The Arabian camel (Camelus dromedarius) is the most important livestock animal in arid and semi-arid regions and provides basic necessities to millions of people. In the current context of climate change, there is renewed interest in the mechanisms that enable camelids to survive in arid conditions. Recent investigations described genomic signatures revealing evolutionary adaptations to desert environments. We now present a comprehensive catalogue of the transcriptomes and proteomes of the dromedary kidney and describe how gene expression is modulated as a consequence of chronic dehydration and acute rehydration. Our analyses suggested an enrichment of the cholesterol biosynthetic process and an …

Department of Pathology, Anatomy, and Cell Biology Faculty Papers

Impaired liver regeneration has been considered as a hallmark of progression of alcohol-associated liver disease. Our previous studies demonstrated that in vivo inhibition of the microRNA (miRNA) miR21 can restore regenerative capacity of the liver in chronic ethanol-fed animals. The present study focuses on the role of microRNA regulatory networks that are likely to mediate the miR-21 action. Rats were chronically fed an ethanol-enriched diet along with pair-fed control animals and treated with AM21 (anti-miR-21), a locked nucleic acid antisense to miR-21. Partial hepatectomy (PHx) was performed and miRNA expression profiling over the course of liver regeneration was assessed. Our …

The shortcomings of current anti-human cytomegalovirus (HCMV) drugs has stimulated a search for anti-HCMV compounds with novel targets. We screened collections of bioactive compounds and identified a range of compounds with the potential to inhibit HCMV replication. Of these compounds, we selected bisbenzimide compound RO-90-7501 for further study. We generated analogues of RO-90-7501 and found that one compound, MRT00210423, had increased anti-HCMV activity compared to RO-90-7501. Using a combination of compound analogues, microscopy and biochemical assays we found RO-90-7501 and MRT00210423 interacted with DNA. In single molecule microscopy experiments we found RO-90-7501, but not MRT00210423, was able to compact DNA, …

Automatic Camera Trap Classification Using Wildlife-Specific Deep Learning In Nilgai Management, Matthew Kutugata, Jeremy Baumgardt, John A. Goolsby, Alexis Racelis

School of Integrative Biological & Chemical Sciences (Formerly Dept. of Biology)

Camera traps provide a low-cost approach to collect data and monitor wildlife across large scales but hand-labeling images at a rate that outpaces accumulation is difficult. Deep learning, a subdiscipline of machine learning and computer science, has been shown to address the issue of automatically classifying camera trap images with a high degree of accuracy. This technique, however, may be less accessible to ecologists, to small scale conservation projects, and has serious limitations. In this study, a simple deep learning model was trained using a dataset of 120,000 images to identify the presence of nilgai Boselaphus tragocamelus, a regionally specific …

Transactions of the Nebraska Academy of Sciences and Affiliated Societies

The Central Platte River Valley (CPRV) is a unique and biologically important ecoregion, and several studies have conducted localized herpetofauna species inventories. However, there has been significantly less effort to quantify the habitat associations or activity patterns of these species. Habitat associations and activity patterns vary regionally and provide important information for ecosystem management. We deployed a total of 38 traps of 5 different types (pitfall, funnel, hoop, box, and coverboard) across 11 sites (7 terrestrial, 4 aquatic) for an 8-week period in June and July 2019. Our exploratory analyses used generalized linear models with a quasibinomial distribution to examine …

The Role Of Chemokines In Vertebrate Physiology And Disease: A Concise Review, Hebatallah Mahgoub Dec 2021

The Role Of Chemokines In Vertebrate Physiology And Disease: A Concise Review, Hebatallah Mahgoub

Mansoura Veterinary Medical Journal

Chemokines are classified as proteins with chemoattractant activities that have multiple crucial roles in health and disease, where they participate in the processes of development, angiogenesis, hematopoiesis and tumor metastasis. Multiple cells are involved in the production of chemokines. However, the main cells involved in such role are blood monocytes, macrophages and polymorphonuclear leukocytes, where they exert their role in the inflammatory reaction following antigen recognition by tissue phagocytes. Due to their important role in the establishment of successful immune response, several microbes are incriminated in the production of proteins that mimic chemokines. In addition, their receptors could be used …

