BACKGROUND: Epigenetic modifications such as DNA methylation play a critical role in hypoxic cell programs. However, no previous studies have investigated the epigenetic regulation of gene expression in peripheral artery disease (PAD), a condition characterized by intermittent ischemia. Our study aims to examine how PAD affects the DNA methylome in skeletal muscle of patients with PAD with intermittent claudication or critical limb ischemia compared with non-PAD controls. METHODS: This was a cross-sectional study evaluating differences in the DNA methylome and transcriptome using reduced representation bisulfite sequencing, and small and bulk RNA sequencing, in skeletal muscle from non-PAD controls, intermittent claudication, …
High-Order Adaptive Solutions For Coupled Fractional Riccati Equations Via Daubechies Redundant Frames, Mutaz Mohammad, Alexander Trounev
High-Order Adaptive Solutions For Coupled Fractional Riccati Equations Via Daubechies Redundant Frames, Mutaz Mohammad, Alexander Trounev
Mathematical Modelling and Numerical Simulation with Applications
Coupled fractional Riccati equations play a fundamental role in modeling complex systems with memory effects and anomalous diffusion, frequently arising in engineering, control theory, finance, and quantum mechanics. Their analytical and numerical treatment remains highly challenging due to the nonlocal nature of fractional-order derivatives and the presence of nonlinear coupling terms. This study introduces an adaptive numerical framework that combines the Caputo fractional derivative with redundant Daubechies wavelet frames. The method leverages multi-resolution analysis, compact support, and controlled redundancy to achieve accurate approximation of both localized and global solution features, particularly in scenarios characterized by singular behavior and long-range memory …

Stabilized Weak-Gradient Discontinuous Finite Elements With Optimal Error Estimates For Second-Order Elliptic Pdes, Aymen Laadhari
Mathematical Modelling and Numerical Simulation with Applications
This work introduces an accurate finite element approach employing a new stabilized discrete weak gradient, designed for second-order elliptic problems on arbitrary conforming meshes. We formulate the approach within a discontinuous Galerkin framework and derive a consistent and coercive bilinear form. Appropriate error analysis on a model problem confirms optimal convergence. Building on the core analysis, we extend the method to more challenging settings, including time-dependent heterogeneous scenarios and a biophysically realistic optimal-control model of photobleaching in the budding yeast cell. We further illustrate the versatility of the weak-gradient construction by applying it to an unsteady level-set equation relevant to …

Global Stability And Bifurcation Analysis Of A Predator-Prey Model Involving Allee Effect And Monod-Haldane Functional Response, Resmawan Resmawan, Agus Suryanto, Isnani Darti, Hasan S. Panigoro
Mathematical Modelling and Numerical Simulation with Applications
In this paper, the complexity of the dynamic behavior of the interaction between prey and predator is studied. The predator-prey relationship involves Allee effects and Monod-Haldane functional response. The constructed model has been shown to have validity in several respects, including the existence and uniqueness of the solution, as well as its non-negativity and boundedness. Three equilibrium points, namely trivial, axial, and coexistence points, are found, including their global dynamics using the Lyapunov function together with the LaSalle's invariance principle. The effect of the predation conversion rate causes changes in the dynamic behavior of predators and prey, which is characterized …

Solution Of Fractional Order Diffusion Equations With Clique Neural Network, Merve Zeynep Kaya, Mesut Karabacak, Ercan Çelik
Mathematical Modelling and Numerical Simulation with Applications
In this paper, the clique artificial neural network method is used to solve the fractional diffusion equation, which is a subclass of partial differential equations. The clique neural network architecture is constructed using input, hidden, and output layers. Several degrees of clique polynomials were used as activation functions, and the output layer was obtained by multiplying them with weight coefficients. Subsequently, the optimization equation was derived, and the exact solution, numerical solution, and error function graphs were obtained using a specialized algorithm. Analysis of the results demonstrates that the clique artificial neural network method provides quicker and more accurate results …

