sea Urchin Husbandry: Tank Effects, Gamete Production, And Co-Culture, Raven Amber Edwards
Standardizing Sea Urchin Husbandry: Tank Effects, Gamete Production, And Co-Culture, Raven Amber Edwards
All ETDs from UAB
This dissertation seeks to further knowledge of sea urchin husbandry in three major areas that have so far not been investigated substantially: optimal conditions for housing sea urchins in a lab-based recirculating aquaculture systems (RAS), impacts of repetitive spawning regimens on reproductive output, and co-culture utilizing juvenile oysters. All studies utilize a commercial recirculating aquaculture system that is relatively novel for culturing sea urchins. The first chapter investigated the influence of housing conditions on sea urchin growth outcomes. Under the conditions of this study, the size of the tank showed limited influence on growth; however, stocking density was found to …
Structural And Functional Investigation Of The Influenza Nuclear Export Protein, Andrew James Staup
Structural And Functional Investigation Of The Influenza Nuclear Export Protein, Andrew James Staup
All ETDs from UAB
The influenza A nuclear export protein (NEP) is a multi-functional protein that mediates the export of progeny viral ribonucleoprotein (vRNP) complexes from the host nucleus, regulates the activity of the RNA-dependent RNA polymerase (RdRp), and is implicated in zoonosis. It is therefore critical to understand the mechanisms of NEP’s functions at the molecular level to inform its roles in the viral lifecycle and influenza zoonosis. In this dissertation, we report the structure of NEP in complex with the host nuclear export holo-complex as well as investigate the biophysical effects of the adaptive M16I mutation on NEP. We solved the cryo-electron …
Pao Induces Er Stress In Endothelial Cells Leading To Barrier Function Dysfunction And Inhibition Of Brd4 Prevents Lewisite-Induced Peribronchial Fibrosis, Huaxiu Zeng
All ETDs from UAB
Arsenicals are members of one of the most important global environmental vesicants and are a persistent bio accumulative carcinogen. One of the vesicants and chemical warfare agents is lewisite, which causes lung injury with pulmonary edema, hemorrhage, and respiratory failure. A single cutaneous exposure to lewisite in mice induces systemic lung injury through arsenical-mediated mechanisms. Some subjects develop Bronchiolitis Obliterans Syndrome (BOS) where there is evidence of airway dysfunction and pathologic narrowing of the airways with extracellular protein deposition and fibroblast invasion. In vitro for human lung endothelial cells, Phenyl arsine Oxide (PAO) exposure, as surrogate of lewisite, leads to …
Proteasome Augmentation Is Protective Against Alzheimer’S Disease And Agerelated Cognitive Decline, Kanisa Davidson
Proteasome Augmentation Is Protective Against Alzheimer’S Disease And Agerelated Cognitive Decline, Kanisa Davidson
All ETDs from UAB
Proteostasis is essential for the degradation of oxidized, misfolded proteins and proper function of neuronal plasticity. Proteasome inhibition has been shown to exacerbate cognitive and behavioral deficits in aging and Alzheimer’s Disease (AD) models. Proteasomal activity is decreased across human and animal models of AD. We hypothesize that aging and AD markers, β-amyloid and/or hyperphosphorylated tau, impair proteasome function, thereby preventing the proteasome’s capacity to degrade critical synaptic proteins essential for memory formation. We investigated the role of pharmacological and genetic augmentation on proteasomal function, to rescue aging and AD-like cognitive deficits in cell culture, Drosophila, and mice. We demonstrate …

Strength Of Self-Adhesive Resin Cements To Shortened Zirconia Crowns With And Without An Additional Coat Of Adhesive, Abraham John
All ETDs from UAB
Self-adhesive cements have gained popularity due to their ease of manipulation as they do not require a separate etch, primer, or bonding agent to bond to the tooth or crown surfaces. However, the use of an adhesive bonding agent with a self-adhesive resin cement has been claimed to improve its bond strength. The objective of this study is to compare the retention bond strength of 2 self-adhesive cements on to the tooth surface with and without the use of bonding agents with a known control. Materials and Methods: A total of 50 extracted non-carious second premolar teeth were collected. The …

