The breast and ovarian tumor suppressor BRCA1 is a cell cycle-regulated protein and tumors with reduced BRCA1 protein level may share molecular features of BRCA1-mutant tumor and respond to PARPi therapy. Here, we identify that BRCA1 protein stability is controlled through ubiquitin lysine 11 (K11)-linkage modification under the regulation of Cezanne deubiquitinating enzyme, APC/C E3 ligase, and Ube2S E2 conjugating enzyme in a cell cycle-dependent manner. Cezanne-deficiency leads to increased BRCA1 K11-ubiquitination, decreased BRCA1 protein level, and increased cellular sensitivity to PARPi. Black and brown carbon are byproducts of the incomplete combustion of biomass or fossil fuels. These light-absorbing aerosols influence climate by altering the radiative balance through the absorption and scattering of sunlight, exacerbating global climate change. Their subsequent deposition on terrestrial and marine environments affects surface albedo, snow and ice melt, and biogeochemical processes, leading to further environmental consequences. In contrast, green and blue carbon refer to carbon stored in terrestrial vegetation and marine ecosystems, respectively. Purpose: Stroke survivors may develop spasticity (post-stroke spasticity [PSS]) that can challenge activity and participation. Recognising the needs and expectations of people with PSS is crucial for enhancing care. This study is the first to employ social media listening to explore the experiences, unmet needs, and quality of life (QoL) of people with PSS.

Materials and methods: A subset of 417 patient-centric PSS-related posts published on major social media platforms was identified for analysis from 31 600 retrieved.

UNLABELLED: Metastasis is the main cause of prostate cancer-associated deaths, highlighting the urgent need to determine the mechanisms underlying prostate cancer progression. TROP2 (also known as tumor-associated calcium signal transducer 2) is an oncogenic transmembrane surface protein that is highly expressed in metastatic prostate cancer. Naturally occurring cleavage of TROP2 leads to a release of the TROP2 extracellular domain (TECD) into the extracellular environment. In this study, we identified an important functional role of TECD in prostate cancer metastasis. Background: Monocytes contribute to the initiation and propagation of venous thrombosis. Little is known about the roles monocytes play in arterial thrombosis, the cause of stroke and myocardial infarction.

Methods: We investigated how CCR2 (CC chemokine receptor 2) knockout (-/-)-mediated monocyte deficiency affects platelet function, blood coagulation, thrombus volume, and thrombolytic susceptibility in 666 male mice with FeCl3-mediated carotid arterial thrombosis, including 365 C57BL/6 wild type (WT) mice, 295 CCR2-/- mice, and 6 CX3CR1-GFP (CX3C chemokine receptor 1-green fluorescent protein) mice.

OxaliTEX (NOVO-111) is a novel gadolinium(III) texaphyrin-platinum(IV) complex that is under development for clinical trials. It was designed as a prodrug of oxaliplatin (1,2-diaminocyclohexane-platinum(II) oxalate) tethered to a tumor-affinic texaphyrin moiety to improve drug delivery to the tumor. However, oxaliTEX was not only more effective as an antitumor agent but also better tolerated in mice. Background: Ultra-high dose rate radiotherapy (typically defined >40 Gy/s) has shown promise for enhanced normal tissue sparing while maintaining tumor control, termed the FLASH effect when this biological response is observed. However, precise dose rate control remains a technical challenge in proton therapy systems, particularly for research applications investigating dose rate effects.

Purpose: To develop and validate a passive beam modulation technique using tungsten scatterers for controllable dose rate adjustment in a synchrotron-based proton therapy beamline without modifying accelerator parameters.

Purpose: To characterize the newly released GafChromicTM EBT4 film for transit dose measurement in high-dose rate brachytherapy (HDR-BT) surface applicators and to systematically investigate the relationship between transit dose and 192Ir source activity.