Synergistic Modeling Of Hydrogel Gelation Via Time-Delay Dynamics And Machine Learning Algorithms, Mine Babaoglu, Dipesh ., Pankaj Kumar, Jagjit Singh Dhatterwal, Mansoor Alsulami
Mathematical Modelling and Numerical Simulation with Applications
This paper presents an integrated framework in which delay differential equation (DDE) modeling and machine learning (ML) approaches are coupled to study hydrogel formation kinetics, with emphasis on delayed crosslinker addition. Conventional mechanistic models disclose many physical and kinetic complexities of reacting mixtures; they seldom depict the nonlinear and time-evolving complexities inherent in developing polymer networks. To address this, a mathematical model is developed that examines how the insertion of crosslinkers affects system stability and equilibrium. Analytical and numerical results show that delays nearing critical levels cause bifurcation behavior with substantial implications on gelation kinetics. Sophisticated machine learning systems, including …

Dynamics And Optimal Intervention Strategies In A Shigellosis Transmission Model, Mehmet Gümüs, Shewafera Wondimagegnhu Teklu, Kemal Türk
Mathematical Modelling and Numerical Simulation with Applications
This research explores how water treatment contributes to limiting the transmission of Shigellosis, an infection caused by bacteria from the Shigella genus. A mathematical framework is formulated to evaluate the influence of protective strategies and water purification on the spread of the disease. To confirm the model's biological relevance, its well-posedness is investigated. The basic reproduction number $(\mathfrak{R}_0)$, a critical indicator of disease behavior, is derived using the matrix operator method. Findings indicate that if $\mathfrak{R}_01$, the infection persists, with the endemic equilibrium exhibiting local asymptotic stability. A comprehensive cost-effectiveness analysis reveals that combining environmental protection with water treatment represents …

Human T-cell leukemia virus type 1 (HTLV-1), the first oncogenic human retrovirus, causes adult T-cell leukemia/lymphoma (ATLL), an aggressive neoplasm of mature CD4+ T-cells that is incurable in most patients and is associated with a median survival of less than 1 year. HTLV-1 also causes inflammatory disorders, including HTLV-associated myelopathy/tropical spastic paraparesis (HAM/TSP) and uveitis. The estimated lifetime risks of ATLL and HAM/TSP in HTLV-1 carriers are 3-5% and 0.25-1.8%, respectively. Although there is uncertainty about other health effects of HTLV-1, a recent meta-analysis showed an association between HTLV-1 and cardiovascular, cerebrovascular, and metabolic diseases and a 57% increased risk …

The Lived Experiences Of Mothers Facing Phubbing From Their Adolescent Children, Mye Alexis
Walden Dissertations and Doctoral Studies
Widespread use of technology, particularly smartphones, introduced new challenges involving parent–adolescent attachment. Although existing literature primarily examines parental phubbing of adolescents, there is a significant gap in research exploring the experiences of mothers who are phubbed by their adolescent children. To address this gap, the central research question guiding this study was: What are the lived experiences of mothers who have been phubbed by their adolescent (ages 14-18) children? Grounded in Bowlby’s attachment theory, this phenomenological qualitative study aimed to deepen understanding of the emotional impacts of phubbing on mothers’ emotional wellbeing, emotional bonding, and mother–adolescent attachment. A homogeneous convenience …

Covalent inhibitors are an emerging class of therapeutics, but methods to comprehensively profile their binding kinetics and selectivity across the proteome have been limited. Here we introduce COOKIE-Pro (COvalent Occupancy KInetic Enrichment via Proteomics), an unbiased method for quantifying irreversible covalent inhibitor binding kinetics on a proteome-wide scale. COOKIE-Pro uses a two-step incubation process with mass spectrometry-based proteomics to determine kinact and KI values for covalent inhibitors against both on-target and off-target proteins. We validated COOKIE-Pro using BTK inhibitors spebrutinib and ibrutinib, accurately reproducing known kinetic parameters and identifying both expected and unreported off-targets. The method revealed that …

Social learning enables a subject to make decisions by observing the actions of another. How neural circuits acquire relevant information during observation to guide subsequent behavior is unknown. Utilizing an observational spatial working memory task, we show that neurons in the rat anterior cingulate cortex (ACC) associated with spatial trajectories during self-running in a maze are reactivated when observing another rat running the same maze. The observation-induced ACC activities are reduced in error trials and are correlated with activities of hippocampal place cells representing the same trajectories. The ACC activities during observation also predict subsequent hippocampal place cell activities during …