Psychedelics And Psychosis: An Interdisciplinary Re-Examination, Haley Maria Dourron
All ETDs from UAB
Psychedelics have long been described as potential drug models of psychosis. There is also concern that psychedelics might worsen psychotic symptoms in individuals with a history of psychotic disorders. This dissertation examines the overlap between psychedelic-induced states and non-affective psychotic disorders (NAPD) from both subjective and neurological perspectives, as well as the perceived impact of psychedelic use in people with a history of NAPD. Nineteen people with a history of NAPD who had used psychedelics were interviewed about their experiences. Participants generally described psychedelic and psychotic experiences as distinct, though both affected similar experiential domains, such as sensory alterations. Altered …
Murder On The Axonal Express: Synucleinopathy Disruption Of 14-3-3 Protein-Protein Interactions Impairs Axonal Trafficking, Frank Sanders Pair
Murder On The Axonal Express: Synucleinopathy Disruption Of 14-3-3 Protein-Protein Interactions Impairs Axonal Trafficking, Frank Sanders Pair
All ETDs from UAB
14-3-3s are a family of proteins highly expressed in the brain. These proteins exert their functions mainly through protein-protein interactions (PPIs) and have thousands of binding partners. 14-3-3s have been shown to be present in pro-teinaceous aggregates of alpha-synuclein (αsyn), a pathological hallmark of synucleinopathies. This includes such diseases as Parkinson’s Disease (PD) and Dementia with Lewy Bodies (DLB). 14-3-3 proteins, particularly the 14-3-3θ isoform, have been shown to be protective in multiple synucleinopathy models. However, reduced soluble levels of 14-3-3s and increased phosphorylation of 14-3-3θ at S232 observed in synucleinopathies like DLB may impair their protec-tive ability, potentially due …

Origin And Differentiation Of Uterine Natural Killer Cells, Morgan Elizabeth-Carolyn Greene
Origin And Differentiation Of Uterine Natural Killer Cells, Morgan Elizabeth-Carolyn Greene
All ETDs from UAB
Uterine natural killer (uNK) cells are specialized tissue-resident lymphocytes that are essential for successful pregnancy, yet the molecular pathways governing their development remain poorly defined. This knowledge gap hinders our ability to understand and treat immunological complications of pregnancy, such as recurrent pregnancy loss and pre-eclampsia. This dissertation addresses the fundamental question of how human uNK cells arise and differentiate, first within the non-pregnant endometrium and subsequently within the decidua of early pregnancy. Using single-cell RNA sequencing of matched human blood and endometrial samples, we first delineate the developmental trajectory of endometrial NK (eNK) cells. We demonstrate that eNK cells …

Schizophrenia is a complex psychiatric disorder characterized by hallucinations, delusions, disorganized thinking, and abnormalities in emotional expression, social interaction, and motivation. Cognitive deficits and alterations in perception are core features of psychosis spectrum disorders, though the neurobiological bases of these impairments are not thoroughly understood. Examination of the structures responsible for cognition, along with how their connectivity and neurochemistry are altered in psychosis, is crucial to understanding and treating schizophrenia. Gamma-aminobutyric acid (GABA) is the primary inhibitory neurotransmitter in the central nervous system and plays a crucial role in the regulation of neuronal activity. Evidence from animal models and postmortem …
Mechanistic Rationale For Toxic Aerosol Formation From Vaping And Dabbing With A Focus On Ketene Formation From Semi-Synthetic Cannabinoids, Kaelas Ryan Munger
Mechanistic Rationale For Toxic Aerosol Formation From Vaping And Dabbing With A Focus On Ketene Formation From Semi-Synthetic Cannabinoids, Kaelas Ryan Munger
Dissertations and Theses
Cannabis vaping is a relatively new route of administration that is increasingly common among youth and young adults. Cannabis vaping has been associated with cases of acute lung injury, especially among youth, and most often involves the heating and aerosolization of high-potency products having cannabinoid concentrations of ~ 50 % - 98 % w/w. Proponents argue that vaping is safer than smoking due to the characteristically lower temperatures and fewer plant materials. However, the extreme cannabinoid concentrations, along with non-natural formulations having undisclosed additives, require studies of the health effects of these products. Despite the popularity of cannabis concentrate vaping, …

