Trichloroethylene Remediation By Engineered Soil Bacteria, Madeline Hannah Mclaughlin Armond
Trichloroethylene Remediation By Engineered Soil Bacteria, Madeline Hannah Mclaughlin Armond
Theses and Dissertations
Trichloroethylene (TCE) is a toxic pollutant that has become a widespread problem by seeping into groundwater across the developed world. Clean-up of sites contaminated with TCE is extremely difficult due to the absence of an efficient and cost-effective method for clean-up. Bioremediation efforts include a variety of potential microbial candidates with various metabolic capabilities as clean up options of contaminated sites. Cupriavidus necator, a soil bacterium was found to possess the ability to degrade TCE via a phenol-dependent pathway. Previous research by Ayoubi and Harker (1998) created a strain (MM02) capable of constitutive TCE degradation but the underlying genetic alteration …

Single-Molecule Localization Microscopy Of 3d Orientation And Anisotropic Wobble Using A Polarized Vortex Point Spread Function, Tianben Ding, Matthew D. Lew
Electrical & Systems Engineering Publications and Presentations
Within condensed matter, single fluorophores are sensitive probes of their chemical environments, but it is difficult to use their limited photon budget to image precisely their positions, 3D orientations, and rotational diffusion simultaneously. We demonstrate the polarized vortex point spread function (PSF) for measuring these parameters, including characterizing the anisotropy of a molecule’s wobble, simultaneously from a single image. Even when imaging dim emitters (∼500 photons detected), the polarized vortex PSF can obtain 12 nm localization precision, 4°–8° orientation precision, and 26° wobble precision. We use the vortex PSF to measure the emission anisotropy of fluorescent beads, the wobble dynamics …

We have previously shown that the αvβ6 integrin plays a key role in promoting prostate cancer (PrCa) and it can be transferred to recipient cells via small extracellular vesicles (sEVs). Furthermore, we have reported in a proteomic analysis that αvβ6 integrin down-regulation increases the expression of IFIT3 (interferon induced protein with tetratricopeptide repeats 3) in PrCa cells and their derived sEVs. IFIT3 is a protein well known for being an antiviral effector, but recently its role in cancer has also been elucidated. To study the relationship between IFIT3 and STAT1 (signal transducer and activator of transcription 1), an upstream regulator …

Antibiotic resistance is a major public health concern. The shrinking selection of effective antibiotics and lack of new development is making the situation worse. Gram-negative bacteria more specifically pose serious threat because of their double layered cell envelope and effective efflux systems, which is a challenge for drugs to penetrate. One promising approach to breach this barrier is the “Trojan horse strategy”. In this technique, an antibiotic molecule is conjugated with a nutrient molecule that helps the antibiotic to enter the cell through dedicated transporters for the nutrient. Here, we explored the approach using biotin conjugation with a florescent molecule …

Investigating The Growth Of Algae Under Low Atmospheric Pressures For Potential Food And Oxygen Production On Mars, Leena M. Cycil, Elisabeth M. Hausrath, Douglas W. Ming, Christopher T. Adcock, James Raymond, Daniel Remias, Warren P. Ruemmele
Geoscience Faculty Research
With long-term missions to Mars and beyond that would not allow resupply, a self-sustaining Bioregenerative Life Support System (BLSS) is essential. Algae are promising candidates for BLSS due to their completely edible biomass, fast growth rates and ease of handling. Extremophilic algae such as snow algae and halophilic algae may also be especially suited for a BLSS because of their ability to grow under extreme conditions. However, as indicated from over 50 prior space studies examining algal growth, little is known about the growth of algae at close to Mars-relevant pressures. Here, we explored the potential for five algae species …

The Reward Positivity In Individuals With Alcohol Use Disorder, Garima Singh
Psychology ETDs
The Reward Positivity (RewP) is a positive deflection in the EEG after receiving a reward. Recent evidence suggests that the RewP is modulated by both reward probability aside from affective liking. Here we examined the sensitivity of the RewP to affective and alcohol images in individuals with alcohol use disorder (AUD). We recruited 55 participants (AUD =28, Control= 27). Participants completed an image rating task and a reinforcement learning task with picture feedback (puppy or alcohol images over a green or red screen). Although there was no between group differences in puppy image ratings, there was a significant group difference …