Effects Of Il-10 On Local Cell Populations And Functional Recovery Following Vml Injury, Zain Blackwell Dec 2021

Effects Of Il-10 On Local Cell Populations And Functional Recovery Following Vml Injury, Zain Blackwell

Biomedical Engineering Undergraduate Honors Theses

Volumetric muscle loss (VML) injuries are prevalent in both military personnel suffering from battlefield related incidents, and civilians following severe motor accidents. Despite its prevalence, VML has no pro-regenerative clinical treatments in place to recover some of the functional capabilities of the damaged muscle. Free flap grafting, debridement of damaged tissue, and physical therapy are the only clinical standards available that offer little functional recovery benefits, even after years of consistent treatment. In this study, anti-inflammatory cytokine interleukin-10 in conjunction with autologous minced muscle was assessed as a possible treatment for VML injuries and its influences on cellular behavior within …

Role Of Hsp70 And The Tlr4-Md2 Complex In The Vasculature: From Function To Dysfunction In Diabetes, Amanda Almeida De Oliveira Dec 2021

Role Of Hsp70 And The Tlr4-Md2 Complex In The Vasculature: From Function To Dysfunction In Diabetes, Amanda Almeida De Oliveira

Theses and Dissertations

Diabetes impairs vascular functionality, which contributes to the pathophysiology of its associated complications. Recently, it was suggested that innate immune receptors assist in the development of diabetic vasculopathies, which sustains the pro-inflammatory state of the disease. Among these receptors, Toll-like receptor 4 (TLR4) is a strategic player as its activation prime the release of inflammatory mediators and reactive oxygen species (ROS), which are well-known pathological mechanisms linked to diabetic complications. The TLR4-Myeloid differentiation factor 2 (MD2) complex has gained much attention over the last decade, primarily because it interacts with self-produced molecules. In vascular tissues, the Heat-shock protein 70 (HSP70) …

Rice Quality And Its Impacts On Food Security And Sustainability In Bangladesh, Indrani Saha, Alvaro Durand-Morat, Lawton Lanier Nalley, Mohammad Jahangir Alam, Rodolfo M. Nayga Jr. Dec 2021

Rice Quality And Its Impacts On Food Security And Sustainability In Bangladesh, Indrani Saha, Alvaro Durand-Morat, Lawton Lanier Nalley, Mohammad Jahangir Alam, Rodolfo M. Nayga Jr.

Agricultural Economics and Agribusiness Faculty Publications and Presentations

Rice market efficiency is important for food security in countries where rice is a staple. We assess the impact of rice quality on rice prices, food security, and environmental sustainability in Bangladesh. We find that while price varies as expected for most quality attributes, it is unaffected by a broken percentage below 24.9 percent. This reveals a potential inefficiency, considering the average 5 percent broken rate observed in the market. An increase in the broken rate of milled rice within the limits supported by our findings can, ceteris paribus, increase rice rations by 4.66 million a year, or conversely, …

Sex Differences In The Co-Expression Of Estrogen Receptor Alpha With Corticotropin Releasing Factor, Deborah Ariyibi Dec 2021

Sex Differences In The Co-Expression Of Estrogen Receptor Alpha With Corticotropin Releasing Factor, Deborah Ariyibi

Biological Sciences

Women are far more likely to develop anxiety and depression than men. It is believed that the dysregulation of the HPA axis by the binding of corticotropin -releasing factor (CRF) to the corticotropin-releasing factor receptor 1 (CRFR1) contributes to the likelihood of these stress- related disorders. Estrogens acting through Estrogen receptor alpha (ERa) have been shown to increase anxiety production upon activating the HPA axis. In this current study, we explored whether CRF-expressing neurons in various regions of the brain express ERa. The levels of ERa were counted in the bed nucleus of the stria terminalis (BST), the medial preoptic …