Scallops (order Pectinida) are well-known for their robust and beautiful calcitic shells that protect them from external impacts and predators. Scallops respond to environmental conditions, including water temperature, salinity, and food supply, which are reflected in the shell growth rates and patterning. The bay scallop (Argopecten irradians) is a species of high ecological and aquacultural value in North America, and its habitat may expand towards higher latitudes with inevitable global warming. To investigate the effect of water temperature on the accretion rate and the polycrystalline microstructure of the Bay scallop shell, we conducted a controlled growth experiment on juveniles, 4 …

Combined Crystallographic Study Of King Scallop (Pecten Maximus) Shells Using Sem, Ebsd And Raman Spectroscopy, Lise Guichaoua, Natalie Reznikov, Bryce D. Stewart, Roland Kröger, Raynald Gauvin
School of Biological and Marine Sciences
The shells of Pecten maximus (king scallop) are composed primarily of polycrystalline calcitic calcium carbonate, with a crucial aragonitic prismatic myostracum layer that facilitates soft tissue attachment and contributes to mechanical strength. Despite its importance, the impact of environmental stressors, such as metal contamination, on the myostracum remains underexplored. Hence, this study's main goal was to shed light on the microstructure and crystallography of king scallop shells, particularly the myostracum region, using a combination of scanning electron microscopy (SEM), electron backscatter diffraction (EBSD) analysis, and Raman spectroscopy. This approach was chosen to develop new protocols that integrate imaging techniques for …

Neurotransmitter Signaling In Molecular And Behavioral Immune Responses To Pathogens In C Elegans, Benson Otarigho, Alejandro Aballay
Neurotransmitter Signaling In Molecular And Behavioral Immune Responses To Pathogens In C Elegans, Benson Otarigho, Alejandro Aballay
Faculty, Staff and Student Publications
Neurotransmitter signaling pathways play major roles in both molecular and behavioral defenses against pathogen invasion, shaping the ability of Caenorhabditis elegans to sense and respond to environmental challenges. Given the conservation of neurotransmitter signaling pathways, their understanding may not only provide insights into the neurobiology of C. elegans but also has broader implications for our understanding of neural-immune interactions and host defense mechanisms in higher organisms. In this review, we discussed the literature on various neurotransmitter signaling pathways, including serotonergic, dopaminergic/octopaminergic, GABAergic, and glutamatergic pathways, and how these pathways modulate molecular and behavioral immune defense against pathogens.

International Journal of Exercise Science 18(1): 1010-1029, 2025. The conflation of sex and gender in sport and exercise science research has led to gaps in representation and understanding of gender-inclusive outcomes. This invited editorial presents a framework to guide researchers in conducting statistical analyses that account for multiple gender identities beyond the traditional binary classification. The invited editorial guides deliberation on whether to test for sex or gender differences in sport or exercise science research. It prompts investigators to answer the question, “Is there a valid, literature-supported rationale for investigating sex or gender differences?” If “yes”, we propose approaches that …

A Model Of The Determinants Of Maternal Mortality In Indigenous Women, Jennifer L. Heck, Judy Goforth Parker, Kathryn M. L. Konrad, Ingrid Svenson, Annika Jennings
International Nursing Research Congress (INRC)
Despite increasing attention to the well-documented maternal health crisis in the U.S., stark disparities persist between groups of women. In December 2021, Indigenous mothers experienced 118.7 maternal deaths per 100,000 live births, whereas their White counterparts had 26.6 [1]. Maternal mortality is a multifaceted, complex issue that complicates the health and wellbeing of Indigenous women, yet the root causes of this disparity are not well-described. We need a deeper understanding of the determinants of health (social, structural, biological, political) that contribute to Indigenous maternal mortality. Indeed, the existing literature lacks adequate theoretical understanding to address this phenomenon. Therefore, the purpose …

Online, asynchronous professional development (PD) offers flexibility for learners, yet what is the most effective way to provide online PD? While current research suggests online platforms can be effective for learning, there is a lack of studies examining differences between fully autonomous, asynchronous modules and those that are asynchronous but still collaborative. This mixed-method study examined the learning differences and perceptions between two cohorts of special education teachers as they went through an eight-unit online course on dyslexia. One cohort received an autonomous platform without interaction among the students or instructor. The other cohort received the same content, but online …