Methods: EBT4 film was used to measure dose at 3 mm depth from a horizontally oriented Leipzig-style surface applicator connected to a 192Ir HDR-BT remote afterloading unit. Transit dose was systematically characterized across eight different source activities ranging from 3.372 to 9.716 Ci (13,724 U to 39,544 U) using measurements with seven nominal dwell times (5-100 seconds, scaled to 10 Ci (40,700 U)). Background: Matrix metalloproteinase 7 (MMP-7) is a novel biomarker for diagnosis of biliary atresia (BA), the most common cholestatic liver disease in infancy. There is a pressing need to determine the utility of MMP-7 levels in infants with congenital heart disease (CHD) to avoid unnecessary invasive diagnostic procedures in this high-risk population. We investigated the utility of MMP-7 in discriminating BA from non-BA cholestasis in infants with CHD and whether MMP-7 elevation was present in infants requiring treatment for clinically significant PH.

Methods: This is a single-center cross-sectional study including infants < 180 days of age with cholestasis and serum MMP-7 levels collected from 2019 to 2023. Demographic data and descriptive statistics were summarized with medians with interquartile ranges and frequencies with percentages. Median MMP-7 levels were assessed via Wilcoxon rank-sum test.

Aims/hypothesis: Pancreatic ductal adenocarcinoma-related diabetes mellitus (PDAC-DM) is a paraneoplastic syndrome with a poorly understood pathophysiology. PDAC-DM is often clinically confused with type 2 diabetes, resulting in delayed cancer detection and poorly individualised hyperglycaemia treatment. We investigated whether these forms of diabetes can be distinguished at the metabolic level.

Methods: Adults with either cancer treatment-naive PDAC-DM (n=28) or type 2 diabetes (n=97), and with diabetes onset within 3 years, underwent mixed-meal tolerance tests to investigate glucose metabolism. Vision-threatening retinal diseases, such as diabetic retinopathy (DR) and diabetic macular edema (DME), are major causes of preventable blindness worldwide. Early screening and follow-up are essential, but advanced imaging like Optical Coherence Tomography (OCT) is expensive and not always available outside large clinics. In contrast, color fundus photography is inexpensive and common, yet it only shows a surface view and can miss subtle changes inside the retina. Aneurysmal subarachnoid hemorrhage (aSAH) is a devastating neurological disease, and one of the primary drivers of morbidity after aneurysm rupture is the phenomenon of delayed cerebral ischemia (DCI). SARS-CoV-2 is the betacoronavirus causing the COVID-19 pandemic. Although the SARS-CoV-2 genome and transcriptome were reported previously, the function of individual viral proteins is largely unknown. Utilizing biochemical and molecular biology methods, we identified that four SARS-CoV-2 RNA-binding proteins (RBPs) regulate the host RNA metabolism by direct interaction with mature miRNA let-7b revealed by Nuclear Magnetic Resonance spectroscopy (NMR). SARS-CoV-2 RBP Nsp9 primarily binds mature miRNA let-7b, a direct ligand of the Toll-like Receptor 7 (TLR7), one of the potential SARS-CoV-2 therapeutics. Background: Apparent Diffusion Coefficient (ADC) as measured by diffusion weighted imaging is known to negatively correlate with prostate tumor aggressiveness. Heterogeneity in system and protocol performance causes potential variability in ADC acquired across a large scanner network, prompting a need to evaluate quantitative ADC from a prostate-specific MR diffusion protocol as part of quality assurance (QA). Due to the temperature dependence of ADC, repeatability and reproducibility assessments typically require phantoms to maintain a temperature of 0°C, imposing a considerable burden when assessing large numbers of scanners.