Light Level Intensity Alters Anxiety, But Not Memory, During Open-Field And Novel Object Location Tasks In Male Rats, Angelina Tassone Dec 2021

Light Level Intensity Alters Anxiety, But Not Memory, During Open-Field And Novel Object Location Tasks In Male Rats, Angelina Tassone

Biological Sciences

Appropriate design and control of testing conditions during assessment of animal behavior is critical to maximize generalizability, replicability, and translational relevance. Some sensory stimuli are often controlled: for instance, during rodent behavioral testing, efforts are commonly made to reduce or eliminate olfactory and auditory distractions. However, less attention is paid to the ambient light level intensity (lux), which may vary even between rooms in the same facility. We sought to explore whether behavior is influenced by the standard illumination intensity in one of our behavioral testing rooms. To this end, we measured anxiety-like behaviors, exploration, and spatial memory performance in …

How Salivary Glands Recover From Fibrotic Injury, Gabriella Majka Dec 2021

How Salivary Glands Recover From Fibrotic Injury, Gabriella Majka

Biological Sciences

Extracellular matrix deposition is required for repair after injury, but if left unresolved can result in fibrosis. Fibrosis is an excess deposition of extracellular matrix proteins following injury or aging that leads to eventual organ dysfunction. One surgical model that we have used to study mechanisms of fibrosis is the salivary gland ductal ligation model. During the process of ligation, a clip is placed on the main ducts of the submandibular (SMG) and sublingual gland (SLG) which leads from the salivary glands to the mouth. This causes the gland to atrophy, or waste away, resulting in decreased acinar cell differentiation …

Identifying The Molecular Cause Of Extreme Endoplasmic Reticulum Dilation In Pediatric Osteosarcoma And Its Relationship To The Disease, Rachael Wood Dec 2021

Identifying The Molecular Cause Of Extreme Endoplasmic Reticulum Dilation In Pediatric Osteosarcoma And Its Relationship To The Disease, Rachael Wood

Theses and Dissertations (ETD)

Pediatric osteosarcoma tumors are characterized by an unusual abundance of grossly dilated endoplasmic reticulum and an immense genomic instability that has complicated identifying new effective molecular therapeutic targets. Here we report a novel molecular signature that encompasses the majority of 108 patient tumor samples, PDXs and osteosarcoma cell lines. These tumors exhibit reduced expression of four critical COPII vesicle proteins that has resulted in the accumulation of procollagen-I protein within ‘hallmark’ dilated ER. Using CRISPR activation technology, increased expression of only SAR1A and SEC24D to physiologically normal levels was sufficient to restore both collagen-I secretion and resolve dilated ER morphology …

Characterizing And Overcoming Resistance To Aminomethylspectinomycins In Gram-Negative Bacteria, Nisha Das Dec 2021

Characterizing And Overcoming Resistance To Aminomethylspectinomycins In Gram-Negative Bacteria, Nisha Das

Theses and Dissertations (ETD)

Spectinomycin (SPC) is a broad-spectrum aminocyclitol antibiotic. Its use in agriculture has led to widespread resistance in enteric bacteria, necessitating the development of more effective analogs. Aminomethyl spectinomycins (amSPC) are modified spectinomycins with increased potency against many bacterial species. These species include Legionella pneumophila, which harbors a chromosomally encoded aminoglycoside modifying enzyme (AME). In this study, we follow up on this observation and examine the extent to which the amSPCs are substrates for AMEs through adenylation (ANTs) and phosphorylation (APH). APH(9)-Ia and ANT(3")(9) were expressed in E. coli BL21(DE3) and purified using the Ni-affinity chromatography. The ability of AMEs to …

Examination Of Antiviral Resistance In Venezuelan Equine Encephalitis Virus, Jasper Lee

Theses and Dissertations (ETD)