Alterations in lipid profiles have been shown in patients with chronic obstructive pulmonary disease (COPD), but the underlying molecular mechanisms remain unclear. In this study, we aimed to investigate the role of cytochrome P450 family-1 subfamily B member 1 (CYP1B1) in cigarette smoke (CS)-induced lipid accumulation in alveolar type II epithelial (AT2) cells. We observed a steady increase in CYP1B1 protein levels in AT2 cells from COPD patients. Additionally, CS exposure induced CYP1B1 expression in AT2 cells of murine lungs. In vitro, treatment with cigarette smoke extract (CSE) not only upregulated CYP1B1 expression but also triggered lipid accumulation in AT2-like …

The lymphatic system is critical to the body's immune and circulatory system, and lymphangiogenesis, the development of new lymphatic vessels from pre-existing ones, is a significant process capitalized upon by cancer during tumorigenesis. Decorin is a small leucine-rich proteoglycan which we have previously shown to be anti-tumorigenic and a suppressor of lymphangiogenesis. We have also shown that decorin exercises its anticancer properties through its ability to evoke autophagy. Through a comprehensive and unbiased proteomic analysis, we explored the implications of decorin exposure on protein expression within mouse lymphatic endothelial cells. We discovered that decorin enriches several protein pathways, notably proteasomal …

Poultry provides both direct and indirect employment opportunities to individuals. Every farm owner desires a reduced investment with more earnings. Over 50 % of the overall cost of chicken rearing is allocated to feed stuff. Among all feed ingredients, protein sources are the most expensive, yet protein is essential for production and growth. This exorbitant price cannot be sustained by the small-scale farmers. Consequently, contemporary scientists are attempting to substitute the protein supply with black soldier fly in their formulations. This review study examines the advantageous effect of utilizing Black Soldier Fly powder, larvae or oil, as demonstrated in scientific …

Leucine-rich repeat containing 15 (LRRC15) has emerged as an attractive biomarker and target for cancer therapy. Transforming growth factor-β (TGFβ) induces the expression of this plasma membrane protein specifically in aggressive and treatment resistant tumor cells derived from mesenchymal stem cells, with minimal expression observed in non-neoplastic tissues. We have developed a humanized monoclonal antibody, DUNP19, that specifically binds with high affinity to a phylogenetically conserved LRRC15 epitope and is rapidly internalized upon LRRC15 binding. In multiple subcutaneous and orthotopic tumor xenograft mouse models, Lutetium-177 labeled DUNP19 ([

Urban densification threatens biodiversity, yet conventional greenspace expansion is con-strained by limited land availability. Living wall systems (LWS) offer potential biodiversity enhancement through vertical green infrastructure, though their ecological value re-mains underexplored. This study evaluated the biodiversity performance of three LWS in Plymouth, UK, using multi-taxa surveys to assess invertebrate communities, bird assemblages, and bat activity. A scoping review of 2,638 publications revealed limited research on LWS biodiversity, with only 27% of biodiversity-focused papers referencing specific species. Field surveys employed standardized protocols including flower-visiting pollinator observations, spider assessments, soil invertebrate extraction using Tullgren funnels, acoustic bird monitoring, and bat emergence …

Background: Multimodality treatment {surgery + pre-operative chemoradiation [chemoradiotherapy for oesophageal cancer followed by surgery (CROSS)] or perioperative chemotherapy [fluorouracil, leucovorin, oxaliplatin and docetaxel (FLOT)]} is the standard of care for locally advanced oesophageal cancer. Whilst highly tolerated, clinically, patients may experience toxicities which negatively impact their functional status. Exercise prehabilitation aims to improve functional status in advance of oesophagectomy to minimise postoperative morbidities. The PRE-HIIT trial examined the ability of a high-intensity interval training (HIIT) exercise programme to improve cardiorespiratory fitness in advance of complex cancer surgery. This cohort study charts the physical functioning of a sub-cohort of participants recruited …

Trombidium grandissimum is also known by names, red velvet mite, real velvet mite, giant red velvet mite and rain bug, belongs to the family Trombidiidae and subclass Acari. Red velvet mite has four life stages: egg, larva, nymph, and adult. Adult red velvet mites were collected from agricultural farm area of Khwaja Fareed University of Engineering & Information Technology. Important taxonomic features were revealed by morphological study, including a body length of around half of an inch, velvety, unridged skin with dense hairs, stalked eyes, and specialized organs on the anterior legs including sensilla and claws. Carotene-induced bright red coloring …