Ubiquitous Yet Inconspicuous: Quantifying Trophic Impact Of A Widespread Oceanic Comb Jelly (Ctenophore), Elizabeth Potter
Ubiquitous Yet Inconspicuous: Quantifying Trophic Impact Of A Widespread Oceanic Comb Jelly (Ctenophore), Elizabeth Potter
USF Tampa Graduate Theses and Dissertations
The oceanic lobate ctenophore Ocyropsis spp. has a widespread distribution throughout the world’s tropical and sub-tropical oceans. While patchy, Ocyropsis spp. population densities are known to exceed one individual per m3. Studies of coastal ctenophores have shown that some species are capable of exerting strong predatory impacts on their ecosystems, but little is known about the potential trophic impacts of their oceanic relatives. Many oceanic genera such as Ocyropsis, Bolinopsis, and Eurhamphaea exhibit morphologies and prey capture mechanisms that are different from the more well-studied coastal species. Thus, existing data on coastal taxa may not apply to oceanic species due …

A Multimodal Literacy Exploration: Lived Experiences Of Haitian Immigrant Adolescent Girls In The Bahamas, Natasha Swann
USF Tampa Graduate Theses and Dissertations
As the geographic landscape of our world increasingly changes, a constant migration is occurring, with people seeking refuge and asylum. Many of those seeking a better way of life are children and adolescents. It is important not only to consider and accommodate how children and adolescents develop and grow, but how they do so in a country that is not their home and with language and literacy practices that are often new to them. Adjusting and acculturating into host countries might compound issues youth experience during child and adolescence development. This study provided a space and a platform for immigrant …

Raspberry ketone (RK) is a natural compound with anti-inflammatory properties that may benefit tissue healing. Effects of RK on inflammation and healing were evaluated by examining macrophage polarization in response to guided bone regeneration (GBR) membranes loaded with 0,100 or 500µg RK over 4 weeks in a rat calvarial model. Initially, pro-inflammatory M1 macrophage phenotypes remove damaged tissues and bacteria and then transition to pro-healing M2 phenotypes to coordinate tissue repair and formation in wounds. Immunohistochemical staining was used to characterize M1 and M2 macrophage phenotypes using CD68 as a PAN macrophage marker, iNOS for M1 and CD206 for M2 …

Chronological age (CA) is determined by time of birth, whereas biological age (BA) is based on changes on a cellular level and strongly correlates with morbidity, mortality, and longevity. Type 2 diabetes (T2D) associates with increased morbidity and mortality; thus, we hypothesized that BA would be increased and calculated it from biomarkers collected at routine clinical visits. Deidentified data was obtained from three cohorts of patients (20-80 years old)-T2D, type 1 diabetes (T1D), and prediabetes-and compared to gender- and age-matched non-diabetics. Eight clinical biomarkers that correlated with CA in people without diabetes were used to calculate BA using the Klemera …