Venezuelan equine encephalitis virus (VEEV) is a New World Alphavirus that causes Venezuelan equine encephalitis (VEE), which is characterized by a febrile illness that can progress to neurological disease and death. While no major outbreaks of VEE have occurred since 1995, VEEV is a virus of concern as, in addition to its spread through mosquitos, it can be aerosolized and used as a bioweapon. Unfortunately, there are currently no FDA-approved vaccines or antivirals against VEEV. Efforts have been made to discover small molecules with an inhibitory effect on VEEV, but the potential for emergence of antiviral resistance to these compounds …

The Effect Of Cancer Cachexia Progression On The Feeding Regulation Of Skeletal Muscle Protein Turnover, Brittany R. Franch Dec 2021

The Effect Of Cancer Cachexia Progression On The Feeding Regulation Of Skeletal Muscle Protein Turnover, Brittany R. Franch

Theses and Dissertations (ETD)

Cancer cachexia is defined as the unintentional loss of skeletal muscle mass with or without fat loss that cannot be reversed by conventional nutritional support. Cachexia occurs in ~20% of cancer patients. More specifically, 50% of lung cancer patients, the most common cancer worldwide, develop cachexia. Cachexia occurs most often in lung and gastrointestinal cancers, whereas breast and prostate have the lowest rate of cachexia. Cancer-induced cachexia disrupts skeletal muscle protein turnover (decreasing protein synthesis and increasing protein degradation). Skeletal muscle’s capacity for protein synthesis is highly sensitive to local and systemic stimuli that are controlled by mTORC1 and AMPK …

The purpose of this descriptive phenomenological study was to gain more knowledge about leadership dispositions of school principals who could give insight into helping failing schools succeed. High-poverty, low-performing (HPLP) environments with chronically underperforming schools are identified as schools needing a “turnaround.” By learning more about the experiences of teachers and administrative leaders working with school principals in low-performing schools, a better understanding emerged of the leadership dispositions that can engage employees in a successful school turnaround. Three themes emerged based on the experiences from six teachers and four supervisors of school principals regarding the principals’ leadership dispositions and their …

Detecting Gsr Indicative Particles On Decayed Bones Using A Novel Field Kit, Sven Engling Dec 2021

Detecting Gsr Indicative Particles On Decayed Bones Using A Novel Field Kit, Sven Engling

Student Theses

Decomposed human remains are complex forensic puzzles, escalating in difficulty as the remains’ age obscures evidence, like trauma. Research has shown that scanning electron microscopes with energy dispersive X-ray spectrometers (SEM-EDX) are capable of detecting and identifying gunshot residue (GSR) particles on bones. However, SEM-EDX work is time consuming, expensive, and not accessible to every forensic department. Therefore, a preliminary field test capable of detecting GSR indicative particles, like lead, could save departments money and assist in trauma identification. This study examines the viability of using either the 3M Lead Check Test swabs or a sodium rhodizonate solution as part …

Support For Militant U.S. Foreign Policy: The Role Of Authoritarianism And International Xenophobia, Brandon M. Garcia Dec 2021

Support For Militant U.S. Foreign Policy: The Role Of Authoritarianism And International Xenophobia, Brandon M. Garcia

Student Theses

Right-Wing Authoritarianism (RWA) has been studied extensively in the domestic socio-political context, but its potential impact on foreign policy is still poorly understood. The current study replicated the putative two-factor model of the construct (Authoritarian Aggression/Submission and Conventionalism) and examined the associations of each factor with perceptions of overall danger to the U.S. posed by other countries and with the support for more militant U.S. foreign policy. As expected, both RWA factors correlated with self-reported levels of political conservatism (r = 0.32, r = 0.33; p < .001) Additionally, Authoritarian Aggression and Submission (but not Conventionalism) were correlated with increased perceived danger from foreign countries (r = .35; p < .001) and increased support for a more militant U.S. foreign policy (r = .25; p < .001). Participants higher in RWA were more likely to view the world as a hostile and threatening place, and in turn support more aggressive military action in response to those fears.