The F. nucleatum group (sensu lato) is historically composed of four subspecies (F. nucleatum subsp. animalis, F. nucleatum subsp. nucleatum, F. nucleatum subsp. polymorphum, F. nucleatum subsp. vincentii). Although F. nucleatum sensu lato members are typically associated with oral disease, they have recently been shown to disseminate to the gastrointestinal tract and are associated with adverse health conditions such as inflammatory bowel disease and colorectal cancer (CRC). Cancerous tissue is a largely unexplored microbial niche that provides a unique environment for the colonization and growth of specific bacterial communities, and with it, the opportunity to identify novel bacterial species. Here, we report distinct features of a novel Fusobacterium species, F. sphaericum sp. nov. (Fs), isolated from primary colon adenocarcinoma tissue. We acquire the complete closed genome and associated methylome of this organism and phylogenetically confirm its classification into the Fusobacterium genus, with F. perfoetens as its closest neighbor. Fecal Microbiota Transplant (FMT) is a treatment for recurrent Clostridium difficile infections and is being explored for other clinical applications, from alleviating digestive and neurological disorders, to restoring microbiomes impacted by cancer treatment. Quantifying the extent of engraftment following an FMT is important in understanding a recipient's response to treatment. Engraftment and clinical response need to be investigated independently to evaluate an FMT's role (or lack thereof) in achieving a clinical response. Applied muscular strain and hamstring strain capacity have a joint interaction on hamstring strain injury (HSI) with modifiable risk factors frequently assessed. However, to date there is limited observations on the interaction between these factors. The purpose of the present study was to observe if spatiotemporal characteristics, running kinematics and muscle activation were related to modifiable risk factors of HSI. Twenty-two competitive team sport athletes (24.7 ± 4.3 years, 1.82 ± 0.07 m, 84.9 ± 8.5 kg) participated whereby the Bicep femoris long head (BFLH) fascicle length assessed via ultrasound and isokinetic eccentric hamstring strength was assessed. Communication And Privacy Management Between Female Veterans, Their Military Voluntary Kin, And Biolegal Family Members, Carol Lee Tschampl-Diesing

Military voluntary kin (MVK) refer to relationships between Service members and/or Veterans that they consider to be “like family,” despite lacking blood or legal ties. The purpose of this qualitative/interpretive communication study was to explore how female Veterans, who are the largest growing population of U.S. Veterans, discursively construct MVK relationships, navigate communication, privacy management, and boundary turbulence with their MVK and biolegal families (BLF). The researcher undertook in-depth interviews with nine female U.S. Veterans who reported having at least one MVK. The researcher posed four research questions, engaging in an interpretive analysis.

Although variation in effect sizes and predicted values among studies of similar phenomena is inevitable, such variation far exceeds what might be produced by sampling error alone. One possible explanation for variation among results is differences among researchers in the decisions they make regarding statistical analyses. A growing array of studies has explored this analytical variability in different fields and has found substantial variability among results despite analysts having the same data and research question. Climate-driven shifts in species distributions can undermine the effectiveness of protected areas. We present a framework to facilitate climate change adaptation planning by identifying where highly migratory species habitats will persist (climate refugia), emerge (bright spots), disappear (dark spots), or remain unsuitable based on model analysis by 2100. When applied to eight species in the California Current System, we found that, on average, 37% of habitats are expected to be climate refugia, 9% are bright spots, and 13% are dark spots within National Marine Sanctuaries by 2100. Halophila stipulacea (Forsskål & Niebuhr) is a tropical seagrass native to the Red Sea, Persian Gulf, and Indian Ocean. In 2002, invasive populations of H. stipulacea were identified in the Caribbean Sea where it now forms large meadows, displaces native seagrasses, alters invertebrate and fish communities, and potentially affects organic matter dynamics in invaded meadows. This study investigated differences in organic matter dynamics between meadows of H. stipulacea and native Thalassia testudinum within the Jobos Bay National Estuarine Research Reserve (JBNERR) on the southern coast of Puerto Rico. Urbanization alters landscapes in ways that can reshape wildlife populations by changing habitat connectivity, resource availability, and selective pressures. To investigate how urbanization influences both genetic differentiation and morphology in small mammals, I examined Peromyscus leucopus (PELE) across an urban–rural gradient in northern Georgia. Using RADseq SNP data from PELE individuals across 13 sites, I quantified pairwise genetic differentiation (FST) and tested how various environmental variables, including impervious surface cover, human population density, and tree canopy cover, predict genetic differentiation after accounting for geographic distance. Peripheral Artery Disease (PAD) affects an estimated 200 million people worldwide and is the leading cause of amputation in the United States. PAD is characterized by atherosclerotic narrowing of arteries, resulting in ischemia from reduced blood flow to distal tissues. Although surgical and percutaneous interventions can restore perfusion, fewer than 50% of patients experience sustained benefit, highlighting the need for alternative therapeutic strategies. One promising approach is promoting arteriogenesis, the growth and outward remodeling of pre-existing collateral vessels that improve blood flow around the occluded arteries. Arteriogenesis is regulated by regenerative, M2-polarized macrophages, making macrophage polarization a key therapeutic target. …