Multifaceted Approach To Understanding Acinetobacter Baumannii Biofilm Formation And Drug Resistance, Jessie L. Allen
Multifaceted Approach To Understanding Acinetobacter Baumannii Biofilm Formation And Drug Resistance, Jessie L. Allen
USF Tampa Graduate Theses and Dissertations
Acinetobacter baumannii is a multi-drug resistant nosocomial pathogen known for causing wound related- and respiratory-infections. It is currently on the WHO’s list of critical pathogens due to its broadly drug resistant nature and the constant appearance of pan-resistant isolates. A majority of the infections caused by this organism are biofilm associated, however there is limited existing knowledge regarding the mechanisms used to engage in this multicellular lifestyle. As such, we set out to explore the factors influencing this behavior using an 10,000+ isolate transposon mutant library of A. baumannii strain AB5075. Of the strains tested, 6.45% demonstrated some level of …
The Ecological Role Of Cassiopea In Shallow Mangrove Habitats, David Marcel Durieux
The Ecological Role Of Cassiopea In Shallow Mangrove Habitats, David Marcel Durieux
USF Tampa Graduate Theses and Dissertations
Upside-down jellyfish, Cassiopea sp., can be locally abundant in shallow mangrove habitats throughout the tropics and subtropics. Unlike other jellyfish, Cassiopea sp. are epibenthic. Due to this unique lifestyle and their ability to achieve high population densities, an understanding is needed of their ecological impacts both individually and in aggregations, particularly given that Cassiopea sp. ranges are expanding poleward due to global climate change. We quantified the fluid flow produced by Cassiopea sp. feeding currents and found that an average-sized Cassiopea sp. can transport over 200 l/h of water in the vertical excurrent jet of their feeding current. Populations of …
Mechanistic Insight Into Β-Lactamase Catalysis, Inhibitor Design And Resistance, Michael Trent Kemp
Mechanistic Insight Into Β-Lactamase Catalysis, Inhibitor Design And Resistance, Michael Trent Kemp
USF Tampa Graduate Theses and Dissertations
The emergence of antibiotic resistance and spread of Gram negative bacteria poses a very real health threat to the public. The main mode of resistance within Gram negative bacteria is the production of β-lactamase enzymes that catalyze the breakdown of β-lactam antibiotics through a hydrolysis mechanism. Once the β-lactam ring is hydrolyzed and opened, the drug loses its efficacy, which allows for the bacteria to grow and proliferate uninhibited. These β-lactamase enzymes are organized into four categories based on the Ambler classification, with classes A, C and D being denoted as serine-based β-lactamase enzymes. Class B is composed of metalloenzymes …
Moving In Fluid: Exploring How Fishes Manipulate Water To Swim Efficiently, Nils B. Tack
Moving In Fluid: Exploring How Fishes Manipulate Water To Swim Efficiently, Nils B. Tack
USF Tampa Graduate Theses and Dissertations
Moving through a dense fluid such as water presents some unique challenges to minimizing energy use and maximizing performance (i.e., speed). Due to animal-fluid interactions during swimming (drag and thrust production) fish have evolved a variety of morphological structures and locomotor mechanisms. For instance, fish rely on body bending and/or fins to interact with the surrounding water such that energy can be transferred to generate thrust. Typically, this synergy promotes morphologies and behaviors aimed at enhancing propulsive efficiency and/or minimizing metabolic activity to lessen the cost of transport (COT). This work focuses on quantifying the energetic and hydromechanical benefits of …

Mitogen-activated protein kinase 6 (MAPK6) is an atypical MAPK. Its function in regulating cancer growth remains elusive. Here, we reported that MAPK6 directly activated AKT and induced oncogenic outcomes. MAPK6 interacted with AKT through its C34 region and the C-terminal tail and phosphorylated AKT at S473 independent of mTORC2, the major S473 kinase. mTOR kinase inhibitors have not made notable progress in the clinic. Our identified MAPK6-AKT axis may provide a major resistance pathway. Besides repressing growth, inhibiting MAPK6 sensitized cancer cells to mTOR kinase inhibitors. MAPK6 overexpression is associated with decreased overall survival and the survival of patients with …
Responses Of Wetland Soil Microbial Communities To Different Types Of Anthropogenic Stress, Grace Ann Cagle
Responses Of Wetland Soil Microbial Communities To Different Types Of Anthropogenic Stress, Grace Ann Cagle
LSU Doctoral Dissertations
Microorganisms are the major drivers of biogeochemical transformations of carbon and nitrogen in wetlands. These processes are crucial for ecological sustainability and are heavily impacted by human activities. How anthropogenic stress affects the microbial drivers of carbon storage and nutrient cycling in wetlands is not well understood. I investigated the response of wetland soil microbial communities to three different types of anthropogenic stress using DNA sequencing. First, I studied the effect of warming on microbial carbon processing in a peatland underlaid by permafrost by comparing the abundances of specific genes between sites subjected to five years of warming and controls. …
Vehicular Design And Resource Allocation Policies For Equitable Road Safety, Alyssa Ryan
Vehicular Design And Resource Allocation Policies For Equitable Road Safety, Alyssa Ryan
PSU Transportation Seminars
The diversity of road users is not equitably accounted for in vehicular and infrastructure design and funding allocation policies, among other areas that impact mobility. This impedes the ability for all road users to experience the same level of safety while traveling. Moving towards equitable road safety for all road users is critical to improve the quality of life and save lives of those that are most underserved in the transportation sector.
Professor Alyssa Ryan discusses two strategies to increase safety for vulnerable road users. First, a study on road injury differences between drivers of different biological sex is presented. …

Hello Traitor: An Examination Of Individual Differences In Perceptions Of Technology-Related Incivility, David J. Howard
USF Tampa Graduate Theses and Dissertations
Workplace incivility is unfortunately common among employees in today’s workplace. The increase in usage of email, texting, smartphones, and social media for interpersonal workplace communication has led to an increase of these mediums being used in an uncivil manner. While there has been a growth of general workplace incivility research conducted in the past two decades, the extant literature lacks sufficient primary studies that examine technology-related workplace incivility. This research project aims to add to the burgeoning literature in the technology-related incivility content domain. First, it examined the prevalence of email incivility reported by workers and found a much lower …

Decisions And How Doctors Make Them: Modeling Multilevel Decision-Making Within Diagnostic Medicine, Michelle S. Kaplan
Decisions And How Doctors Make Them: Modeling Multilevel Decision-Making Within Diagnostic Medicine, Michelle S. Kaplan
USF Tampa Graduate Theses and Dissertations
Effective decision-making is critical and necessary for organizational success across a wide range of occupations, situations, and industries. However, decision making, by its nature, is not always a direct process of a single decision leading to a direct outcome. Rather, it can often become a multilevel process whereby one decision’s outcome leads to information that is used in subsequent larger or other types of decisions. The decision-making process then becomes progressively more complex and more difficult to navigate as these decisions compound within one another. Thus, decision-makers must find an appropriate way to approach such decisions. Understanding the multilevel nature …

Light Harvesting In Photoactive Guest-Based Metal-Organic Frameworks, Christopher R. Mckeithan
USF Tampa Graduate Theses and Dissertations
Harvesting sunlight as a clean and renewable energy source has long been a goal of the scientific community. One potential route for developing artificial light harvesters is using a bioinspired approach which utilizes the critical features found in these natural systems. Such features include a way in which to capture a broad range of the solar spectrum, fixed distance and orientation of redox pairs to facilitate efficient directional transfer of electrons, transfer of electrons to a terminal acceptor, and recovery of the systems for repeated use. Metal organic frameworks (MOFs) are attractive candidates for developing light harvesting systems due their …
Computationally Driven Design Of Shape-Controlled Polymeric Nanostructures, Davindra Tulsi
Computationally Driven Design Of Shape-Controlled Polymeric Nanostructures, Davindra Tulsi
USF Tampa Graduate Theses and Dissertations
In the realm of shape control involving synthetic soft materials, block copolymers are ubiquitously exploited due to their ability to spontaneously self-assemble into a plethora of morphologies. The growing need in areas such as drug delivery, lithography, and microelectronics to generate new nanostructures demands ever finer control of copolymer self-assembly. It is desirable to better control the shape and size of soft nanostructures, with minimal post processing techniques to assert stability.
While there is an abundance of literature on diblock copolymer assembly, few relationships exist for sequence-controlled copolymers. It is well known that biological macromolecules are highly sequence specific and …

Library Philosophy and Practice (e-journal)
This paper will discuss catastrophe recovery and likelihood development for digital library structures. The article establishes a foundation for establishing Library continuity and disaster recovery strategies through the use of best practices. Library continuity development and catastrophe recovery are critical modules of the planning stage for a virtual digital library. Few institutions that experience a catastrophic disaster occurrence are powerless to recover always, but libraries can suggestively boost the possibility of long-term recovery of institutional digital resources by drafting a continuity and recovery strategy in preparation. This article is intended for system designers and administrators, as well as high-ranking library …