“Conservationist” and “environmentalist” are two prominent environmental social identities often perceived as conflicting, particularly on wedge issues like hunting. While these groups may hold differing philosophies, their beliefs could overlap, revealing opportunities for collaboration in conservation. We examined environmental identities among U.S. college students across 22 states (n = 17,203) from 2018 to 2020, assessing identity, wildlife values, and positions on polarizing issues. Students were classified into four groups: conservationists (8 %), environmentalists (9 %), pluralists (59 %) who expressed both identities, and eco-agnostics (24 %) who expressed neither. Resseliella maxima Gagné (Diptera: Cecidomyiidae) is a recently described and economically significant pest of soybean (Glycine max (L.) Merr.) in the Midwestern United States. Since its identification in 2019, this species has become widespread and is now associated with substantial yield losses in parts of the region. Prior to its formal description, orange larvae were observed in soybean associated with plants damaged by hail, suggesting that plant injury may influence infestation. Given the pest’s economic importance, understanding the role of hail injury and developing effective, integrated management strategies are critical for mitigating its impact.

The Singapore Court of Appeal held a law prohibiting male–male sexual intercourse (s 377A of the Penal Code) unenforceable on non-rights-based grounds. The court's remarks about the rights to life and liberty (including whether sexual orientation is mutable), freedom of expression, and equality are obiter. But they are significant, since in Singapore, there are few constitutional rights cases and the courts focus on spelling out the limits to executive and legislative power without necessarily striking down exercises of such power on rights grounds. Introduction: Healthy Longevity Medicine (HLM) offers a strategy to reduce the healthspan-lifespan gap, yet public perspectives remain unclear. This study refines the Healthy Longevity (HELO) framework through a qualitative exploration of public views towards lifespan, healthspan, and HLM.Methods: Individuals living in Singapore participated in semi-structured group or individual discussions to explore (a) their understanding of lifespan and healthspan, (b) motivational factors for health behaviours, and (c) their awareness of HLM. Sampling maximised variation across age, sex, and ethnicity. Gregarious desert locusts produce stage-specific pheromones that facilitate cohesive behavior in juveniles and synchronize maturation and mating in sexually mature adults. During locust outbreaks, merging populations result in cross-stage interactions, yet their impact on locust biology remains poorly understood. This study tested the hypothesis that cross-stage interactions influence juvenile cohesion and physiological traits. Using behavioral assays and gas chromatography-mass spectrometry, we examined short- and long-term interactions between juvenile and adult desert locusts. The push-pull technology (PPT) has often been presented as a management strategy for stemborers and witchweed. However, its value as an agronomic practice and an agroecological approach remains largely underappreciated. This review aims to appraise the PPT used in eastern Africa, synthesize evidence for its ecological and economic benefits, and identify barriers to its adoption and opportunities for its expansion to other crops and farming systems in sub-Saharan Africa.
